Expressions of Thymidylate Synthase, Thymidine Phosphorylase, Class Ⅲ β-tubulin, and Excision Repair Cross-complementing Group 1 Predict Response in Advanced Gastric Cancer Patients Receiving Capecitabine Plus Paclitaxel or Cisplatin

Objective: To evaluate the role of class III β-tubulin (TUBB3), thymidylate synthase (TS), thymidine phosphorylase (TP), and excision repair cross-complementing group 1 (ERCC1) in clinical outcome of advanced gastric cancer patients receiving capecitabine plus paclitaxel or cisplatin. Methods: The clinical data and tumor specimens from 57 advanced gastric cancer patients receiving first-line capecitabine plus paclitaxel (cohort 1, n=36) and capecitabine plus cisplatin (cohort 2, n=21) were retrospectively collected, and TUBB3, TS, TP, and ERCC1 expressions were detected by real-time quantitative PCR. The associations between expressions of biomarkers and response or survival were analyzed statistically. Results: The median age of 57 patients was 57 years (range: 27–75 years) with 38 males and 19 females. Of all patients, the response rates of patients with high TP, low TP and high TS, low TS expressions were 57.1%, 27.6% (P=0.024), and 55.2%, 28.6% (P=0.042), respectively. Among cohort 1, the response rates and median overall survivals of patients with low and high TUBB3 expressions were 61.1% vs. 33.3% (P=0.095) and 13.8 months vs. 6.6 months (P=0.019), respectively; the response rate (87.5%) of patients with low TUBB3 and high TP expressions was higher than that (14.3%) of patients with high TUBB3 and low TP expressions (P=0.01). Among cohort 2, the response rates of patients with low ERCC1 and high ERCC1 expressions were 45.5% and 20.0% respectively (P=0.361). Conclusion: TUBB3, TS and TP expressions could predict the response of advanced gastric cancer patients receiving capecitabine-based and paclitaxel-based chemotherapy. These results will be further confirmed in future large samples.

Microstructure and mechanical properties of in situ(TiC+SiC)/FeCrCoNi high entropy alloy matrix composites

In situ(TiC+SiC)particles(5 vol.%and 10 vol.%,respectively)-reinforced FeCrCoNi high entropy alloy matrix composites were fabricated via vacuum inductive melting method,with equal volume fractions of TiC and SiC particles.X-ray diffraction,scanning electron microscope and energy diffraction spectrum were employed to analyze the microstructure and composi-tion of the *** results manifested that the FeCrCoNi matrix is composed of FCC phase,and the in situ particles are homogeneously scattered in the *** presence of reinforcements augmented the ultimate tensile strength from 452 to 783 MPa,and raised the yield strength from 162 to 466 MPa at room temperature,whereas the elongation to fracture was reduced from 70.6%to 28.6%.All the tensile fracture surfaces consisted of numerous tiny dimples,indicating that the composites exhibited ductile ***,the enhancement of strength ascribes to a combination of thermal mis-match strengthening,load-bearing effect,grain refinement,Orowan strengthening and solid solution strengthening effect,which contribute about 58.0%,2.4%,12.3%,11.1%and 16.2%to the improvement of yield tensile strength,respectively.

Functional changes in synaptic transmission from the lateral entorhinal cortex to the dentate gyrus(LEC-DG)are considered responsible for the chronification of ***,the underlying alterations in fan cells,which are the predominant neurons in the LEC that project to the DG,remain ***,we investigated possible mechanisms using a rat model of complete Freund’s adjuvant(CFA)-induced inflammatory *** found a substantial increase in hyperpolarization-activated/cyclic nucleotide-gated currents(Ih),which led to the hyperexcitability of LEC fan cells of CFA *** phenomenon was attenuated in CFA slices by activating dopamine D2,but not D1,*** activation of the ventral tegmental area-LEC projection had a D2 receptor-dependent analgesic ***-LEC microinjection of a D2 receptor agonist also suppressed CFA-induced behavioral hypersensitivity,and this effect was attenuated by pre-activation of the *** findings suggest that down-regulating the excitability of LEC fan cells through activation of the dopamine D2 receptor may be a strategy for treating chronic inflammatory pain.

Bionic tracking method by hand ＆ eye-vergence visual servoing

Animals rotate their eyes to gaze at the target prey, enhancing the ability of measuring the distance to the target precisely for catching it. These animals, visual tracking includes the triangular eye-vergence control and their body＇s motion control by visual servoing. The research aims to realize a bionic robot tracking perfor- mance, in which the body links moves together with eyes＇ view orientation. This paper proposed a hand ＆ eye-ver- gence dual control system which included two feedback loops： an outer loop for conventional visual servoing to direct a manipulator toward a target object and an inner loop for active motion control of binocular cameras to change the viewpoint along with the moving object to give an accurate and broad observation. This research also foused on how to compensate a fictional motion of the target seen by camera images in an eye-in-hand system, where the camera was fixed on the end-effector and moved together with the hand motion. A robust motion-feedfor- ward （MFF） recognition method is proposed to compensate the fictional motion of the target based on the manipula- tor＇s joint velocity, then the real motion of the target seen by camera images is extracted, which can improve the feedback image sensing unit to make the whole servoing system dynamically stable. The effectiveness of the pro- posed hand ＆ eye-vergence visual servoing method is shown by tracking experiments using a 6-DoF robot manipulator and a 3-DoF binocular vision system.

