Habitat-specific changes of plant and soil microbial community composition in response to fairy ring fungus Agaricus xanthodermus on the Qinghai-Tibet Plateau

Fairy rings are common in diverse global biomes and often appear as lush vegetation in one to three concentric zones caused by the spread of mycelia in grassland ***,the underlying mechanisms and environmental adaptation of fairy rings remain largely *** this study,two fairy rings(A and B)caused by Agaricus xanthodermus were sampled on the Qinghai-Tibet Plateau during a time when fairy rings are most *** conducting a vegetation survey and high-throughput sequencing,the changes of plants and soil microorganisms to fairy ring fungi were *** above-ground biomass at both fairy rings was greatly increased by fairy ring fungi,but the response of dominant plant species is different at two fairy ring *** addition,bacterial and fungal communities significantly varied within distinct sampling zones across the fairy rings,and showed variable genus-specific responses at two fairy ring *** fairy ring A,soil available N:P ratio was essential in shaping the structure of plant and microbial community,while soil available N concentration was the most important predictor at fairy ring *** together,our results indicated Agaricus xanthodermus fairy rings have variable effects on alpine meadow plants and soil microbes at different *** propose that the impacts of fairy ring fungi on plants and microbes are determined by the level of soil available N concentration and available N:P *** results contribute to a better understanding of the mechanisms through which fairy rings affect the vegetation of alpine meadows.

NIR-IIb-triggered photodynamic therapy combined with chemotherapy platform based on rare-earth-doped nanoparticles

Photodynamic therapy(PDT),with high spatiotemporal selectivity,minimal side effects and less drug resistance,has emerged as a promising approach for the treatment of ***,inadequate penetration depth and poor efficacy seriously restrict its practical *** address these challenges,we developed a combined therapy platform(RENP@C/D-FA)based on rare-earth-doped nanoparticles(RENPs)with a structure of NaErF_(4):Tm(0.5%)@NaYF_(4),on which photosensitizer Ce6(chlorin e6)and chemo-therapeutic drug(DOX)(doxorubicin)were loaded by encapsulating with an amphiphilic polymer distearoyl phosphatidylethanolamine-polyethylene glycol-folic acid(DSPE-PEG-FA).RENPs with NaErF_(4) as the matrix possess multiple excitation/emission peaks,enabling activation of PDT under near-infrared-Ⅱb(NIR-Ⅱb,1500-1700 nm)excitation and luminescence imaging in NIR-Ⅱb region by absorbing 808 nm near-infrared ***,DOX can be selectively released in the acidic microenvironment of *** from the tumor targeting capability of folic acid,this platform can successfully ablate tumor cells and inhibit tumor growth by combining PDT and *** innovative approach holds great promise as a powerful tool for cancer treatment,showing the potential to revolutionize PDT.

Comparative DNA methylation reveals epigenetic adaptation to high altitude in snub-nosed monkeys

DNA methylation plays a crucial role in environmental ***,using whole-genome bisulfite sequencing,we generated comprehensive genome-wide DNA methylation profiles for the high-altitude yunnan snub-nosed monkey(Rhinopithecus bieti)and the closely related golden snub-nosed monkey(***).Our findings indicated a slight increase in overall DNA methylation levels in golden snub-nosed monkeys compared to yunnan snub-nosed monkeys,suggesting a higher prevalence of hypermethylated genomic regions in the *** genomic methylation analysis demonstrated that genes associated with differentially methylated regions were involved in membrane fusion,vesicular formation and trafficking,hemoglobin function,cell cycle regulation,and neuronal *** results suggest that the high-altitude-related epigenetic modifications are extensive,involving a complete adaptation process from the inhibition of single Ca^(2+)channel proteins to multiple proteins collaboratively enhancing vesicular function or inhibiting cell differentiation and *** assays demonstrated that overexpression or down-regulation of candidate genes,such as SNX10,TIMELESS,and CACYBP,influenced cell viability under stress ***,this research suggests that comparing DNA methylation across closely related species can identify novel candidate genomic regions and genes associated with local adaptations,thereby deepening our understanding of the mechanisms underlying environmental adaptations.

Fully distributed dynamic event-triggered formation-containment tracking for multiagent systems with multiple types of disturbances

This paper is dedicated to addressing the time-varying output formation-containment tracking(TVOFCT) problem for heterogeneous linear multiagent systems(MASs) with multiple types of disturbances under directed communication *** agents are divided into tracking leaders,formation leaders,and *** the output space,the formation leaders are required to form an expected geometric formation while tracking the tracking leader,and the followers are required to enter the convex hull spanned by formation ***,a disturbance observer is designed to observe the disturbance with deterministic ***,by combining adaptive technology with event-triggered technology,an independent fully distributed dynamic event-triggered(FDDET) compensator is designed to estimate the state of the tracking *** main advantages of this compensator lie in the following three aspects:(1) the adaptive weight does not increase unboundedly;(2) the interval time between events is expanded;(3) the design does not rely on any global *** to the design of this compensator,an independent FDDET convex hull observer is further designed to observe the convex hull spanned by formation ***,based on the designed disturbance observer,compensator,and convex hull observer,the adaptive disturbance rejection control input which can make the MASs achieve TVOFCT is ***,a numerical simulation is provided to clearly verify the validity of the theoretical results.

