BACKGROUND Thrombocytopenia 2,an autosomal dominant inherited disease characterized by moderate thrombocytopenia,predisposition to myeloid malignancies and normal platelet size and function,can be caused by 5’-untranslated region(UTR)point mutations in ankyrin repeat domain containing 26(ANKRD26).Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1)have been identified as negative regulators of ***,the positive regulators of ANKRD26 are still *** To prove the positive regulatory effect of GATA binding protein 2(GATA2)on ANKRD26 *** human induced pluripotent stem cells derived from bone marrow(hiPSC-BM)INTRODUCTION Ankyrin repeat domain containing protein 26(ANKRD26)acts as a regulator of adipogenesis and is involved in the regulation of feeding behavior[1-3].The ANKRD26 gene is located on chromosome 10 and shares regions of homology with the primate-specific gene family *** to the human Protein Atlas database,the ANKRD26 protein is localized to the Golgi apparatus and vesicles,and its expression can be detected in nearly all human tissues[4].Moreover,UniProt annotation revealed that ANKRD26 is localized in the centrosome and contains coiled-coil domains formed by spectrin helices and ankyrin repeats[5,6].The most common disease related to ANKRD26 is thrombocytopenia 2(THC2),which is a rare autosomal dominant inherited disease characterized by lifelong mild-to-moderate thrombocytopenia and mild bleeding[7-9].Caused by the variants in the 5’-untranslated region(UTR)of ANKRD26,THC2 is defined by a decrease in the number of platelets in circulating blood and results in increased bleeding and decreased clotting ability[8,10].Due to the point mutations that occur in the 5’-UTR of ANKRD26,its negative transcription factors(TFs),Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1),lose their repression effect[11].The persistent expression of ANKRD26 increases the activi

Application of wastewater-based epidemiological monitoring of COVID-19 for disease surveillance in the city

Wastewater-based surveillance serves as a supplementary approach to clinical surveillance of COVID-19 during the *** study aimed to track the prevalence of the disease and the viral genetic variability through wastewater-based surveillance in the post-epidemic *** January to December 2023,samples were collected from the influent lines of two wastewater treatment plants(WWTPs),concentrated using PEG8000,and subjected to detection of the target genes ORF 1ab and N of SARS-CoV-2 via reverse transcriptional quantitative PCR(RT-qPCR).For next-generation sequencing(NGS),high-quality samples from both wastewater and clinical patients were *** analysis were performed using R software to evaluate the correlation between the SARSCoV-2 RNA concentrations in wastewater and positive rate of reported cases,indicating a positive *** diversity patterns of SARS-CoV-2 in wastewater resembled those in the patient source based on Principal Coordinates Analysis(PCoA)with three clusters for different *** rise of RNA concentration in wastewater indicates the growth of cases and the emergence of new variants,serving as an early warning of potential viral mutations,disease outbreaks even possible ***,the genomic surveillance of wastewater could help identify new variants that may not be captured through population monitoring,especially when sample sizes are ***,surveillance of SARS-CoV-2 in municipal wastewater has emerged as a reliable,earlywarning monitoring system for COVID-19 in the post-epidemic era.

A Facile Li2TiO3 Surface Modification to Improve the Structure Stability and Electrochemical Performance of Full Concentration Gradient Li-Rich Oxides

Full concentration gradient lithium-rich layered oxides are catching lots of interest as the next generation cathode for lithium-ion batteries due to their high discharge voltage,reduced voltage decay and enhanced rate performance,whereas the high lithium residues on its surface impairs the structure stability and long'term cycle performance. Herein,a facile multifunctional surface modification method is implemented to eliminate surface lithium residues of full concentration gradient lithium-rich layered oxides by a wet chemistry reaction with tetrabutyl titanate and the post-annealing process. It realizes not only a stable Li2TiO3coating layer with 3D diffusion channels for fast Li+ions transfer,but also dopes partial Ti4+ions into the sub-surface region of full concentration gradient lithium-rich layered oxides to further strengthen its crystal structure. Consequently,the modified full concentration gradient lithium-rich layered oxides exhibit improved structure stability,elevated thermal stability with decomposition temperature from 289.57℃to321.72℃,and enhanced cycle performance (205.1 mAh g-1after 150 cycles)with slowed voltage drop （1.67 mV per cycle）. This work proposes a facile and integrated modification method to enhance the comprehensive performance of full concentration gradient lithium-rich layered oxides,which can facilitate its practical application for developing higher energy density lithium-ion batteries.

Chronic hepatitis B(CHB)is consistently challenging to conquer with numerous complications and *** the effectiveness of antiviral therapies in suppressing hepatitis B virus(HBV)replication,there is an urgent need for novel and more effective treatment *** strategies predominantly emphasize immune activation but still face challenges in sufficiently eliciting T cell *** into account the targeted delivery of nanoparticles to the liver and spleen via intravenous injection,we have proposed a dual-pronged therapeutic strategy based on antigen-presenting cells(APCs)-derived ***,exosomes targeted to the spleen can activate specific immune responses,while those targeted to the liver can modulate or reverse the liver’s immunosuppressive *** immunization,exosome formulations exhibit the remarkable ability to effectively activate APCs,thereby triggering the proliferation of CD8^(+)T ***,they also play an immunoregulatory role by converting M2 macrophages into M1 *** two-pronged therapeutic strategy precisely addresses the issues of T cell dysfunction and immune suppression,both characteristic features of CHB *** combined with Aluminum(Alum)-adjuvanted vaccine,these exosome formulations not only demonstrate a high level of cellular immune response but also secrete specific antibodies comparable to those induced by Alum *** combined approach effectively enhances both cellular and humoral immunity,offering a promising avenue for the development of therapeutic hepatitis B vaccines based on exosome formulations.
