Identification of a novel planthopper resistance gene from wild rice(Oryza rufipogon Griff.)

Rice planthoppers,including brown planthopper(BPH)and white-backed planthopper(WBPH),are the most destructive pests in Asian rice cultivation *** resistance genes that have been mapped and characterized advance our understanding of underlying resistance mechanisms and facilitate the breeding of resistant varieties,thereby contributing to an efficient pest management *** this study,a novel resistance gene Bph38 derived from the wild rice species Oryza rufipogon *** found to confer high resistance to BPH and *** mapping was performed to identify regions associated with BPH and WBPH resistance,and two mapping efforts led to the same region on chromosome 4 flanked by markers RM16563 and ***-segregant analysis and next-generation sequencing were performed using the same population to detect the resistance *** mapping narrowed the region to a 12.3-Mb segment,and fine mapping using BC1 F2 recombinants identified a 79-kb segment flanked by markers YM112 and ***-isogenic lines(NILs)carrying Bph38 in the 9311(indica)and BR54(japonica)genetic backgrounds were developed by crossing and backcrossing with marker-assisted *** agronomic traits and BPH and WBPH resistance of the NILs were similar to those of the recurrent *** feeding and host-choice tests revealed that Bph38 showed both antibiotic and antixenotic effects in both insects,with stronger effects in indica-background *** fine mapping and characterization of the major gene may result in map-based cloning of the gene and allow its application in breeding insectresistant rice varieties.

[目的]利用全基因组选择信号检测方法探索中国地方猪基因组中与体型相关的候选基因,分析中国地方猪在进化和驯化过程中的受选择情况。[方法]基于310头地方猪的全基因组重测序数据,利用跨群体扩展单倍型纯合(cross population extended haplotype homozygosity,XP-EHH)和固定分化指数F ST统计量(fixation index)两种方法进行全基因组选择信号检测。取两种方法各前0.1%位点的交集作为候选位点,分别向上、下游延伸200 kb作为潜在受选择区域。通过富集分析进一步探索选择信号的生物学功能。[结果]使用XP-EHH和F ST两种方法共检测到633个潜在受选择位点,获得633个潜在区域,共注释到133个基因。数量性状基因座(quantitative trait locus,QTL)富集分析发现,不同体型中国地方猪之间的选择信号与肉质和胴体性状相关;染色质状态富集结果发现,不同体型中国地方猪群体之间的差异组织为内脏、消化系统和小脑。GO功能和KEGG通路富集分析发现16个基因在6个功能条目(P3)上显著富集,主要集中在生长代谢相关通路,其中ACSM 5、ACSM 4、FXR 1和FOXA 2作为候选基因主要富集于脂肪酸合成与代谢、肌肉生长发育等信号通路。[结论]本研究采用两种选择信号检测方法对不同体型的中国地方猪进行了分析,筛选到一系列候选位点和基因,重点挖掘了参与猪生长发育的候选基因,如ACSM 5、ACSM 4、FXR 1和FOXA 2。

ZBTB7A governs 2-DG-inhibited glycolysis by regulating GLUT1 in nasopharyngeal carcinoma

Our previous studies suggested a potential interaction between the POK erythroid myeloid ontogenic factor ZBTB7A and glucose transporter 1(GLUT1)in nasopharyngeal carcinoma(NPC).This study was designed to confirm the interaction and further evaluate the precise mechanism by which ZBTB7A and GLUT1 regulate NPC *** binding sites between ZBTB7A and the promoter of GLUT1 were predicted by *** expression was measured by quantitative real-time polymerase chain reaction(qPCR),western blotting,and *** activities of key glycolysis enzymes,including hexokinase(HK),pyruvate kinase(PK),lactate dehydrogenase(LDH),and lactate,were detected using specific enzyme-linked immunosorbent assay *** connection between ZBTB7A and GLUT1 was analyzed by dual-luciferase reporter assay and chromatin *** vitality,proliferation,and tumorigenicity of the cells expressing different levels of ZBTB7A were tested by adding the glycolysis inhibitor 2-deoxy-D-glucose(2-DG),followed by MTT,colorimetric focus forming,and xenograft assays,*** results showed that high expression of GLUT1 was associated with late-stage *** constructing stably transfected cells with lentiviruses,ZBTB7A was effectively knocked down in 5-8F cells(RNAi-5-8F)and overexpressed in 6-10B cells(ZBTB7A-6-10B).The up-or downregulation of GLUT1 secondary to ZBTB7A changes was also *** vitality and proliferation of the cells expressing low ZBTB7A were notably blocked by *** cells expressing high ZBTB7A were not very sensitive to *** growth of RNAi-5-8F xenografts was strongly suppressed by *** activities of HK,PK,and LDH were suppressed by 2-DG in the cells expressing low ***-5-8F cells had the lowest 2-DG-induced lactate ***7A directly suppressed the promoter region of GLUT1 to regulate GLUT1 ***,ZBTB7A controls the 2-DG-induced inhibition of glycolysis by affecting GLUT1.