Targeting HMGCS1 restores chemotherapy sensitivity in acute myeloid leukemia

Acute myeloid leukemia(AML)is a common hematological malignancy with overall poor *** novel targets is urgent and necessary to improve the clinical outcome of relapsed and refractory(RR)AML *** clinical specimens,animal models and cell-level studies,we explored the specific mechanism of 3-hydroxy-3-methylglutaryl coenzyme A synthase 1(HMGCS1)in AML and the mechanism of targeting HMGCS1 to attenuate cell proliferation,increase chemotherapy sensitivity and improve the occurrence and development of ***,we reveal that HMGCS1 is overexpressed in RR patients and negatively related to overall survival(OS).Knocking out HMGCS1 in AML cells attenuated cell proliferation and increased chemotherapy sensitivity,while stable overexpression of HMGCS1 had the opposite ***,we identified that knockout of HMGCS1 suppressed mitogen-activated protein kinase(MAPK)pathway activity,while overexpression of HMGCS1 could remarkably enhance the pathway.U0126,a MEK1 inhibitor,offset the effects of HMGCS1 overexpression,indicating that HMGCS1 promotes RR AML through the MAPK ***,we verified that hymeglusin,a specific inhibitor of HMGCS1,decreases cell growth both in AML cell lines and primary bone marrow cells of AML ***,combination of hymeglusin and the common chemotherapeutic drug cytarabine and adriamycin(ADR)had synergistic toxic effects on AML *** study demonstrates the important role of HMGCS1 in AML,and targeting this protein is promising for the treatment of RR AML.

Impact of Annealing on the Melt Recrystallization of a-PDLA/α-PLLA Double-layered Films

Poly(lactic acid)(PLA) as a bio-based polymer with biodegradability and biocompatibility has attracted much attention. To manipulate its properties for different applications, regulation of crystal structure and crystalline morphology becomes an attractive research topic. In this work, the structure evolution of layered samples containing an amorphous poly(D-lactide)(PDLA) layer and a crystalline poly(L-lactide)(PLLA)layer with highly oriented edge-on α lamellar crystals after annealing at 150 ℃ or/and after melt-recrystallization has been studied by AFM, FTIR,and TEM combined with electron diffraction. The results demonstrate that melt recrystallization of the as-prepared sample leads to the formation of abundant randomly oriented PLA stereo-complex(PLA SC) crystals. Annealing at 150 ℃ results in the formation of a small amount of oriented PLA SC crystals at the interface. These PLA SC crystals show great impact on the recrystallization behavior of sample after melting at 190 ℃ and then crystallizing at 90 ℃. First, they impede the mutual diffusion of the overlying PDLA and underlying PLLA, and thus reduce their stereocomplexation ability as manifested by the decreased amount of PLA SC crystals. Second, they act as substrate to initiate the epitaxial crystallization of the overlying PDLA and underlying PLLA, which ensures the production of a highly oriented structure of PDLA and PLLA after melt recrystallization again.

Acute pancreatitis(AP)is a potentially fatal condition with no targeted treatment *** inhibiting xanthine oxidase(XO)in the treatment of AP has been studied in several experimental models and clinical trials,whether XO is a target of AP and what its the main mechanism of action is remains ***,we aimed to re-evaluate whether XO is a target aggravating AP other than merely generating reactive oxygen species that trigger *** first revealed that XO expression and enzyme activity were significantly elevated in the serum and pancreas of necrotizing AP *** also found that allopurinol and febuxostat,as purine-like and non-purine XO inhibitors,respectively,exhibited protective effects against pancreatic acinar cell death in vitro and pancreatic damage in vivo at different doses and treatment time ***,we observed that conditional Xdh overexpression aggravated pancreatic necrosis and *** mechanism analysis showed that XO inhibition restored the hypoxia-inducible factor 1-alpha(HIF-1α)-regulated lactate dehydrogenase A(LDHA)and NOD-like receptor family pyrin domain containing 3(NLRP3)signaling pathways and reduced the enrichment of^(13)C_(6)-glucose to^(13)C_(3)-***,we observed that clinical circulatory XO activity was significantly elevated in severe cases and correlated with C-reactive protein levels,while pancreatic XO and urate were also increased in severe AP *** results together indicated that proper inhibition of XO might be a promising therapeutic strategy for alleviating pancreatic necrosis and preventing progression of severe AP by downregulating HIF-1α-mediated LDHA and NLRP3 signaling pathways.

Size-dependent thermomechanical vibration characteristics of rotating pre-twisted functionally graded shear deformable microbeams

A three-dimensional(3D)thermomechanical vibration model is developed for rotating pre-twisted functionally graded(FG)microbeams according to the refined shear deformation theory(RSDT)and the modified couple stress theory(MCST).The material properties are assumed to follow a power-law distribution along the chordwise *** model introduces one axial stretching variable and four transverse deflection variables including two pure bending components and two pure shear *** complex modal analysis and assumed mode methods are used to solve the governing equations of motion under different boundary conditions(BCs).Several examples are presented to verify the effectiveness of the developed *** coupling the slenderness ratio,gradient index,rotation speed,and size effect with the pre-twisted angle,the effects of these factors on the thermomechanical vibration of the microbeam with different BCs are *** is found that with the increase in the pre-twisted angle,the critical slenderness ratio and gradient index corresponding to the thermal instability of the microbeam increase,while the critical material length scale parameter(MLSP)and rotation speed *** sensitivity of the fundamental frequency to temperature increases with the increasing slenderness ratio and gradient index,and decreases with the other increasing ***,the size effect can suppress the dynamic stiffening effect and enhance the Coriolis ***,the mode transition is quantitatively demonstrated by a modal assurance criterion(MAC).
