Validation of a radioimmunoassay-based fecal corticosteroid assay for Richardson＇s ground squirrels Urocitellus richardsonii and behavioural correlates of stress

We validated a radioimmunoassay-based method quantifying fecal glucocorticoid metabolites （FGMs） from captive male and female Richardson＇s ground squirrels Urocitellus richardsonii. Blood samples were drawn to explore the correlation between plasma cortisol and FGM concentrations. We also injected groups of squirrels with normal saline （CTL; control）, adre- nocorticotropic hormone （ACTH; stimulating adrenal activity）, or dexamethasone （DEX; suppressing adrenal activity）. Potential correlations between stress and behaviour were explored through quantification of fecal pellet production and the intervention necessary to elicit defecation, as well as the behaviour of subjects in the context of handling. Changes in plasma cortisol concen- tration between capture （baseline）, and following handling （stress-induced） were also quantified for free-living squirrels. While glucocorticoid concentrations recovered from feces during our captive-animal study were not well correlated with plasma eortisol concentrations, and uncorrelated with defecation or behaviour, FGM concentrations did reflect the activation of the hypothalamic- pituitary-adrenal （HPA） axis. FGM concentrations increased significantly during initial captivity, but declined to baseline level as individuals acclimated to the novel environment. Injection of subjects with ACTH increased FGMs above baseline, confirming activation of the HPA axis. Plasma cortisol concentrations increased significantly with induced stress, indicating that capture and handling activated the glucocorticoid stress response even among previously handled, free-living subjects. Our findings validate a non-invasive tool that will afford new insight into the physiological processes underlying social, reproductive and antipredator behaviour of Richardson＇s ground squirrels [Current Zoology 60 （5）： 591-601, 2014 ].

Climate change induces profound changes in mountain territories that affect the activities pursued *** is one activity that is strongly impacted by these *** research on tourism in mountain regions has focused on winter sports tourism,but glacier tourism,a summer activity,is also severely affected by climate change-induced phenomena,including glacier retreat and other geomorphological *** that Alpine glacier tourism has been pursued since the 18th century and glaciers have been changing throughout this period,this article aims to examine the adaptations implemented by glacier tourism operators since the activity’s *** analysis of historical documents,glaciological surveys and semi-structured interviews,this paper reveals how tourism activities around the Mer de Glace(the largest French glacier)have been impacted by glacier fluctuations since the first visit to the area in *** study reveals that these developments have mainly been driven by glacier retreat and associated paraglacial *** recently,the shortening of the period of snow cover on the glacier has also impacted tourism *** of the adaptation strategies implemented by tourism operators during the period in question are found to be reactive,consisting in the installation of safety equipment,the renovation of access points or the building of new structures to allow tourist activities to continue despite the consequences of glacier *** strategies,such as the adoption of new activities by workers previously specialised in another,are identified that could be considered transformative strategies,although these were only adopted by workers when their activity was no longer *** analysis of materials from 1741 to the present also reveals that the recent impacts of climate change on glacier tourism are much more significant and diverse than those produced by past ***,it is important that stakeholders take suit

Phosphorus(P)is an essential nutrient for maize production,but in temperate areas the P uptake during early growing stages can be limited due to low soil temperature,even though the soil was tested high in *** objective of this study was to assess the effects of nitrogen and phosphorous(nP)starter fertilisation during early growth stages and its carryover until maize harvest,in mineral-fertilised or manured systems.A field experiment was carried out in north-west Italy during the 2019 and 2020 growing *** trial compared sub-surface placement of nP(diammonium phosphate)or n alone(ammonium nitrate)in bands close to the maize seed furrows,in differing long-term(LT)fertilisation managements:two doses of urea(Min-L and Min-H),two doses of bovine slurry(Slu-L and Slu-H)or two doses of farmyard manure(Fym-L and Fym-H).The two rates,low(L)and high(H),corresponded to 170 and250 kg n ha^(-1)year^(-1) *** to n fertilisation,nP starter fertilisation improved early maize growth assessed by leaf area index(LAI)and shoot dry weight(SDW)in all *** effects differed between the two years(2019:LAI+63%,SDW+67%;2020:LAI+36%,SDW+38%),as 2019was cool during the first *** LAI and SDW values were confirmed at crop flowering in the mineral-fertilised systems *** shoot growth was enhanced by nP starter fertilisation,anthesis occurred 1 day earlier in all ***,a response to nP starter fertilisation at harvest was recorded in mineral-fertilised systems only(+1.3 and+3.2 t ha^(-1) in Min-L and Min-H,respectively).The uptake of P,used as a true indicator of soil nutrient availability,increased with increasing soil Olsen P until 39 mg kg^(-1).These results suggest that soil test thresholds should be revised for points above which P fertilisation should be suspended.

