Total glucosides of paeony alleviates cGAS-STING-mediated diseases by blocking the STING-IRF3 interaction

In the realm of autoimmune and inflammatory diseases,the cyclic GMP-AMP synthase(cGAS)stimulator of interferon genes(STING)signaling pathway has been thoroughly investigated and established.Despite this,the clinical approval of drugs targeting the cGAS-STING pathway has been limited.The Total glucosides of paeony(TGP)is highly anti-inflammatory and is commonly used in the treatment of rheumatoid arthritis(RA),emerged as a subject of our study.We found that the TGP markedly reduced the activation of the cGAS-STING signaling pathway,triggered by various cGAS-STING agonists,in mouse bone marrow-derived macrophages(BMDMs)and Tohoku Hospital Pediatrics-1(THP-1)cells.This inhibition was noted alongside the suppression of interferon regulatory factor 3(IRF3)phosphorylation and the expression of interferon-beta(IFN-β),C-X-C motif chemokine ligand 10(CXCL10),and inflammatory mediators such as tumor necrosis factor-alpha(TNF-α)and interleukin-6(IL-6).The mechanism of action appeared to involve the TGP’s attenuation of the STING-IRF3 interaction,without affecting STING oligomerization,thereby inhibiting the activation of downstream signaling pathways.In vivo,the TGP hindered the initiation of the cGAS-STING pathway by the STING agonist dimethylxanthenone-4-acetic acid(DMXAA)and exhibited promising therapeutic effects in a model of acute liver injury induced by lipopolysaccharide(LPS)and D-galactosamine(D-GalN).Our findings underscore the potential of the TGP as an effective inhibitor of the cGAS-STING pathway,offering a new treatment avenue for inflammatory and autoimmune diseases mediated by this pathway.

Joint Optimization of Resource Allocation and Trajectory Based on User Trajectory for UAV-Assisted Backscatter Communication System

The Backscatter communication has gained widespread attention from academia and industry in recent years. In this paper, A method of resource allocation and trajectory optimization is proposed for UAV-assisted backscatter communication based on user trajectory. This paper will establish an optimization problem of jointly optimizing the UAV trajectories, UAV transmission power and BD scheduling based on the large-scale channel state signals estimated in advance of the known user trajectories, taking into account the constraints of BD data and working energy consumption, to maximize the energy efficiency of the system. The problem is a non-convex optimization problem in fractional form, and there is nonlinear coupling between optimization variables.An iterative algorithm is proposed based on Dinkelbach algorithm, block coordinate descent method and continuous convex optimization technology. First, the objective function is converted into a non-fractional programming problem based on Dinkelbach method,and then the block coordinate descent method is used to decompose the original complex problem into three independent sub-problems. Finally, the successive convex approximation method is used to solve the trajectory optimization sub-problem. The simulation results show that the proposed scheme and algorithm have obvious energy efficiency gains compared with the comparison scheme.

Stabilizing iridium sites via interface and reconstruction regulations for water oxidation in alkaline and acidic media

Exploring effective iridium(Ir)-based electrocatalysts with stable iridium centers is highly desirable for oxygen evolution reaction(OER).Herein,we regulated the incorporation manner of Ir in Co_(3)O_(4)support to stabilize the Ir sites for effective OER.When anchored on the surface of Co_(3)O_(4)in the form of Ir(OH)_6 species,the created Ir-OH-Co interface leads to a limited stability and poor acidic OER due to Ir leaching.When doped into Co_(3)O_(4)lattice,the analyses of X-ray absorption spectroscopy,in-situ Raman,and OER measurements show that the partially replacement of Co in Co_(3)O_(4)by Ir atoms inclines to cause strong electronic effect and activate lattice oxygen in the presence of Ir-O-Co interface,and simultaneously master the reconstruction effect to mitigate Ir dissolution,realizing the improved OER activity and stability in alkaline and acidic environments.As a result,Ir_(lat)@Co_(3)O_(4)with Ir loading of 3.67 wt%requires 294±4 mV/285±3 mV and 326±2 mV to deliver 10 mA cm^(-2)in alkaline(0.1 M KOH/1.0 M KOH)and acidic(0.5 M H_(2)SO_(4))solution,respectively,with good stability.

