Centennial Analysis of Human Activity Intensity and Associated Historical Events in Heilongjiang River Sino-Russo Watershed

Human activities in a transborder watershed are complex under the influence of domestic policies,international relations,and global events.Understanding the forces driving human activity change is important for the development of transborder watershed.In this study,we used global historical land cover data,the hemeroby index model,and synthesized major historical events to analyze how human activity intensity changed in the Heilongjiang River(Amur River in Russia)watershed(HLRW).The results showed that there was a strong spatial heterogeneity in the variation of human activity intensity in the HLRW over the past century(1900-2016).On the Chinese side,the human activity intensity change shifted from the plain areas for agricultural reclamation to the mountainous areas for timber extraction.On the Russian side,human activity intensity changes mostly concentrated along the Trans-Siberian Railway and the Baikal-Amur Mainline.Localized variation of human activity intensity tended to respond to regional events while regionalized variation tends to reflect national policy change or broad international events.The similarities and differences between China and Russia in policies and positions in international events resulted in synchronous and asynchronous changes in human activity intensity.Meanwhile,policy shifts were often confined by the natural features of the watershed.These results reveal the historical origins and fundamental connotations of watershed development and contribute to formulating regional management policies that coordinate population,eco-nomic,social,and environmental activities.

Immunotherapy represents a promising strategy for cancer treatment that utilizes immune cells or drugs to activate the patient's own immune system and eliminate cancer cells.One of the most exciting advances within this field is the targeting of neoantigens,which are peptides derived from non-synonymous somatic mutations that are found exclusively within cancer cells and absent in normal cells.Although neoantigen-based therapeutic vaccines have not received approval for standard cancer treatment,early clinical trials have yielded encouraging outcomes as standalone monotherapy or when combined with checkpoint inhibitors.Progress made in high-throughput sequencing and bioinformatics have greatly facilitated the precise and efficient identification of neoantigens.Consequently,personalized neoantigen-based vaccines tailored to each patient have been developed that are capable of eliciting a robust and long-lasting immune response which effectively eliminates tumors and prevents recurrences.This review provides a concise overview consolidating the latest clinical advances in neoantigen-based therapeutic vaccines,and also discusses challenges and future perspectives for this innovative approach,particularly emphasizing the potential of neoantigen-based therapeutic vaccines to enhance clinical efficacy against advanced solid tumors.

Circulating tumor cells(CTCs)are important biomarkers in the development and progression of lung cancer because they can reach other organs through the blood circulation and form distant metastases,exacerbating lung cancer progression.The presence of CTCs is also the main reason for the failure of nanomedicine-based lung cancer treatments.Therefore,magnetic MoSe_(2) nanosheets loaded with programmed death-ligand 1(PD-L1),named PD-L1-MFP NS,were employed here to precisely capture lung cancer CTCs in the blood circulation through the tumor-targeting effect of PD-L1 killing CTCs with highly effective photothermal therapy(PTT).In addition,by increasing the expression of cytomegalovirus UL16-binding protein(ULBP)ligands on tumor cells,the PD-L1-MFP NS further activated natural killer(NK)cells and triggered NK cell-induced cancer immunotherapy,thereby enhancing the overall tumor-killing effect.In summary,this material designed to capture CTCs provides a substantial advancement for personalized PTT-triggered immunotherapy and has great clinical translational potential.

OTDR-based optical fiber bending and tensile loss analysis

When optical fiber is deployed in practical engineering,bending and stretching of fiber optics is inevitable,which will affect optical communication.The fiber losses of different bending radii are simulated by COMSOL software.In order to verify the accuracy of simulation results,an experiment was designed to measure the losses of single-mode fiber under different bending radii and tension forces.The results show that the sensitive bending diameter of fiber loss is between 5 mm and 10 mm.The tensile effect has little influence on the fiber loss,but when the tensile force is greater than 160 N,the fiber breaks.This study provides an important reference for fiber layout in practical engineering.

