Baicalin Prevents Colon Cancer by Suppressing CDKN2A Protein Expression

Objective:To observe the therapeutic effects and underlying mechanism of baicalin against colon ***:The effects of baicalin on the proliferation and growth of colon cancer cells MC38 and *** were observed and predicted potential molecular targets of baicalin for colon cancer therapy were studied by network ***,molecular docking and drug affinity responsive target stability（DARTS）analysis were performed to confirm the interaction between potential targets and ***,the mechanisms predicted by in silico analyses were experimentally verified in-vitro and ***:Baicalin significantly inhibited proliferation,invasion,migration,and induced apoptosis in MC38 and CT26 cells（all P<0.01）.Additionally,baicalin caused cell cycle arrest at the S phase,while the G0/G1phase was detected in the tiny portion of the *** network pharmacology analysis identified 6 therapeutic targets associated with baicalin,which potentially affect various pathways including 39 biological processes and 99 sugnaling *** addition,molecular docking and DARTS predicted the potential binding of baicalin with cyclin dependent kinase inhibitor 2A（CDKN2A）,protein kinase B（AKT）,caspase 3,and mitogen-activated protein kinase（MAPK）.In vitro,the expressions of CDKN2A,MAPK,and p-AKT were suppressed by baicalin in MC38 and CT26 *** vivo,baicalin significantly reduced the tumor size and weight（all P<0.01） in the colon cancer mouse model via inactivating p-AKT,CDKN2A,cyclin dependent kinase 4,cyclin dependent kinase 2,interleukin-1,tumor necrosis factor α,and activating caspase 3 and mouse double minute 2 homolog signaling（all P<0.05）.Conclusion:Baicalin suppressed the CDKN2A protein level to prevent colon cancer and could be used as a therapeutic target for colon cancer.

Preparation of Chemically Recyclable Poly(ether-alt-ester) by the Ring Opening Polymerization of Cyclic Monomers Synthesized by Coupling Glycolide and Epoxides

Polyester and polyether are two key oxygenated polymers, and completely alternative sequence of poly(ester-alt-ether) could efficiently combine the advantages(including flexibility, degradability, etc.) of both segments. Currently, despite their copolymers could be synthesized from one-pot mixture of cyclic esters and epoxides, perfectly alternative microstructure is very challenging to realize and typically restricted to certain monomer pairs. Moving forward, synthesizing poly(ester-alt-ether) from commercially available and largescale monomers would be a significant advance. For example, successfully commercialized poly(glycolic acid)(PGA), which is not easily soluble in polymers due to its high crystallinity and is brittle and difficult to control the degradation cycle, would encounter a new paradigm if engineered into poly(ester-altether). In this work, starting from the design of monomer with hybrid structures, we successfully synthesized a series of 1,4-dioxan-2-one containing different substituents based on glycolide(GA) and epoxides using commercially available Salen-Cr(III) and PPNCl catalytic *** new monomers underwent ring-opening polymerization(ROP) to form a series of poly(ester-alt-ether) with perfectly alternating glycolic acid and propylene glycol repeat units under catalytic system of thiourea/base. The poly(ester-alt-ether) have significantly lower glass-transition temperature than PGA. Additionally, the poly(ester-alt-ether) can be chemically recovered to monomer using Sn(Oct)2 or 1,8-diazabicyclo[5.4.0]undecane-7-ene(DBU) as a catalyst in solution, thus establishing a closed-loop life cycle. From monomers derived from GA and epoxides, this work furnishes a novel strategy for the synthesis of poly(ester-alt-ether) with chemical recyclability.

Crude oil scheduling optimization is an effective method to enhance the economic benefits of oil *** uncertainties,including uncertain demands of crude distillation units(CDUs),might make the production plans made by the traditional deterministic optimization models infeasible.A data-driven Wasserstein distributionally robust chance-constrained(WDRCC)optimization approach is proposed in this paper to deal with demand uncertainty in crude oil ***,a new deterministic crude oil scheduling optimization model is developed as the basis of this *** Wasserstein distance is then used to build ambiguity sets from historical data to describe the possible realizations of probability distributions of uncertain demands.A cross-validation method is advanced to choose suitable radii for these ambiguity *** deterministic model is reformulated as a WDRCC optimization model for crude oil scheduling to guarantee the demand constraints hold with a desired high probability even in the worst situation in ambiguity *** proposed WDRCC model is transferred into an equivalent conditional value-at-risk representation and further derived as a mixed-integer nonlinear programming *** case studies from a real-world refinery are conducted to show the effectiveness of the proposed ***-of-sample tests demonstrate that the solution of the WDRCC model is more robust than those of the deterministic model and the chance-constrained model.

