Network rewiring,adaptive resistance and combating strategies in breast cancer

Resistance to targeted anti-cancer drugs is a complex phenomenon and a major challenge in cancer *** is becoming increasingly evident that a form of acquired drug resistance known as“adaptive resistance”is a common cause of treatment failure and patient relapse in many *** classical resistance mechanisms that are acquired via genomic alterations,adaptive resistance is instead driven by non-genomic changes involving rapid and dynamic rewiring of signalling and/or transcriptional networks following therapy,enabled by complex pathway crosstalk and feedback *** network rewiring allows tumour cells to adapt to the drug treatment,circumvent the initial drug challenge and continue to survive in the presence of the *** its great clinical importance,adaptive resistance remains largely under-studied and poorly *** review is focused on recent findings which provide new insights into the mechanisms underlying adaptive resistance in breast cancer,highlighting how breast tumour cells rewire intracellular signalling pathways to overcome the stress of initial targeted *** particular,we investigate adaptive resistance to targeted inhibition of two major oncogenic signalling axes frequently dysregulated in breast cancer,the PI3k-AkT-mTOR and RAS-MAPk signalling pathways;and discuss potential combination treatment strategies that overcome such *** addition,we highlight application of quantitative and computational modelling as a novel integrative and powerful approach to gain network-level understanding of network rewiring,and rationally identify and prioritise effective drug combinations.

The metal-organic framework(MOF)derived Ni–Co–C–N composite alloys(NiCCZ)were“embedded”inside the carbon cloth(CC)strands as opposed to the popular idea of growing them upward to realize ultrastable energy storage and conversion *** NiCCZ was then oxygen functionalized,facilitating the next step of stoichiometric sulfur anion diffusion during hydrothermal sulfurization,generating a flower-like metal hydroxysulfide structure(NiCCZOS)with strong partial implantation inside *** obtained NiCCZOS shows an excellent capacity when tested as a supercapacitor electrode in a three-electrode ***,when paired with the biomass-derived nitrogen-rich activated carbon,the asymmetric supercapacitor device shows almost 100%capacity retention even after 45,000 charge–discharge cycles with remarkable energy density(59.4 Wh kg^(-1)/263.8μWh cm^(–2))owing to a uniquely designed ***,the same electrode performed as an excellent bifunctional water-splitting electrocatalyst with an overpotential of 271 mV for oxygen evolution reaction(OER)and 168.4 mV for hydrogen evolution reaction(HER)at 10 mA cm−2 current density along with 30 h of unhinged chronopotentiometric stability performance for both HER and ***,a unique metal chalcogenide composite electrode/substrate configuration has been proposed as a highly stable electrode material for flexible energy storage and conversion applications.

Background In Viet Nam, tuberculosis (TB) represents a devastating life-event with an exorbitant price tag, partly due to lost income from daily directly observed therapy in public sector care. Thus, persons with TB may seek care in the private sector for its flexibility, convenience, and privacy. Our study aimed to measure income changes, costs and catastrophic cost incurrence among TB-affected households in the public and private *** Between October 2020 and March 2022, we conducted 110 longitudinal patient cost interviews, among 50 patients privately treated for TB and 60 TB patients treated by the National TB Program (NTP) in Ha Noi, Hai Phong and Ho Chi Minh City, Viet Nam. Using a local adaptation of the WHO TB patient cost survey tool, participants were interviewed during the intensive phase, continuation phase and post-treatment. We compared income levels, direct and indirect treatment costs, catastrophic costs using Wilcoxon rank-sum and chi-squared tests and associated risk factors between the two cohorts using multivariate *** The pre-treatment median monthly household income was significantly higher in the private sector versus NTP cohort (USD 868 vs USD 578;P=0.010). A Scalable Pillar Layered Metal-Organic Framework for Natural Gas Purification

