The recent emergence and application of engineered endonucleases have led to the development of genome editing tools capable of rapidly implementing various targeted genome editions in a wide range of ***,these novel tools have become easier to use and have resulted in a great increase of *** gene knockout（KO） or knockin（KI） animal models are relatively easy to achieve,there is a bottleneck in the detection and analysis of these *** several methods exist to detect these targeted mutations,we developed a heteroduplex mobility assay on an automated microfluidic capillary electrophoresis system named hMA-CE in order to accelerate the genotyping *** hMA-CE method uses a simple PCR amplification of genomic DNA（gDNA） followed by an automated capillary electrophoresis step which reveals a heteroduplexes（hD） signature for each *** allows efficient discrimination of wild-type and genome-edited animals down to the single base pair level.

Background In children, surveys on Staphylococcus aureus have focused on specific infections, situations or strains but no study has so far given an overview on S. aureus isolation without any selection. here, we describe the overall bacteriologi-cal and clinical characteristics of S. aureus isolation in children, with a special focus on isolates harbouringtst,sea, and/or luk-PV genes, respectively, encoding the three clinically relevant toxins: toxic shock syndrome toxin-1, enterotoxin A and Panton–Valentine leukocidin. Methods Data associated with S. aureus isolation were reviewed: isolation site, infection status,tst,sea andluk-PV genes, antimicrobial susceptibility pattern,agr typing. Results Three hundred and seventy-seven isolates retrieved from 328 children during S. aureus infection (55.2%) or colo-nisation (44.8%) were ***,sea and luk-PV genes were amplified in 14.3, 9.5 and 5.8% of the isolates, respectively. These isolates were significantly more frequently retrieved during infection (69.1%) than colonisation but differences were observed according to isolation site. Methicillin-resistance was found in 7.2% of the isolates, 78% of which harboured ≥ 1 of the targeted toxin-encoding genes. Conclusions This first comprehensive study of S. aureus in children showed S. aureus to be mainly retrieved during infection and a high rate of colonisation, not limited to the nasopharynx. Predominant infections were skin and soft tissue infections wheretst was most frequently ***-PV was most commonly detected during bone and joint infections. Isolates harbouring targeted toxin-encoding genes were significantly associated with infections but a quarter of children were asymp-tomatic carriers representing a reservoir for dissemination of isolates with virulence potency.

Over the last decades and centuries,European mountain landscapes have experienced substantial *** and anthropogenic lUlC changes(land use and land cover changes), especially agro-pastoral activities,have directly influenced the spatial organization and composition of European mountain *** the past sixty years, natural reforestation has been occurring due to a decline in both agricultural production activities and rural ***, to better anticipate future changes,need spatially and temporally explicit models to identify areas at risk of land change and possible *** paper presents an integrated approach combining forecasting scenarios and a lUlC changes simulation model to assess where lUlC changes may occur in the Pyrenees Mountains,based on historical lUlC trends and a range of future socio-economic *** proposed methodologyconsiders local specificities of the Pyrenean valleys,sub-regional climate and topographical properties,and regional economic *** indicate that some regions are projected to face strong abandonment, regardless of the scenario ***, high rates of change are associated with administrative regions where land productivity is highly dependent on socio-economic drivers and climatic and environmental conditions limit intensive(agricultural and/or pastoral) production and *** combination of the results for the four scenarios allows assessments of where encroachment(*** by shrublands) and reforestation are the most *** assessment intends to provide insight into the potential future development of the Pyrenees to help identify areas that are the most sensitive to change and to guide decision makers to help their management decisions.

To gain better insight into the influence of the anion size and symmetry on the transport properties and thermal stability of an electrolyte based on lithium(fluorosulfonyl)(trifluoromethanesulfonyl)-imide(FTFSI)salt,we performed the physical and electrochemical characterization of an electrolyte based on FTFSI incorporated in standard binary(3 EC/7 EMC)and ternary(EC/PC/3 DMC)alkylcarbonate *** applying the Jones-Dole-Kaminsky(JDK),Eyring and Arrhenius empirical models to the electrolyte viscosity we show that the activation enthalpy and entropy energy barriers(Δh≠,ΔS≠)for viscous flow are between 12 and 15 kJ·mol^(-1).They are strongly dependent on the solvent nature and are significantly lower than their symmetric anions liFSI and liTFSI(19-20 kJ·mol^(-1))in the binary ***,the hydrodynamic radius,rs,calculated by JDK,and the ionicity behavior illustrated by the Walden role,showed that the FTFSI anion is outside the solvation sphere(rs>0.6 nm)which is smaller in the case of an EC/EMC solvent *** the 3 EC/7 EMC solvent mixture,liFTFSI is less conductive than in the ternary mixture i.e.,σ_(max)=8.9 mS cm^(-1) at C_(max)=1.1 mol l^(-1) for 3 EC/7 EMC and,σ_(max)=10.5 mS cm^(-1) at max=0.7 mol l^(-1) for EC/PC/3 DMC,due to a strong solvation and a greater association of FTFSI ions in the binary solvent *** thermal stability of FTFSI based electrolytes was determined by the shift of the evaporation temperature of the volatile solvents(DMC,EMC)in the presence of salt,towards the higher *** feature is visible on the thermograms obtained by DSC both with the liquid electrolyte and with charged lMO cathodes in presence of *** consequences of these properties on the electrochemical behavior of a graphite(Gr)half-cell,a lithium metal(li)anode and a manganese lithium oxide(lMO)cathode demonstrated on the one hand the formation of a thick solid electrolyte interphase(SEI)on graphite that consumed a significant amou

