●AIM:To identify disease-causative mutations in families with congenital cataract.●METHODS:Two Chinese families with autosomaldominant congenital cataract(ADCC)were recruited and underwent comprehensive eye *** panel next-generation sequencing of common pathogenic genes of congenital cataract was performed in the proband of each *** sequencing was used to valid the candidate gene mutations and sequence the other family members for co-segregation *** effect of sequence changes on protein structure and function was predicted through bioinformatics *** intrinsic protein(MIP)-wildtype and MIP-G29R plasmids were constructed and microinjected into zebrafish single-cell stage *** embryonic lens phenotypes were screened using confocal microscopy.●RESULTS:A novel heterozygous mutation(c.85G>A;p.G29R)in the MIP gene was identified in the proband of one family.A known heterozygous mutation(c.97C>T;p.R33C;rs864309693)in MIP was found in the proband of another ***-silico prediction indicated that the novel mutation might affect the MIP protein *** embryonic lens was uniformly transparent in both wild-type PCS2+MIP and mutant PCS2+MIP.●CONCLUSION:Two missense mutations in the MIP gene in Chinese cataract families are identified,and one of which is *** findings expand the genetic spectrum of MIP mutations associated with *** functional studies suggest that the novel MIP mutation might not be a gain-of-function but a loss-of-function mutation.

目的:回顾性研究呼吸重症监护室(RICU)住院患者肺部多重耐药菌(MDRO)感染的临床特点,进一步探讨RICU患者肺部多重耐药菌感染的危险因素。方法:选取2021年1月至2021年12月在武汉大学中南医院RICU收治的502例患者为研究对象,收集患者临床资料,根据药敏试验结果分为多重耐药菌组(n=166)与非多重耐药菌组(n=336),采用SPSS 25.0系统分别行单因素和多因素分析RICU患者肺部多重耐药菌感染的危险因素。结果:502例患者中MDRO感染者166例。单因素分析显示,两组患者RICU住院时间>14 d、气切或气管插管、有创机械通气、中心静脉导管、使用3种及以上抗生素、多器官功能衰竭的差异均有统计学意义(P14 d及多器官功能衰竭是肺部MDRO感染的危险因素(P<0.05)。结论:RICU患者肺部多重耐药菌感染率很高,抗生素的合理使用、尽量减少有创操作及缩短RICU住院时间可能有利于减少多重耐药菌感染的发生率。

With the acceleration of urbanization and the improvement of people’s living standards,people have increasingly higher requirements for the living environment,especially in the design of outdoor ***,existing landscape balconies generally suffer from insufficient shading and rainwater drainage problems,which affects people’s living *** response to this problem,this article proposes a new design scheme for building landscape balconies to solve the existing problems of landscape *** design plan fully considers the user’s usage needs and scenarios,improves the use value and comfort of the balcony through technical means,and achieves intelligence and functionality.

Redox-altered plasticity refers to redox-dependent reversible changes in synaptic plasticity via altering functions of key proteins, such as N-methyl-D-aspartate receptor(NMDAR). Age-related cognitive disorders includes Alzheimer’s disease(AD), vascular dementia(VD), and age-associated memory impairment(AAMI). Based on the critical role of NMDAR-dependent long-term potentiation(LTP) in memory, the increase of reactive oxygen species in cognitive disorders, and the sensitivity of NMDAR to the redox status, converging lines have suggested the redox-altered NMDAR-dependent plasticity might underlie the synaptic dysfunctions associated with cognitive disorders. In this review, we summarize the involvement of redox-altered plasticity in cognitive disorders by presenting the available evidence. According to reports from our laboratory and other groups, this "redox-altered plasticity" is Hydrogen sulfidemore similar to functional changes rather than organic injuries, and strategies targeting redox-altered plasticity using pharmacological agents might reverse synaptic dysfunctions and memory abnormalities in the early stage of cognitive disorders. Targeting redox modifications for NMDARs may serve as a novel therapeutic strategy for memory deficits.

Rice(Oryza sativa L.)is a staple cereal for more than two thirds of the world's *** salinity severely limits rice growth,development,and grain *** is desirable to elucidate the mechanism of rice's salt-stress *** the major source of H_(2)O_(2),NADPH oxidase(Rboh)is believed to be involved in salt-stress ***,the function and mechanism of rice Rboh in salt stress response remain *** this study,we found that the expression of OsRbohA was up-regulated by NaCl treatment in the shoots and roots of rice *** of OsRbohA reduced the tolerance of rice to salt *** of OsRbohA blocked NaCl-induced increases of NADPH activity and H_(2)O_(2) content in *** A knockout inhibited root growth and disrupted K^(+)homeostasis by reducing the expression of K^(+) transporters and channel-associated genes(OsGORK,OsAKT1,OsHAK1,and OsHAK5)in roots under NaCl *** NaCl treatment,OsRbohA knockout also reduced subcellular K^(+) contents of the plasma membrane and soluble *** of OsRbohA increased the expression of K^(+) transporters and channel-associated genes and reduced the loss of K^(+) ions in *** results indicate that OsRboh A-mediated H_(2)O_(2) accumulation modulates K^(+) homeostasis,thereby increasing salt tolerance in rice.

Cadmium(Cd)pollution affects plant growth and poses a serious threat to food safety and human ***-contaminated rice is assumed to be the main source of Cd exposure to humans,with grave health *** is an efficient,cost-effective,and eco-friendly approach to minimize Cd accumulation in ***,research on the effect of rice intercropping with wetland plants that exhibit great capacity for phytoremediation in decreasing Cd concentrations in paddies is limited.A pot experiment in greenhouse was conducted to evaluate the effect of rice intercropping with two wetland plants(Pontederia cordata and Canna indica)under different soil Cd levels(0,1,10,and 25 mg kg^(-1))on rice growth and Cd accumulation and *** results showed that rice intercropping with the two wetland plants significantly improved plant height,tiller,and biomass of rice,but reduced Cd bioconcentration factors(BCFs)and Cd concentrations in rice and wetland *** with rice monocropping,the BCF in rice grain under 25 mg kg^(-1)soil Cd level significantly decreased by 54.39%and 59.65%in the systems of rice intercropping with *** and ***,*** addition,intercropping inhibited the increase in BCFs in rice and wetland plants under increasing soil Cd *** conclusion,the systems of rice intercropping with *** and *** can be considered two new and effective approaches for ameliorating Cd pollution in paddies,as well as minimizing Cd accumulation in rice and improving food *** the two intercropping systems,rice intercropping with *** performed better than intercropping with ***.

Mitochondrial superoxide overproduction is believed to be responsible for the neurotoxicity associated with ***-targeted antioxidants,such as MitoQ,have emerged as potentially effective antioxidant *** sulfoxide reductase A(MsrA)is a key mitochondrial-localized endogenous antioxidative enzyme and it can scavenge oxidizing species by catalyzing the methionine(Met)-centered redox cycle(MCRC).In this study,we observed that the natural L-Met acted as a good scavenger for antimycin A-induced mitochondrial superoxide overproduction in PC12 *** antioxidation was largely dependent on the Met oxidase activity of MsrA.S-methyl-L-cysteine(SMLC),a natural analogue of Met that is abundantly found in garlic and cabbage,could activate the Met oxidase activity of MsrA to scavenge free ***,SMLC protected against antimycin A-induced mitochondrial membrane depolarization and alleviated 1-methyl-4-phenylpyridinium(MPP+)-induced ***,our data highlighted the possibility for SMLC supplement in the detoxication of mitochondrial damage by activating the Met oxidase activity of MsrA.
