Human umbilical cord mesenchymal stem cells(hUC-MSCs)are a promising candidate for spinal cord injury(SCI)repair owing to their advantages of low immunogenicity and easy accessibility over other MSC ***,modest clinical efficacy hampered the progression of these cells to clinical *** discrepancy may be due to many variables,such as cell source,timing of implantation,route of administration,and relevant efficacious cell dose,which are critical factors that affect the efficacy of treatment of patients with ***,we have evaluated the safety and efficacy of 4×10^(6) hUC-MSCs/kg in the treatment of subacute SCI by intrathecal implantation in rat *** search for a more accurate dose range for clinical translation,we compared the effects of three different doses of hUC-MSCs-low(0.25×10^(6) cells/kg),medium(1×10^(6) cells/kg)and high(4×10^(6) cells/kg)-on subacute SCI repair through an elaborate combination of behavioral analyses,anatomical analyses,magnetic resonance imaging-diffusion tensor imaging(MRI-DTI),biotinylated dextran amine(BDA)tracing,electrophysiology,and quantification of mRNA levels of ion channels and neurotransmitter *** study demonstrated that the medium dose,but not the low dose,is as efficient as the high dose in producing the desired therapeutic ***,partial restoration of theγ-aminobutyric acid type A(GABAA)receptor expression by the effective doses indicates that GABAA receptors are possible candidates for therapeutic targeting of dormant relay pathways in injured spinal ***,this study revealed that intrathecal implantation of 1×10^(6) hUC-MSCs/kg is an alternative approach for treating subacute SCI.

Human umbilical cord mesenchymal stem cells(hUC-MSCs)support revascularization,inhibition of inflammation,regulation of apoptosis,and promotion of the release of beneficial ***,they are regarded as a promising candidate for the treatment of intractable spinal cord injury(SCI).Clinical studies on patients with early chronic SCI(from 2 months to 1 year post-injury),which is clinically common,are rare;therefore,we will conduct a prospective,multicenter,randomized,placebo-controlled,single-blinded clinical trial at the Third Affiliated Hospital of Sun Yat-sen University,West China Hospital of Sichuan University,and Shanghai East Hospital,Tongji University School of Medicine,*** trial plans to recruit 66 early chronic SCI *** patients will undergo randomization at a 2:1 ratio to two arms:the observation group and the control *** in the observation group will receive four intrathecal transplantations of stem cells,with a dosage of 1×106/kg,at one calendar month *** in the control group will receive intrathecal administrations of 10 mL sterile normal saline in place of the stem cell *** safety will be assessed by the analysis of adverse events and laboratory *** American Spinal Injury Association(ASIA)total score will be the primary efficacy endpoint,and the secondary efficacy outcomes will be the following:ASIA impairment scale,International Association of Neural Restoration-Spinal Cord Injury Functional Rating Scale,muscle tension,electromyogram,cortical motor and cortical sensory evoked potentials,residual urine volume,magnetic resonance imaging–diffusion tensor imaging,T cell subtypes in serum,neurotrophic factors and inflammatory factors in both serum and cerebrospinal *** evaluations will be performed at 1,3,6,and 12 months following the final intrathecal *** the entire study procedure,all adverse events will be reported as soon as they are *** trial is d

Different Acupuncture Therapies for Postherpetic Neuralgia:An Overview of Systematic Reviews and Meta-analysis

Background: Postherpetic neuralgia(PHN) is the most common complication of herpes zoster infection and affects patients' quality of life. Acupuncture therapy is regarded as a competitive method of treatment for analgesia. Objective: To summarize evidence from systematic reviews(SRs) and evaluate the effectiveness and safety of different acupuncture therapies for treating PHN. Methods: Eight electronic databases were searched from their inception to August 5, 2022, including 4 international electronic databases(PubMed, EMBASE, the Cochrane library, and Web of Science) and 4 Chinese databases(Chinese Biomedical Database, China National Knowledge Infrastructure, VIP Database and Wanfang Database).Methodological quality was assessed by A Measurement Tool to Assess Systematic Reviews 2(AMSTAR 2).The Risk of Bias in Systematic Review(ROBIS) tool was used to assess the risk of bias in SRs. Evidence level was assessed by the Grading of Recommendations Assessment, Development, and Evaluation(GRADE)approach. Results: Totally, 7 SRs were included, including 128 studies and 9,792 patients. In AMSTAR 2, most of the SRs were of low or critically low levels since they had more than 1 critical deficiency. In ROBIS, 1 SR(14.29%)was rated as high risk, and the other 6(85.71%) were rated as low risk. In the GRADE system, 9 outcomes(28.13%)were valued as high level, 5(15.63%) as moderate level, 1(3.13%) as low, and 17(53.13%) as very low. In the effectiveness of acupuncture therapy, the group "moxibustion vs. original medical treatment" [mean difference(MD)=–1.44, 95% confidence interval(CI): –1.80 to –1.08, I2=99%, P<0.00001] was of the highest heterogeneity and the group "bloodletting vs. original medical treatment"(MD=–2.80, 95% CI: –3.14 to –2.46, I2=0, P<0.00001)was of the lowest heterogeneity. Effective background infusion rate of ropivacaine 0.2% for patient-controlled interscalene brachial plexus analgesia after rotator cuff repair surgery

