Background:Hypertension and non-alcoholic fatty liver disease(NAFLD)share several pathophysiologic risk factors,and the exact relationship between the two remains unclear.Our study aims to provide evidence concerning the relationship between hypertension and NAFLD by analyzing data from the National Health and Nutrition Examination Survey(NHANES)2017-2018 and Mendelian randomization(MR)analyses.Methods:Weighted multivariable-adjusted logistic regression was applied to assess the relationship between hypertension and NAFLD risk by using data from the NHANES 2017-2018.Subsequently,a two-sample MR study was performed using the genome-wide association study(GWAS)summary statistics to identify the causal association between hypertension,systolic blood pressure(SBP),diastolic blood pressure(DBP),and NAFLD.The primary inverse variance weighted(IVW)and other supplementary MR approaches were conducted to verify the causal association between hypertension and NAFLD.Sensitivity analyses were adopted to confirm the robustness of the results.Results:A total of 3144 participants were enrolled for our observational study in NHANES.Weighted multivariable-adjusted logistic regression analysis suggested that hypertension was positively related to NAFLD risk(odds ratio[OR]=1.677;95%confidence interval[CI],1.159-2.423).SBP≥130 mmHg and DBP≥80 mmHg were also significantly positively correlated with NAFLD.Moreover,hypertension was independently connected with liver steatosis(β=7.836[95%CI,2.334-13.338]).The results of MR analysis also supported a causal association between hypertension(OR=7.203[95%CI,2.297-22.587])and NAFLD.Similar results were observed for the causal exploration between SBP(OR=1.024[95%CI,1.003-1.046]),DBP(OR=1.047[95%CI,1.005-1.090]),and NAFLD.The sensitive analysis further confirmed the robustness and reliability of these findings(all P>0.05).Conclusion:Hypertension was associated with an increased risk of NAFLD.

Adjusting agronomic measures to alleviate the kernel position effect in maize is important for ensuring high yields.In order to clarify whether the combined application of organic fertilizer and chemical fertilizer(CAOFCF)can alleviate the kernel position effect of summer maize,field experiments were conducted during the 2019 and 2020 growing seasons,and five treatments were assessed:CF,100%chemical fertilizer;OFCF1,15%organic fertilizer+85%chemical fertilizer;OFCF2,30%organic fertilizer+70%chemical fertilizer;OFCF3,45%organic fertilizer+55%chemical fertilizer;and OFCF4,60%organic fertilizer+40%chemical fertilizer.Compared with the CF treatment,the OFCF1 and OFCF2 treatments significantly alleviated the kernel position effect by increasing the weight ratio of inferior kernels to superior kernels and reducing the weight gap between the superior and inferior kernels.These effects were largely due to the improved filling and starch accumulation of inferior kernels.However,there were no obvious differences in the kernel position effect among plants treated with CF,OFCF3,or OFCF4 in most cases.Leaf area indexes,post-silking photosynthetic rates,and net assimilation rates were higher in plants treated with OFCF1 or OFCF2 than in those treated with CF,reflecting an enhanced photosynthetic capacity and improved postsilking dry matter accumulation(DMA)in the plants treated with OFCF1 or OFCF2.Compared with the CF treatment,the OFCF1 and OFCF2 treatments increased post-silking N uptake by 66.3 and 75.5%,respectively,which was the major factor driving post-silking photosynthetic capacity and DMA.Moreover,the increases in root DMA and zeatin riboside content observed following the OFCF1 and OFCF2 treatments resulted in reduced root senescence,which is associated with an increased post-silking N uptake.Analyses showed that post-silking N uptake,DMA,and grain yield in summer maize were negatively correlated with the kernel position effect.In conclusion,the combined application of 15-30%organic fertilizer and 70-85%chemical fer

