A novel long non-coding RNA, DIR, increases drought tolerance in cassava by modifying stress-related gene expression

Cassava is an important tropical cash crop. Severe drought stresses affect cassava productivity and quality, and cause great economic losses in agricultural production. Enhancing the drought tolerance of cassava can effectively improve its yield. Long non-coding RNAs(lncRNAs) are present in a wide variety of eukaryotes. Recently, increasing evidence has shown that lncRNAs play a critical role in the responses to abiotic stresses. However, the function of cassava lncRNAs in the drought response remains largely unknown. In this study, we identified a novel lncRNA, DROUGHT-INDUCED INTERGENIC lncRNA(DIR). Gene expression analysis showed that DIR was significantly induced by drought stress treatment, but did not respond to abscisic acid(ABA) or jasmonic acid(JA) treatments. In addition, overexpression of the DIR gene enhanced proline accumulation and drought tolerance in transgenic cassava. RNA-seq analysis revealed that DIR preferentially affected drought-related genes that were linked to transcription and metabolism. Moreover, RNA pull-down mass spectrometry analysis showed that DIR interacted with 325 proteins. A protein–protein interaction(PPI)analysis found a marked enrichment in proteins associated with the mRNA export and protein quality control pathways.Collectively, these results suggest that DIR and its interacting proteins that regulate mRNA or protein metabolism are involved in mediating the drought stress response. Thus, regulating DIR expression has potential for improving cassava yield under drought conditions.

应用被动控制技术建成的隔震建筑,其隔震性能缺乏检测手段,因此进行隔震结构动力特性的现场测试具有重要意义。现场测试1栋4层基础隔震幼儿园,展示了试验装置、方法及所得结果,将结果与同条件下非隔震结构模型进行对比分析,探究实际隔震结构动力响应规律及减震效果。用液压千斤顶将建筑物推开使隔震层产生98 mm(对应LNR500剪应变102%)水平初位移,安装混凝土顶杆支撑建筑物;利用炸药将顶杆爆破卸载使建筑物做自由振动;最后测试和分析其动力响应等参数。分析结果表明:水平初位移条件下,隔震结构的一阶自振周期比非隔震结构显著延长,阻尼比增大;隔震层滞回曲线饱满;各楼层动力响应控制效果明显,但观察到屋顶层相比底层加速度略有放大;卸载后隔震层瞬间复位,表明隔震层具有瞬间复位特性。

Ignition processes and characteristics of charring conductive polymers with a cavity geometry in precombustion chamber for applications in micro/nano satellite hybrid rocket motors

The arc ignition system based on charring polymers has advantages of simple structure,low ignition power consumption and multiple ignitions,which bringing it broadly application prospect in hybrid propulsion system of micro/nano satellite.However,charring polymers alone need a relatively high input voltage to achieve pyrolysis and ignition,which increases the burden and cost of the power system of micro/nano satellite in practical application.Adding conductive substance into charring polymers can effectively decrease the conducting voltage which can realize low voltage and low power consumption repeated ignition of arc ignition system.In this paper,a charring conductive polymer ignition grain with a cavity geometry in precombustion chamber,which is composed of PLA and multiwall carbon nanotubes(MWCNT)was proposed.The detailed ignition processes were analyzed and two different ignition mechanisms in the cavity of charring conductive polymers were revealed.The ignition characteristics of charring conductive polymers were also investigated at different input voltages,ignition grain structures,ignition locations and injection schemes in a visual ignition combustor.The results demonstrated that the ignition delay and external energy required for ignition were inversely correlated with the voltages applied to ignition grain.Moreover,the incremental depth of cavity shortened the ignition delay and external energy required for ignition while accelerated the propagation of flame.As the depth of cavity increased from 2 to 6 mm(at 50 V),the time of flame propagating out of ignition grain changed from 235.6 to 108 ms,and values of mean ignition delay time and mean external energy required for ignition decreased from 462.8 to 320 ms and 16.2 to 10.75 J,respectively.The rear side of the cavity was the ideal ignition position which had a shorter ignition delay and a faster flame propagation speed in comparison to other ignition positions.Compared to direct injection scheme,swirling injection provided a more favorable flow field

Immunomodulatory Function of Pien Tze Huang in T Cell-Mediated Anti-tumor Activity against B16-F10,MC38 and Hep1-6 Tumor Models

