Screening biomarkers for spinal cord injury using weighted gene co-expression network analysis and machine learning

Immune changes and inflammatory responses have been identified as central events in the pathological process of spinal co rd injury.They can greatly affect nerve regeneration and functional recovery.However,there is still limited understanding of the peripheral immune inflammato ry response in spinal cord inju ry.In this study.we obtained microRNA expression profiles from the peripheral blood of patients with spinal co rd injury using high-throughput sequencing.We also obtained the mRNA expression profile of spinal cord injury patients from the Gene Expression Omnibus(GEO)database(GSE151371).We identified 54 differentially expressed microRNAs and 1656 diffe rentially expressed genes using bioinformatics approaches.Functional enrichment analysis revealed that various common immune and inflammation-related signaling pathways,such as neutrophil extracellular trap formation pathway,T cell receptor signaling pathway,and nuclear factor-κB signal pathway,we re abnormally activated or inhibited in spinal cord inju ry patient samples.We applied an integrated strategy that combines weighted gene co-expression network analysis,LASSO logistic regression,and SVM-RFE algorithm and identified three biomarke rs associated with spinal cord injury:ANO10,BST1,and ZFP36L2.We verified the expression levels and diagnostic perfo rmance of these three genes in the original training dataset and clinical samples through the receiver operating characteristic curve.Quantitative polymerase chain reaction results showed that ANO20 and BST1 mRNA levels were increased and ZFP36L2 mRNA was decreased in the peripheral blood of spinal cord injury patients.We also constructed a small RNA-mRNA interaction network using Cytoscape.Additionally,we evaluated the proportion of 22 types of immune cells in the peripheral blood of spinal co rd injury patients using the CIBERSORT tool.The proportions of naive B cells,plasma cells,monocytes,and neutrophils were increased while the proportions of memory B cells,CD8^(+)T cells,resting natural killer cells,resti

The nitrile butadiene rubber(NBR)hardness effect on the sealing characteristics of hydraulic O-ring rod seals is analyzed based on a mixed lubrication elastohydrodynamic model.Parameterized studies are conducted to reveal the mechanism of the influence of rubber hardness on the static and dynamic behavior of seals.The optimized selections of rubber hardness are then investigated under different conditions.Results show that the low hardness seal is prone to stress concentration due to the extrusion effect under high pressure conditions;it is also more prone to leaking.A high hardness seal can better prevent leakage by reducing film thickness but it will cause large frictional power loss and increase the probability of wear failure.The choice of low hardness is recommended to reduce friction with the premise that leakage requirements are met.

Dry eye disease(DED)is a major ocular pathology worldwide,causing serious ocular discomfort and even visual impairment.The incidence of DED is gradually increasing with the highfrequency use of electronic products.Although inflammation is core cause of the DED vicious cycle,reactive oxygen species(ROS)play a pivotal role in the vicious cycle by regulating inflammation from upstream.Therefore,current therapies merely targeting inflammation show the failure of DED treatment.Here,a novel dual-atom nanozymes(DAN)-based eye drops are developed.The antioxidative DAN is successfully prepared by embedding Fe and Mn bimetallic single-atoms in N-doped carbon material and modifying it with a hydrophilic polymer.The in vitro and in vivo results demonstrate the DAN is endowed with superior biological activity in scavenging excessive ROS,inhibiting NLRP3 inflammasome activation,decreasing proinflammatory cytokines expression,and suppressing cell apoptosis.Consequently,the DAN effectively alleviate ocular inflammation,promote corneal epithelial repair,recover goblet cell density and tear secretion,thus breaking the DED vicious cycle.Our findings open an avenue to make the DAN as an intervention form to DED and ROSmediated inflammatory diseases.

Ginkgo biloba extract alleviates fatty liver hemorrhagic syndrome in laying hens via reshaping gut microbiota