Inevitable geometric variations significantly affect the performance of turbines or even that of entire engines;thus,it is necessary to determine their actual characteristics and accurately estimate their impact on performance.In this study,based on 1781 measured profiles of a typical turbine blade,the statistical characteristics of the geometric variations and the uncertainty impact are analyzed,and some commonly used uncertainty modelling methods based on Principal-Component Analysis(PCA)are verified.The geometric variations are found to be evident,asymmetric,and non-uniform,and the non-normality of the random distributions is non-negligible.The performance is notably affected,which is manifested as an overall offset,a notable scattering,and significant deterioration in several extreme cases.Additionally,it is demonstrated that the PCA reconstruction model is effective in characterizing major uncertainty characteristics of the geometric variations and their impact on the performance with almost the first 10 PCA modes.Based on a reasonable profile error and mean geometric deviation,the Gaussian assumption and stochasticprocess-based model are also found to be effective in predicting the mean values and standard deviations of the performance variations.However,they fail to predict the probability of some extreme cases with high loss.Finally,a Chi-square-based correction model is proposed to compensate for this deficiency.The present work can provide a useful reference for uncertainty analysis of the impact of geometric variations,and the corresponding uncertainty design of turbine blades.

IGF2-NR4A2 Signaling Regulates Macrophage Subtypes to Attenuate Liver Cirrhosis

Background and Aims:Liver cirrhosis can lead to liver failure and eventually death.Macrophages are the main contributors to cirrhosis and have a bidirectional role in regulating matrix deposition and degradation.Macrophage-based cell therapy has been developed as an alternative to liver transplantation.However,there is insufficient evidence regarding its safety and efficacy.In this study,we aimed to explore the effect of combining insulin-like growth factor 2(IGF2)with bone marrow-derived macrophages(BMDMs)to treat mice with liver cirrhosis.Methods:We assessed liver inflammation,fibrosis regression,liver function,and liver regeneration in mice with CCl4-induced cirrhosis and treated with BMDM only or IGF2+BMDM.We performed in vitro experiments in which activated hepatic stellate cells(HSCs)were co-cultured with macrophages in the presence or absence of IGF2.The polarity of macrophages and the degree of inhibition of HSCs were examined.The effect of IGF2 on macrophages was also verified by the overexpression of IGF2.Results:Combining IGF2 with BMDM reduced liver inflammation and fibrosis and increased hepatocyte proliferation.Combining IGF2 with BMDM was more effective than using BMDM alone.In vitro experiments demonstrated that IGF2 inhibited HSCs activation by upregulating NR4A2 to promote the anti-inflammatory macrophages phenotype.IGF2 also increased the synthesis of matrix metalloproteinases(MMPs)by macrophages,which may explain why administering IGF2 combined with BMDM was more effective than administering BMDM only.Conclusions:Our study provides a theoretical basis for the future use of BMDM-based cell therapy to treat liver cirrhosis.

在涡轮的典型工作剖面内,采用计算流体力学和两种多学科耦合求解方式对某二级低压涡轮部件进行仿真,比较不同求解方式所预测的涡轮气动热力参数之间的差异,分析工作剖面内局部几何的形变规律以及局部形变对流场的影响,结果表明:多学科耦合作用对低叶高位置叶型负荷分布产生一定的影响,具体影响规律与不同工况密切相关。考虑固体域热传导和形变的多学科耦合求解方式所预测的效率值更高。随着涡轮功率的提升,级间轮缘间歇和封严篦齿间隙的改变会导致更多封严冷气聚集在前腔中,影响盘腔内的流动结构,削弱了燃气入侵现象,实现盘腔内更好的冷却和封严效果。

教育评价改革是提高科研创新人才培养质量的指挥棒。基于“双一流”建设高校博士后的实证调查发现,博士后对评价指标柔性化和激励性评价较为满意,但对学术评价环境和留校转聘制度的评价较低;“操作层”的指标合理性、激励性评价及留校转聘评价均对博士后科研创新能力及其子维度发挥显著正向影响,而“环境层”的唯论文倾向、功利化导向则分别产生负向和正向影响;学术评价指标对博士后科研创新能力的影响效应在性别、学科类型、聘任类型、毕业院校方面存在异质性。高校管理者应科学认识学术评价的“双刃剑”效应,将评价导向转向激励性、赋能性、使动性;重塑“生态评价观”,系统构建人本取向、贡献导向的分类评价体系;根据博士后的“最近发展区”,设置具有挑战性和激励性的留校转聘标准。

