Rigidity Symmetry Line for Thermodynamic Fluid equations-of-State

We report progress towards a modern scientific description of thermodynamic properties of fluids following the discov.ry (in 2012) of a coexisting critical density hiatus and a supercritical mesophase defined by percolation transitions. The state functions density ρ(p,T), and Gibbs energy G(p,T), of fluids, e.g. CO2, H2O and argon exhibit a symmetry characterised by the rigidity, ω = (dp/dρ)T, between gaseous and liquid states along any isotherm from critical (Tc) to Boyle (TB) temperatures, on either side of the supercritical mesophase. Here, using experimental data for fluid argon, we inv.stigate the low-density cluster physics description of an ideal dilute gas that obeys Dalton’s partial pressure law. Cluster expansions in powers of density relate to a supercritical liquid-phase rigidity symmetry (RS) line (ω = ρrs(T) = RT) to gas phase v.rial coefficients. We show that it is continuous in all deriv.tiv.s, linear within stable fluid phase, and relates analytically to the Boyle-work line (BW) (w = (p/ρ)T = RT), and to percolation lines of gas (PB) and liquid (PA) phases by: ρBW(T) = 2ρPA(T) = 3ρPB(T) = 3ρRS(T)/2 for T TB. These simple relationships arise, because the higher v.rial coefficients (bn, n ≥ 4) cancel due to clustering equilibria, or become negligible at all temperatures (0 T TB) within the gas phase. The Boyle-work line (p/ρBW)T is related exactly at lower densities as T → TB, and accurately for liquid densities, by ρBW(T) = −(b2/b3)T. The RS line, ω(T) = RT, defines a new liquid-density ground-state physical constant (ρRS(0) = (2/3)ρBW(0) for argon). Background: Immune checkpoint ligand-receptor interactions appear to be associated with multiple myeloma (MM) progression. Simultaneously, prev.ous studies showed the possibility of PD-1 and TIM-3 expression on T cells upon stimulation with common γ-chain family cytokines in v.tro and during homeostatic proliferation. The aim of the present work was to study the impact of homeostatic proliferation on the expansion of certain T cell subsets up-regulating PD-1 and TIM-3 checkpoint molecules. Methods: The expression of CD25, CD122, CD127 common γ-chain cytokine receptors, phosphorylated signal transducer and activ.tor of transcription-5 (pSTAT5) and eomesodermin (eOMeS) was comparativ.ly assessed with flow cytometry in PD-1-and TIM-3-negativ. and positiv. T cells before the conditioning and during the first post-transplant month in peripheral blood samples of MM patients. Results: Substantial proportions of PD-1-and TIM-3-positiv. T lymphocytes expressed common γ-chain cytokine receptors and pSTAT5. Frequencies of cytokine receptor expressing cells were significantly higher within TIM-3+ T cells compared to PD-1+TIM-3− subsets. Considerable proportions of both PD-1-/TIM-3-negativ. and positiv. CD8+ T cells express eOMeS, while only moderate frequencies of CD4+ PD-1+ /TIM-3+ T cells up-regulate this transcription factor. Besides, the surface presence of CD25 and intranuclear expression of eOMeS in CD4+ T cells were mutually exclusiv. regardless of PD-1 and TIM-3 expression. The stimulation with common γ-chain cytokines up-regulates PD-1 and TIM-3 during the proliferation of initially PD-1/TIM-3-negativ. T cells but fails to expand initially PD-1+ and TIM-3+ T cell subsets in v.tro. Conclusions: Both PD-1 and TIM-3 expressing T cells appear to be able to respond to homeostatic cytokine stimulation. A paper analytical dev.ce (PAD) was dev.loped for the colourimetric estimation of alkaline phosphatase (ALP). ALP catalyzes the hydrolysis of disodium phenyl phosphate (DSPP) to phenol, which then undergoes oxidativ. coupling with4-aminoantipyrine (4-AAP) producing a reddish brown-coloured quinone imine. The colourimetric reaction tested in solution has been translated onto a PAD for the point of care (PoC) testing of ALP. A uniform colour was obtained in 25 min with the introduction of ALP onto the reagent drop cast paper dev.ce. The images obtained by scanning the PAD were processed using ImageJ software. The colour intensity obtained by image processing was proportional to the concentration of ALP present in the sample. ALP in serum and saliv. samples were tested using the PAD in the range of 0 to 528 U/L and0 to 187 U/L, respectiv.ly. The interference studies rev.aled that the PAD was selectiv. to ALP in the presence of other biomolecules. The PAD exhibited a limit of detection (LOD) of 3.3 U/L and a sensitiv.ty of 48.36 a.u/Log(U/L) for serum ALP and LOD of 1.69 U/L and a sensitiv.ty of 28.5 a.u/Log (U/L) for saliv.ry ALP. The PAD showed good agreement with the clinical methods for real samples.
