Objective Ovarian cancer(OC)is one of the most common and most lethal gynecological *** has an age-dependent incidence and occurs more commonly in females older than 50 years *** OC patients are diagnosed at an advanced stage and have a poor *** mutations in the BRCA1 Dna repair associated gene(BRCA1)and the BRCA2 Dna repair associated gene(BRCA2)account for 20%–25%of epithelial ovarian cancer(EOC).BRCA1 germline mutations are more common in Chinese EOC *** This study reported a three-generation Han-Chinese family containing four EOC patients and a rectal adenocarcinoma ***-exome sequencing was performed on two EOC patients and an unaffected *** validation was also performed in all available members by Sanger *** A heterozygous splice site variant,c.4358-2A>G in the BRCA1 gene,was *** analysis showed that the variant may change the splicing *** The BRCA1 splice site variant,c.4358-2A>G was identified as the likely genetic cause for EOC,and may also be associated with the increased risk of rectal adenocarcinoma in the *** findings were beneficial for genetic counseling,helpful for cancer prevention in other family members,and may facilitate therapy decision-making in the future to reduce cancer lethality.

Coniopteris simplex is a common component in many Jurassic ***,due to morphological variations in sterile fronds and incomplete preservation of reproductive organs,its classification has been controversial for a long ***,we present new material collected from the Middle Jurassic Yan’an Formation in the Ordos Basin,Inner *** new material reveals important morphological characters,including hemi-dimorphic fronds,two forms of sterile pinnae,short-stalked sori and sporangia on fertile fronds,annulus of sporangium composed of about 30 cells,in situ rounded triangular spores with protruded suture,and the lips of trilete laesura encircled by one or two rows of small *** on these observations,we emended the diagnosis of this common *** spatio-temporal distribution of Coniopteris simplex shows that the iconic species was restricted to the paleolatitudes ranging from 19.5°N to 48.9°N in the Northern Hemisphere during the Early—Late Jurassic,mainly distributed in the tropical to paratropical zones,and the paleolongitude ranging from 9.6°E to 129.3°E,which may be limited by the opening of the Viking Corridor and the splitting of the Central Atlantic Ocean.

3D imaging technology is pivotal in monitoring the functional dynamics and morphological alterations in living cells and ***,conventional volumetric imaging associated with mechanical z-scanning encounters challenges in limited 3D imaging speed with inertial ***,we present a unique phase-modulated multifoci microscopy (PM^(3)) technique to achieve snapshot 3D imaging with the advantages of extended imaging depths and adjustable imaging intervals between each focus in a rapid *** accomplish the tasks,we utilize a spatial light modulator (SLM) to encode the phases of the scattered or fluorescence light emanating from a volumetric sample and then project the multiple-depth images of the sample onto a single charge-coupled device camera for rapid 3D *** demonstrate that the PM^(3)technique provides~55-fold improvement in imaging depth in polystyrene beads phantom compared to the depth of field of the objective lens ***^(3)also enables the real-time monitoring of Brownian motion of fluorescent beads in water at a 15 Hz volume *** precisely manipulating the phase of scattered light on the SLM,PM^(3)can pinpoint the specific depth information in living zebrafish and rapidly observe the 3D dynamic processes of blood flow in the zebrafish *** work shows that the PM^(3)technique developed is robust and versatile for fast 3D dynamic imaging in biological and biomedical systems.

We present a novel time-of-flight resolved Bessel light bullet-enabled stimulated Raman scattering(B2-SRS)microscopy for deeper tissue 3D chemical imaging with high resolution without a need for mechanical *** accomplish the tasks,we conceive a unique method to enable optical sectioning by generating the counterpropagating pump and Stokes Bessel light bullets in the sample,in which the group velocities of the Bessel light bullets are made ultraslow(e.g.,vg≈0.1c)and tunable by introducing programmable angular dispersions with a spatial light *** theoretically analyze the working principle of the collinear multicolor Bessel light bullet generations and velocity controls with the relative time-of-flight resolved detection for SRS 3D deep tissue *** have also built the B2-SRS imaging system and present the first demonstration of B2-SRS microscopy with Bessel light bullets for 3D chemical imaging in a variety of samples(e.g.,polymer bead phantoms,biological samples such as spring onion tissue and porcine brain)with high *** B2-SRS technique provides a>2-fold improvement in imaging depth in porcine brain tissue compared to conventional SRS *** method of optical sectioning in tissue using counter-propagating ultraslow Bessel light bullets developed in B2-SRS is generic and easy to perform and can be readily extended to other nonlinear optical imaging modalities to advance 3D microscopic imaging in biological and biomedical systems and beyond.