Objective To investigate the anti-tumor effects of Pien Tze Huang(PZH)in mouse models of B16–F10 melanoma,MC38 colorectal cancer,Hep1-6 hepatocellular carcinoma and chemically induced hepatocellular carcinoma model.Methods Various tumor models,including B16–F10,MC38 and Hep1-6 tumor hypodermic inoculation models,B16–F10 and Hep1-6 pulmonary metastasis models,Hep1-6 orthotopic implantation model,and chemically induced hepatocellular carcinoma model,were utilized to evaluate the anti-tumor function of PZH.Tumor growth was assessed by measuring tumor size and weight of solid tumors isolated from C57BL/6 mice.For cell proliferation and death of tumor cells in vitro,as well as T cell activation markers,cytokine production and immune checkpoints analysis,single-cell suspensions were prepared from mouse spleen,lymph nodes,and tumors after PZH treatment.Results PZH demonstrated significant therapeutic efficacy in inhibiting tumor growth(P0.05).Nevertheless,PZH exhibited a remarkable ability to enhance T cell activation and the production of interferon gamma,tumor necrosis factor alpha,and interleukin 2 in CD4^(+)T cells in vitro(P<0.01 or P<0.05).Importantly,PZH substantially inhibited T cell exhaustion and boosted cytokine production by tumor-infiltrating CD8^(+)T cells(P<0.01 or P<0.05).Conclusion This study has confirmed a novel immunomodulatory function of PZH in T cell-mediated anti-tumor immunity,indicating that PZH holds promise as a potential therapeutic agent for cancer treatment.

Ferrihydrite transformation impacted by coprecipitation of lignin:Inhibition or facilitation?

Lignin is a common soil organic matter that is present in soils,but its effect on the transformation of ferrihydrite(Fh)remains unclear.Organic matter is generally assumed to inhibit Fh transformation.However,lignin can reduce Fh to Fe(Ⅱ),in which Fe(Ⅱ)-catalyzed Fh transformation occurs.Herein,the effects of lignin on Fh transformation were investigated at 75℃ as a function of the lignin/Fh mass ratio(0-0.2),pH(4-8)and aging time(0-96 hr).The results of Fh-lignin samples(mass ratios=0.1)aged at different pH values showed that for Fh-lignin the time of Fh transformation into secondary crystalline minerals was significantly shortened at pH 6 when compared with pure Fh,and the Fe(Ⅱ)-accelerated transformation of Fh was strongly dependent on pH.Under pH 6,at low lignin/Fh mass ratios(0.05-0.1),the time of secondary mineral formation decreased with increasing lignin content.For high lignosulfonate-content material(lignin:Fh=0.2),Fh did not transform into secondary minerals,indicating that lignin content plays a major role in Fh transformation.In addition,lignin affected the pathway of Fh transformation by inhibiting goethite formation and facilitating hematite formation.The effect of coprecipitation of lignin on Fh transformation should be useful in understanding the complex iron and carbon cycles in a soil environment.

Mussel-inspired PTW@PDA composites for developing high-energy gun propellants with reduced erosion and enhanced mechanical strength

The severe erosion and inadequate mechanical strength are prominent challenges for high-energy gun propellants.To address it,novel PTW@PDA composites was prepared by polydopamine(PDA)-modifying onto potassium titanate whisker(PTW,K_(2)Ti_(6)O_(13)),and after was incorporated into gun propellant as erosion-reducing and mechanical-reinforcing fillers.The interfacial characterizations results indicated that as-prepared PTW@PDA composites exhibits an enhanced surface compatible with propellant matrix,thereby facilitating their dispersion into propellants more effectively than raw PTW materials.Compared to original propellants,PTW@PDA-modified propellants exhibited significant less erosion,with a Ti-Kbased protective coating being detected on the eroded steel.And 0.5 wt%and 1.0 wt%addition of PTW@PDA significantly improved impact,compressive and tensile strength of propellants.Despite the inevitably reduction in relative force,PTW@PDA slightly increase propellant burning rate while exerting little adverse impact on propellant dynamic activity.This strategy can provide a promising alternative to develop high-energy gun propellant with less erosion and more mechanical strength.

Two-dimensional bifunctional electrocatalyst(Mo-NiFe-LDH)with multilevel structure for highly efficient overall water splitting

To effectively address energy challenges,it is crucial to explore efficient and stable bifunctional nonprecious metal catalysts.In this study,a Mo-doped nickeliron layered double hydroxide with flower-cluster architecture was successfully prepared by a one-step hydrothermal method,which demonstrated a good water splitting performance.After an appropriate amount of Mo doping,some lattice distortions in the material provided reactive sites for the adsorption and conversion of intermediates,thus optimising the charge distribution of the material.Moreover,the multidimensional void structures formed after doping had a larger specific surface area and accelerated the penetration of the electrolyte,which significantly improved the activity of the catalyst in alkaline media.At 10 mA·cm-2,the hydrogen and oxygen evolution overpotentials of Mo-doped nickel-iron double hydroxides（Mo-NiFe LDH/NF-0.2） were 167 and 220 mV,respectively,with an excellent durability up to 24 h.When the Mo-NiFe LDH/NF-0,2 catalyst was used as the cathode and anode of an electrolytic cell,the catalyst achieved a current density of 10 mA·cm-2at an applied voltage of 1.643 V.This study provides a novel approach for designing excellent bifunctional electrocatalysts containing nonprecious metals.

