A Systems Biology Approach for Studying Heterotopic Ossification： Proteomic Analysis of Clinical Serum and Tissue Samples

Heterotopic ossification （HO） refers to the abnormal formation of bone in soft tissue. Although some of the underlying processes of HO have been described, there are currently no clinical tests using validated biomarkers for predicting HO formation. As such, the diagnosis is made radiographically after HO has formed. To identify potential and novel biomarkers for HO, we used isobaric tags for relative and absolute quantitation （iTRAQ） and high-throughput antibody arrays to produce a semi-quantitative proteomics survey of serum and tissue from subjects with （HO ＋） and without （HO-） heterotopic ossification. The resulting data were then analyzed using a systems biology approach. We found that serum samples from subjects experiencing traumatic injuries with resulting HO have a different proteomic expression controls. Subsequent quantitative eLISA identified profile compared to those from the matched five blood serum proteins that were differentially regulated between the HO-- and HO- groups. Compared to HO- samples, the amount of insulin-like growth factor I （IGF1） was up-regulated in HO＋ samples, whereas a lower amount of osteopontin （OPN）, myeloperoxidase （MPO）, runt-related transcription factor 2 （RUNX2）,and growth differentiation factor 2 or bone morphogenetic protein 9 （BMP-9） was found in HO ＋ samples （Welch two sample t-test; P 〈 0.05）. These proteins, in combination with potential serum biomarkers previously reported, are key candidates for a serum diagnostic panel that may enable early detection of HO prior to radiographic and clinical manifestations.

The crystalline materials Ca3Sc2Si3O12 and Ca3Y2Si3O12 Were characterized by different crystal structures, as the fonmer is a cubic garnet, while the latter is an orthorhombic compound. We investigated the optical spectroscopy of these materials doped with several trivalent lanthanide ions and compared the results for the two hosts. PolycrystaUine samples were prepared by solid state reaction, both undoped and doped with the trivalent lanthanide ions eu3＋, Tb3＋ and Sm3＋. emission, excitation and Raman spectra of these materials were measured at temperatures ranging from 300 to 10 K. The optical spectra were assigned and discussed, and the effects of the crystal structure of the host on the spectroscopic behaviour were addressed. The technological potential of these compounds in the field of optical materials and devices was discussed,

Measurement methods and analysis tools for rail irregularities:a case study for urban tram track

Rail irregularities, in particular for urban rail- way infrastructures, are one of the main causes for the generation of noise and vibrations. In addition, repetitive loading may also lead to decay of the structural elements of the rolling stock. This further causes an increase in main- tenance costs and reduction of service life. Monitoring these defects on a periodic basis enables the network rail managers to apply proactive measures to limit further damage. This paper discusses the measurement methods for rail corrugation with particular regard to the analysis tools for evaluating the thresholds of acceptability in re- lation to the tramway Italian transport system. Further- more, a method of analysis has been proposed： an application of the methodology used for treating road profiles has been also utilized for the data processing of rail profilometric data.

Cardiorespiratory fitness,white blood cell count,and mortality in men and women

Background:We examined the associations of cardiorespiratory fitness(CRF) and white blood cell count(WBC) with mortality ***: A total of 52,056 apparently healthy adults completed a comprehensive health examination,including a maximal treadmill test and blood chemistry *** was categorized as high,moderate,or low by age and sex;WBC was categorized as sex-specific ***:During 17.8± 9.5 years(mean± SD) of follow-up,a total of 4088 deaths *** regressed jointly,significantly decreased allcause mortality across CRF categories was observed within each quartile of WBC in *** WBC Quartile 1,all-cause mortality hazard ratios(HRs) with a 95% confidence interval(95%CI) were 1.0(referent),1.29(95%CI:1.06-1.57),and 2.03(95%CI:1.42-2.92) for high,moderate,and low CRF categories,respectively(p for trend ends were observed in the remaining 3 *** the exception of cardiovascular disease(CVD) mortality within Quartile 1(p for trend=0.743),there were also similar trends across CRF categories within WBC quartiles in men for both CVD and cancer mortality(p for trend en,there were no significant trends across CRF categories for mortality outcomes within Quartiles ***,we observed significantly decreased all-cause mortality across CRF categories within WBC Quartile 4(HR=1.05(95%CI:0.76-1.44),HR=1.63(95%CI:1.20-2.21),and HR=1.87(95%CI:1.29-2.69) for high,moderate,and low CRF,respectively(p for trend=0.002)).Similar trends in women were observed for CVD and cancer mortality within WBC Quartile 4 ***:There are strong joint associations between CRF,WBC,and all-cause,CVD,and cancer mortality in men;these associations are less consistent in women.

