The interactions of cnoidal waves with a submerged quartercircular breakwater are investigated by a ReynoldsAveraged Navier–Stokes(RANS) flow solver with a Volume of Fluid(VOF) surface capturing scheme(RANSVOF) model. The vertical variation of the instantaneous velocity indicates that flow separation occurs at the boundary layer near the breakwater. The temporal evolution of the velocity and vorticity fields demonstrates vortex generation and shedding around the submerged quartercircular breakwater due to the flow separation. An empirical relationship between the vortex intensity and a few hydrodynamic parameters is proposed based on parametric analysis. In addition, the instantaneous and time-averaged vorticity fields reveal a pair of vortices of opposite signs at the breakwater which are expected to have significant effect on sediment entrainment, suspension, and transportation,therefore, scour on the leeside of the breakwater.

Tissue engineered skeletal muscle is expected to treat muscle defects caused by trauma and ***,designing and manufacturing thick and complex tissue engineered skeletal muscle requires vascularization to ensure its internal cell viability and nutrient supply in *** this article,we developed a set of Direct-Writing(DW)bio-printing procedure to manufacture a prevascularized composite construct with Human Umbilical Vein Endothelial Cell(HUVEC)and C2C12 cells for muscle tissue engineering *** put the cells into the construct during the DW process to obtain the prevascularization and intend to promote its vascularization in vivo *** constructs with cells or without cells were implanted respectively into nude mice back for 3 weeks,after which the mice healthily live for all the time and all the implants are tightly bonded to the *** immunohistochemical analysis,CD31-positive blood vessels existed in the implanted samples with cells are more substantial than those without cells,but the implanted samples with HUVEC and C2C12 cells have much more number of small blood vessels distributing ***,the implants with cells,especially that with HUVEC and C2C12 cells,are able to get better fusion with the host skin and subcutaneous *** analysis demonstrates that our DW-based constructs have the potential to be getting to vascularize the tissue engineered muscle.

Stereoselective metabolic disruption of cypermethrin by remolding gut homeostasis in rat

Cypermethrin(CYP), a prototypical synthetic pyrethroid, reportedly causes metabolic disruption, while its stereoselective impact remains elusive. This study initially revealed that only α-CYP caused significant weight loss at 8.5 mg/(kg·day) in rats. All three CYP isomers caused the accumulation of hepatic glycogen, and hyperlipemia phenotype as the increment of total triglyceride. Rats treated with α-CYP had markedly high blood glucose levels and homeostasis model assessment of insulin resistance index. The systematic inflammation of θ-CYP group rats was evidenced by high lipopolysaccharide-binding protein levels and abnormalities of leukocytes indices. By examining the gut microbiome, we found thatα-CYP-treated rats had low contents of Firmicutes and high levels of Verrucomicrobia while Elusimicrobia was enriched in the β-CYP group. The increasing alpha diversity in the θ-CYP group may be due to the dominance of pathogenic bacteria and the increase of probiotics to counteract adverse effects. Exclusively, the α-CYP group enriched total short-chain fatty acids(SCFAs), whereas most SCFAs depleted in the θ-CYP group. The correlation analysis further found Firmicutes, an energy storage modulator, was positive to body weight(BW),while SCFAs exerted the opposite, confirming the low BW in α-CYP. Blood glucose that correlated well with SCFAs and Verrucomicrobia can be accounted for the discrepancy betweenα-CYP and θ-CYP. Overall, the three isomers exerted stereoselective glycolipid disruption in rats, and gut homeostasis acted as vital indicators.

CRYPTOCHROMES （CRYs） are photolyase-like ultraviolet-A/blue light photoreceptors that mediate various light responses in plants. The signaling mechanism of Arabidopsis CRYs （CRY1 and CRY2） involves direct CRY-COP1 interaction. Here, we report that CRY1G380R, which carries a Gly-to-Arg substitution of the highly conserved G380 in the photo-lyase-related （PHR） domain of Arabidopsis CRY1, shows constitutive CRY1 photoreceptor activity in Arabidopsis. Transgenic plants overexpressing CRY1G380R display a constitutively photomorphogenic （COP） phenotype in darkness, as well as a dramatic early flowering phenotype under short-day light conditions （SD）. We further demonstrate that CRY1G380R expression driven by the native CRY1 promoter also results in a COP phenotype in darkness. Moreover, over- expression of either the Arabidopsis homolog CRY2G377R or the rice ortholog OsCRYlbG388R of CRY1G380R in Arabidopsis results in a COP phenotype in darkness. Cellular localization studies indicate that CRY1G380R co-localizes with COP1 in the same nuclear bodies （NBs） in vivo and inhibits the nuclear accumulation of COP1 in darkness. These results suggest that the conserved G380 may play a critical role in regulating the photoreceptor activity of plant CRYs and that CRY1G380R might constitutively phenocopy the photo-activated CRY1 in darkness and thus constitutively mediate CRY1 signaling.

Hydrogen evolution reaction(HER) is crucial for achieving sustainable development and carbon neutrality, and thus demands efficient catalysts, which necessitates fundamental theory to relieve trial-and-error experiment. To fast screen HER candidates, most studies focus on d-band center(ε)associated with the Gibbs energy of H* adsorption(ΔG). Unfortunately, εrule is not applicable to Pt single atoms on transition metal disulfides(Pt_(1)/TMDs) because of the additional contributions from p states of S atom. Here, we propose a new HER descriptor — d-band frontier(d) by defining the weight of d-band in the energy range of [-1.0 eV, 1.0 eV] of Pt single atoms. This dis exactly correlated with the ΔGof Pt_(1)/TMDs, and thus perfectly describes the structure–activity relationship, as validated by systematical experimental evidences. Moreover, this ddescriptor can be extended to Pt single atoms anchored on other supports(e.g., CN, C, MoO, and CoO), indicating its promising generality.