Opportunity to harmonize aspects of oxygen reduction reaction(ORR) performance and structure, and morphology, as well as composition, is urgent for the commercialization of proton exchange membrane fuel cells. Herein, we demonstrate the design and synthesis of a functionalized-supported-Pt catalyst(Pt@HNC) featuring a hollow nitrogen-modified dodecahedral carbon substrate obtained by a stress-induced-shrink tailoring route. The asobtained Pt@HNC catalyst possesses enhanced ORR performance, in particular with half-wave potential, mass activity(MA) and specific activity, which greatly exceed the commercial Pt/C. The density functional theory(DFT)calculations further confirmed that the charge redistribution induced by the electronegativity differences improved the electron interaction between Pt and HNC support. The optimized electronic structure of Pt weakens the reaction energy barrier on the Pt@HNC surface and adsorption of*OH species, thus cooperatively improving the intrinsic activity toward ORR. Additionally, our work indeed provides a guide for the future design of functional nanomaterials in the field of catalysts and clean energy.

The co-transport of Cd(Ⅱ) and nZnO in saturated soil packed column:effects of ionic strength and pH

The rapid development and widespread use of ZnO nanoparticles(nZnO) in various industries have raised concerns about their potential environmental impact.Therefore,understanding the fate and role of nZnO in the natural environment is crucial for mitigating their hazardous effects on the environment and human safety.The purpose of the present study was to provide scientific support for understanding and eliminating the joint risk of nanoparticle and heavy metal pollution in the soil environment by revealing the co-transport characteristics of Cd(Ⅱ) and ZnO nanoparticles(nZnO) in soil under different ionic strength(IS) and pH.The impacts of different IS and pH on the co-transport of Cd(Ⅱ) and nZnO in a20 cm long with an inner diameter of 2.5 cm acrylic column packed with 10 cm high soil samples were investigated in the present study.In the above system,a500 μg L^(-1) Cd(Ⅱ) loaded nZnO suspension pulse with varying IS or pH was introduced into the soil column for leaching over 5 PVs,followed up by 5 PVs background solutions without nZnO.The IS was 1,10,or 50 mM NaCl,with pH6,or the pH was 6,7 or 8 with 1 mM NaCl.Meanwhile,Sedimentation experiments for nZnO,adsorption of Cd(Ⅱ) on soil,and nZnO,DLVO theory calculation for the same background condition were conducted.The presence of nZnO significantly increased the mobility of Cd(Ⅱ) as a result of its strong adsorption capacity for nZnO-associated Cd(Ⅱ).However,with the increase of IS,the co-transport of nZnO and Cd(Ⅱ) was decreased and the retention of nZnO in the soil column due to more nZnO attended to aggregate and sediment during the transport and the decrease in the adsorption capacity of nZnO for Cd(Ⅱ) by competition of Na^(+).When pH was 6,7,and 8,the co-transport of nZnO and Cd(Ⅱ) increased with higher pH due to the lower electrostatic attraction between nZnO and soil under higher pH.Meanwhile,the DLVO theory was fitted to describe the above co-transport process of nZnO and Cd(Ⅱ).More attention should be paid to the presence of nZnO on the migration of Cd(Ⅱ) i

Idiopathic mesenteric phlebosclerosis missed by a radiologist at initial diagnosis: A case report

BACKGROUND Idiopathic mesenteric phlebosclerosis(IMP)is a rare type of ischemic colitis characterized by thickening of the wall of the right hemicolon and calcification,sclerosis,and fibrosis of mesenteric veins.The diagnosis of IMP is based on typical clinical features and imaging findings.We report a case of IMP that was initially missed by the radiologist.CASE SUMMARY A 77-year-old woman was admitted to the hospital due to chronic diarrhea for over 2 months.She had been consuming Chinese patent medicines(CPM)containing fructus gardeniae for more than 15 years.Colonoscopy revealed an edematous mucosa,bluish-purple discoloration,erosions,and ulcerations throughout the colorectal area.Abdominal computed tomography(CT)showed diffuse mural thickening of the entire colorectum,with tortuous thread-like calcifications in the right hemicolon,left hemicolon,and rectum.Most of the calcifications were located in the mesenteric vein.The diagnosis of IMP was established based on medical history,colonoscopy,CT findings,and histopathological examination.The patient was treated conservatively with papaverine and rifaximin,and CPM was stopped.Her diarrhea symptoms improved,indicating the effectiveness of the treatment.Over the next several years,she took opium alkaloids for an extended period and did not require hospitalization for the aforementioned gastrointestinal disorder.CONCLUSION IMP is a rare gastrointestinal disease affecting Asian populations,possibly related to long-term herbal medicine intake.Accurate imaging analysis is crucial for diagnosis,but insufficient understanding of the disease can lead to misdiagnosis or missed diagnosis.Treatment strategies should be personalized.