大型闪烁体γ探测阵列在核物理和核天体物理等领域的实验研究中发挥着重要作用。传统的光电倍增管(PMT)体积较大,对设计带来了诸多限制,进而影响阵列的效率、紧凑性和颗粒度等关键性能指标的提升,可采用体积更小的硅光电倍增器(SiPM)作为替代方案。然而,与PMT相比,SiPM的性能对温度的敏感性更显著,这一效应和闪烁体自身的温度效应相互叠加,进一步增加了探测器的使用复杂性。本工作将6 cm×6 cm×12 cm的大尺寸锗酸铋(BGO)闪烁体和8×8通道的SiPM阵列耦合形成BGO-SiPM探测器,并对其在不同温度下的工作条件进行了测试,确定了其最佳工作电压,得到了最佳能量分辨率。同时,还采用PMT对同一BGO闪烁体进行读出(BGO-PMT探测器)并在相同温度下进行了对比测量。实验结果显示,当温度高于20℃时,SiPM迅速上升的暗电流会导致探测器能量分辨变差;20℃以下,BGO-SiPM和BGO-PMT探测器性能接近,但BGO-SiPM探测器的能量分辨率随温度降低的提升速度略优于BGO-PMT探测器,这主要归因于SiPM暗电流随温度降低而减小。此外,本工作还研究了20℃附近BGO-SiPM探测器峰道址随温度的变化关系。研究表明,大尺寸BGO-SiPM探测器性能相较BGO-PMT探测器具有一定的优势,但峰道址的温度敏感性更强,为SiPM在BGO探测阵列的应用提供重要参考。

Self-adaptive behavior of nunchakus-like tracer induced by active Brownian particles

We design a nunchakus-like tracer and investigate its self-adaptive behavior in an active Brownian particle(ABP)bath via systematically tuning the self-propelled capability and density of ***,the nunchakus-like tracer will have a stable wedge-like shape in the ABP bath when the self-propelled force is high *** analyze the angle between the two arms of the tracer and the velocity of the joint point of the *** angle exhibits a non-monotonic phenomenon as a function of active ***,it increases with density of ABPs increasing monotonically.A simple linear relationship between the velocity and the self-propelled force is found under the highly active *** other words,the joint points of the tracer diffuse and the super-diffusive behavior can make the relation between the self-propelled force and the density of ABPs persist *** addition,we find that the tracer can flip at high density of *** results also suggest the new self-adaptive model research of the transport properties in a non-equilibrium medium.

Rapid Inactivation of Chloroplastic Ascorbate Peroxidase is Responsible for Oxidative Modification to Rubisco in Tomato (Lycopersicon esculentum) under Cadmium Stress

To investigate the sensitive site of antioxidant systems in chloroplast under cadmium stress and its consequence on reactive oxygen species production and action, the sub-organellar localization of chloroplast superoxide dismutases （SOD, EC 1.***.1.1） and ascorbic peroxidase （APX, EC 1.***.1.11） isoenzymes and changes of enzymes activities under cadmium stress were investigated in tomato seedlings. Two APX isoforms, one thylakoid-bound and one stromal, were detected. Cd at 50 μM induced a moderate increase of SOD activities but a rapid inactivation of both APX isoenzymes. APX inactivation was mainly related to the decrease of ascorbate concentration, as supported by in vitro treatment of exogenous ascorbate and APX kinetic properties under Cd stress. H2O2 accumulation in chloroplast, as a consequence of APX inactivation, was associated with a 60% loss of Rubisco （EC 4.***.1.39） activity, which could be partially accounted for by a 10% loss of Rubisco content. Protein oxidation assay found that the Rubisco large subunit was the most prominent carbonylated protein; the level of carbonylated Rubisco large subunit increased fivefold after Cd exposure. Thiol groups in the Rubisco large subunit were oxidized, as indicated by non-reducing electrophoresis. Treating crude extract with H2O2 resulted in a similar pattern of protein oxidation and thiols oxidation with that observed in Cd-treated plants. Our study indicates that APXs in the chloroplast is a highly sensitive site of antioxidant systems under Cd stress, and the inactivation of APX could be mainly responsible for oxidative modification to Rubisco and subsequent decrease in its activity.

The new synthesis of sesquiterpenoids 10-bromo-α-chamigrene

The new synthesis of 10-bromo-α-chamigrene was achieved as follows; 6-methyl-5-heptene-2-one was transformed into corresponding thioacetals, and then successively treated with Cp2Ti（P（OEt）3）2. The intermediate reacted with mono-ketal of cyclohexane-1,4-dione, and gave the carbonyl coupling product. It was then transformed into the key intermediate γ-bisabolene via deketalization, Grignard reaction, dehydration and then furnished the target molecule by polyene cyclization, with total yield 2%. All structures were confirmed by ^1H NMR and ^13C NMR. The final compound was confirmed by ^1H NMR, ^13C NMR and MS.
