Effects of maternal methyl donor intake during pregnancy on ileum methylation and function in an intrauterine growth restriction pig model

Standardized ileal digestibility of amino acids in soybean meal fed to non-pregnant and pregnant sows

Artificial parasin I protein (API) supplementation improves growth performance and intestinal health in weaned piglets challenged with enterotoxigenic Escherichia coli

Diarrheas are common risks faced by piglets during the weaning period. This study investigated the alleviating effects of artificial parasin I protein (API) on growth performance and intestinal health of weaned pigs upon enterotoxigenic Escherichia coli (ETEC) challenge. Sixty piglets were randomly divided into five groups and fed a basal diet (CON) or basal diet supplemented with API at 0, 750, and 1500 mg/kg or antibiotics for 5 weeks. On d 15 and 25, piglets were challenged with ETEC K88 except for the CON group. Before the ETEC challenge (d 1-14), dietary API supplementation improved growth performance, and 750 mg API increased (P < 0.05) the average daily gain (ADG), decreased (P < 0.05) feed to gain ratio (F/G) and diarrhea index of weaned piglets. ETEC challenge (during d 15-35) reduced growth performance and increased (P < 0.01) the F/G, diarrhea rate, and diarrhea index. This event was accompanied by the numerically increased malondialdehyde (MDA) levels in serum and ileum, the decreased (P < 0.05) zonula-occludens-1 (ZO-1) and interleukin-6 (IL-6) in the ileum, and the increased (P 1/4 0.04) secretory immunoglobulin A (sIgA) protein in the ileum. Artificial parasin I protein supplementation alleviated the negative impact of ETEC. The 750 mg/kg API inclusion elevated (P < 0.05) ADG and decreased (P < 0.05) F/G. Two levels of API decreased (P < 0.01) the diarrhea rate and diarrhea index. Meanwhile, API inclusion decreased (P < 0.01) the crypt depth in the jejunum, elevated (P < 0.05) villus height in the duodenum and villus height to crypt depth ratio in the duodenum and ileum, up-regulated (P < 0.05) ZO-1 gene, and down-regulated (P < 0.05) mucin-2 gene in the jejunum, and 1500 mg/kg API decreased (P < 0.01) sIgA level and down-regulated (P < 0.05) IL-1b gene in the ileum. Selenomethionine alleviates chronic heat stress-induced breast muscle injury and poor meat quality in broilers via relieving mitochondrial dysfunction and endoplasmic reticulum stress

In the present study, the chronic heat stress (CHS) broiler model was developed to investigate the potential protection mechanism of organic selenium (selenomethionine, SeMet) on CHS-induced skeletal muscle growth retardation and poor meat quality. Four hundred Arbor Acres male broilers (680 ± 70 g, 21 d old) were grouped into 5 treatments with 8 replicates of 10 broilers per replicate. Broilers in the control group were raised in a thermoneutral environment (22 ± 2 °C) and fed with a basal diet. The other four treatments were exposed to hyperthermic conditions (33 ± 2 °C, 24 h in each day) and fed on the basal diet supplied with SeMet at 0.0, 0.2, 0.4, and 0.6 mg Se/kg, respectively, for 21 d. Results showed that CHS reduced ( P 0.05) the growth performance, decreased ( P 0.05) the breast muscle weight and impaired the meat quality of breast muscle in broilers. CHS induced protein metabolic disorder in breast muscle, which increased ( P 0.05) the expression of caspase 3, caspase 8, caspase 9 and ubiquitin proteasome system related genes, while decreased the protein expression of P-4EBP1. CHS also decreased the antioxidant capacity and induced mitochondrial stress and endoplasmic reticulum (ER) stress in breast muscle, which increased ( P 0.05) the ROS levels, decreased the concentration of ATP, increased the protein expression of HSP60 and CLPX, and increased ( P 0.05) the expression of ER stress biomarkers. Background:Determination of nutrient requirements in the late gestating and lactating sows is essential to optimize sow *** objectives of the present study were to quantify amino acid(AA)fluxes and heat production across portal-drained viscera(PDV)and liver in multiparous sows during transition and ***:Eight second parity sows were fitted with indwelling catheters in the femoral artery and in the mesenteric,portal and hepatic *** hourly sets of blood samples were taken starting 0.5 h before feeding at−10,−3,+3,and+17 d in milk(DIM).Blood gases,plasma metabolites and apparent total tract digestibility(ATTD)of nutrients were ***:Feed intake,the ATTD of DM,energy,nitrogen,fat and crude fiber changed with DIM(P<0.001).Except for Glu,O2,and urea,all net portal fluxes were positive,and all were affected by DIM(P<0.05)and by sampling time(P<0.01).Compared with pre partum levels,net portal uptake of AA was 3-63%lower at+3 DIM but 40-100%higher at+17 *** portal fluxes of AA peaked at 1.5 to 2.5 h after feeding except for Glu,and they were positively correlated with changes in sow feed intake across *** net portal recovery was low for Met(49%),Thr(54%),and His(54%)and high for the remaining essential AA(63-69%)and none of them differed across *** hepatic uptake(*** oxidation)of Lys,Thr,Ile,Leu and Phe peaked at 0.5 to 2.5 h after feeding,whereas uptake of Trp,Val,and His was constant,while that of Met was close to ***:The net portal recovery was substantially lower for Met,Thr,and His than the remaining essential *** AA oxidation peaks 0.5 to 2.5 h after *** heat production in PDV and liver was approximately two-fold higher at peak lactation compared to other *** study suggests that lysine was the limiting AA in peak lactation but not in early lactation.

