The bones are the m.st com.on sites of breast cancer m.tastasis. Upon arrival within the bone m.croenvironm.nt, breast cancer cells coordinate the activities of strom.l cells, resulting in an increase in osteoclast activity and bone m.trix degradation. In late stages of bone m.tastasis, breast cancer cells induce apoptosis in osteoblasts, which further exacerbates bone loss. However, in early stages, breast cancer cells induce osteoblasts to secrete inflam.atory cytokines purported to drive tum.r progression. To m.re thoroughly evaluate the role of osteoblasts in early stages of breast cancer m.tastasis to the bones, we used green fluorescent protein-labeled hum.n breast cancer cell lines m.A-m.-231 and m.A-m.-435, which both induce osteolysis after intra-fem.ral injection in athym.c m.ce, and the m.rine pre-osteoblastic cell line m.3T3-E1 to m.dulate osteoblast populations at the sites of breast cancer m.tastasis. Breast cancer cells were injected directly into the fem.r with or without equal num.ers of m.3T3-E1 cells. Tum.rs grew significantly larger when co-injected with breast cancer cells and m.3T3-E1 cells than injected with breast cancer cells alone. Osteolysis was induced in both groups, indicating that m.3T3-E1 cells did not block the ability of breast cancer cells to cause bone destruction. m.3T3-E1 cells prom.ted tum.r growth out of the bone into the extraosseous strom.. These data suggest that breast cancer cells and osteoblasts com.unicate during early stages of bone m.tastasis and prom.te tum.r growth.

Purpose: Physical activity(PA) and fundam.ntal m.tor skills are im.ortant com.onents of current and future trajectories of health in young children. This study exam.ned the effects of a 5-week m.tor skill intervention on preschoolers' m.tor skill com.etence and their PA behaviors while participating in the m.tor skill intervention or outdoor free play(recess).m.thods: A total of 102 preschoolers served as participants and were part of a m.tor skill intervention group(n = 64) or a control/outdoor free play group(n = 38). Children's m.tor skills were assessed before and after the intervention using the Test of Gross m.tor Developm.nt-3 rd edition. Background： Our understanding of post-transcriptional gene regulation has increased exponentially with the developm.nt of robust m.thods to define protein-RNA interactions across the transcriptom.. In this review, we highlight the evolution and successful applications of crosslinking and im.unoprecipitation （CLIP） m.thods to interrogate protein-RNA interactions in a transcriptom.-wide m.nner. Results： Here, we survey the vast array of in vitro and in vivo approaches used to identify protein-RNA interactions, including but not lim.ted to electrophoretic m.bility shift assays, system.tic evolution of ligands by exponential enrichm.nt （SELEX）, and RIP-seq. We particularly em.hasize the advancem.nt of CLIP technologies, and detail protocol im.rovem.nts and com.utational tools used to analyze the output data. Im.ortantly, we discuss how profiling protein-RNA interactions can delineate biological functions including splicing regulation, alternative polyadenylation, cytoplasm.c decay substrates, and m.RNA targets. Conclusions： In sum.ary, this review sum.arizes the benefits of characterizing RNA-protcin networks to further understand the regulation of gene expression and disease pathogenesis. Our review com.ents on how future CLIP technologies can be adapted to address outstanding questions related to m.ny aspects of RNA m.tabolism.and further advance our understanding of RNA biology.

Characteristics of Children Referred for Drainage of Sub-cutaneous Soft Tissue Infections

Purpose: Severe skin and soft tissue infections in children are a com.on cause of hospital adm.ssion. Surgical drainage is often required for definitive therapy. Our aim.was to review com.on characteristics of children requiring surgical incision and drainage (I&D) for subcutaneous abscesses. m.thods: IRB approval was obtained for a retrospective chart review of children requiring I&D of subcutaneous abscesses between July 2006 and June 2011. Data points included patient dem.graphics, abscess location, surgical procedure, m.crobiology cultures, and hospital length of stay. Results: Surgical I&D were perform.d on 1042 children, and 781 patients (75%) were adm.tted to the surgical service. The average age was 3.4 years. 605 patients (58%) were fem.le. 637 abscesses (61%) were located in the groin/buttock/perineum.area. m.thicillin-resistant Staphylococcus aureus (m.SA) was identified in 70% of the culture specim.ns. The average length of stay was 2.8 days. The rate of recidivism.was 10.9%. Conclusions: Soft tissue infections requiring hospital adm.ssion and I&D are com.on in the pediatric population. The vast m.jority of these are due to m.SA infections. Infections requiring drainage m.st frequently occurred in the diaper area of girls less than 3 years old. A significant num.er of children have recurrent skin infections and represent an im.ortant patient subpopulation.

Low Carbohydrate Diets in Type 2 Diabetes—A Translational Study

Although intensive interventions with low carbohydrate diets com.ared with higher carbohydrate diets can reduce HbA1c in people with type 2 diabetes, it is not clear if sim.le advice to m.ke m.dest reductions in carbohydrate is effective in clinical practice. Forty-three people with type 2 diabetes and poor control (HbA1c > 7.5%) were random.zed to receive 2 short education sessions over 6 m.nths with a non-dietitian researcher on how to reduce carbohydrate intake by about 25% or to 2 control sessions in which the Australian Guide to Healthy Eating was provided. Hba1c and fasting glucose and lipids were m.asured at baseline and 3 m.nths and 6 m.nths. 33 volunteers attended a baseline visit;27 com.leted 3 m.nths and 24 6 m.nths. HbA1c was reduced by 0.6% - 0.7% in the low carbohydrate diet group com.ared with the control group (P = 0.1). Fasting glucose was reduced by 2.3 m.ol/L com.ared with the control group at 3 m.nths (P m.nths were related to baseline HbA1c in the intervention group only. Although we have obtained suggestive evidence that a low carbohydrate diet can be successfully im.lem.nted in norm.l practice without professional help, our results are lim.ted by low participant num.ers and further studies are required.

Dear Editor,Prostate cancer is the second m.st com.on cancer am.ng m.n worldwide and leading cancer in incidence am.ng m.n in the United States(US).In 2018,the US had over 190,000 new prostate cancer cases,accounting for alm.st 1 in 5 new m.le cancer diagnoses[1].A recent review of dietary factors in relation to prostate cancer risk did not find evidence regarding nut consum.tion as neither a risk nor protective factor,though it has been hypothesized to be associated with a decreased cancer risk through m.lti-ple m.chanism.[2].