The current study evaluated the effects of oxalic acid(OA)application on the growth and Mn phytoremediation efficiency of Polygonum pubescens Blume cultivated in three different manganese(Mn)-contaminated soils sampled from an unexplored area(US),an explored area(ES)and a tailing area(TS)of the Ertang Mn mine,South *** supplied levels of OA were 0(control),1(low level),3(medium level),and 9(high level)mmol/kg,referred to as CK,OA1,OA3 and OA9,*** results revealed that the average water-extractable Mn concentrations US,ES and TS amended with OA increased by 214.13,363.77 and 266.85%,*** OA supply levels increased plant growth and Mn concentrations in *** low OA supply level increased plant growth in ES and TS;however,contrasting results were found for the medium and high OA supply *** Mn concentrations and total Mn increased in ES and TS in response to all OA supply *** Mn in the aerial parts increased by 81.18,44.17 and 83.17%in US,ES and TS,respectively;the corresponding percentages for the whole plants were 81.53,108.98 and 77.91%,*** rate of·O_(2)^(−)production and malondialdehyde(MDA)concentrations increased in response to OA amendment,especially the medium and high OA supply levels in ES and *** general,antioxidant enzymes might play a vital role in alleviating Mn stress in plants cultivated in US,while non-enzymatic antioxidants might be the main factor for plants cultivated in ES and TS.

Total flavonoids of hawthorn leaves promote motor function recovery via inhibition of apoptosis after spinal cord injury

Flavonoids have been reported to have therapeutic potential for spinal cord *** leaves have abundant content and species of total flavonoids,and studies of the effects of the total flavonoids of hawthorn leaves on spinal cord injury have not been published in or outside ***,Sprague-Dawley rats were used to establish a spinal cord injury model by Allen's *** were intraperitoneally injected with 0.2 m L of different concentrations of total flavonoids of hawthorn leaves(5,10,and 20 mg/kg)after spinal cord *** were administered once every 6 hours,three times a day,for 14 *** treatment with various concentrations of total flavonoids of hawthorn leaves,the Basso,Beattie,and Bresnahan scores and histological staining indicated decreases in the lesion cavity and number of apoptotic cells of the injured spinal cord tissue;the morphological arrangement of the myelin sheath and nerve cells tended to be regular;and the Nissl bodies in neurons *** Basso,Beattie,and Bresnahan scores of treated spinal cord injury rats were *** blot assays showed that the expression levels of pro-apoptotic Bax and cleaved caspase-3 were decreased,but the expression level of the anti-apoptotic Bcl-2 protein was *** improvement of the above physiological indicators showed a dose-dependent relationship with the concentration of total flavonoids of hawthorn *** above findings confirm that total flavonoids of hawthorn leaves can reduce apoptosis and exert neuroprotective effects to promote the recovery of the motor function of rats with spinal cord *** study was approved by the Ethics Committee of the Guangxi Medical University of China(approval No.201810042)in October 2018.

QSAR Models for Predicting Additive and Synergistic Toxicities of Binary Pesticide Mixtures on Scenedesmus Obliquus

Pesticides released into the environment may pose potential risks to the ecological system and hu-man ***,existing toxicity data on pesticide mixtures still lack,especially regarding the toxic interac-tions of their *** study aimed to determine the toxic interactions of binary mixtures of pesticides on Scenedesmus Obliquus(***)and to build quantitative structure-activity relationship models(QASR)for predicting the mixture *** applying direct equipartition ray method to design binary mixtures of five pes-ticides(linuron,dimethoate,dichlorvos,trichlorfon and metribuzin),the toxicity of a single pesticide and its mix-ture was tested by microplate toxicity analysis on *** QASR models were built for combined toxicity of binary mixtures of pesticides at the half-maximal effective concentration(EC_(50)),30%maximal effective concen-tration(EC_(30))and 10%maximal effective concentration(EC_(10)).The results showed that the single toxicity follows:metribuzin>linuron>dichlorvos>trichlorfon>*** mixtures of linuron and trichlorfon,dichlorvos and metribuzin,dimethoate and metribuzin induced synergetic effects,while the remaining binary mixtures exhib-ited *** developed QSAR models were internally validated using the leave-one-out cross-validation(LOO),leave-many-out cross-validation(LMO),bootstrapping,and y-randomization test,and externally validated by the test *** three QSAR models satisfied well with the experimental values for all mixture toxicities,and presented high internally(R^(2)and Q^(2)>0.85)and externally(Q^(2)_(F1),Q^(2)_(F2),and Q^(2)_(F3)>0.80)predictive *** developed QSAR models could accurately predict the toxicity values of EC_(50),EC_(30)and EC_(10)and were superior to the concentration addition model's results(CA).Compared to the additive effect,the QSAR model could more ac-curately predict the binary mixture toxicities of pesticides with synergistic effects.

Quantitative Evaluation of Leaf Inclination Angle Distribution on Leaf Area Index Retrieval of Coniferous Canopies

Both leaf inclination angle distribution(LAD)and leaf area index(LAI)dominate optical remote sensing *** G-function,which is a function of LAD and remote sensing geometry,is often set to 0.5 in the LAI retrieval of coniferous canopies even though this assumption is only valid for spherical *** uncertainties are thus ***,because numerous tiny leaves grow on conifers,it is nearly impossible to quantitatively evaluate such uncertainties in LAI *** this study,we proposed a method to characterize the possible change of G-function of coniferous canopies as well as its effect on LAI ***,a Multi-Directional Imager(MDI)was developed to capture stereo images of the branches,and the needles were *** accuracy of the inclination angles calculated from the reconstructed needles was ***,we analyzed whether a spherical distribution is a valid assumption for coniferous canopies by calculating the possible range of the G-function from the measured LADs of branches of Larch and Spruce and the true G-functions of other species from some existing inventory data and threedimensional(3D)tree *** show that the constant G assumption introduces large errors in LAI retrieval,which could be as large as 53%in the zenithal viewing direction used by spaceborne *** a result,accurate LAD estimation is *** the absence of such data,our results show that a viewing zenith angle between 45 and 65 degrees is a good choice,at which the errors of LAI retrieval caused by the spherical assumption will be less than 10%for coniferous canopies.
