Hedgehog signaling pathway gene variant influences bronchopulmonary dysplasia in extremely low birth weight infants

Background Genome wide association study identified hedgehog interacting protein gene(HHIP)variants with chronic obstructive pulmonary disease and *** of HHIP,a key regulator of the hedgehog signaling pathway,leads to impaired lung morphogenesis and lethality in animal models,through unimpeded sonic hedgehog expression blocking mesenchymal-expressed fibroblast growth factor 10(FGF10).Since bronchopulmonary dysplasia(BPD)is also associated with altered lung development and worsens with stimuli including mechanical ventilation,reactive oxygen species,and inflammation,HHIP and FGF10 may be candidate *** This was an observational,cohort study including extremely low birth weight infants that who developed BPD and those who did *** was isolated from buccal swabs and subjected to allelic discrimination,using specific HHIP and FGF10 *** levels were measured in tracheal ***'s t test,Chi-square,z test and logistic regression were *** Demographic characteristics did not differ except that birth weight(715±153 vs.835±132 g)and gestational age(25 vs.26 weeks)were less in babies with *** variant rs 13147758(GG genotype)was found to be independently protective for BPD(odds ratio 0.35,95%confidence interval 0.15-0.82,P=-0.02).Early airway HHIP protein levels were increased in infants with BPD compared to those without[median(interquartile range)130.6(55.6-297.0)and 41.2(22.1-145.6)pg/ml,respectively;P=0.05].The FGF10 single nucleotide polymorphisms were not associated with *** HHIP,as a regulator of lung bud formation,affects BPD susceptibility,and may be valuable in understanding the specific mechanisms for this disease as well as for identifying therapeutic targets in the era of personalized medicine.

Human hematopoiesis was evaluated using the techniques of controlled stem cell differentiation, two-dimensional gel electrophoresis-based proteomics, and functional genomics. We provide the first report that glia maturation factor gamma （GMFG） is a cytokine-responsive protein in erythropoietin-induced and granulocyte-colony stimulating factor-induced hematopoietic lineage development. Results from global functional genomics analysis indicate that GMFG possesses several other features： hematopoietic tissue-specific gene expression, a promoter concentrated with high-score hematopoiesis-specific transcription factors, and possible molecular coevolution with a rudimentary blood/immune system. On the basis of our findings, we hypothesize that GMFG is a hematopoietic-specific protein that may mediate the pluripotentiality and lineage commitment of human hematopoietic stem cells.

The effects of operation parameters of combined blowing converter on the volumetric mass transfer coefficient between slag and steel are studied with a cold model with water simulating steel, oil simulating slag and benzoic acid as the transferred substance between water and oil. The results show that, with lance.l.vel of 2.1m and the top blowing rate of 25000Nm3/h, the volumetric mass transfer coefficient changes most significantly when the bottom blowing rate ranges from 384 to 540Nm3/h. The volumetric mass transfer coefficient reaches its maximum when the lance.l.vel is 2.1m, the top blowing rates is 30000Nm3/h, and the bottom blowing rate is 384Nm3/h with tuyeres located symmetrically at 0.66D of the converter bottom.

在水热条件下用氯化锰和咪唑基配体合成了配合物Mn[(IPB)_2Cl_2]·4H_2O(IPB=4-(1H-imidazo[4,5-f][1,10]phenanthrolin-2-yl)benzaldehyde),并通过元素分析、红外光谱、紫外衍射、X射线单晶衍射法和z-扫描技术对其结构和性质进行了表征。X射线单晶衍射结果表明,该化合物晶体属正交晶系,空间群Pnna。采用z-扫描技术测量了该化合物的非线性光学特性,结果显示,以45μJ的脉冲能量,在焦点处的透光率为93%,非线性吸收系数为-4.5×10-11 m·W-1,非线性折射率为1.0×10^(-19) m^2·W^(-1)。该化合物显示了自聚焦的性质。在紫外-可见光谱区,该化合物的最大吸收波长为282nm。

Hand,foot and mouth disease(HFMD)is a major public health problem among children in the Asia-Pacific *** optimal specimen for HFMD virological diagnosis remains *** A71(EV-A71)neutralizing antibody titres detected in paired sera were considered the reference standard for calculating the sensitivity,specificity,positive and negative predictive value of throat swabs,rectal swabs,stool,blood samples and cerebrospinal fluid(CSF)by RT-PCR or ElISA *** this study,clinical samples from 276 HFMD patients were collected for analysing the sensitivity of different kind of *** results showed that stool had the highest sensitivity(88%,95%CI:74%–96%)and agreement with the reference standard(91%).The order of diagnostic yield for EV-A71 infection was stool samplerectal swab>throat swab>blood sample>CSF sample,and using a combination of clinical samples improved sensitivity for enterovirus *** sensitivity of ElISA for IgM antibody detection in sterile-site specimens was significantly higher than that of RT-PCR(serum/plasma:62%vs.2%,CSF:47%vs.0%)(Plusion,our results suggest that stool has the highest diagnostic yield for EV-A71-infected *** stool is unavailable,rectal swabs can be collected to achieve a similar diagnostic ***,throat swabs may be useful in detecting positive *** IgM in blood or CSF is diagnostically accurate,it lacks sensitivity,missing 40%–50%of *** higher proportion of severe cases and shorter interval between onset and sampling contributed to the increase in congruency between clinical testing and the serological reference standard.

Powdery mildews present specific challenges to phenotyping systems that are based on *** previously developed lowthroughput,quantitative microscopy approaches for phenotyping resistance to Erysiphe necator on thousands of grape leaf disk samples for genetic analysis,here we developed automated imaging and analysis methods for *** severity on leaf *** pairing a 46-megapixel CMOS sensor camera,a long-working distance lens providing 3.5×magnification,X-Y sample positioning,and z-axis focusing movement,the system captured 78%of the area of a 1-cm diameter leaf disk in 3 to 10 focus-stacked images within 13.5 to 26 *** image pixel represented 1.44 m2 of the leaf disk.A convolutional neural network(CNN)based on GoogleNet determined the presence or absence of *** hyphae in approximately 800 subimages per leaf disk as an assessment of severity,with a training validation accuracy of 94.3%.For an independent image set the CNN was in agreement with human experts for 89.3%to 91.7%of *** live-imaging approach was nondestructive,and a repeated measures time course of infection showed differentiation among susceptible,moderate,and resistant *** over one thousand samples per day with good accuracy,the system can assess host resistance,chemical or biological efficacy,or other phenotypic responses of grapevine to *** addition,new CNNs could be readily developed for phenotyping within diverse pathosystems or for diverse traits amenable to leaf disk assays.