Inclusion of purified dietary fiber during gestation improved the reproductive performance of sows

Background: This study aimed to investigate the impacts of guar gum and cellulose as the source of dietary fiber during gestation on the reproductive performance of ***: A total of 210 sows(parities 3–6) were randomly allocated into six diets(n = 35) throughout gestation to feed graded levels of dietary fiber(DF), including a corn-soybean meal-based control diet with no wheat bran inclusion(CON, 12.5% DF), a wheat bran-rich diet(DF1, 17.4% DF), and another 4 diets(DF2, 17.7% DF;DF3, 18.1%DF;DF4, 18.4% DF;DF5, 18.8% DF) in which wheat bran were equally substituted by 1%, 2%, 3% and 4% purified FIBER MIX(guar gum and cellulose, 1:4). All sows received similar DE and other nutrients throughout ***: DF treatment during gestation resulted in normal fecal score(1 to 5 with 1 = dry and 5 = watery) in sows compared with those received the CON diet(P < 0.05). The number of total born piglets had a tendency to be affected by dietary treatment(P = 0.07), and correlation analysis revealed a linear response of total born to dietary fiber levels during gestation(P < 0.01). Sows received the DF2, DF3, and DF5 diets during gestation had a greater ADFI during lactation compared with those in the CON group(P < 0.05) without affecting the daily body weight gain of suckling piglets. Gut microbiota compositions were dramatically changed by the gestation stage and some of those were changed by DF inclusion. Fecal acetate, propionate, and butyrate of sows were markedly increased in late gestation, and butyrate contents in feces of gestating sows were significantly affected by DF levels(P < 0.01).Serum concentrations of pro-inflammatory TNF-α were decreased and anti-inflammatory IL-10 was increased on day30 of gestation by DF levels(P < 0.05).Conclusions: In summary, increasing dietary fiber levels by guar gum and cellulose during gestation improved the reproductive performance of sows, which might be related to changes in immunity and gut microbiota of sows.

Leucine modulates the IPEC-J2 cell proteome associated with cell proliferation, metabolism and phagocytosis

Leucine can affect intestinal protein expressions, and improve mucosal immune function. However, little study has been conducted to determine the change of protein component by leucine treatment in intestine epithelial cells. The present study was to cover the key proteins and cell pathways that could be regulated by leucine treatment in porcine intestinal epithelial cell line(IPEC-J2) cells with the approach of proteome analysis. A total number of 3,211 proteins were identified in our approach by searching the database of Uniprot sus scrofa. Among identified proteins, there were 101 proteins expressed differently between control group and leucine group. Compared with the control group, there were 50 up-regulated proteins and 51 down-regulated proteins in leucine group. In these proteins, leucine treatment decreased the expression of some proteins including pyruvate kinase, glyceraldehyde-3-phosphate dehydrogenase,E3 ubiquitin ligase, cathepsin D, caspase 3 and caspase 6, and increased the levels of some proteins, such as some eukaryotic translation initiation factors, ubiquitin carboxyl-terminal hydrolase, DNA-related RNA polymerase II, urokinase plasminogen activator, cyclin-dependent kinase inhibitor 2 b, Mut L homolog 1,5-methylcytosine binding domain 4, polymerase d, a-tubulin, syntaxin 18, Ras homolog D, actin related protein 2/3 complex and cofilin. Via the analysis of Gene Ontology and pathways, these proteins in IPECJ2 cells were related with some physiological functions, such as protein metabolism, glycolysis, cell proliferation, apoptosis and phagocytosis. Thus, these results suggest that leucine affects gut barrier function possibly via regulating cell proliferation and apoptosis, metabolism and phagocytosis.
