Design,synthesis of novel antifungal triazole derivatives with high activities against Aspergillus fumigatus

Based on the active site of Aspergillus fumigatus lanosterol 14α-demethylase （AF-CYP51）, novel triazole compounds were designed. Their chemical synthesis and the antifungal activities were reported. The results showed that all the target compounds exhibited excellent activities with broad spectrum; in which compounds 4, 12 and 15 showed comparable activities against A. fumigatus to the control drug itraconazole.

AIM To compare the efficacy,improved quality of life,and prognosis in patients undergoing either subtotal colonic bypass with antiperistaltic cecoproctostomy(SCBAC) or subtotal colonic bypass plus colostomy with antiperistaltic cecoproctostomy(SCBCAC) for the treatment of slow transit *** Between October 2010 and October 2014,aged patients with slow transit constipation who were hospitalized and underwent laparoscopic surgery in our institute weredivided into two groups: the bypass group,15 patients underwent SCBAC,and the bypass plus colostomy group,14 patients underwent SCBCAC. The following preoperative and postoperative clinical data were collected: gender,age,body mass index,operative time,first flatus time,length of hospital stay,bowel movements(BMs),Wexner fecal incontinence scale,Wexner constipation scale(WCS),gastrointestinal quality of life index(GIQli),numerical rating scale for pain intensity(NRS),abdominal bloating score(ABS),and ClavienDindo classification of surgical complications(CD) before surgery and at 3,6,12,and 24 mo after *** All patients successfully underwent laparoscopic surgery without open surgery conversion or surgeryrelated death. The operative time and blood loss were significantly less in the bypass group than in the bypass plus colostomy group(P = 0.007). No significant differences were observed in first flatus time,length of hospital stay,or complications with CD > 1 between the two groups. No patients had fecal incontinence after surgery. At 3,6,and 12 mo after surgery,the number of BMs was significantly less in the bypass plus colostomy group than in the bypass group. Hybrid rice has contributed greatly to the self-sufficiency of food supply In China. To meet the future demand for rice production, a national program on super rice breeding was established In China In 1996. The corresponding targets, breeding strategies and most significant advances are reviewed In this paper. New plant type models have been modified to adjust to various rice growing regions. In recognition of the Importance of applying parents with Intermediate subspecies differentiation In Increasing F1 yield, medium type parental lines were selected from populations derived from Inter-subspecies crosses with the assistance of DNA markers for subspecies differentiation. Results also indicate that a substantial increase of blomass Is the basis for further enhancement of the grain yield potential, and amelioration of leaf characteristics Is helpful In Increasing the photosynthetic rate. Thirty-four super hybrid rice varieties have been released commercially, growing In a total area of 13.5 million hm2 and producing 6.7 thousand million kg more rice In 1998-2005. Anaerobic digestion(AD)is widely employed for sludge stabilization and waste ***,the slow hydrolysis process hinders methane production and leads to prolonged sludge *** this study,an efficient and eco-friendly lysozyme pre-treatment method was utilized to address these *** optimizing lysozyme dosage,hydrolysis and cell lysis were ***,lysozyme combined with hydrothermal pretreatment enhanced overall *** indicate that:(1)When lysozyme dosage reached 90 mg/g TS after 240 min of pretreatment,SCOD,soluble polysaccharides,and protein content reached their maxima at 855.00,44.09,and 204.86 mg/L,*** represented an increase of 85.87%,365.58%,and 259.21%compared to the untreated *** fluorescence spectroscopy revealed the highest fluorescence intensity in the IV region(soluble microbial product),promoting microbial metabolic activity.(2)Lysozyme combined with hydrothermal pretreatment significantly increased SCOD,soluble proteins,and polysaccharide release from sludge,reducing SCOD release *** experiments identified Group 3 as the most effective for SCOD and soluble polysaccharide release,while Group 9 released the most soluble *** significance order of factors influencing SCOD,soluble proteins,and polysaccharide release is hydrothermal temperature>hydrothermal time>enzymatic digestion time.(3)The lysozyme-assisted hydrothermal pretreatment group exhibited the fastest release and the highest SCOD concentration of 8,135.00 mg/L during anaerobic *** SCOD consumption and cumulative gas production increased by 95.89%and 130.58%,respectively,compared to the control group,allowing gas production to conclude 3 days earlier.

Spinal cord injury typically causes corticospinal tract disruption. Although the disrupted corticospinal tract can self-regenerate to a certain degree, the underlying mechanism of this process is still unclear. N6-methyladenosine(m^(6)A) modifications are the most common form of epigenetic regulation at the RNA level and play an essential role in biological processes. However, whether m^(6)A modifications participate in corticospinal tract regeneration after spinal cord injury remains unknown. We found that expression of methyltransferase 14 protein(METTL14) in the locomotor cortex was high after spinal cord injury and accompanied by elevated m^(6)A levels. Knockdown of Mettl14 in the locomotor cortex was not favorable for corticospinal tract regeneration and neurological recovery after spinal cord injury. Through bioinformatics analysis and methylated RNA immunoprecipitation-quantitative polymerase chain reaction, we found that METTL14 regulated Trib2 expression in an m^(6)A-regulated manner, thereby activating the mitogen-activated protein kinase pathway and promoting corticospinal tract regeneration. Finally, we administered syringin, a stabilizer of METTL14, using molecular docking. Results confirmed that syringin can promote corticospinal tract regeneration and facilitate neurological recovery by stabilizing METTL14. Findings from this study reveal that m^(6)A modification is involved in the regulation of corticospinal tract regeneration after spinal cord injury.

Pelagic fish are the most abundant species in upwelling regions,contributing 25%of total global fisheries ***-driven changes in the marine environment play a crucial role in their population *** Chilean jack mackerel(Trachurus murphyi)as an example,this study conducted simulations to quantify the impacts of environmental variations on the stock assessment.A habitat-based surplus production model was developed by integrating suitable habitat area into the model parameters carrying capacity(K)and intrinsic growth rate(r),with a suitable habitat area serving as the proxy for the environmental conditions for Chilean jack mackerel in the Southeast Pacific *** dynamics of Chilean jack mackerel stock and fisheries data were simulated,and four assessment models with different configurations were built to fit simulated data,with or without considering environmental *** results indicated that Joint K-r model,which integrated both parameters with the suitable habitat area index,outperformed the others by coming closest to the‘true'population *** habitat variations in the estimation model tended to overestimate biomass and underestimate harvest rate and reference *** observation and process error,the results were estimated with bias,while FMSY is relatively *** research illustrates the importance to consider random errors and environmental influences on populations,and provides foundation guidelines for future stock assessment.