In this study,a top cover system is investigated as a control for emissions during the aftercare of new landfills and for old landfills where biogas energy production might not be *** materials were studied as landfill cover system in lab-scale columns：mechanical–biological pretreated municipal solid waste（MBP）;mechanical–biological pretreated biowaste（PB）;fine（PBSf）and coarse（PBSc）mechanical–biological pretreated mixtures of biowaste and sewage sludge,and natural soil（nS）.The effectiveness of these materials in removing methane and sulphur compounds from a gas stream was tested,even coupled with activated carbon *** of CO2,CH4,O2,n2,H2S and mercaptans were analysed at different depths along the *** degradation was assessed using mass balance and the results were expressed in terms of methane oxidation rate（MOR）.The highest maximum and mean MOR were observed for MBP（17.2 g CH4/m^2/hr and 10.3 g CH4/m^2/hr,respectively）.Similar values were obtained with PB and *** lowest values of MOR were obtained for nS（6.7 g CH4/m^2/hr）and PBSf（3.6 g CH4/m^2/hr）,which may be due to their low organic content and void index,*** membranes with high load capacity did not seem to have an influence on the methane oxidation process：MBP coupled with 220 g/m^2and 360 g/m^2membranes gave maximum MOR of 16.5 g CH4/m^2/hr and 17.4 g CH4/m^2/hr,*** carbon membranes proved to be very effective on H2S ***,carbonyl sulphide,ethyl mercaptan and isopropyl mercaptan seemed to be easily absorbed by the filling materials.

Loco-regional recurrences and distant metastases represent the main cause of head and neck squamous cell carcinoma(HnSCC) mortality. The overexpression of chemokine receptor 4(CXCR4) in HnSCC primary tumors associates with higher risk of developing loco-regional recurrences and distant metastases, thus making CXCR4 an ideal entry pathway for targeted drug delivery. In this context, our group has generated the self-assembling protein nanocarrier T22-GFP-H6, displaying multiple T22 peptidic ligands that specifically target CXCR4. This study aimed to validate T22-GFP-H6 as a suitable nanocarrier to selectively deliver cytotoxic agents to CXCR4^(+)tumors in a HnSCC model. Here we demonstrate that T22-GFP-H6 selectively internalizes in CXCR4^(+)HnSCC cells, achieving a high accumulation in CXCR4^(+)tumors in vivo, while showing negligible nanocarrier distribution in non-tumor bearing organs. Moreover, this T22-empowered nanocarrier can incorporate bacterial toxin domains to generate therapeutic nanotoxins that induce cell death in CXCR4-overexpressing tumors in the absence of histological alterations in normal organs. Altogether, these results show the potential use of this T22-empowered nanocarrier platform to incorporate polypeptidic domains of choice to selectively eliminate CXCR4^(+)cells in HnSCC. Remarkably, to our knowledge, this is the first study testing targeted proteinonly nanoparticles in this cancer type, which may represent a novel treatment approach for HnSCC patients.

AIM： To evaluate the efficacy and safety of glaucoma drainage device Ex-PRESS~ P-50 for combined cataract surgery and glaucoma.·METHODS： Patients having cataract and open angle glaucoma or patients with open advanced glaucoma which needed two or more antiglaucoma medications were included. Combined cataract surgery and glaucoma with Ex-PRESS~ P-50 model placed under scleral flap was performed.·RESULTS： Out of 40 eyes of 40 patients（55% male and45% female） completed the study during one-year follow-up. The mean of age was 76.6 ±11.02 y. The intraocular pressure（IOP） decreased significantly during the 12-month follow-up from 23.5 mm Hg to 16.8 mm Hg（Wilcoxon signed ranks test, P 〈0.001）. A 59.5% of patients did not need any topical treatment, 10.8% of them needed one active principle, 27% needed two active principles, and 2.7% of them needed three active principles for successful IOP control（〈21 mm Hg）.·COnCLUSIOn：Combinedsurgeryofphacoemulsification with Ex PRESS~ P-50 lowers IOP from the preoperative baseline and reduces significantly the number of antiglaucoma active principles for IOP control after the operation.