Carbon mitigation of buildings is critical to promote a net-zero *** international society has vigorously promoted‘‘Net Zero Carbon Buildings”across the globe,and accounting for building carbon emissions is critical to support this *** carbon,which represents carbon emissions from the entire lifecycle of the buildings,is fundamental for realizing the idea of zero ***,only limited studies have been conducted so far that take into account the city *** paper aimed to act as a first try to account for the embodied carbon emissions in buildings in 2020 for the Guangdong-Hong Kong Macao Greater Bay Area in China(GBA).We integrated remote sensing techniques such as night-time light data(NLT)and building material flows analysis to calculate and spatialize the newly generated building material stocks(MS).Based on the MS data,we further applied life cycle assessment(LCA)to assess the embodied carbon in the *** results highlighted that over 163 million tons of embodied carbon in buildings of GBA are expected to be generated,from 497 million tons of newly generated building MS in *** embodied carbon in each life cycle stage is valuable for further lifecyclebased policy designs for:(i)supporting the updating of the green building certification system with consideration of the embodied carbon;(ii)promoting the green building material application and certification;and(iii)reducing the embodied carbon intensity from compact urban planning policy,such as the urban agglomeration policies in *** goal of this paper was to shed a light on reducing carbon emissions from the perspective of the entire lifecycle and promote the development of net zero carbon buildings in China and Asia-Pacific.

Lead-free BiFeO_(3)-BaTiO_(3) based high-Tc ferroelectric ceramics:Antiferroelectric chemical modification leading to high energy storage performance

Dielectric capacitors have been widely used in pulsed power devices owing to their ultrahigh power density,fast charge/discharge speed,and excellent ***,developing lead-free dielectric materials with a combination of high recoverable energy storage density and efficiency remains a ***,a high energy storage density of 7.04 J/cm^(3) as well as a high efficiency of 80.5%is realized in the antiferroelectric Ag(Nb_(0.85)Ta_(0.15))O_(3)-modified BiFeO3-BaTiO3 ferroelectric *** achievement is mainly attributed to the combined effect of a high saturation polarization(Pmax),increased breakdown field(Eb),and reduction of the remnant polarization(Pr).The modification of pseudotetragonal BiFeO3 by Ag(Nb_(0.85)Ta_(0.15))O_(3) leads to a high Pmax,and the enhanced relaxor behavior gives rise to a small *** promoted microstructure(such as a dense structure,fine grains,and compact grain boundaries)after modification results in a high breakdown ***,both the recoverable energy density and efficiency exhibit high stability over a broad range of operating frequencies(1–50 Hz)and working temperatures(25–120℃).These results suggest that the(0.67–x)BiFeO_(3)-0.33BaTiO_(3)-xAg(Nb_(0.85)Ta_(0.15))O_(3) ceramics can be highly competitive as a lead-free relaxor for energy storage applications.

Microstructure evolution and phase transformation of the mushy zone in a quenchedβ-solidifying TiAl alloy

Calcium-chelating peptides from rabbit bone collagen:characterization,identification and mechanism elucidation

This study aimed to characterize and identify calcium-chelating peptides from rabbit bone collagen and explore the underlying chelating *** peptides and calcium were extracted from rabbit bone by instant ejection steam explosion(ICSE)combined with enzymatic hydrolysis,followed by chelation reaction to prepare rabbit bone peptide-calcium chelate(RBCP-Ca).The chelating sites were further analyzed by liquid chromatography-tandem mass(LC-MS/MS)spectrometry while the chelating mechanism and binding modes were *** structural characterization revealed that RBCP successfully chelated with calcium ***,LC-MS/MS analysis indicated that the binding sites included both acidic amino acids(Asp and Glu)and basic amino acids(Lys and Arg),Interestingly,three binding modes,namely Inter-Linking,Loop-Linking and Mono-Linking were for the first time found,while Inter-Linking mode accounted for the highest proportion(75.1%),suggesting that chelation of calcium ions frequently occurred between two ***,this study provides a theoretical basis for the elucidation of chelation mechanism of calcium-chelating peptides.

Theoretical and experimental study on multiscale coupled Mohr-Coulomb shear strength criterion of fibre-reinforced sand

Fibre-reinforced sand(FRS)is a multiphase and multiscale geo-material,which is widely used in geotechnical engineering as supporting structure of excavation of underground space and reinforcement of foundation of underground structures,and its strength is determined by the properties of the heterogeneous substances of the FRS and their coupling mechanical *** order to investigate the influence of fibre characteristics and mechanical properties on the shear strength of the FRS,according to the microscopic interface slip effect generated by the interaction between sand particles and the interaction between these particles and fibre,the material phase of the FRS is divided to conceptualize a micro cell element of the FRS that is capable of reflecting the internal material characteristic information of the ***,based on the coordinated deformation condition between fibres and sand particles at the microscale and the couple stress theory that is capable of effectively describing the discontinuous mechanical responses at the sand-fibre interface,a mesomechanism-based multiscale Mohr–Coulomb shear strength criterion of the FRS is derived,and the yield locus of the FRS is also drawn on the p ***,a series of FRS samples with different fibre content and fibre length were prepared by adopting the freezing method,and consolidated and drained triaxial compression tests were conducted on these samples to validate the proposed multiscale coupled Mohr–Coulomb shear strength *** show that the multiscale coupled Mohr–Coulomb shear strength criterion is capable of effectively reproducing and predicting the yield strength of the *** yield locus of the FRS extends outwards as fibre content and fibre length *** yield stress of the FRS predicted by the proposed multiscale coupled Mohr–Coulomb shear strength criterion is in good agreement with that of the test result.