Validation of a radioimmunoassay-based fecal corticosteroid assay for Richardson＇s ground squirrels Urocitellus richardsonii and behavioural correlates of stress

We validated a radioimmunoassay-based method quantifying fecal glucocorticoid metabolites （FGMs） from captive male and female Richardson＇s ground squirrels Urocitellus richardsonii. Blood samples were drawn to explore the correlation between plasma cortisol and FGM concentrations. We also injected groups of squirrels with normal saline （CTL; control）, adre- nocorticotropic hormone （ACTH; stimulating adrenal activity）, or dexamethasone （DeX; suppressing adrenal activity）. Potential correlations between stress and behaviour were explored through quantification of fecal pellet production and the intervention necessary to elicit defecation, as well as the behaviour of subjects in the context of handling. Changes in plasma cortisol concen- tration between capture （baseline）, and following handling （stress-induced） were also quantified for free-living squirrels. While glucocorticoid concentrations recovered from feces during our captive-animal study were not well correlated with plasma eortisol concentrations, and uncorrelated with defecation or behaviour, FGM concentrations did reflect the activation of the hypothalamic- pituitary-adrenal （HPA） axis. FGM concentrations increased significantly during initial captivity, but declined to baseline level as individuals acclimated to the novel environment. Injection of subjects with ACTH increased FGMs above baseline, confirming activation of the HPA axis. Plasma cortisol concentrations increased significantly with induced stress, indicating that capture and handling activated the glucocorticoid stress response even among previously handled, free-living subjects. Our findings validate a non-invasive tool that will afford new insight into the physiological processes underlying social, reproductive and antipredator behaviour of Richardson＇s ground squirrels [Current Zoology 60 （5）： 591-601, 2014 ].

The Impact of a Bicuspid Aortic Valve on Aortic Geometry and Function in Patients with Aortic Coarctation:A Comprehensive CMR Study

Background:An isolated bicuspid aortic valve(BAV)is associated with structural and functional abnormalities of the aorta and the left ventricle(LV).Although~50%of patients with aortic coarctation(CoA)have a BAV,less is known about its impact on LV function and aortic geometry and function in CoA *** this cardiovascular magnetic resonance imaging(CMR)study,we analysed markers of LV and aortic function as well as aortic geometry in a large cohort of CoA patients with a BAVand compared them with CoA patients with a tricuspid aortic valve(TAV).Methods:We included 48 patients with a BAV(18.4±9.3 years)and 45 patients with TAV(20.7±9.9 years).LV volumes,mass and ejection fraction as well as aortic distensibilty,pulse wave velocity(PWV)were measured from standard cine CMR and phase-contrast CMR images.2-dimensional CMR feature tracking(2DCMR-FT)was performed to measure longitudinal,circumferential and radial strain and strain rate of the *** arch geometry was classified as romanic,gothic and ***:LV volumes,mass and ejection fraction as well as aortic distensibility and PWV did not significantly differ between the BAV and the TAV *** was also no significant difference for LV global longitudinal,radial and circumferential strain and strain rate between both *** with a BAV had more commonly a gothic aortic arch compared to TAV patients,but this difference was not statistically significant(22 vs.14,p=0.2).Ascending and descending aortic distensibility correlated with LV mass in the entire patient group(perential and radial strain(GLS,GCS,GRS)and global longitudinal and circumferential strain rate(GLSR,GCSR)correlated with LV ejection fraction(pest that the presence of a BAV does not adversely impact LV and aortic function in children and young adults with *** correlation of global circumferential,longitudinal and radial strain values with LV ejection fraction dem