Background: A large number of studies have addressed whiplash injury, and many meta-analyses have sought to highlight chronicity factors;the implicated processes, however, remain a matter of debate. The present study used data from the ESPARR cohort (an on-going prospective study of a representative cohort of road accident victims in the Rh?ne administrative département of France). The objectives were to describe the consequences of whiplash injury and to determine prognostic factors for poor recovery and persistent pain at 1 year post-accident. Methods: The cohort included 255 “pure” whiplash victims, 173 of whom responded to the 1-year follow-up questionnaire. Correlations between explanatory variables and health and pain status were explored by modified Poisson regression to provide adjusted relative risk (RR) values. Results: half of the victims had not fully recovered health status by 1 year. The main factor associated with non-recovery was pain (RR = 1.3;1.0-1.7). A birth in the family preceding the accident emerged as another factor (RR=1.5;1.2-1.9). Victims responsible for their accident were twice as likely to report being free of pain as those not responsible (RR = 0.5;0.3-0.8). No correlation emerged with accident-related characteristics or PTSD. Conclusions: The present results extend our understand- ing of whiplash injury. Residual pain is the fundamental factor causing whiplash victims to feel that they have not recovered good health. Our findings suggest this may be bound up with physical factors (gender susceptibility);external factors such as having to carry weights (such as a baby) and with perceiving oneself as a victim are not incompatible with this hypothesis.

European mountains are particularly sensitive to climatic disruptions and land use *** latter leads to high rates of natural reforestation over the last 50 years. Faced with the challenge of predicting possible impacts on ecosystem services,lUCC models offer new opportunities for land managers to adapt or mitigate their *** the spatial uncertainty of future lUCC is crucial for the definition of sustainable land use strategies. however, the sources of uncertainty may differ, including the input parameters, the model itself, and the wide range of possible futures. The aim of this paper is to propose a method to assess the probability of occurrence of future lUCC that combines the inherent uncertainty of model parameterization and the ensemble uncertainty of the future based scenarios. For this purpose, we used the land Change Modeler tool to simulate future lUCC on a study site located in the Pyrenees Mountains(France) and two scenarios illustrating two land use strategies. The model was parameterized with the same driving factors used for its calibration. The definition of ‘static vs. dynamic' and ‘quantitative ***(discretized)' driving factors, and their combination resulted in four parameterizations. The combination of model outcomes produced maps of the spatial uncertainty of future lUCC. This work involves adapting the definition of spatial uncertainty in the literature to future-based lUCC studies. It goes beyond the uncertainty of simulation models by integrating the uncertainty of the future to provide maps to help decision makers and land managers.

Arsenic trioxide(As2O3)is recently found to have therapeutic potential in systemic sclerosis(SSc),a life-threatening multi-system fibrosing autoimmune disease with type I interferon(IFN-I)*** activated plasmacytoid dendritic cells(pDCs)are responsible for IFN-I secretion and are closely related with fibrosis establishment in *** this study,we showed that high concentrations of As2O3 induced apoptosis of pDCs via mitochondrial pathway with increased BAX/BCl-2 ratio,while independent of reactive oxygen species ***,at clinical relevant concentrations,As2O3 preferentially inhibited IFN-αsecretion as compared to other cytokines such as TNF-α,probably due to potent down-regulation of the total protein and mRNA expression,as well as phosphorylation of the interferon regulatory factor7(IRF7).In addition,As2O3 induced a suppressive phenotype,and in combination with cytokine inhibition,it down-regulated pDCs’capacity to induce CD4+T cell proliferation,Thl/Th22 polarization,and B cell differentiation towards ***,chronically activated pDCs from SSc patients were not resistant to the selective IFN-αinhibition,and regulatory phenotype induced by ***,our data suggest that As2O3 could target pDCs and exert its treatment efficacy in SSc,and more autoimmune disorders with IFN-I signature.
