Silencing transformer and transformer-2 in Zeugodacus cucurbitae causes defective sex determination with inviability of most pseudomales

transformer is a switch gene for sex determination in many insects, which cooperates with transformer-2 that is expressed in both sexes to regulate female differentiation, particularly in dipterans. Zeugodacus cucurbitae(Coquillett) is a very destructive pest worldwide, however, its sex determination pathway remains largely uncharacterized. Here, we show that the female sex ratio is sharply reduced with knockdown of either transformer or transformer-2 by RNA interference in early embryos of Z. cucurbitae. Most of the males grown from the embryos with transient transformer and transformer-2 suppression mated with wild-type females and produced mixed sex progeny, with one exception that produced only female progeny, and all of the few remaining males failed to mate with wild-type females and produced no progeny. The exceptional male and those males with mating failure were XX pseudomales as determined by the detection of Y chromosome-linked Maleness-on-the-Y, indicating that most XX pseudomales are not viable. The phenotypes of transformer and transformer-2 suggest that they play a key role in regulating sex determination and are required for female sexual development of Z. cucurbitae. Our results will be beneficial to the understanding of sex determination in Z. cucurbitae and can facilitate the development of genetic sexing strains for its biological control.

[目的]为了更方便地计算附加质量和提高无环流理论的严谨性,基于涡动力学理论重新推导了一种基于物面滑移速度分布计算附加质量的方法.[方法]首先基于涡动力学理论给出以流场涡量和速度表征的流体动力和力矩表达式,然后指出其收敛于无环流的条件:1)用滑移速度代替物面速度;2)力矩参考点要与刚体固连;3)令流场中的涡量强度为零.利用刚体运动可用平动参考点速度和旋转角速度来表征的特性,进一步建立平动和旋转速度与流体动力和力矩的关系,并导出附加质量新算法.基于这种新算法,分析了物体尺寸、平动参考点位置以及参考坐标系对附加质量的影响及转换方法.[结果]针对经典外形,本文方法与数值算法得到的附加质量完全一致.[结论]本文提出的新算法在附加质量计算和转换方面具有正确性和简便性.在数值计算过程中,对于采用面元法计算滑移边界速度的讨论,指出了目前面涡法在实际应用中失败的根本原因,并提出解决方案,为面元法的实际应用提供了新的思路.

Influenza viruses not only cause respiratory illness,but also have been reported to elicit neurological manifestations following acute viral *** central nervous system(CNS)has a specific defense mechanism against pathogens structured by cerebral microvasculature lined with brain endothelial cells to form the blood–brain barrier(BBB).To investigate the response of human brain microvascular endothelial cells(hBMECs)to the Influenza A virus(IAV),we inoculated the cells with the A/WSN/33(H1N1)*** then conducted an RNAseq experiment to determine the changes in gene expression levels and the activated disease pathways following *** analysis revealed an effective activation of the innate immune defense by inducing the pattern recognition receptors(PRRs).Along with the production of proinflammatory cytokines,we detected an upregulation of interferons and interferon-stimulated genes,such as IFN-β/λ,ISG15,CXCL11,CXCL3 and IL-6,***,infected hBMECs exhibited a disruption in the cytoskeletal structure both on the transcriptomic and cytological *** RNAseq analysis showed different pathways and candidate genes associated with the neuroactive ligand-receptor interaction,neuroinflammation,and neurodegenerative diseases,together with a predicted activation of the ***,some genes linked with the mitochondrial structure and function displayed a significantly altered *** masse,this data supports that hBMECs could be infected by the IAV,which induces the innate and inflammatory immune *** results suggest that the influenza virus infection could potentially induce a subsequent aggravation of neurological disorders.

An analysis on the airship dynamic model linearization

The existing results on airship dynamic model linearization are usually achieved under the assumption that the airship trim state has a linear velocity with nonzero axial and normal components. This means that the direction of airship velocity is not coincident with the axis in the trim state and the deflection angles of the air rudders are different from their attack angles. However, these differences are not explicitly distinguished when calculating the air rudders＇ aerodynamic force. In this paper, to build a more accurate airship dynamic model, aerodynamic forces and moments produced by the air rudders are deduced for the case that the direction of airship velocity is not coincident with the trim axis. A special trim state that all linear and angular velocities are zeros except axial and normal linear velocity, adopted by most research on airship dynamic model linearization, is selected to linearize the six degree airship dynamic model for simplicity and for comparison. Both the control forces of the propelling system and the air rudders are linearized to achieve more control inputs for the linear model. The extra control inputs provided by the propelling system can be used to stabilize the airship when the air rudders are invalid due to low speed of the airship or the constraint of air rudder mechanical strength. The nuance between the new linearized model and those deduced in the literature is discussed.

The temperature distribution on the skin of the stratospheric airship plays an important role in its thermal behavior. In order to analyze the temperature distribution on the skin of the stratospheric airship, a 3-parameter thermal model is ***, a 3-parameter thermal model for the skin is established under the condition that the attitude of the stratospheric airship changes slowly. Then, based on the net heat method, the calculation method of the 3-parameter thermal model is introduced. At last, the temperature distribution on the skin of the airship is obtained by numerical simulation of the 3-parameter thermal model,and the effectiveness of the 3-parameter thermal model is verified by analyzing the simulation results, which lays the foundation for further simplifying the thermal model of the airship skin.

A model considering water vapor condensation process is built in this paper to simulate the process of airship ascending more accurately. In order to consider the difference of skin temperature around the airship surface, the skin of airship is divided into small grids. The radiation heat absorbed by the internal gas when the skin is semitransparent is calculated with an improved net heat method. With the assumption that the water vapor density of the airship in the air bag is saturated, the calculation method of the latent heat generated by the vapor condensation process and the attenuation coefficient of the thermal radiation under the corresponding vapor density are analyzed. By uniting the dynamical model,thermal model and ideal gas law, the simulation model for the ascending process is built. Numerical result is proposed for a concept airship. The results show that the condensation process does affect the ascending process and should be considered.

Power supply is a critical problem for stratospheric airship so it’s important to operate airship in the optimal motion mode. This paper studies the optimal trajectory for position correction when airship is drifted away to spot hovering boundary. The optimization model is firstly built under the assumptions of constant wind field and constant propeller efficiency. Then, by using the minimal principle of Pontryagin, the trajectory optimization problem is converted into a Hamiltonian boundary value problem （HBVP）. Finally, the optimal control policy and the optimal trajectory are given by solving the HBVP analytically.

为统一有粘和无粘不可压缩流场中的流体动力表达式,在允许滑移边界条件的情况下,重新推导了不可压缩流场中运动体的流体动力表达式.根据流场动量定理将流体动力表示为无穷远边界上流场应力的积分与流场动量变化率积分的代数和;再利用导数矩转换(DMT)公式分别将这两个积分结果进行分解,并根据速度在无穷远处的渐近特性将这些分解结果组合并简化成流场第一涡量矩的积分及流场在物面边界上切向速度矩的积分之和,得到了新的流体动力表达式.理论分析和算例证明了这种新的流体动力表达式在无粘流的情况下收敛于Lamb的流体动力表达式,而在有粘流的情况下与涡动力学理论一致.
