An overview of autophagy in the differentiation of dental stem cells

Dental stem cells(DSCs)have attracted significant interest as autologous stem cells since they are easily accessible and give a minimal immune response.These properties and their ability to both maintain self-renewal and undergo multi-lineage differentiation establish them as key players in regenerative medicine.While many regulatory factors determine the differentiation trajectory of DSCs,prior research has predominantly been based on genetic,epigenetic,and molecular aspects.Recent evidence suggests that DSC differentiation can also be influenced by autophagy,a highly conserved cellular process responsible for maintaining cellular and tissue homeostasis under various stress conditions.This comprehensive review endeavors to elucidate the intricate regulatory mechanism and relationship between autophagy and DSC differentiation.To achieve this goal,we dissect the intricacies of autophagy and its mechanisms.Subsequently,we elucidate its pivotal roles in impacting DSC differentiation,including osteo/odontogenic,neurogenic,and angiogenic trajectories.Furthermore,we reveal the regulatory factors that govern autophagy in DSC lineage commitment,including scaffold materials,pharmaceutical cues,and the extrinsic milieu.The implications of this review are far-reaching,underpinning the potential to wield autophagy as a regulatory tool to expedite DSC-directed differentiation and thereby promote the application of DSCs within the realm of regenerative medicine.

The deuteration of organic compounds has attracted more attentions in recent years for the potential applications in new drug discovery and synthetic chemistry.For this purpose,many efficient deuterium labeling methodologies have been developed,including hydrogen isotope exchange(HIE),reductive deuteration,and dehalogenative deuteration that allow for the synthesis of selectively deuterated compounds.In the last few years,great breakthroughs in selective isotope labeling have been achieved and the interest in new methodologies for the deuteration of organic molecules is rising.In this review,we summarized the recent developments in the selective deuteration of organic molecules since 2021.Several types of key processes in deuterium incorporation reactions,including H/D exchange,reductive deuteration and dehalogenative deuteration,are introduced and discussed.

Method for triangular fuzzy multiple attribute decision making based on two-dimensional density operator method

Aiming at the triangular fuzzy(TF)multi-attribute decision making(MADM)problem with a preference for the distribution density of attribute(DDA),a decision making method with TF number two-dimensional density(TFTD)operator is proposed based on the density operator theory for the decision maker(DM).Firstly,a simple TF vector clustering method is proposed,which considers the feature of TF number and the geometric distance of vectors.Secondly,the least deviation sum of squares method is used in the program model to obtain the density weight vector.Then,two TFTD operators are defined,and the MADM method based on the TFTD operator is proposed.Finally,a numerical example is given to illustrate the superiority of this method,which can not only solve the TF MADM problem with a preference for the DDA but also help the DM make an overall comparison.

Effect of droplet characteristics on liquid-phase distribution in spray zone of internal mixing air-mist nozzle

In continuous casting production,droplet characteristics are important parameters for evaluating the nozzle atomization quality,and have a significant impact on the secondary cooling effect and the slab quality.In order to study the behavior of atomized droplets after reaching the slab surface and to optimize the spray cooling effect,the influence of droplet diameter and droplet velocity on the migration behavior of droplets in the secondary cooling zone was analyzed by FLUENT software.Results show that the droplets in the spray zone and on the slab surface are mainly concentrated in the center,thus,the liquid volume fraction in the center is higher than that of either side.As the droplet diameter increases,the region of high liquid volume fraction on the slab surface becomes wider,and the liquid phase distribution in the slab width direction becomes uneven.Although increasing the droplet velocity at the nozzle exit has little effect on droplet diffusion in the spray zone,the distribution becomes more uneven due to more liquid reaches the slab surface per unit time.A prediction formula of the maximum water flow rate on the slab surface for specific droplet characteristics was proposed based on dimensionless analysis and validated by simulated data.A nozzle spacing of 210 mm was recommended under the working conditions in this study,which ensures effective coverage of the spray water over the slab surface and enhances the distribution uniformity of water flow rate in the transverse direction.

Three-stage niobium mineralization at Bayan Obo, China

The Chinese Bayan Obo deposit is a world-class rare earth element(REE) deposit with considerable niobium(Nb) and iron(Fe) resources. A complete genetic understanding on all metals is fundamental for establishing genetic models at Bayan Obo. With extensive research being focused on REE enrichment, the timing and controls of Nb enrichment remain unresolved at Bayan Obo, which is mainly due to the challenges in dating, i.e. multistage thermal events, fine-grained minerals with complex textures and the rare occurrence of uranium-enriched minerals with mature dating methods. Based on robust geological and petrographic frameworks, here we conducted ion probe uranium-lead(U-Pb) dating of ferrocolumbite to unravel the timing, hence the genesis of Nb mineralization. Three types of hydrothermal ferrocolumbites—key Nb-bearing minerals—are identified based on their textures and mineral assemblages. They yield U-Pb ages of 1312 ± 47 Ma(n = 99), 438 ± 7 Ma(n = 93), and 268 ± 5 Ma(n = 19), respectively. In line with deposit geology, we tentatively link the first, second and third stage Nb mineralization to Mesoproterozoic carbonatite magmatism, ubiquitous early Paleozoic hydrothermal activity, and Permian granitic magmatism, respectively. While quantifying the contribution of metal endowment from each stage requires further investigation, our new dates highlight that multi-stage mineralization is critical for Nb enrichment at Bayan Obo, which may also have implications for the enrichment mechanism of Nb in REE deposits in general.