Interstitial cystitis(IC)is a severely debilitating and chronic disorder with unclear etiology and pathophysiology,which makes the diagnosis difficult and treatment *** investigate the role of immunity in IC bladders,we sequenced 135,091 CD45^(+) immune cells from 15 female patients with IC and 9 controls with stress urinary incontinence using single-cell Rna sequencing(scRna-seq).22 immune subpopulations were identified in the constructed *** them,M2-like macrophages,inflammatory CD14^(+) macrophages,and conventional dendritic cells had the most communications with other immune ***,a significant increase of central memory CD4^(+)T cells,regulatory T cells,GZMK^(+)CD8^(+)T cells,activated B cells,un-switched memory B cells,and neutrophils,and a significant decrease of CD8+effector T cells,Th17 cells,follicular helper T cells,switched memory B cells,transitional B cells,and macrophages were noted in IC *** enrichment analysis identified a virus-related response during the dynamic change of cell proportion,furthermore,the human polyomavirus-2 was detected with a positive rate of 95%in urine of patients with *** integrating the results of scRna-seq with spatial transcriptomics,we found nearly all immune subpopulations were enriched in the urothelial region or located close to fibroblasts in IC bladders,but they were discovered around urothelium and smooth muscle cells in control *** findings depict the immune landscape for IC and might provide valuable insights into the pathophysiology of IC.

Electroreduction of nitrate has been gaining wide attention in recent years owing to it's beneficial for converting nitrate into benign N_(2) from the perspective of electrocatalytic denitrification or into value-added ammonia from the perspective of electrocatalytic NH_(3) *** reason of the undesired formation of ammonia is dominant during electroreduction of nitrate-containing wastewater,chloride has been widely used to improve N_(2) ***,selective electroreduction of nitrate to N2 gas in chloride-containing system poses several *** this review,we focus on the key strategies for efficiently enhancing N_(2) selectivity of electroreduction of nitrate in chloride-free system,including optimal selection of elements,combining an active metal catalyst with another metal,manipulating the crystalline morphology and facet orientation,constructing core–shell structure catalysts,*** summarizing the strategies,four possible reaction pathways of electro-reduction of nitrate to N_(2) are ***,this review attempts to provide practical strategies for enhancing N2 selectivity without the aid of electrochlorination and highlight directions for future research for designing appropriate electrocatalyst for final electrocatalytic denitrifi-cation.

The role of the auxin-response genes MdGH3.1 and MdSAUR36 in bitter pit formation in apple

Apples often exhibit bitter pits in response to metabolic disorders during ripening and storage;however, the mechanisms underlying the bitter pit(BP) development remain unclear. Here, metabolome and transcriptome analyses were performed to investigate BP pulp of 'Fuji'. Two auxin-response genes, MdGH3.1 and MdSAUR36, were screened. Their expression as well as the auxin content in BP pulp were found to be higher than those in healthy pulp(P lization. On Day 30 before harvest, the two genes were transiently transferred to the fruit, and 20.69% and 23.21% of BP fruits were harvested. After 10 μmol·L-1auxin was infiltrated at low pressure into postharvest fruit, the increase in MdGH3.1 expression occurred earlier than that in MdSAUR36. MdGH3.1 increased the expression of MdSAUR36, but MdSAUR36 did not increase expression of MdGH3.1. Therefore, we suggest that MdGH3.1 acts upstream of MdSAUR36 during BP formation and that these genes induce BP formation by regulating auxin and phenylpropanoid biosynthesis.

From risk control to resilience: developments and trends of urban roads designed as surface flood passages to cope with extreme storms

Urban roads can be designated as surface flood passages to transport excess runoff during extreme storms, thereby preventing local flooding, which is known as the major drainage system. However, this practice poses significant risks, including human loss and property damage, due to the high flow rate and velocity carried by roads. Moreover, urban roads with low flood-resilience may significantly hamper the transportation function during severe storms, leading to dysfunction of the city. Therefore, there is an urgent need to transform risk-oriented flood passages into resilient urban road-based flood passages. This paper presents a systematic review of existing methodologies in designing a road network-based flood passage system, along with the discussion of new technologies to enhance system resilience. The study also addresses current knowledge gaps and future directions. The results indicate that flood management measures based on the urban road network should integrate accessibility assessment, lifeline and emergency planning to ensure human well-being outcomes. Furthermore, the special needs and features of vulnerable groups must be taken into serious consideration during the planning stage. In addition, a data-driven approach is recommended to facilitate real-time management and evaluate future works.
