Effects of <i>Pereskia aculeata</i>Miller on the biochemical Profiles and body Composition of Wistar Rats

Pereskia aculeata Miller commonly known as “ora-pro-nobis” has been used extensively in folclore medicine. Plant belongs to the family Cactacea and it is found in the brazil (reaching bahia and Rio Grande do Sul states). The purpose was to evaluate the effects of plant on biochemical and corporal profiles of Wistar rats. The 80 animals were divided randomly into 4 groups by sex (n = 10 females and 10 males per group) as follows: G1 group (water/rat food ad libitum);G2 group (drink A: commercial condensed milk, sugar and water/rat food ad libitum);G3 group (drink b: P. aculeata juice/rat food ad libitum);G4 group (drink C: commercial condensed milk, sugar and P. aculeate juice/rat food ad libitum). After 35 days their biochemical and corporal profiles were analyzed (cholesterol, glycemia, triglycerides, HDL-c, AST, ALT, Lee Index, weight and visceral fat). Intake of the plant caused no changes in the lipid profile of Wistar rats. However, intake in male rats of the same prevented the increase in triglycerides front of a hypercaloric diet. There were significant changes in glycemic index due to the high carbohydrate content of the plant. The plant was effective in reducing the levels of AST and ALT in male rats and ALT only in females. The use of the plant did not cause significant changes in the index of Lee but the Pereskia aculeata Miller may reduce visceral fat gain in female with a regular diet.

Role of serial multiparametric magnetic resonance imaging in prostate cancer active surveillance

AIM:To examine whether addition of 3T multiparametric magnetic resonance imaging(mp MRI)to an active surveillance protocol could detect aggressive or progressive prostate ***:Twenty-three patients with low risk disease were enrolled on this active surveillance study,all of which had Gleason score 6 or less *** patients had clinical assessments,including digital rectal examination and prostate specific antigen(PSA)testing,every 6 mo with annual 3T mp MRI scans with gadolinium contrast and minimum sextant prostate *** MRI images were anonymized of patient identifiers and clinical information and each scan underwentradiological review without the other results *** statistics for demographics and follow-up as well as the sensitivity and specificity of mp MRI to identify prostate cancer and progressive disease were ***:During follow-up(median 24.8 mo)11 of 23 patients with low-risk prostate cancer had disease progression and were taken off study to receive definitive *** progression was identified through upstaging of Gleason score on subsequent biopsies for all 11 patients with only 2 patients also having a PSA doubling time of less than 2 *** 23 patients had biopsy confirmed prostate cancer but only 10 had a positive index of suspicion on mp MRI scans at baseline(43.5% sensitivity).Aggressive disease prediction from baseline mpM RI scans had satisfactory specificity(81.8%)but low sensitivity(58.3%).Twentytwo patients had serial mp MRI scans and evidence of disease progression was seen for 3 patients all of whom had upstaging of Gleason score on biopsy(30% specificity and 100% sensitivity).CONCLUSION:Addition of mp MRI imaging in active surveillance decision making may help in identifying aggressive disease amongst men with indolent prostate cancer earlier than traditional methods.

Sensibility Analysis of the brazilian Standards for Energy Efficiency Regarding the Variation of Internal Load Density in Office buildings in brazilian bioclimatic Zones 1 and 7

The RTQ-C (Technical Requirements of Quality for the Energy Performance Level of Commercial buildings) publication classified the buildings in five efficiency levels. In RTQ-C, the evaluation can be done with two methods: a prescriptive method and a simulation one. This paper aims to identify the sensibility of the prescriptive method RTQ-C regarding the variation of equipment internal load density in office buildings in bioclimatic Zones 1 and 7 of the brazilian bioclimatic zoning. The research results show that the building with walls and roof configured to meet specific prerequisites for energy efficiency Levels b and C had a lower consumption than buildings that meet the prerequisites to Level A. The study also showed that buildings with high internal load density of equipment, maximum shape factor and high, with walls and roofs with higher thermal transmittance, have lower power consumption than constructions with an envelope with greater thermal resistance. The increase in internal load density causes an increase in the internal heat generated by the large amount of equipment. In buildings with higher thermal insulation (Level A), the internal heat is maintained in the environment, causing overheating and the need for an air conditioning system.

肺癌是全球肿瘤发病率和病死率的主因,且预后很差。加强对肿瘤生物学的认识对肺癌研究至关重要。被誉为"下一代测序技术"的NGS技术(next-generation sequencing)是一种针对全基因组鉴定的有力工具,可以对致癌体细胞突变进行全面检测。大多数的NGS技术是基于平台特异性DNA文库进行多重聚合酶链反应(polymerase chain reaction,PCR),从而对目的基因扩增后测序。这种技术适用于高通量测序,可以检测出肿瘤中出现的全部基因组变异。缺点是这种技术需要在时间、实验设备、计算机数据分析、生物信息技术等各方面的大量投入。NGS技术已广泛应用于全基因组、外显子组、转录组和表观基因组的研究中,为肺癌研究和医疗模式带来改变。这项新技术的开展将转变当前对致癌信号通路的认识,可为癌症诊疗提供新的分子靶点。肺癌体细胞突变已有NGS技术的分析报道,但大规模基因组研究仍在进行中。个体化治疗策略将改善那些潜在获益患者的治疗方式,避免"无辜"患者受无效治疗带来的高额费用和不良反应。NGS的组织化、计算机化和生物信息化技术推动了科技的进步,同时,患者知情权和数据发布的相关伦理问题也浮现出来。信号通路中,驱动基因(driver gene)突变和传递基因(passenger gene)突变的区别,需要对测序结果进行细致解读。解读准确与否取决于DNA提取的样本类型、样本处理技术和样本含量。肿瘤异质性也会降低肿瘤基因突变的检测效能。NGS技术将推动对肿瘤基因突变的基础和临床研究,而且,也可应用于单细胞和游离的循环DNA,未来还将用于从体液和肿瘤亚群中获取的DNA样本。如果能进一步降低费用、提高检验速度和精度,NGS技术无疑将会成为肺癌研究的绝佳选择。
