Dehydrating stresses trigger the accumulation of abscisic acid(ABA),a key plant stress-signaling hormone that activates Snf1-Related Kinases(SnRK2s)to mount adaptive ***,the regulatory circuits that terminate the SnRK2s signal relay after acclimation or post-stress conditions remain to be ***,we show that the desensitization of the ABA signal is achieved by the regulation of OST1(SnRK2.6)protein stability via the E3-ubiquitin ligase *** ABA signal,HOS15-induced degradation of OST1 is inhibited and stabilized OST1 promotes the stress *** the ABA signal terminates,protein phosphatases ABI1/2 promote rapid degradation of OST1 via ***,we found that even in the presence of ABA,OST1 levels are also depleted within hours of ABA signal *** unexpected dynamics of OST1 abundance are then resolved by systematic mathematical modeling,demonstrating a desensitizing feedback loop by which OST1-induced upregulation of ABI1/2 leads to the degradation of *** model illustrates the complex rheostat dynamics underlying the ABA-induced stress response and desensitization.

建立了藏茶中88种农药残留的改进QuEChERS/超高效液相色谱-串联质谱(QuEChERS/UPLC-MS/MS)检测方法。藏茶样品以1%乙酸乙腈提取和2 g NaCl盐析处理,经交联聚乙烯基吡咯烷酮(PVPP)、十八烷基键合硅胶(C18)、石墨化炭黑(GCB)净化后,以0.01%甲酸水溶液(含2 mmol/L甲酸铵)和0.01%甲酸甲醇为流动相,采用ACQUITY UPLC BEH C18(50 mm×2.1 mm,1.7µm)色谱柱在15 min内实现分离。在电喷雾正离子模式下,采用多反应监测(MRM)模式检测,基质匹配标准曲线外标法定量。结果表明,藏茶中88种农药在相应质量浓度范围内线性良好(相关系数r≥0.9990),检出限(LOD)和定量下限(LOQ)分别为0.15~3µg/kg和0.5~10µg/kg。在1、2、10倍LOQ 3个加标水平下,平均回收率分别为73.2%~108%、73.2~109%、72.5%~110%,相对标准偏差(RSD)均不大于12%。将该方法应用于12批实际样品检测,所有样品均检出农药,其中有4批次样品中吡虫啉超限量值。该方法灵敏度高、简便快捷,能够满足藏茶中88种农药的快速检测要求。

The international community has made significant efforts to flatten the COVID-19 curve,including predicting transmission[1,2],executing unprecedented global lockdowns and social distancing[3,4],promoting the wearing of facemasks and social distancing measures[5],and isolating confirmed cases and contacts[6].Because of the adverse consequences of these lockdown measures[7],many cities have reopened so they can rebuild their ***,as mobility has gradually returned towards normal,imported cases from unknown sources have disrupted the recovery situation,and cities are continually at high risk of new waves of infection[8,9]since airborne transmission is the dominant transmission route[10].

Reduced corporal fibrosis to protect erectile function by inhibiting the Rho-kinase/lim-kinase/ cofilin pathway in the aged transgenic rat harboring human tissue kallikrein 1

Our previous studies have demonstrated that erectile function was preserved in aged transgenic rats （TGR） harboring the human tissue kallikrein 1 （hKLK1）, while the molecular level of hKLK1 on corporal fibrosis to inhibit age-related erectile dysfunction （ED） is poorly understood. Male wild-type Sprague-Dawley rats （WTR） and TGR harboring the hKLK1 gene were fed to 4- or 18-month-old and divided into three groups： young WTR （yWTR） as the control, aged WTR （aWTR）, and aged TGR （aTGR）. Erectile function of all rats was assessed by cavernous nerve electrostimulation method. Masson＇s trichrome staining was used to evaluate corporal fibrosis in the corpus cavernosum. We found that the erectile function of rats in the aWTR group was significantly lower than that of other two groups. Masson＇s trichrome staining revealed that compared with those of the yWTR and aTGR groups, the ratio of smooth muscle cell （SMC）/collagen （C） was significantly lower in the aWTR group. Immunohistochemistry and Western blotting analysis were performed, and results demonstrated that expression of a-SMA was lower, while expressions of transforming growth factor-β1 （TGF-β1）, RhoA, ROCK1, p-MYPT1, p-limK2, and p-cofilin were higher in the aWTR group compared with those in other two groups. However, limK2 and cofilin expressions did not differ among three groups. Taken together, these results indicated that the RhoA/ROCK1/limK/cofilin pathway may be involved in the corporal fibrosis caused by advanced age, and hKLK1 may reduce this corporal fibrosis by inhibiting the activation of this pathway to ameliorate age-related ED.

