Antibody Levels and Infection Status of Pertussis in the Population under Pertussis Resurgence in Guangxi in 2018:A Cross-Sectional Survey

Objective Pertussis cases have increased markedly since 2018 in *** aim of this study was to evaluate antibody levels and the infection status of pertussis in the resident *** A total of 10,215 serum samples from residents were collected from August-November 2018 and tested for anti-pertussis IgG and toxin IgG using the enzyme-linked immunosorbent assay(ELISA).Results Of the collected samples,1,833(17.94%)tested positive for anti-pertussis IgG,with the median concentration of 16.06 IU/*** level<10 IU/mL accounted for more than 60%in children under 4 years of age,but declined with age,whereas the percentages of the other three levels(10-40,40-50,and≥50 IU/mL)increased almost with age(P<0.001).Moreover,7,924 samples were selected for anti-pertussis toxin IgG,of which 653(8.24%)tested positive(≥40 IU/mL)with the median concentration of 5.89 IU/mL,and 204 participants(2.56%)had recent pertussis infection(≥100 IU/mL).Among the different age groups,the highest rates of positivity and recent infection were observed at 11-20 years of age,the lowest positivity rate at 5 years of age,and the lowest recent infection rate at 4 years of age(P<0.001,P=0.005,respectively).Conclusion The survey results showed that all age groups in Guangxi lacked immunity against pertussis,which was one of the main factors contributing to the resurgence of pertussis in *** addition,the prevalence of pertussis is relatively high in Guangxi,and its incidence is seriously underestimated,especially in adolescents and adults.

Myeloid-derived Suppressor Cells Activate Liver Natural Killer Cells in a Murine Model in Uveal Melanoma

Objective Elevated myeloid-derived suppressor cells(MDSCs)in many malignancies are associated with the increased risk for metastases and poor ***,a mouse model of intraocular melanoma was established to explore how MDSCs influence liver *** In this study,murine B16LS melanoma cells were transplanted into the posterior compartment(PC)of the eye of C57BL/6 *** from the liver of naive mice and mice bearing melanoma liver metastasis were isolated using isotonic Percoll centrifugation,examined by flow cytometry for their expression of Gr1,CD11b,F4/80,RAE-1,and Mult-1,and further isolated for MDSCs and natural killer(NK)*** effects of MDSCs on NK cells were tested by coculturing and assessing the ability of NK cells to produce interferon-gamma(IFN-γ)by ELISA and NK cell cytotoxicity by 3H-thymidine incorporation *** impact of IFN-γon liver metastases was examined via selectively depleting IFN-γin *** The results showed that mice with liver metastases had increased levels of CD11b+Gr1+F4/80+as well as CD11b+Gr1+F4/80−*** significantly enhanced the generation of IFN-γtogether with the cytotoxicity of the NK ***,these effects were cell-cell *** IFN-γwas not of a toxic nature to the melanoma cells,it profoundly inhibited B16LS cell *** IFN-γin vivo led to increased liver *** All these findings first revealed that MDSCs accumulated in liver metastasis of intraocular melanoma could activate the NK cells to produce an effective anti-tumor immune ***,the MDSCs’performance in different tumor models would need more investigation to boost current immunotherapy modalities.

Relationship between autophagy and perineural invasion, clinicopathological features, and prognosis in pancreatic cancer

AIM To investigate the relationship between autophagy and perineural invasion(PNI), clinical features, and prognosis in patients with pancreatic cancer. METHODS Clinical and pathological data were retrospectively collected from 109 patients with pancreatic ductal adenocarcinoma who underwent radical resection at the First Affiliated Hospital of Zhengzhou University from January 2011 to August 2016. Expression levels of the autophagy-related protein microtubuleassociated protein 1 A/1 B-light chain 3(LC3) and PNI marker ubiquitin carboxy-terminal hydrolase(UCH) in pancreatic cancer tissues were detected by immunohistochemistry. The correlations among LC3 expression, PNI, and clinical pathological features in pancreatic cancer were analyzed. The patients were followed for further survival analysis. RESULTS In 109 cases of pancreatic cancer, 68.8%(75/109) had evidence of PNI and 61.5%(67/109) had high LC3 expression. PNI was associated with lymph node metastasis, pancreatitis, and CA19-9 levels(P < 0.05). LC3 expression was related to lymph node metastasis(P < 0.05) and was positively correlated with neural invasion(P < 0.05, r = 0.227). Multivariate logistic regression analysis indicated that LC3 expression, lymph node metastasis, pancreatitis, and CA19-9 level were factors that influenced neural invasion, whereas only neural invasion itself was an independent factor for high LC3 expression. Univariate analysis showed that LC3 expression, neural invasion, and CA19-9 level were related to the overall survival of pancreatic cancer patients(P < 0.05). Multivariate COX regression analysis indicated that PNI and LC3 expression were independent risk factors for poor prognosis in pancreatic cancer(P < 0.05). CONCLUSION PNI in patients with pancreatic cancer is positively related to autophagy. Neural invasion and LC3 expression are independent risk factors for pancreatic cancer with a poor prognosis.

Determination of Trace Elements in Edible Nuts in the Beijing Market by ICP-MS

Nuts have received increased attention from the public in recent years as important sources of some essential elements, and information on the levels of elements in edible nuts is useful to consumers. Determination of the elemental distributions in nuts is not only necessary in evaluating the total dietary intake of the essential elements, but also useful in detecting heavy metal contamination in food. The aim of this study was to determine the mineral contents in edible nuts, and to assess the food safety of nuts in the Beijing market. Levels of Li, Cr, Mn, Co, Cu, Zn, As, Se, Rb, Sr, Mo, Cd, Cs, Ba, Pb, Th, and U in 11 types of edible nuts and seeds （macadamia nuts, lotus nuts, pistachios, sunflower seeds, pine nuts, almonds, walnuts, chestnuts, hazelnuts, cashews, and ginkgo nuts） as well as raisins were determined by inductively coupled plasma mass spectrometry （ICP-MS）. The accuracy of the method was validated using standard reference materials GBWlO014 （cabbage） and GBWlO016 （tea）. Our results provide useful information for evaluating the levels of trace elements in edible nuts in the Beijing market, will be helpful for improving food safety, and will aid in better protecting consumer interests.

Effects of elevation on spring phenological sensitivity to temperature in Tibetan Plateau grasslands

Vegetation phenology is an important indicator of climate change impacts on the seasonal dynamics of the biosphere. However, little is known about the influence of elevation on spring phenological sensitivity to temperature in an alpine ecosystem. Based on remotely sensed land surface phenology and temperature data from 2001 to2010, this study investigated the rate of spring phenological change of the Tibetan Plateau(TP) grasslands in response to interannual temperature variations at different elevations. Results suggest that spring phenology in the TP grasslands exhibits a stronger response to changes in temperature at higher elevations than at lower ones. In particular, spring phenology advanced by 1–2 days in response to a 1 °C increase in May average temperature at elevations from 3,000 to 3,500 m, while the rate was up to8–9 days/°C at 5,000–5,500 m. Analysis using accumulated growing degree days(AGDD) from January 1through May 31 showed the same general trend with increased elevation associated with increased sensitivity(as measured by phenological change per unit of AGDDchange). Such temperature sensitivity gradients in the TP grasslands could be partly explained by the growth efficiency hypothesis which suggests that vegetation adapted to colder climates likely requires less heat energy for the onset of growing season and vice versa in warmer ***, accumulated growing degree days from January 1 to the greenup date were found to decrease with increasing elevations, which provided evidence to support the applicability of the growth efficiency hypothesis in an alpine grassland ecosystem.