Background Continuous interscalene nerve block （CISB） is considered to be the most effective method for postoperative analgesia after shoulder surgery with prolonged severe *** study was performed to evaluate the minimum effective background infusion rate and the effective background infusion rate of ropivacaine 0.2％ for CISB after arthroscopic rotator cuff repair surgery in 95％ of *** Patients scheduled for arthroscopic rotator cuff repair surgery under general anesthesia at Peking University Third Hospital were prospectively enrolled from December 2011 to May ***,an interscalene catheter （ISC） was placed under the guidance of ultrasound and nerve stimulation in each ***,30 patients with successful nerve block were included.A continuous infusion of ropivacaine 0.2％ with a 5 ml patient-controlled bolus available hourly was started at postoperative anesthesia care unit （PACU） after completion of *** initial background infusion rate was 6 ml/h,which was subsequently varied for each consecutive patient according to the analgesic effects of the previous *** minimum effective background rate was determined using the Dixon and Massey up-and-down *** effective background rate in 95％ of patients was calculated using isotonic *** The minimum effective background rate based on the Dixon and Massey up-and-down method was 2.8 ml/h （95％ CI,2.3-3.3 ml/h）.The effective background rate in 95％ of patients calculated with the isotonic regression analysis was 4.4 ml/h （95％ CI,3.8-6.5 ml/h）.Conclusion The effective background rate for patient-controlled interscalene brachial plexus analgesia after shoulder surgery in 50％ and 95％ of the patients was 2.8 and 4.4 ml/h,respectively.

Node ranking based on graph curvature and PageRank

Identifying key nodes in complex networks is crucial for understanding and controlling their dynamics. Traditional centrality measures often fall short in capturing the multifaceted roles of nodes within these networks. The Page Rank algorithm, widely recognized for ranking web pages, offers a more nuanced approach by considering the importance of connected nodes. However, existing methods generally overlook the geometric properties of networks, which can provide additional insights into their structure and functionality. In this paper, we propose a novel method named Curv-Page Rank(C-PR), which integrates network curvature and Page Rank to identify influential nodes in complex networks. By leveraging the geometric insights provided by curvature alongside structural properties, C-PR offers a more comprehensive measure of a node's influence. Our approach is particularly effective in networks with community structures, where it excels at pinpointing bridge nodes critical for maintaining connectivity and facilitating information flow. We validate the effectiveness of C-PR through extensive experiments. The results demonstrate that C-PR outperforms traditional centrality-based and Page Rank methods in identifying critical nodes. Our findings offer fresh insights into the structural importance of nodes across diverse network configurations, highlighting the potential of incorporating geometric properties into network analysis.

Effects of Aging and Advanced Glycation on Gene Expression in Cerebrum and Spleen of Mice

Objective To analyze the effects of aging or advanced glycation on gene expression in the cerebrum and spleen of female C57BL/6J mice. Methods The gene expression profile was determined by using cDNA expression arrays containing 588 cDNA. Results Aging and advanced glycation resulted in differential gene expression patterns of cerebrum and spleen compared with young mice. Among the 80 genes detected in cerebrum, 43 exhibited a change in mRNA ratios with aging or treatment. Thirty-four changes (79%) were common in aged and D-galactose treated mice, whereas the cerebrum from aged and AGE-lysine treated mice showed common changes in expression of 38 genes (88%). Of the 86 genes detected in spleen, 29 (34%) displayed an age-related decrease in expression, whereas 3 (3%) displayed an increase in expression levels with aging. Eighteen genes from the detectable genes exhibited expression changes in both cerebrum and spleen of mice. Conclusions The gene expression profiles of D-galactose and AGE-lysine treated mice resemble those of aged mice. Use of cDNA hybridization arrays may provide a promising tool to explore the mechanism of aging at a molecular level.

Study on Haploid Inducing and Its Meiotic Abnormality in Maize

The haploid-inducing line Stock 6 was used to produce haploid maize and expected to obtain maize haploid plants successfully. The detailed meiotic studies on selected haploid maize （n = x = 10） were conducted. Cytogenetic analysis revealed a high frequency of meiotic abnormality occurred in both meiosis Ⅰ and meiosis Ⅱ. During the prophase Ⅰ, univalents were common configurations, and there were bivalents or trivalents in some pollen mother cells, however, a few cells containing five bivalents were also observed. After prophase Ⅰ, chromosomes did not congregate in a single metaphase plate but they were scattered in the cytoplasm. At anaphase Ⅰ, the chromosome distribution was highly irregular with almost all possible combinations. In some cells, chromosomes were grouped into the three or four masses and several spindles appeared. At the tetrad stage of meiosis Ⅱ, eytokinesis splitting abnormality occurred, and a variety of diad, triad, tetrad, pentad, hexad, as well as decury microspores were easily observed. As a consequence of abnormalities of the two meiotic stages, various microspores and the pollen were almost completely sterile. grains with different size were formed, and its pollen grains
