Signal transducer and activator of transcription factor 3(STAT3)is a transcription factor that regulates various cellular processes such as proliferation,survival and angiogenesis in cancer *** the present study,we investigated the mechanisms whereby isothiocyanate sulforaphane(SFN)suppresses STAT3 activation in DU145 prostate cancer *** significantly inhibited SFN-inhibited STAT3 phosphorylation at Tyr705 as well as the deoxyribonucleic acid(DNA)binding capability in electrophoresis mobility shift assay(EMSA)in time-and concentration-dependent *** also abrogated the Janus activated kinase 2(JAK2)*** addition,SFN down-regulated STAT3-related gene products including Bcl-2,Bcl-xL,and cyclin D1 and vascular endothelial growth factor(VEGF),and inhibited the proliferation and induced ***,SFN mediated reactive oxygen species(ROS)production at the early ***,these results demonstrate that ROS generation may be involved in the inhibition of JAK2/STAT3 activation and apoptosis in DU145 cells.

Sojucktang (SJT) has long been used for the treatment of endometrial diseases in Korea. However, the mechanisms responsible for the SJT-induced apoptosis in endometrial cancer cells remain unclear. In the present study, SJT was demonstrated to show cytotoxic effect and induce apoptotic cell death via mitochondrial regulation in KLE endometrial cancer cells. Linderae Radix, Glycyrrhizae Radix, Zedoariae Rhizoma, Trogopterorum Faeces and Agelicae Gigantis Radix were found to be the potent constituent herbs of SJT to significantly decrease the viability of KLE cells by a tetra zolium salt (XTT) assay. Apoptotic bodies were observed in SJT-treated KLE cells by 4′-6-diamidino-2-phenylindole (DAPI) and TdT-mediated-dUTP nick-end labeling (TUNEL) assay. SJT also increased sub-G1 DNA contents of the cell cycle undergoing apoptosis in a dose-dependent manner. Furthermore, it was observed that SJT activated caspase-3 and cleaved poly (ADP-ribose) polymerase (PARP), and decreased mitochondrial membrane potential in a dose-dependent manner. Taken together, this study shows that SJT exerts anti-tumor activity against KLE endometrial cancer cells via mitochondrial dependent apoptosis induction.

Oriental herbal medicines have been widely used for the prevention or treatment of various diseases including cancer in Asia. However, to prove their chemo preventive efficacies in modern times, scientific evidence for those herbal medicines is required. Thus, in the present study, an effective herbal cocktail Bojungbangdocktang (BJBDT) was investigated to elucidate antiangiogenic mechanism in vitro and in vivo. BJBDT significantly inhibited vascular endothelial growth factor (VEGF) induced proliferation in HUVECs at nontoxic concentrations, despite weak cytotoxicity against human umbilical vein endothelial cells (HUVECs). BJBDT also significantly suppressed VEGF-induced migration and tube formation of HUVECs. Furthermore, BJBDT treatment resulted in pale color and low hemoglobin level in Matrigel plugs, as well as dark red color and high hemoglobin level in untreated control. Interestingly, BJBDT specifically inhibited the binding of VEGF to vascular endothelial growth factor receptor 2 (VEGFR2), but not VEGFR1. In addition, friedelin, formononetin, ginsenoside Rb1, naringin, atractyloside, diosgenin, and allantonin were identified from BJBDT by high-performance liquid chromatography (HPLC) analysis as a quality of control. Taken together, these results suggest that BJBDT is a potent angiogenesis inhibitor blocking the VEGF/VEGFR2 signaling pathway in HUVECs.

Elucidating the complex dynamic cellular organization in the hypothalamus is critical for understanding its role in coordinating fundamental body functions. Over the past decade, single-cell and spatial omics technologies have significantly evolved, overcoming initial technical challenges in capturing and analyzing individual cells. These high-throughput omics technologies now offer a remarkable opportunity to comprehend the complex spatiotemporal patterns of transcriptional diversity and cell-type characteristics across the entire hypothalamus. Current single-cell and single-nucleus RNA sequencing methods comprehensively quantify gene expression by exploring distinct phenotypes across various subregions of the hypothalamus. However, single-cell/single-nucleus RNA sequencing requires isolating the cell/nuclei from the tissue, potentially resulting in the loss of spatial information concerning neuronal networks. Spatial transcriptomics methods, by bypassing the cell dissociation, can elucidate the intricate spatial organization of neural networks through their imaging and sequencing technologies. In this review, we highlight the applicative value of single-cell and spatial transcriptomics in exploring the complex molecular-genetic diversity of hypothalamic cell types, driven by recent high-throughput achievements.