Sucrose-associated SnRK1a1-mediated phosphorylation of Opaque2 modulates endosperm filling in maize

During maize endosperm filling,sucrose not only serves as a source of carbon skeletons for storage-reserve synthesis but also acts as a stimulus to promote this process.However,the molecular mechanisms underlying sucrose and endosperm filling are poorly understood.In this study,we found that sucrose promotes the expression of endosperm-filling hub gene Opaque2(O2),coordinating with storage-reserve accumulation.We showed that the protein kinase SnRK1a1 can attenuate O2-mediated transactivation,but sucrose can release this suppression.Biochemical assays revealed that SnRK1a1 phosphorylates O2 at serine 41(S41),negatively affecting its protein stability and transactivation ability.We observed that mutation of SnRK1a1 results in larger seeds with increased kernel weight and storage reserves,while overexpression of SnRK1a1 causes the opposite effect.Overexpression of the native O2(O2-OE),phospho-dead(O2-SA),and phospho-mimetic(O2-SD)variants all increased 100-kernel weight.Although O2-SA seeds exhibit smaller kernel size,they have higher accumulation of starch and proteins,resulting in larger vitreous endosperm and increased test weight.O2-SD seeds display larger kernel size but unchanged levels of storage reserves and test weight.O2-OE seeds show elevated kernel dimensions and nutrient storage,like a mixture of O2-SA and O2-SD seeds.Collectively,our study discovers a novel regulatory mechanism of maize endosperm filling.Identification of S41 as a SnRK1-mediated phosphorylation site in O2 offers a potential engineering target for enhancing storage-reserve accumulation and yield in maize.

Motor imagery-based EEG signals classification by combining temporal and spatial deep characteristics

Purpose-In order to improve the weak recognition accuracy and robustness of the classification algorithm for brain-computer interface(BCI),this paper proposed a novel classification algorithm for motor imagery based on temporal and spatial characteristics extracted by using convolutional neural networks(TSCNN)model.Design/methodology/approach-According to the proposed algorithm,a five-layer neural network model was constructed to classify the electroencephalogram(EEG)signals.Firstly,the author designed a motor imagery-based BCI experiment,and four subjects were recruited to participate in the experiment for the recording of EEG signals.Then,after the EEG signals were preprocessed,the temporal and spatial characteristics of EEG signals were extracted by longitudinal convolutional kernel and transverse convolutional kernels,respectively.Finally,the classification of motor imagery was completed by using two fully connected layers.Findings-To validate the classification performance and efficiency of the proposed algorithm,the comparative experiments with the state-of-the-arts algorithms are applied to validate the proposed algorithm.Experimental results have shown that the proposed TS-CNN model has the best performance and efficiency in the classification of motor imagery,reflecting on the introduced accuracy,precision,recall,ROC curve and F-score indexes.Originality/value-The proposed TS-CNN model accurately recognized the EEG signals for different tasks of motor imagery,and provided theoretical basis and technical support for the application of BCI control system in the field of rehabilitation exoskeleton.

Effect of Dangfei liganning capsule(当飞利肝宁胶囊) on liver X receptor α/steroid regulatory element binding protein-1/fatty acid synthase signal pathway in rats with metabolic-associated fatty liver disease

OBJECTIVE: To study the mechanism of Dangfei liganning capsule(当飞利肝宁胶囊) in the treatment of rats with metabolic associated fatty liver disease(MAFLD). METHODS: Totally 48 specific pathogen free SpragueDawley male rats were randomly divided into normal Group, model group, Dangfei liganning high, moderate, and low-dose groups and Essentiale group which were fed with high fat diet for 8 weeks, and gavage and molding were carried out simultaneously. Dangfei liganning high, middle and low-dose group were given 0.27, 0.135 and 0.0675 g·kg-1·d-1 respectively by gavage, Essentiale group was given 0.123 g·kg-1·d-1 by gavage, the same amount of distilled water was given by gavage in the normal group and the model group. The rats were weighed at the 0th week, 2nd week, 4th week, 6th week and 8th weekend respectively. The rats were sacrificed at the end of the 8th week. Serum levels of alanine aminotransferase(ALT), alanine aminotransferase(AST),triglyceride(TG), total cholesterol(CHO), high-density lipoprotein cholesterol(HDL-C), low-density lipoprotein (LDL-C), total protein(TP), albumin(Alb), globulin(GLB), total bilirubin(TBIL), direct bilirubin(DBIL), tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6) were measured. The levels of liver tumor necrosis factor-α(TNF-α), interleukin-6(IL-6) and liver pathology [hematoxylin and eosin(HE) staining, oil red O staining] were detected. The expression levels of liver X receptor α(LXRα), steroid regulatory element binding protein-1(SREBP-1) and fatty acid synthase(FAS) were detected by immunohistochemistry, Western blot and reverse transcription-polymerase chain reaction reverse transcription-polymerase chain reaction. RESULTS: From the beginning to the 8th week, the growth rate of body weight in the Dangfei liganning highdose group was slower than all other groups. There was no significant difference in ALB level in all groups(P > 0.05). Compared with the model group, the levels of ALT, AST, LDL-C, TG, CHO, TP, GLB, TBIL, DBIL, IL-6, TNF-α were significantly decreased and HDL

