SARS-CoV-2 spike protein binds to bacterial lipopolysaccharide and boosts proinflammatory activity

There is a link between high lipopolysaccharide(LPS)levels in the blood and the metabolic syndrome,and metabolic syndrome predisposes patients to severe ***,we define an interaction between SARS-CoV-2 spike(S)protein and LPS,leading to aggravated inflammation in vitro and in *** gel electrophoresis demonstrated that SARS-CoV-2 S protein binds to *** thermophoresis yielded a of〜47 nM for the *** modeling and all-atom molecular dynamics simulations further substantiated the experimental results,identifying a main LPS-binding site in SARS-CoV-2 S protein.S protein,when combined with low levels of LPS,boosted nuclear factor-kappa B(NF-k B)activation in monocytic THP-1 cells and cytokine responses in human blood and peripheral blood mononuclear cells,*** in vitro inflammatory response was further validated by employing NF-kB reporter mice and in vivo *** light scattering,transmission electron microscopy,and LPS-FITC analyses demonstrated that S protein modulated the aggregation state of LPS,providing a molecular explanation for the observed boosting *** together,our results provide an interesting molecular link between excessive inflammation during infection with SARS-CoV-2 and comorbidities involving increased levels of bacterial endotoxins.

A Randomized,Double-Blind,Placebo-Controlled Pilot Clinical Study on ColdZyme^(■) Mouth Spray against Rhinovirus-Induced Common Cold

Common colds incur significant costs in terms of sick leave and personal discomfort for affected individuals. This study investigated the performance of ColdZyme? Mouth Spray (ColdZyme), a protective barrier against common cold, in rhinovirus-inoculated healthy volunteers. This randomized, doubleblind, placebo-controlled pilot study was conducted on 46 healthy volunteers inoculated with rhinovirus 16 via the nose. Subjects self-administered ColdZyme or placebo 6 times daily for 11 days. Symptoms were recorded daily in a diary. Rhinovirus 16 in nasal and oropharyngeal samples at days 0, 3, 4, 6, 7 and 10 were quantified by RT-qPCR. The primary outcome measure was the reduction in viral load in oropharyngeal samples. Rhinovirus 16 was only detected in 35 out of 46 inoculated subjects. Exploratory analysis measuring the total viral load (i.e., area under the curve (AUC)) for days 3 - 10 in successfully inoculated subjects found that ColdZyme treatment resulted in a lower total viral load in the oropharynx (p = 0.023). In subjects who experienced symptomatic common cold, irrespectively, if virus were detected, treatment with ColdZyme resulted in a reduction in the number of days with common cold symptoms from 6.5 to 3.0 days (p = 0.014) in comparison to placebo. ColdZyme reduced virus infection in the oropharynx and reduced the number of days with common cold symptoms and highlights the possible importance of the oropharynx in common cold infections. Suitable outcome measures for a feasible study on ColdZyme are total viral load in the oropharynx in subjects having detectable virus present in nasal or oropharyngeal samples.

Paleoproterozoic,High-Metamorphic,Metasedimentary Units of Siberian Craton

Sensitive, high-resolution ion microprobe zircon U-Pb ages of Paleoproterozoic, high-grade, metasedimentary rocks from the south-western part of the Siberian Craton are reported. Early Precambrian, high-grade complexes, including garnet-biotite, hypersthene-biotite, and cordierite- bearing gneisses compose the Irkut terrane of the Sharyzhalgay Uplift. Protoliths of studied gneisses correspond to terrigenous sediments, ranging from greywacke to shale. The paragneiss model Nd ages of 2.4-3.1 Ga indicate Archean-to-Paleoproterozoic source provinces. Zircons from gneisses show corerim textures in cathodoluminescence （CL） image. Round or irregular shaped cores indicate detrital origin. Structureless rims with low Th/U are metamorphic in origin. The three age groups of detrital cores are：〉2.7, -2.3, and 1.95-2 Ga. The ages of metamorphic rims range from 1.86 to 1.85 Ga; therefore, the sediments were deposited between 1.95 and 1.86 Ga and derived from Archean and Paleoproterozoic source rocks. It should be noted that Paleoproterozoic metasedimentary rocks of the Irkut Block are not unique. High-grade metaterrigenous sediments, with model Nd ages ranging from 2.3 to 2.5 Ga, are widely distributed within the Aldan and Anabar Shields of the Siberian Craton. The same situation is observed in the North China Craton, where metasedimentary rocks contain detrital igneous zircon grains with ages ranging from 3 to 2.1 Ga （Wan et al., 2006）. All of these sedimentary units were subjected to Late Paleoproterozoic metamorphism. In the Siberian Craton, the Paleoproterozoic sedimentary deposits are possibly marked passive margins of the Early Precambrian crustal blocks, and their high-grade metamorphism was related to the consolidation of the Siberian Craton.