Background Ginkgo biloba extract(GBE)is evidenced to be effective in the prevention and alleviation of metabolic disorders,including obesity,diabetes and fatty liver disease.However,the role of GBE in alleviating fatty liver hemorrhagic syndrome(FLHS)in laying hens and the underlying mechanisms remain to be elucidated.Here,we investigated the effects of GBE on relieving FLHS with an emphasis on the modulatory role of GBE in chicken gut microbiota.Results The results showed that GBE treatment ameliorated biochemical blood indicators in high-fat diet(HFD)-induced FLHS laying hen model by decreasing the levels of TG,TC,ALT and ALP.The lipid accumulation and pathological score of liver were also relieved after GBE treatment.Moreover,GBE treatment enhanced the antioxidant activity of liver and serum by increasing GSH,SOD,T-AOC,GSH-PX and reducing MDA,and downregulated the expression of genes related to lipid synthesis(FAS,LXRα,GPAT1,PPARγand Ch REBP1)and inflammatory cytokines(TNF-α,IL-6,TLR4 and NF-κB)in the liver.Microbial profiling analysis revealed that GBE treatment reshaped the HFD-perturbed gut microbiota,particularly elevated the abundance of Megasphaera in the cecum.Meanwhile,targeted metabolomic analysis of SCFAs revealed that GBE treatment significantly promoted the production of total SCFAs,acetate and propionate,which were positively correlated with the GBE-enriched gut microbiota.Finally,we confirmed that the GBE-altered gut microbiota was sufficient to alleviate FLHS by fecal microbiota transplantation(FMT).Conclusions We provided evidence that GBE alleviated FLHS in HFD-induced laying hens through reshaping the composition of gut microbiota.Our findings shed light on mechanism underlying the anti-FLHS efficacy of GBE and lay foundations for future use of GBE as additive to prevent and control FLHS in laying hen industry.

Originating but free from chromosomal DNA,extrachromosomal circular DNAs(eccDNAs)are organized in circular form and have long been found in unicellular and multicellular eukaryotes.Their biogenesis and function are poorly understood as they are characterized by sequence homology with linear DNA,for which few detection methods are available.Recent advances in high-throughput sequencing technologies have revealed that eccDNAs play crucial roles in tumor formation,evolution,and drug resistance as well as aging,genomic diversity,and other biological processes,bringing it back to the research hotspot.Several mechanisms of eccDNA formation have been proposed,including the breakage-fusion-bridge(BFB)and translocation-deletion-amplification models.Gynecologic tumors and disorders of embryonic and fetal development are major threats to human reproductive health.The roles of eccDNAs in these pathological processes have been partially elucidated since the first discovery of eccDNA in pig sperm and the double minutes in ovarian cancer ascites.The present review summarized the research history,biogenesis,and currently available detection and analytical methods for eccDNAs and clarified their functions in gynecologic tumors and reproduction.We also proposed the application of eccDNAs as drug targets and liquid biopsy markers for prenatal diagnosis and the early detection,prognosis,and treatment of gynecologic tumors.This review lays theoretical foundations for future investigations into the complex regulatory networks of eccDNAs in vital physiological and pathological processes.

Numerical Analysis on the Static Performance of Gas Journal Bearing by Using Finite Element Method

In this paper,finite element method is used to calculate the static performance of gas journal bearing,in which rotation speed term is introduced into the stiffness matrix of linear triangular element to realize the performance calculation of the bearing with rotation speed.The results indicate that the average gas film thicknesses corresponding to the maximum load capac-ity and stiffness,and the minimum attitude angle increase with the growth of orifice diameter.Load capacity and stiffness significantly improved with the increase of rotation speed,eccentricity ratio and supply pressure when the bearing has thin average gas film thickness.Attitude angle increases with the growth of rotation speed,while the growth rate slows down or even decreases at high speed.The most effective way of reducing attitude angle is to increase supply pressure.It can be found that rotation speed affects attitude angle through changing gas pressure difference between two orifices,while other parameters have the same effect by changing gas pressure at orifice outlet.

Comparison of 2-acetylfuran,2-ethylfuran,and 2-methylfuran spherically expanding flame intrinsic instabilities

The intrinsic instability of three furan derivatives, namely, 2-acetylfuran(AF), 2-ethylfuran(EF), and 2-methylfuran(MF), is experimentally and theoretically investigated and compared. A deep-learning-based end-to-end system is used to segment the cells and measure the crack length on the cellular flame front of the spherically expanding flame to quantitively investigate the flame morphology. Moreover, the flame front radius profile is quantitively investigated to shed light on the correlation between disturbance amplitude and flame radius. With the use of the linear stability model, the theoretical critical flame radius and Peclet number are obtained and compared to their corresponding experimental results to characterize the outset of cellularization and evaluate the flame instability propensity. Our analysis shows that compared with AF and EF, MF is prone to cellularization at a smaller flame radius and has a faster cell proliferation rate and crack length growth rate before reaching the hydrodynamic saturation limit. Meanwhile, the average area of saturated cells tends to be similar for the three furan derivatives. Furthermore,AF premixed flame does not exhibit cellularization before the flame radius is less than 45 mm in low initial pressure.