Background:Bariatric surgery represents an important treatment option for severely obese patients with nonalcoholic fatty liver disease(NAFLD).However,there remains inadequate data regarding the effects of different bariatric procedures on various NAFLD parameters,especially for histological ***,this meta-analysis aimed to compare the effects of restrictive bariatric procedures and foregut bypass on the metabolic,biochemical,and histological parameters for patients with ***:Medline and Embase were searched for articles relating to bariatric procedures and *** meta-analysis was conducted to compare efficacy of bariatric procedures ***-procedure with subgroup analysis to further compare restrictive against foregut bypass ***:Thirty-one articles involving 3,355 patients who underwent restrictive bariatric procedures(n=1,460)and foregut bypass(n=1,895)were *** foregut bypass(P<0.01)and restrictive procedures(P=0.03)significantly increased odds of fibrosis *** to restrictive procedures,foregut bypass resulted in a borderline non-significant decrease in fibrosis score(P=0.06)and significantly lower steatosis score(P<0.001).For metabolic parameters,foregut bypass significantly lowered body mass index(P=0.003)and low-density lipoprotein(P=0.008)compared to restrictive *** significant differences were observed between both procedures for aspartate aminotransferase(P=0.17)and alkaline phosphatase(P=0.61).However,foregut bypass resulted in significantly lower gamma-glutamyl transferase than restrictive procedures(P=0.01)while restrictive procedures resulted in significantly lower alanine transaminase than foregut bypass(P=0.02).Conclusions:The significant histological and metabolic advantages and comparable improvements in biochemical outcomes support the choice of foregut bypass over restrictive bariatric procedures in NAFLD management.

Decreased mitochondrial deoxyribonucleic acid and increased oxidative damage in chronic hepatitis C

AIM： To determine whether alteration of the mito- chondria DNA （mtDNA） copy number and its oxidative damage index （mtDNA△CT） can be detected by analysis of peripheral blood cells in hepatitis C virus （HCV）- infected patients. METHODS： This study enrolled two groups of pa- tients aged 40-60 years： a control group and an HCV- infected group in Department of Gastroenterology and Hepatology in Changhua Christian Hospital. Patients with co-infection with hepatitis B virus or human im- munodeficiency virus, autoimmune disease, malignant neoplasia, pregnancy, thyroid disease, or alcohol con- sumption 〉 40 g/d were excluded. HCV-infected pa- tients who met the following criteria were included：（1） positive HCV antibodies for 〉 6 mo; （2） alanine aminotransferase （ALT） levels more than twice the upper lim- it of normal on at least two occasions during the past 6 mo; and （3） histological fibrosis stage higher than F1. The mtDNA copy number and oxidative damage index of HCV mtDNA （mtDNA△CT） were measured in periph- eral blood leukocytes. The association between mtDNA copy number and mtDNA△CT was further analyzed using clinical data. RESULTS： Forty-seven normal controls （male/female： 26/21, mean age 50.51 ± 6.15 years） and 132 HCV- infected patients （male/female： 76/61, mean age 51.65 ± 5.50 years） were included in the study. The geno- types of HCV-infected patients include type 1a （n = 3）, type 1b （n = 83）, type 2a （n = 32）, and type 2b （n = 14）. Liver fibrosis stages were distributed as follows： F1/F2/F3/F4 = 1/61/45/25 and activity scores were A0/ A1/A2/A3 = 7/45/55/25. There were no age or gender differences between the two groups. HCV-infected pa- tients had higher hepatitis activity （aspartate transami- nase levels 108.77 ± 60.73 vs 23.19 ± 5.47, P 〈 0.01; ALT levels 168.69 ± 93.12 vs 23.15 ± 9.45, P 〈 0.01） and lower platelet count （170.40±58.00 vs 251.24 ± 63.42, P 〈 0.01） than controls. Purpose: To report the identification of a novel heterozygous mutation of CRYGC in a Chinese proband with congenital nuclear ***: The proband and several family members underwent a complete ophthalmic examination. The genomic DNA was extracted from peripheral blood leukocytes. 134 nuclear cataracts related genes, GJA8, MIP, lim2, CRYGC, were screened for causative mutations by directly ***: Three mutations were observed in the proband. Two affected individuals(proband and daughter) showed a sequence variation in the candidate gene CRYGC encoding γ-Crystallin a c.470 G>A transversion in exon 3 of CRYGC, predicted to introduce a translation stop codon tryptophan 157(W157 X). This substitution, consider as primary mutation,was not absent from an unaffected family members(parents and wife).Conclusion: The W157 X mutant of CRYGC is association with autosomal dominant congenital cataracts.