随着区域协同发展政策战略地位的不断提高,区域协同研究正如火如荼,但现阶段学界尚有待阐明区域协同瓶颈之因并提出解决之法及相应的区域协同新机制。本研究试图聚焦于此,在理论层面梳理区域协同瓶颈产生机制与区域协同新机制,并对成渝地区双城经济圈协同推进机关事务标准化工作进行案例分析与问卷调查,在实证层面检验本研究的两个理论框架。案例分析发现,以机关事务为例,成渝地区双城经济圈存在治理尺度差异,这与中央政策调控共同构成协同的内外动机,驱动成渝地区深化合作意愿,卷入区域协同;与此同时,地方保护主义现象在两地机关事务标准化协同工作中亦有所披露,并阻碍了成渝地区双城经济圈建设的协同发展。问卷调查数据进一步验证案例分析结果:推进协同地区加强行政治理协同、推进治理尺度统一具有紧要性,成渝地区双城经济圈建设推进标准化执行助推区域协同发展,标准化的行政治理协同增强了成渝地区双城经济圈协同互信意识,在一定程度上消解行政壁垒和地方保护主义;此外,当前成渝地区双城经济圈建设干部协同卷入的奖惩机制不完善,行政治理协同还需大力推进,两地协同仍有较强的地方本位意识。基于此,本研究认为,地方保护现象是长久以来中国实行区域分治所衍生的治理产物,消弭地方保护主义、强化区域一体化思维成为协同瓶颈破局之必然要求,而基于行政尺度视角出发的行政协同治理则可能通过为地区卷入协同建构与区域空间相适配的治理尺度,从而成为区域协同必然的路径选择。并且,协同是基于时间矢量上不断发展变换的过程,地方政府应当通过牢牢把握不同阶段中区域协同“动机—路径—目的”三核心元素以寻求合理的区域协同合作。

目的探讨1例Baraitser-Winter综合征(BWS)患儿的临床特征及遗传学病因。方法选取2022年4月8日就诊于临沂市人民医院儿科的1例BWS患儿作为研究对象。收集患儿的临床资料,采集患儿及其父母的外周血样,对患儿进行全外显子组测序(WES),用Sanger测序对候选变异进行家系验证,并对其进行生物信息学分析。结果患儿为5岁6个月男性,具有BWS的典型临床特征,包括先天性非肌源性睑下垂、拱形眉、宽人中、尖下巴,神经系统症状、小头畸形、发育落后、癫痫、耳聋,头颅MRI示额顶叶脑回粗大、脑白质减少、脑积水。WES显示患儿携带ACTG1基因c.430G>A(p.Asn144Tyr)杂合错义变异,Sanger测序显示其父母未携带相同的变异,提示为新发变异。根据美国医学遗传学与基因组学学会(ACMG)相关指南,该变异被评级为可能致病性(PS2+PM2_Supporting+PP3_Moderate+PP4)。结论ACTG1基因c.430G>A(p.Asn144Tyr)杂合错义变异可能是上述患儿的遗传学病因。新变异的检出丰富了BWS的基因变异谱,为该病的临床诊断与遗传咨询提供了依据。

精浆的功能包括作为精子的运输媒介,以及作为男性向女性生殖组织传递信息的交流手段。它是刺激女性的生殖道向受孕状态转变的重要因素。雌性的生殖道接触到精浆时会发生剧烈的炎症样反应,继而女性多个生殖器官的功能受到调节。在交配过程中可以观察到女性宫颈和子宫腔内对病原体和碎片的清除能力增强。精浆还能够影响子宫内膜上皮细胞和间质细胞的活动,调节子宫的容受性,并通过对免疫细胞系统的调节来建立母体对半异体胚胎的耐受性,为胚胎的有效着床和妊娠提供理想的条件。对于不孕症的治疗,辅助生殖技术已被广泛应用。应用精浆作为一种辅助治疗方法,可能在辅助生殖治疗周期中改善子宫内膜的接受性并调节母体免疫耐受,从而提高胚胎着床及继续发育的潜能。本文对精浆的组成及其对女性生殖道各部分的生理作用进行了综述,并对精浆目前在辅助生殖领域的应用进展进行综述。