Transformer-Aided Deep Double Dueling Spatial-Temporal Q-Network for Spatial Crowdsourcing Analysis

With the rapid development ofmobile Internet,spatial crowdsourcing has becomemore andmore popular.Spatial crowdsourcing consists of many different types of applications,such as spatial crowd-sensing services.In terms of spatial crowd-sensing,it collects and analyzes traffic sensing data from clients like vehicles and traffic lights to construct intelligent traffic prediction models.Besides collecting sensing data,spatial crowdsourcing also includes spatial delivery services like DiDi and Uber.Appropriate task assignment and worker selection dominate the service quality for spatial crowdsourcing applications.Previous research conducted task assignments via traditional matching approaches or using simple network models.However,advanced mining methods are lacking to explore the relationship between workers,task publishers,and the spatio-temporal attributes in tasks.Therefore,in this paper,we propose a Deep Double Dueling Spatial-temporal Q Network(D3SQN)to adaptively learn the spatialtemporal relationship between task,task publishers,and workers in a dynamic environment to achieve optimal allocation.Specifically,D3SQNis revised through reinforcement learning by adding a spatial-temporal transformer that can estimate the expected state values and action advantages so as to improve the accuracy of task assignments.Extensive experiments are conducted over real data collected fromDiDi and ELM,and the simulation results verify the effectiveness of our proposed models.

Uniaxial Compressive Strength Prediction for Rock Material in Deep Mine Using Boosting-Based Machine Learning Methods and Optimization Algorithms

Traditional laboratory tests for measuring rock uniaxial compressive strength(UCS)are tedious and timeconsuming.There is a pressing need for more effective methods to determine rock UCS,especially in deep mining environments under high in-situ stress.Thus,this study aims to develop an advanced model for predicting the UCS of rockmaterial in deepmining environments by combining three boosting-basedmachine learning methods with four optimization algorithms.For this purpose,the Lead-Zinc mine in Southwest China is considered as the case study.Rock density,P-wave velocity,and point load strength index are used as input variables,and UCS is regarded as the output.Subsequently,twelve hybrid predictive models are obtained.Root mean square error(RMSE),mean absolute error(MAE),coefficient of determination(R2),and the proportion of the mean absolute percentage error less than 20%(A-20)are selected as the evaluation metrics.Experimental results showed that the hybridmodel consisting of the extreme gradient boostingmethod and the artificial bee colony algorithm(XGBoost-ABC)achieved satisfactory results on the training dataset and exhibited the best generalization performance on the testing dataset.The values of R2,A-20,RMSE,and MAE on the training dataset are 0.98,1.0,3.11 MPa,and 2.23MPa,respectively.The highest values of R2 and A-20(0.93 and 0.96),and the smallest RMSE and MAE values of 4.78 MPa and 3.76MPa,are observed on the testing dataset.The proposed hybrid model can be considered a reliable and effective method for predicting rock UCS in deep mines.

Numerical simulation of microwave-induced cracking and melting of granite based on mineral microscopic models

This study introduces a coupled electromagnetic–thermal–mechanical model to reveal the mechanisms of microcracking and mineral melting of polymineralic rocks under microwave radiation.Experimental tests validate the rationality of the proposed model.Embedding microscopic mineral sections into the granite model for simulation shows that uneven temperature gradients create distinct molten,porous,and nonmolten zones on the fracture surface.Moreover,the varying thermal expansion coefficients and Young's moduli among the minerals induce significant thermal stress at the mineral boundaries.Quartz and biotite with higher thermal expansion coefficients are subjected to compression,whereas plagioclase with smaller coefficients experiences tensile stress.In the molten zone,quartz undergoes transgranular cracking due to theα–βphase transition.The local high temperatures also induce melting phase transitions in biotite and feldspar.This numerical study provides new insights into the distribution of thermal stress and mineral phase changes in rocks under microwave irradiation.