火力发电的主要能源来自煤炭,而由于燃煤发电过程中产生的煤尘扩散是引起火电厂粉尘爆炸风险和尘肺职业病的主要根源之一,因此必须进行实时有效的检测和控制。当前粉尘检测方法仅使用单一的粉尘质量浓度指标来评估粉尘污染整体状况,缺乏对多种复合因素影响的考量,依靠单一阈值设定进行报警,易出现误报、漏报等现象,以及忽略粉尘爆炸这一重要事故场景,不能建立粉尘污染全面客观的评价方法。研究建立了一种粉尘质量浓度预测模型,基于金豺优化算法对极限学习机的最优初始权重进行寻优,再使用极限学习机对样本数据进行训练学习,提高神经网络模型的精度,可较为准确地预测30 min以内任意时间间隔的粉尘质量浓度,并将现场数据及模拟仿真数据与建立的粉尘质量浓度预测模型进行对比分析。结果显示:建立的粉尘质量浓度预测模型准确度良好,与现场数据及模拟仿真数据对比误差分别为0.72%和2.1%,可加强对火电厂粉尘环境进行预测预警,从而及时采取合理的粉尘控制策略,确保火电厂的生产安全并降低粉尘对作业人员的职业危害。

如何有效调控汇率稳定是推动高质量发展和高水平对外开放的重要现实议题之一。本文构建了包含央行汇率稳定政策的内生理论模型,并基于时变参数结构向量自回归模型(TVP-SVAR),采用1985—2021年我国、日本和英国的季度数据集,检验了价格型货币政策、数量型货币政策和外汇干预三类央行政策对汇率的非线性影响效应和传导效率,得出如下结论:(1)我国货币政策对宏观经济的整体调控效果优于日本和英国,且数量型货币政策优于价格型货币政策;(2)与其他国家不同,我国价格型货币政策对本国汇率的平均调控效果优于数量型货币政策,体现出稳定的逆向调控作用;(3)外汇干预与货币政策协调,能够有效维护人民币汇率稳定,但其调控效果具有短期性和“靶向性”。基于此,本文进一步从优化货币政策调控机制、加强外汇干预的定向性和时机选择、制定长期战略规划等方面提出了政策建议,旨在为有效调控汇率,防范化解重大风险提供政策启示。

目的探讨人脐带间充质干细胞(hUC-MSC)对肝纤维化小鼠的治疗作用及其机制。方法选取18只SPF级6周龄C57BL/6小鼠,使用随机数字表法随机分为3组,分别为对照组、CCl_(4)模型组(CCl_(4)组)和h UC-MSC治疗组(MSC组),每组6只。CCl_(4)组和MSC组腹腔注射CCl_(4)溶液构建小鼠肝纤维化模型,对照组同时注射相同剂量玉米油,MSC组在注射CCl_(4)的过程经尾静脉注射hUC-MSC。于第8周末采取小鼠血清,处死小鼠,取小鼠肝脏固定。使用酶联免疫吸附法测定炎症因子水平,自动生化检测仪检测肝功能相关指标,HE染色、Masson染色、天狼星红染色及α-SMA免疫荧光用于评估肝纤维化情况;TGF-β刺激的肝星状细胞(HSC)分别在有无几丁质酶3样蛋白1(CHI3L1)添加的培养基中与hUC-MSC共培养,Western Blot试验检测蛋白表达水平。计量资料多组间比较采用单因素方差分析,进一步两两比较采用Dunnett-t检验。结果 Masson染色、天狼星红染色提示CCl_(4)组小鼠纤维化较对照组明显(P值均<0.05),MSC组小鼠纤维化较CCl_(4)组减轻(P值均<0.05)。CCl_(4)组IL-1β、IL-6、AST、ALT和ALP水平较对照组明显升高(P值均<0.05),MSC组IL-6、AST、ALT及ALP水平较CCl_(4)组明显降低(P值均<0.05)。CCl_(4)组CHI3L1和α-SMA的表达均高于对照组和MSC组(P值均<0.05)。细胞培养实验显示,MSC+HSC组Bax表达高于HSC组和MSC+CHI3L1组(P值均<0.05),提示CHI3L1逆转了MSC对活化的HSC的促凋亡作用。结论 hUC-MSC治疗可以改善小鼠肝纤维化,其作用机制可能与抑制CHI3L1从而促进HSC的凋亡相关。
