目的用已构建的miR-144-GFP-H1人胚胎干细胞(human embryonic stem cell,hESC)示踪细胞株,以GFP荧光信号报告miR-144表达描述红系发生的进展程度,探究在红系发生过程中lncRNA的潜在功能并初步验证其功能影响。方法利用已构建的miR-144/451-GFP-H1细胞株(下文简称144-H1)进行体外红细胞诱导培养。以CD71,GPA表面分子描述进入红系发生阶段的细胞亚群。以miR-144的GFP报告基因为关键区分,对GFP阳性(高红系发生倾向)和阴性细胞群(低红系发生倾向)分别进行转录组测序,获取差异性表达的lncRNA。选取具备验证价值的lncRNA条目进行初步功能验证。利用CRISPR/Cas9基因编辑技术设计对应lncRNA的功能干扰方案并获取对应lncRNA功能敲除的144-H1细胞株。使用该敲除细胞株与非敲除的144-H1细胞株进行红细胞的平行诱导培养,寻找差异节点,初步验证其对在体外发育模型下对红系发生的影响效应。结果1)构建的144-H1细胞株能够在进入体外红细胞诱导阶段后表达GFP荧光蛋白,提示miR-144的启动。2)在CD71,GPA双阳群中,GFP阳性亚群和GFP阴性亚群存在显著的lncRNA表达差异。3)对多个lncRNA条目设计了能够有效干扰其功能的序列区段删除基因编辑方案。验证编辑成功。在敲除细胞株中,lncRNA的一部分序列被直接删除。4)在红细胞诱导培养的平行验证试验中,LINC01569的功能敲除株对比非敲除株表现出明显的流式亚群差异和细胞增殖能力差异:①敲除株的GFP荧光持续高表达;②敲除株CD71-GPA双阳群在红细胞成熟过中比例持续降低;③敲除株未观察到显著的血红蛋白表达,无明显红色;④敲除株细胞增殖能力远低于非敲除株(P<0.05)。结论成功利用144-H1细胞株对lncRNA在红系发育中的潜在功能进行了探究,可设计更精密的体外发育实验来提高lncRNA功能的抓取精度。在差异性表达的lncRNA条目中,LINC01569经初步验证,对红系发生过程存在调控作用。LINC01569的功能缺失严重影响红细胞的正常分化与增殖。LINC01569在红系发生过程中的具体调控机制有待进一步探索与研究。

Offshore waters provide resources for human beings,while on the other hand,threaten them because of marine *** stations are part of offshore observation networks,and the quality of their data is of great significance for exploiting and protecting the *** used hourly mean wave height,temperature,and pressure real-time observation data taken in the Xiaomaidao station(in Qingdao,China)from June 1,2017,to May 31,2018,to explore the data quality using eight quality control methods,and to discriminate the most effective method for Xiaomaidao *** using the eight quality control methods,the percentages of the mean wave height,temperature,and pressure data that passed the tests were 89.6%,88.3%,and 98.6%,*** the marine disaster(wave alarm report)data,the values failed in the test mainly due to the influence of aging observation equipment and missing data *** mean wave height is often affected by dynamic marine disasters,so the continuity test method is not *** correlation test with other related parameters would be more useful for the mean wave height.

Based on Burgess and Ivani?’s(2010) framework of the discoursal construction of writer identity, this paper explores the interaction between citation practices and identity construction in Chinese EFL learners’ MA thesis writing. A mixed approach of textual analysis and interviews was adopted to examine the citation features of MA theses and the identities Chinese EFL learners constructed through citation selection. It was found that students’ selection of citations reflects their linguistic identity, academic and disciplinary identity, as well as novice identity. Their citation practices project different discursive selves to readers. By adjusting the number and relevance of references, they intend to construct a knowledgeable and credible self. The use of ineffective citations and references not only projects an unfavorable impression on readers but reflects a lack of authorial identity. This study has significant implications for the teaching of academic English and the supervision of thesis writing.