eggplant(Solanum melongena L.) yield is highly sensitive to N fertilization, the excessive use of which is responsible for environmental and human health *** N input together with the selection of improved Nitrogen-Use-efficiency(NUe) genotypes, more able to uptake, utilize, and remobilize N available in soils, can be challenging to maintain high crop yields in a sustainable agriculture. The aim of this study was to explore the natural variation among eggplant accessions from different origins, in response to Low(LN) and High(HN)Nitrate(NO3^-) supply, to identify NUe-contrasting genotypes and their NUe-related traits, in hydroponic and greenhouse pot experiments. Two eggplants, AM222 and AM22, were identified as N-use efficient and inefficient,respectively, in hydroponic, and these results were confirmed in a pot experiment, when crop yield was also evaluated. Overall, our results indicated the key role of Nutilization component(NUt e) to confer high NUe. The remobilization of N from leaves to fruits may be a strategy to enhance NUt e, suggesting glutamate synthase as a key enzyme. Further, omics technologies will be used for focusing on C-N metabolism interacting networks. The availability of RILs from two other selected NUe-contrasting genotypes will allow us to detect major genes/quantitative trait loci related to NUe.

Attosecond twisted beams from high-order harmonicgeneration driven by optical vortices

Optical vortices are structures of the electromagnetic field with a spiral phase ramp about a point-phase singularity,carrying orbital angular momentum(OAM). Recently, OAM has been imprinted to short-wavelength radiation through high-order harmonic generation(HHG), leading to the emission of attosecond twisted beams in the extreme-ultraviolet(XUV) regime. We explore the details of the mapping of the driving vortex to its harmonic spectrum. In particular, we show that the geometry of the harmonic vortices is convoluted, arising from the superposition of the contribution from the short and long quantum paths responsible of HHG. Finally, we show how to take advantage of transverse phase-matching to select twisted attosecond beams with different spatiotemporal properties.

Cambro-Silurian magmatisms at the northern Gondwana margin(Penninic basement of the Ligurian Alps)

The early Paleozoic evolution of the northern margin of Gondwana is characterized by several episodes of bimodal magmatism intruded or outpoured within thick sedimentary basins. These processes are well recorded in the Variscan blocks incorporated in the Ligurian Alps because they experienced low temperature Alpine metamorphism. During the Paleozoic, these blocks, together with the other Alpine basements, were placed between the Corsica-Sardinia and the Bohemian Massif along the northern margin of Gondwana. In this framework, they host several a variegated lithostratigraphy forming two main complexes(Complexs I and II) that can be distinguished by both the protoliths and their crosscutting relationships, which indicate that the acidic and mafic intrusives of Complex II cut an already folded sequence made of sediments, basalts and granitoids of Complex I. Both complexes were involved in the Variscan orogenic phases as highlighted by the pervasive eclogite-amphibolite facies schistosity(foliation II). However, rare relicts of a metamorphic foliation at amphibolite facies conditions(foliation I)is locally preserved only in the rocks of Complex I. It is debatable if this schistosity was produced during the early folding event e occurred between the emplacement of Complex I and II e rather than during an early stage of the Variscan metamorphic *** SHRIMP and LA ICP-MS Ue Pb zircon dating integrated with literature data, provide emplacement ages of the several volcanic or intrusive bodies of both complexes. The igneous activity of Complex I is dated between 507 ± 15 Ma and 494 ± 5 Ma, while Complex II between 467 ± 12 Ma and 445.5 ± 12 *** folding event recorded only by the Complex I should therefore have occurred between 494 ± 5 Ma and 467 ± 12 Ma. The Variscan eclogite-amphibolite facies metamorphism is instead constrained between ~420 Ma and ~300 Ma.