Thiol-mediated transportation pathway:an approach for improving tumor penetration of nanomedicines in vivo

Abnormal tumor microenvironment imposes barriers to tumor penetration of nanomedicine,which remains a major challenge for effective anti-tumor.Herein,we present disulfide-based nanoparticles that actively penetrate deep tumors in vivo through a thiol-mediated transportation pathway.To achieve active tumor accumulation in vivo,disulfide-based nanoparticles are modified with folic acid units(FA-DBNPs).It is gratifying that FA-DBNPs still enter cells via the thiol-mediated pathway,which facilitates transcellular transportation and tumor penetration both in vitro and in vivo.Besides,FA-DBNPs exhibit GSH concentration-dependent depolymerization characterization,indicating that the GSH level in tumor tissues regulates the penetration depth of FA-DBNPs.Benefiting from these advantages,FA-DBNPs showed potent anti-tumor activity in mouse models,leading to the significant regression of tumors.The current study lays a foundation that thiol-mediated transportation is a promising approach in nanomedicine design for solid tumor therapy.

Demagnetization Analysis and Velocity Tracking Control of In-wheel Motor

The error caused by irreversible demagnetization damages the accurate velocity tracking of an in-wheel motor in a mobile robot.A current feedforward vector control system based on ESO is proposed to compensate it for the demagnetization motor.A demagnetization mathematical model is established to describe a permanent magnet synchronous motor,which took the change of permanent magnet flux linkage parameters as a factor to count the demagnetization error in velocity tracking.The uncertain disturbance estimation model of the control system is built based on ESO,which eliminates the system error by the feedforward current compensation.It is compared with the vector control method in terms of control accuracy.The simulation results show that the current feedforward vector control method based on ESO reduces the velocity tracking error greatly in conditions of motor demagnetization less than 30%.It is effective to improve the operation accuracy of the mobile robot.

Distribution law and susceptibility of geohazards across a gradient belt of the Western Sichuan Plateau

Across a gradient belt of the Western Sichuan Plateau,geohazards have seriously limited economic and social development.According to incomplete statistics,15,673 geohazards have been recorded in the study area.In order to mitigate the threat of geohazards to human engineering activities in the region,an overall understanding of the distribution pattern of geohazards and susceptibility assessment are necessary.In this paper,a gradient belt of the Western Sichuan Plateau and its zoning criteria were defined.Subsequently,on the basis of relief amplitude,distance to faults,rainfall,and human activities,three indicators of endogenic process were introduced:Bouguer gravity anomaly gradient,vertical deformation gradient,and horizontal deformation gradient.Thereafter,the distribution patterns of geohazards were investigated through mathematical statistics and ArcGIS software.By randomly selecting 10,449 hazards,a geohazard susceptibility map was generated using the Information Value(IV)model.Finally,the IV model was validated against 5224 hazards using the Area Under Curve(AUC)method.The results show that 47.6%of the geohazards were distributed in the zone of steep slope.Geohazards showed strong responses to distance to faults,human activities,and annual rainfall.The distribution of geohazards in the gradient belt of the Western Sichuan Plateau is more sensitive to vertical internal dynamics factors(such as vertical deformation gradient and Bouguer gravity anomaly gradient)without any apparent sensitivity to horizontal internal dynamics factors.The areas of high and very-high risk account for up to 32.22%,mainly distributed in the Longmenshan and Anning River faults.According to the AUC plot,the success rate of the IV model for generating the susceptibility map is 76%.This susceptibility map and geohazard distribution pattern can provide a reference for geological disaster monitoring,preparation of post-disaster emergency measures,and town planning.

The development of the fifth-generation(5G)mobile communication systems has entered the commercialization stage.5G has a high data rate,low latency,and high reliability that can meet the basic demands of most industries and daily life,such as the Internet of Things(IoT),intelligent transportation systems,positioning,and navigation.The continuous progress and development of society have aroused wide concern.Positioning accuracy is the core demand for the applications,especially in complex environments such as airports,warehouses,supermarkets,and basements.However,many factors also affect the accuracy of positioning in those environments,for example,multipath effects,non-line-of-sight,and clock synchronization errors.This paper provides a comprehensive review of the existing works about positioning for the future wireless network and discusses its key techniques and algorithms,as well as the current development and future directions.We first outline the current traditional positioning technologies and algorithms,which are discussed and analyzed with the relevant literature.In addition,we also discuss application scenarios for wireless localization.By comparing different positioning systems,the challenges and future development directions of existing wireless positioning systems are prospected.