Cyclic-acyclic monomers metathesis polymerization to access photodegradable polydicyclopentadiene and polyethylene-like materials

The development of degradable and recyclable polymers is an attractive strategy to tackle the post-consumption pollution issue of commercial plastics.Cyclic-acyclic monomers metathesis polymerization(CAMMP)has been recently reported to produce degradable thermoset,thermoplastic and elastomeric polymers,and their degradability is enabled with addition of various chemicals.In this contribution,we demonstrate that the utilization of diene comonomers containing photodegradable moiety can give access to thermoset and thermoplastic materials bearing photo-degradation capabilities.The copolymerization of a series easily accessible diene comonomers bearing photodegradable ortho-nitrobenzyl moieties with dicyclopentadiene(DCPD)or cyclooctene(COE)leads to the formation of photodegradable p DCPD thermosets and p COE thermoplastic polymers with mechanical properties comparable to their non-degradable counterparts.Most importantly,their photo-degradation properties can be efficiently tuned with the addition of ultraviolet absorber during in-situ polymerization.

Unconventional Fluorescent and Multi-responsive Polyethyleneimine with LCST and UCST Behavior:Synthesis,Characterization and Biological Applications

Non-aromatic fluorescent and multi-responsive materials,exhibiting inherent fluorescence emission and controlled phase change,have garnered significant attention in recent years.However,the underlying interaction between their fluorescent properties and phase transition remains unclear.In this study,we synthesized a series of catalyst-free aza-Michael addition-based polyethyleneimine(RFPEI)materials by reacting polyethyleneimine(PEI)with N-isopropyl acrylamide(NIPAM).The resulting RFPEI was comprehensively characterized,and demonstrated dual-phase transition behavior(LCST and UCST)in water,which could be finely tuned by adjusting its composition or external factors such as pH.Notably,upon UV irradiation(365 nm),RFPEI exhibited strong fluorescence emission.We further investigated the effects of NIPAM grafting percentage to PEI,polymer concentration,and pH on the LCST/UCST and fluorescent properties of RFPEI aqueous solutions.Moreover,we showcased the great potential of RFPEI as a versatile tool for physiological cell imaging,trace detection,and controlled release of doxorubicin.Our study presents a novel class of stimuli-responsive fluorescent materials with promising applications in the field of biomedicine.

Enhanced hydrogen evolution reaction in FePt film with remanence due to decrease in domain walls

As an effective strategy to improve the properties of electrocatalysts,magnetic field-assisted electrocatalytic water splitting has attracted increasing attention recently.However,the corresponding enhancements mostly depend on the exertion of an external magnetic field during electrochemical reactions,which results in a high cost of industrial production,and makes the magnetic field manipulation of electrocatalysis become a challenging task.In this work,instead of the external magnetic field,a bias magnetic field is self-supplied by the remanence state of a ferromagnetic electrocatalyst of FePt.Owing to the assistance of this bias magnetic field,the FePt film in the remanence state shows the overpotential of 229 mV during hydrogen evolution reaction,which is much lower than that in its demagnetization state(283 mV).Our findings demonstrate that the remanence in ferromagnetic electrocatalysts can improve the catalytic performance,which is attributed to the decrease in domain walls.

Objective Acute myeloid leukemia(AML)is an aggressive hematological malignancy characterized by abnormal myeloid blast expansion.Recent studies have demonstrated that circular RNAs play a role in AML pathogenesis.In this study,we aimed to investigate the clinical significance of circ_0012152 in AML and elucidate its underlying molecular mechanism in the pathogenesis of this condition.Methods Circ_0012152 expression was detected by quantitative real-time polymerase chain reaction in samples obtained from 247 patients with AML and 40 healthy controls.A systematic analysis of clinical characteristics and prognostic factors was also conducted.Cell growth was assessed using the Cell Counting Kit-8(CCK-8)assay,and apoptosis and cell cycle progression were evaluated by flow cytometry.Moreover,RNA pull-down was performed to identify target microRNAs,and transcriptome RNA sequencing and bioinformatics analyses were utilized to identify downstream mRNA targets.Results Circ_0012152 was significantly upregulated in samples from patients with AML and served as an independent adverse prognostic factor for overall survival(OS)(hazard ratio:2.357;95%confidence interval 1.258–4.415).The circ_0012152 knockdown reduced cell growth,increased apoptosis,and inhibited cell cycle progression in AML cell lines.RNA pull-down and sequencing identified miR-652-3p as a target microRNA of circ_0012152.Cell growth inhibition by circ_0012152 knockdown was significantly relieved by miR-652-3p inhibitors.We suggested that miR-652-3p targeted SOX4,as the decrease in SOX4 expression resulting from circ_0012152 knockdown was upregulated by miR-652-3p inhibitors in AML cells.Conclusion Circ_0012152 is an independent poor prognostic factor for OS in AML,and it promotes AML cell growth by upregulating SOX4 through miR-652-3p.

Irreversible Investment under Endowment Constraints

In this paper, we study the problem of irreversible investment under endowment constraints. We first establish the existence and uniqueness of the result and then demonstrate the necessity and sufficient conditions for optimality. Based on this condition, we provide a characterization for optimal investment plans,which can be obtained by the so-called base capacity solving a backward equation. We may obtain explicit solutions for certain typical cases.