To emulate the functionality of the human retina and achieve a neuromorphic visual system,the development of a photonic synapse capable of multispectral color discrimination is of paramount ***,attaining robust color discrimination across a wide intensity range,even irrespective of medium limitations in the channel layer,poses a significant ***,we propose an approach that can bestow the color-discriminating synaptic functionality upon a three-terminal transistor flash memory even with enhanced discriminating *** incorporating the strong induced dipole moment effect at the excitation,modulated by the wavelength of the incident light,into the floating gate,we achieve outstanding RGB color-discriminating synaptic functionality within a remarkable intensity range spanning from 0.05 to 40 mW cm^(-2).This approach is not restricted to a specific medium in the channel layer,thereby enhancing its *** effectiveness of this color-discriminating synaptic functionality is demonstrated through visual pre-processing of a photonic synapse array,involving the differentiation of RGB channels and the enhancement of image contrast with noise ***,a convolutional neural network can achieve an impressive inference accuracy of over 94%for Canadian-Institute-For-Advanced-Research-10 colorful image recognition task after the *** proposed approach offers a promising solution for achieving robust and versatile RGB color discrimination in photonic synapses,enabling significant advancements in artificial visual systems.

Parkinson's disease has long been considered a disorder that primarily affects the brain,as it is defined by the dopaminergic neurodegeneration in the substantia nigra and the brain accumulation of Lewy bodies containingα-synuclein *** recent decades,however,accumulating research has revealed that Parkinson's disease also involves the gut and uncovered an intimate and important bidirectional link between the brain and the gut,called the“gut–brain axis.”Numerous clinical studies demonstrate that gut dysfunction frequently precedes motor symptoms in Parkinson's disease patients,with findings including impaired intestinal permeability,heightened inflammation,and distinct gut microbiome profiles and ***,α-synuclein deposition has been consistently observed in the gut of Parkinson's disease patients,suggesting a potential role in disease ***,individuals with vagotomy have a reduced Parkinson's disease *** these observations,researchers have hypothesized thatα-synuclein accumulation may initiate in the gut and subsequently propagate to the central dopaminergic neurons through the gut–brain axis,leading to Parkinson's *** review comprehensively examines the gut's involvement in Parkinson's disease,focusing on the concept of a gut-origin for the *** also examine the interplay between altered gut-related factors and the accumulation of pathologicalα-synuclein in the gut of Parkinson's disease *** the accessibility of the gut to both dietary and pharmacological interventions,targeting gut-localizedα-synuclein represents a promising avenue for developing effective Parkinson's disease therapies.

This study presents a machine learning-based method for predicting fragment velocity distribution in warhead fragmentation under explosive loading *** fragment resultant velocities are correlated with key design parameters including casing dimensions and detonation *** paper details the finite element analysis for fragmentation,the characterizations of the dynamic hardening and fracture models,the generation of comprehensive datasets,and the training of the ANN *** results show the influence of casing dimensions on fragment velocity distributions,with the tendencies indicating increased resultant velocity with reduced thickness,increased length and *** model's predictive capability is demonstrated through the accurate predictions for both training and testing datasets,showing its potential for the real-time prediction of fragmentation performance.

Advanced OER/HER electrocatalytic alternatives are crucial for the wide adaptation of green hydrogen ***,Ru/NiMnB spherical cluster pillar(SCP),denoted as Ru/NiMnB,is synthesized using a combination of electro-deposition and hydrothermal *** investigation of Ru doping in the NiMnB matrix revealed significant improvements in electrocatalytic *** Ru/NiMnB SCPs demonstrate superior OER/HER activity with low overpotentials of 150 and 103 mV at 50mA/cm^(2)in 1 M KOH,making them highly competitive with state-of-the-art ***,the Ru/NiMnB SCPs exhibit a low 2-E cell voltage of 2.80 V at ultra-high current density of 2,000 m A/cm^(2)in 1 M KOH,outperforming the standard benchmark electrodes of RuO_(2)||Pt/C,thereby positioning Ru/NiMnB as one of the best bifunctional *** SCPs exhibit exceptional high-current characteristics,stability and corrosion resistance,as evidenced by continuous operation at 1,000 mA/cm^(2)high-current density for over 150 h in 6 M KOH at elevated temperatures under harsh industrial *** a small amount of Ru incorporation significantly enhances the electrocatalytic performances of NiMnB,attributed to increased active sites and improved intrinsic properties such as conductivity,adsorption/desorption capability and reaction ***,Ru/NiMnB SCPs present a promising bi-functional electrode concept for efficient green H_(2)production.

Polystyrene nanoparticles pose significant toxicological risks to aquatic ecosystems, yet their impact on zebrafish(Danio rerio) embryonic development, particularly erythropoiesis, remains underexplored. This study used single-cell RNA sequencing to comprehensively evaluate the effects of polystyrene nanoparticle exposure on erythropoiesis in zebrafish embryos. In vivo validation experiments corroborated the transcriptomic findings,revealing that polystyrene nanoparticle exposure disrupted erythrocyte differentiation, as evidenced by the decrease in mature erythrocytes and concomitant increase in immature erythrocytes. Additionally, impaired heme synthesis further contributed to the diminished erythrocyte *** findings underscore the toxic effects of polystyrene nanoparticles on hematopoietic processes, highlighting their potential to compromise organismal health in aquatic environments.