Three New Hydroxytetradecenals from Amomum tsao-ko with Protein Tyrosine Phosphatase 1B and Glycogen Phosphorylase Inhibitory Activity

The fruits of Amomum tsao-ko(Cao-Guo)were documented in Chinese Pharmacopoeia for the treatment of abdominal pain,vomiting,and plague.In our previous study,a series of diarylheptanes and flavonoids withα-glucosidase and protein tyrosine phosphatase 1B(PTP1B)inhibitory activity have been reported from the middle-polarity part of A.tsao-ko,whereas the antidiabetic potency of the low-polarity constituents is still unclear.In this study,three new hydroxytetradecenals,(2E,4E,8Z,11Z)-6R-hydroxytetradeca-2,4,8,11-tetraenal(1),(2E,4E,8Z)-6R-hydroxytetradeca-2,4,8-trienal(2)and(2E,4E)-6R-hydroxytetradeca-2,4-dienal(3)were obtained from the volatile oils of A.tsao-ko.The structures of compounds 1–3 were determined using spectroscopic data involving 1D and 2D nuclear magnetic resonance(NMR),high-resolution mass spectra(HRMS),and specific rotation([α]D).Their hypoglycemic activity was evaluated against glycogen phosphorylase(GPa)and PTP1B.Compounds 1 and 2 displayed moderate activity against PTP1B with inhibition rates of 33.8%−50.3%at 100 and 200μM.Moreover,compound 1 exhibited an obvious inhibitory effect on GPa(IC50=31.7μM),whereas compound 2 was inactive.This study demonstrates hydroxytetradecenals as the characteristic components of A.tsao-ko with therapeutic potential in diabetes.

Divergent contributions of spring and autumn photosynthetic phenology to seasonal carbon uptake of subtropical vegetation in China

Phenological changes play a central role in regulating seasonal variation in the ecological processes,exerting significant impacts on hydrologic and nutrient cycles,and ultimately influencing ecosystem functioning such as carbon uptake.However,the potential impact mechanisms of phenological events on seasonal carbon dynamics in subtropical regions are under-investigated.These knowledge gaps hinder from accurately linking photosynthetic phenology and carbon sequestration capacity.Using chlorophyll fluorescence remote sensing and productivity data from 2000 to 2019,we found that an advancement in spring phenology increased spring gross （GPP） and net primary productivity （NPP） in subtropical vegetation of China by 2.1 gC m-2yr-1and 1.4 gC m-2yr-1,respectively.A delay in autumn phenology increased the autumnal GPP and NPP by 0.4 gC m-2yr-1and 0.2 gC m-2yr-1,respectively.Temporally,the contribution of the spring phenology to spring carbon uptake increased significantly during the study period,while this positive contribution showed a nonsignificant trend in summer.In comparison,the later autumn phenology could significantly contribute to the increase in autumnal carbon uptake;however,this contributing effect was weakened.Path analysis indicated that these phenomena have been caused by the increased leaf area and enhanced photosynthesis due to earlier spring and later autumn phenology,respectively.Our results demonstrate the diverse impacts of vegetation phenology on the seasonal carbon sequestration ability and it is imperative to consider such asymmetric effects when modeling ecosystem processes parameterized under future climate change.

数据产权可以被视为不可分割的一元权利,也可以将所有权和用益权分离,而数据所有权与用益权分离带来的经济意义仍有争议.数字企业提供基于数据的服务激励用户提供数据,同时利用数据进行个性化定价,并且导致用户遭受隐私损失.本文构建了一个垄断厂商模型,考虑了存在个性化定价与隐私损失时,数据收集者缺乏用益权与拥有用益权两种权利界定下的数据收集行为与福利影响.研究发现,数据收集者缺乏用益权时,隐私成本较高则不会有数据收集,隐私成本较低会出现过度的数据收集,体现为收集者耗费大量成本获取所有用户的数据.收集者拥有初始用益权,且用户拥有数据所有权时,反而可以实现有效率的数据收集,提高用户的剩余和社会福利.进一步分析收集者同时拥有数据所有权和用益权的情况,发现数据所有权只影响社会福利在数据收集者与用户之间的分配.因此,应当考虑数据所有权和用益权分离的权利界定架构,并且分场景讨论数据用益权归属:当隐私成本较低时,应赋予数据收集者用益权,并且让用户通过“删除权”等方式行使所有权,以提高社会福利和用户剩余;当隐私成本较高或用户会低估隐私成本时,应采取禁止数据收集而非确权的方式,以保护敏感数据.
