Sequential treatment with bortezomib plus dexamethasone followed by autologous hematopoietic stem cell transplantation in patients with multiple myeloma

Background Whether the sequential treatment with bortezomib plus dexamethasone （BD） followed by autologous hematopoietic stem cell transplantation （ASCT） could extend the overall survival period in multiple myeloma patients is still not clear. Few large case studies about this therapeutics in multiple myeloma were reported in China. Our purpose was to assess the efficacy and adverse effects of sequential treatment with BD chemotherapy and ASCT in patients with multiple myeloma. Methods Fifty-three patients with newly diagnosed or relapsed/refractory multiple myeloma received BD as induction therapy before ASCT. Stem-cell mobilization was undertaken with cyclophosphamide 3-5 g/m2 plus granulocyte colony-stimulating factor 300 IJg/d. Target yield was 2.0x106 CD34＋ cells/kg. Conditioning for ASCT consisted of melphalan 200 mg/m2. Thalidomide and/or Q-interferon was used as post-transplantation maintenance treatment. Results The BD chemotherapy before transplantation was effective in 86.7% of the 53 patients, including 22.6% with complete remission （CR）, 39.6% with near complete remission （nCR）, and 24.5% with partial remission （PR）. The best effect was achieved after two treatment courses. Most bortezomib-related adverse effects were classes 1-2. All patients were successfully mobilized after BD for autologous peripheral blood stem cell transplantation. The ASCT was effective in 96.3% of patients, including 49.1% with CR, 32.1% with nCR, and 15.1% with PR. The CR rate was significantly increased （49.1% vs. 22.6%, P 〈0.05） by sequential ASCT. Within 27 （range, 6-53） months of follow-up, the efficacy of ASCT was maintained in 29 patients and further enhanced by post-transplantation maintenance treatment in four patients. Eleven patients died after transplantation. Among the patients undergoing BD/ASCT treatment, overall survival （OS） was significantly better in newly diagnosed patients in comparison to relapsed/refractory patients （P=0.046）. In this work,a Cu-10Ta alloy with a copper to tantalum mass ratio of 9:1 is prepared using powder metallurgy *** properties of the alloy,including density,microstructure,melting point,and constant-volume specific heat,are *** the split Hopkinson pressure bar(SHPB)and flyerplate impact experiments,the relationship between equivalent stress and equivalent plastic strain of the material is studied at temperatures of 298-823 K and under strain rates of 1×10^(-3)-5.2×10^(3)s^(-1),and the Hugoniot relationship at impact pressures of 1.46-17.25 GPa and impact velocities of 108-942 m/s is *** of the Cu-10Ta microstructure that occurs during high-strain-rate impact is *** indicate that the Cu-10Ta alloy possesses good thermal stability,and at ambient temperatures of up to 50%its melting point,stress softening of less than 15%of the initial strength is observed.A modified J-C constitutive model is employed to accurately predict the variation of yield strength with strain *** strong impact,the copper phase is identified as the primary source of plastic deformation in the Cu-10Ta alloy,while significant deformation of the high-strength tantalum particles is less ***,the longitudinal wave speed D is found to correlate linearly with the particle velocity u upon strong *** reveals that the sound speed and spallation strength of the alloy increase with increasing impact pressure.

The first example of sono-photocatalytic bond formation was reported. With both visible light and ultrasound wave as the energy, various 3-aminoquinoxalin-2(1H)-ones were efficiently obtained with good functional group tolerance in the absence of any additive or external photocatalyst. Compared with the conventional photocatalysis, sono-photocatalysis not only dramatically improved the reaction rates and yields, but also reduced energy consumption.

Objective Abnormal expression of T-lymphokine-activated killer cell-originated protein kinase(TOPK)was reported to be closely related to the resistance of prostate cancer to radiotherapy and to targeted drug resistance in lung ***,the role of TOPK inhibition in enhancing radiosensitivity of colorectal cancer(CRC)cells is *** study aimed to evaluate the radiosensitization of TOPK knockdown in CRC *** The expression of TOPK was detected in CRC tissues by immunohistochemistry,and the effect of TOPK knockdown was detected in CRC cells by Western ***-8 and clonogenic assays were used to detect the growth and clonogenic ability of CRC cells after TOPK knockdown combined with radiotherapy in CRC ***,proteomic analysis showed that the phosphorylation of TOPK downstream proteins changed after *** damage was detected by the comet *** in the DNA damage response signaling pathway were analyzed by Western blotting,and apoptosis was detected by flow *** The expression of TOPK was significantly greater in CRC tissues at grades 2–4 than in those at grade *** irradiation,CRC cells with genetically silenced TOPK had shorter comet tails and reduced expression levels of DNA damage response-associated proteins,including phospho-cyclin-dependent kinase 1(p-CDK1),phospho-ataxia telangiectasia-mutated(p-ATM),poly ADP-ribose polymerase(PARP),and meiotic recombination 11 homolog 1(MRE11).Conclusions TOPK was overexpressed in patients with moderately to poorly differentiated ***,TOPK knockdown significantly enhanced the radiosensitivity of CRC cells by reducing the DNA damage response.

The soil freezing characteristic curve(SFCC)plays a fundamental role in comprehending thermohydraulic behavior and numerical simulation of frozen *** study proposes a dynamic model to uniformly express SFCCs amidst varying total water contents throughout the freezing-thawing ***,a general model is proposed,wherein the unfrozen water content at arbitrary temperature is determined as the lesser of the current total water content and the reference value derived from saturated *** dynamic performance of this model is verified through test ***,in accordance with electric double layer(EDL)theory,the theoretical residual and minimum temperatures in SFCC are calculated to be-14.5℃to-20℃for clay particles and-260℃,*** ensure that the SFCC curve ends at minimum temperature,a correction function is introduced into the general ***,a simplified dynamic model is proposed and investigated,necessitating only three parameters inherited from the general ***,both general and simplified models are evaluated based on a test database and proven to fit the test data exactly across the entire temperature *** recommended parameter values for various types of soils are ***,this study provides not only a theoretical basis for most empirical equations but also proposes a new and more general equation to describe the SFCC.

An improved algorithm based on equivalent sound velocity profile method at large incident angle

With the development of ultra-wide coverage technology,multibeam echo-sounder(MBES)system has put forward higher requirements for localization accuracy and computational efficiency of ray tracing *** classical equivalent sound speed profile(ESSP)method replaces the measured sound velocity profile(SVP)with a simple constant gradient SVP,reducing the computational workload of beam ***,in deep-sea environment,the depth measurement error of this method rapidly increases from the central beam to the edge *** analyzing the positioning error of the ESSP method at edge beam,it is discovered that the positioning error increases monotonically with the incident angle,and the relationship between them could be expressed by polynomial ***,an error correction algorithm based on polynomial fitting is *** simulation experiment conducted on an inclined seafloor shows that the proposed algorithm exhibits comparable efficiency to the original ESSP method,while significantly improving bathymetry accuracy by nearly eight times in the edge beam.