Self-assembly of peptide-based nanostructures： Synthesis and biological activity

Peptide-based nanostructures have received much attention in the field of drug targeting. In fact, peptides have many advantages such as simplicity of the structure, biocompatibility, and chemical diversity. Moreover, some peptides, which are called cell-penetrating peptides, can cross cellular membranes and carry small molecules, small interfering RNA, or viruses inside live cells. These molecules are often covalently or noncovalently linked to cargoes, thus forming amphiphilic conjugates that can self-assemble. Supramolecular nanostructures formed from peptides are used in nanomedicine as a carrier to protect a drug and to target cancer cells. This review explores aliphatic-chain-conjugated peptides and drug-conjugated peptides that can self-assemble. Special emphasis is placed on the synthesis procedure, nanostructure formation, and biological activity.

Joint effects of acetochlor and urea on germinating characteristics of crop seeds

In order to evaluate ecological risk of agrochemicals in common use, joint toxic effects of acetochlor and urea on germinating characteristics of Chinese cabbage (Brassica Pe-kinensis Rupr) seeds were investigated using the water-culture method and the soil-culture method. The results indicated(that excessive application of acetochlor and urea, when the concentrations were higher than 31.3 mg · kg-1 for acetochlor and 500 mg · kg-1 for urea, had strong inhibitory effects on the rate of seed germination, root elongation and hypocotyl length of Chinese cabbage. The inhibitory rate of the germinating characteristics of Chinese cabbage seeds was significantly increased with an increase in the concentration of acetochlor or urea. The two agrochemicals in water had a stronger toxicity than these in the soil at the same concentration. Among the three indexes, hypocotyl length was the most sensitive to the toxicity of acetochlor and urea.

Phenotype manipulation influences microhabitat choice in pygmy grasshoppers

The matching habitat choice hypothesis posits that individuals actively choose those microhabitats that best match their specific phenotype to maximize fitness. Despite the profound implications, matching habitat choice has not been unequivo- cally demonstrated. We conducted two experiments to examine the impact of pigmentation pattern in the color polymorphic pygmy grasshopper Tetrix subulata on habitat choice in a laboratory thermal mosaic arena. We found no behavioral differences in the thermal mosaic among pygmy grasshoppers belonging to either pale, intermediate or dark natural color morphs. However, af- ter manipulating the grasshoppers＇ phenotype, the utilization through time of warmer and colder parts of the arena was different for black-painted and white-painted individuals. White-painted individuals used warmer parts of the arena, at least during the ini- tial stage of the experiment. We conclude that microhabitat choice represents a form of behavioural plasticity. Thus, even if the choice itself is flexible and not genetically determined, it can still lead to spatial genetic structure in the population because the phenotypes themselves may be genetically mediated [Current Zoology 58 （3）： 392--400, 2012].

Tastes of Revolution, Change and Love： Codes of Consumption in Fiction from New China

This paper analyses Socialist Realist novels from the early years of the People＇s Republic of China （PRC）, focusing on scenes of food and drink consumption. While these scenes may appear marginal at first glance, the analysis demonstrates how food and its consumption function as codes to normative values. I am therefore proposing a reading of these texts based on the model of intertextuality （Julia Kristeva） and on an anthropological model on （food） consumption （Mary Douglas）, advocating that acts of consumption reveal social hierarchies and the position of the individual therein. These fictional scenes of everyday activities construct fictional characters as heroes or villains. Given the normative value of this officially endorsed literature, these scenes at the same time prescribe （and, likewise, proscribe） certain behavior to their readers. On another level, however, these codes also convey information that could not be openly spelled out at the time, as when the sharing of food is the only way in which two fictional characters can express their love. Simple food can thus be the source of entertainment, enjoyment, suspense, and even nostalgia for contemporary readers, which, in turn, may be one of the reasons for the lasting popularity of the codes described and of a number of the texts presented in the analysis.

Improving risk reduction potential of weather index insurance by spatially downscaling gridded climate data-a machine learning approach

Open-access gridded climate products have been suggested as a potential source of data for index insurance design and operation in data-limited ***,index insurance requires climate data with long historical records,global geographical coverage and fine spatial resolution at the same time,which is nearly impossible to satisfy,especially with open-access *** this paper,we spatially downscaled gridded climate data(precipitation,temperature,and soil moisture)in coarse spatial resolution with globally available longterm historical records to finer spatial resolution,using satellite-based data and machine learning *** then investigated the effect of index insurance contracts based on downscaled climate data for hedging spring wheat *** study employed countylevel spring wheat yield data between 1982 and 2018 from 56 counties overall in Kazakhstan and *** results showed that in the majority of cases(70%),hedging effectiveness of index insurances increases when climate data is spatially downscaled with a machine learning *** improvements are statistically significant(p≤0.05).Among other climate data,more improvements in hedging effectiveness were observed when the insurance design was based on downscaled temperature and precipitation ***,this study highlights the reasonability and benefits of downscaling climate data for insurance design and operation.