The Bohai Sea is one of the southernmost areas for sea ice formation in the northern *** ice disasters in this body of water severely affect marine activities and the safety of coastal *** this study,we analyze the variation characteristics of the sea ice in the Bohai Sea and establish an annual regression model based on predictable mode analysis *** results show the following:1)From 1970 to 2018,the average ice grade is(2.6±0.8),with a maximum of 4.5 and a minimum of *** Bay(LDB)has the heaviest ice conditions in the Bohai Sea,followed by Bohai Bay(BHB)and Laizhou Bay(LZB).Interannual variation is obvious in all three bays,but the linear decreasing trend is significant only in BHB.2)Three modes are obtained from empirical orthogonal function analysis,namely,single polarity mode with the same sign of anomaly in all of the three bays and strong interannual variability(82.0%),the north–south dipole mode with BHB and LZB showing an opposite sign of anomalies to that in LDB and strong decadal variations(14.5%),and a linear trend mode(3.5%).Critical factors are analyzed and regression equations are established for all the principal components,and then an annual hindcast model is established by synthesizing the results of the three *** model provides an annual spatial prediction of the sea ice in the Bohai Sea for the first time,and meets the demand of operational sea ice forecasting.

2023年9月22日,国内首次采用国产自膨胀式经导管主动脉瓣膜可控弯输送系统(VitaFlow^(■)Ⅲ可控弯输送系统)在1例合并严重心力衰竭、外科手术高危的症状性重度主动脉瓣狭窄患者中行经导管主动脉瓣置换术。患者在全身麻醉下成功完成经导管主动脉瓣置换术,术后瓣膜位置、功能良好。出院前和随访1个月时,患者的症状和心力衰竭程度明显改善。该病例的随访结果初步表明,采用VitaFlow^(■)Ⅲ可控弯输送系统行经导管主动脉瓣置换术安全、可行,其疗效有望在后续的前瞻性、多中心临床试验中进行评估。

Li metal batteries using high-voltage layered oxides cathodes are of particular interest due to their high energy ***,they suffer from short lifespan and extreme safety concerns,which are attributed to the degradation of layered oxides and the decomposition of electrolyte at high voltage,as well as the high reactivity of metallic *** key is the development of stable electrolytes against both highvoltage cathodes and Li with the formation of robust interphase films on the ***,we report a highly fluorinated ether,1,1,1-trifluoro-2-[(2,2,2-trifluoroethoxy)methoxy]ethane(TTME),as a cosolvent,which not only functions as a diluent forming a localized high concentration electrolyte(LHCE),but also participates in the construction of the inner solvation *** TTME-based electrolyte is stable itself at high voltage and induces the formation of a unique double-layer solid electrolyte interphase(SEI)film,which is embodied as one layer rich in crystalline structural components for enhanced mechanical strength and another amorphous layer with a higher concentration of organic components for enhanced *** Li||Cu cells display a noticeably high Coulombic efficiency of 99.28%after 300 cycles and Li symmetric cells maintain stable cycling more than 3200 h at 0.5 mA/cm^(2) and 1.0m Ah/cm^(2).In addition,lithium metal cells using LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2) and Li CoO_(2) cathodes(both loadings~3.0 m Ah/cm^(2))realize capacity retentions of>85%over 240 cycles with a charge cut-off voltage of 4.4 V and 90%for 170 cycles with a charge cut-off voltage of 4.5 V,*** study offers a bifunctional ether-based electrolyte solvent beneficial for high-voltage Li metal batteries.

Sea level rise is a slow-onset *** collected information about the natural and ecological environments,tides and sea levels,and socio-economic aspects to investigate the distribution and zoning of the risks from sea level rise across Shandong *** trends in sea level in different counties of Shandong Province were predicted using moving averages and a random dynamic analysis forecasting model,and the model outputs and socio-economic indicators were combined to assess the *** results show that the risks of sea level rise along the western coast of Bohai Bay and Laizhou Bay in Shandong Province were sufficiently large to warrant attention.
