In recent years,the Internet of Things(IoT)has gradually developed applications such as collecting sensory data and building intelligent services,which has led to an explosion in mobile data ***,with the rapid development of artificial intelligence,semantic c.m.unication has attracted great attention as a new c.m.unication ***,for IoT devices,however,processing image information efficiently in real time is an essential task for the rapid transmission of semantic *** the increase of model parameters in deep learning methods,the model inference time in sensor devices continues to *** contrast,the Pulse coupled Neural Network(PcNN)has fewer parameters,making it more suitable for processing real-time scene tasks such as image segmentation,which lays the foundation for real-time,effective,and accurate image ***,the parameters of PcNN are determined by trial and error,which limits its *** overc.m. this limitation,an Improved Pulse coupled Neural Networks(IPcNN)model is proposed in this *** IPcNN constructs the connection between the static properties of the input image and the dynamic properties of the neurons,and all its parameters are set adaptively,which avoids the inconvenience of manual setting in traditional methods and improves the adaptability of parameters to different types of *** segmentation results demonstrate the validity and efficiency of the proposed self-adaptive parameter setting method of IPcNN on the gray images and natural images from the matlab and Berkeley Segmentation *** IPcNN method achieves a better segmentation result without training,providing a new solution for the real-time transmission of image semantic information.

A biosynthetic gene cluster for the bioactive fungal sesterterpenoids variecolin(1)and variecolactone(2)was identified in Aspergillus aculeatus ATcc *** production of 1 and 2 was achieved in Aspergillus oryzae by expressing the sesterterpene synthase VrcA and the cytochrome P450 ***,the replac.m.nt of VrcB with homologous P450s from other fungal terpenoid pathways yielded three new variecolin analogues(5-7).Analysis of the c.m.ounds’anticancer activity in vitro and in vivo revealed that although 5 and 1 had c.m.arable activities,5 was associated with significantly reduced toxic side effects in cancer-bearing mice,indicating its potentially broader therapeutic *** study describes the first tests of variecolin and its analogues in animals and demonstrates the utility of synthetic biology for creating molecules with improved biological activities.

The global antibiotic resistance crisis necessitates urgent solutions. One innovative approach involves potentiating antibiotics and non-antibiotic drugs with adjuvants or boosters. A major drawback of these membrane-active boosters is their limited bioc.m.atibility, as they struggle to differentiate between prokaryotic and eukaryotic.m.mbranes. This study reports the chemical biology investigation of a dual-action oligoamidine(OA1) booster with a glutathione-triggered dec.m.osition mechanism. OA1, when c.m.ined with other antimicrobial molecules, exhibits a triple-targeting mechanism including cell membrane disruption, DNA targeting, and intracellular enzyme inhibition. This multi-targeting mechanism not only enhances the in vitro and in vivo eradication of antibiotic-resistant “ESKAPE” pathogens, but also suppresses the development of bacterial resistance. Furthermore, OA1 maintains its activity in bacterial cells by creating an oxidative environment, while it quickly dec.m.oses in mammalian cells due to high glutathione levels. These mechanistic insights and design principles may provide a feasible approach to develop novel antimicrobial agents and effective anti-resistance c.m.ination therapies.

As the most c.m.on internal modification of mRNA,Ne-methyladenosine(m^(6)A)and its regulators modulate gene expression and play critical roles in various biological and patholog-ical processes including *** was reported previously that m^(6)A methyltransferase(writer),methyltransferase-like 3(mETTL3)adds m^(6)A in primary microRNAs(pri-miRNAs)and fa-cilitates its processing into precursor miRNAs(pre-miRNAs).However,it is unknown whether m^(6)A modification also plays a role in the maturation process of pre-miRNAs and(if so)whether such a function contributes to ***,we found that YTHDF2 is aberrantly overexpressed in acute myeloid leukemia(AmL)patients,especially in relapsed patients,and plays an onco-genic role in ***,YTHDF2 promotes expression of miR-126-3p(also known as miR-126,as it is the main product of precursor miR-126(pre-miR-126)),a miRNA that was reported as an onc.m.RNA in AmL,through facilitating the processing of pre-miR-126 into mature ***,YTHDF2 recognizes m^(6)A modification in pre-miR-126 and recruits AGO2,a regulator of pre-miRNA processing,to promote the maturation of ***2 posi-tively and negatively correlates with miR-126 and miR-126's downstream target genes,respec-tively,in AmL patients,and forced expression of miR-126 could largely rescue YTHDF2/Ythdf2 depletion-mediated suppression on AmL cell growth/proliferation and leukemogenesis,indi-cating that miR-126 is a functionally important target of YTHDF2 in ***,our studies not only reveal a previously unappreciated YTHDF2/miR-126 axis in AmL and highlight the ther-apeutic potential of targeting this axis for AmL treatment,but also suggest that m^(6)A plays a role in pre-miRNA processing that contributes to tumorigenesis.