One-step harvest of high-purity methane(CH_(4))from ternary propane/ethane/methane(C_(3)H_(8)/C_(2)H_(6)/CH_(4))mixtures remains a desirable yet challenging goal for natural gas ***,adsorbents either suffer from high capacity and selectivity,or are caught in a dilemma of scalable ***,we demonstrate a scalable pillar layered metal-organic framework Ni-MOF for highly efficient one-step CH_(4)***-MOF exhibits high C_(2)H_(6)and C_(3)H_(8)uptakes of 83.3 and 86.1 cm^(3)/g at 298 k and 100 kPa and remarkable C_(2)H_(6)/CH_(4)(50/50,volume ratio,21.5)and C_(3)H_(8)/CH_(4)(50/50,volume ratio,212.0)***,high C_(2)H_(6)(42.2 cm^(3)/g at 10 kPa)and C_(3)H_(8)(64.7 cm^(3)/g at 5 kPa)capacities in the low-pressure region at 298 k were realized on Ni-MOF,suggesting the strong affinities of Ni-MOF towards C_(2)H_(6)and C_(3)H_(8).Furthermore,the dynamic breakthrough experiments revealed that purifying CH_(4)from natural gas in one-step can be achieved in Ni-MOF with high-purity(>99.8%)and productivity(346.0 cm^(3)/g).Most significantly,the production of Ni-MOF can be scalably synthesized at room temperature,rendering it promising potential for industrial *** combined advantages of exceptional separation performance,scalability,and cycle stability of Ni-MOF pave the way for one-step CH_(4)purification from natural gas.

In the present work,a multi-element nanoglass(m-NG)of FeCoCrMoCBY is obtained first time by the laser ablation combined with inert gas condensation(laser-IGC)*** with the conventional rapid-quenched metallic glass(MG)with identical composition,the Fe-based m-NG demonstrates a superior performance as a self-supported electrocatalyst for hydrogen evolution reaction(HER)in acidic *** enhanced HER activity of m-NG is proposed to be closely related to its high en-ergy states,which is originated from the unique inhomogeneous nanostructures with a high density of low-coordinated ***,the Fe-based m-NG exhibits an outstanding comprehensive catalytic performance even beyond the commercial Pt/C catalyst in long-term test due to its self-optimization *** work not only opens the way to the preparation of m-NGs by the novel laser-IGC technique,but also makes a great contribution to developing low-cost,high-efficient,and super-durable HER electrocat-alysts in acidic environment.

Gradient-induced long-range optical pulling force based on photonic band gap

Optical pulling provides a new degree of freedom in optical *** is generally believed that long-range optical pulling forces cannot be generated by the gradient of the incident ***,we theoretically propose and numerically demonstrate the realization of a long-range optical pulling force stemming from a self-induced gradient field in the manipulated *** analogy to potential barriers in quantum tunnelling,we use a photonic band gap design in order to obtain the intensity gradients inside a manipulated object placed in a photonic crystal waveguide,thereby achieving a pulling *** the usual scattering-type optical pulling forces,the proposed gradient-field approach does not require precise elimination of the reflection from the manipulated *** particular,the Einstein-Laub formalism is applied to design this unconventional gradient *** magnitude of the force can be enhanced by a factor of up to 50 at the optical resonance of the manipulated object in the waveguide,making it insensitive to *** developed approach helps to break the limitation of scattering forces to obtain longrange optical pulling for manipulation and sorting of nanoparticles and other *** developed principle of using the band gap to obtain a pulling force may also be applied to other types of waves,such as acoustic or water waves,which are important for numerous applications.

NO_(x) reduction against alkali poisoning over Ce(SO_(4))_(2)-V_(2)O_(5)/TiO_(2) catalysts by constructing the Ce^(4+)-SO_(4)^(2−)pair sites

Commercial V_(2)O_(5)-based catalysts have been successfully applied in NH_(3) selective catalytic reduction(NH_(3)-SCR)of NO_(x) from power stations,but their poor alkali-resistance restrains the wider application in nonelectrical *** this study,NO_(x) reduction against alkali poisoning over V_(2)O_(5)/TiO_(2) is greatly improved via Ce(SO_(4))_(2) *** has been originally demonstrated that Ce^(4+)-SO_(4)^(2−)pair sites play crucial roles in improving NO_(x) reduction against alkali poisoning over V_(2)O_(5)/TiO_(2) *** strong interaction between V species and Ce sites of Ce^(4+)-SO_(4)^(2−)pairs triggers the reaction between NH_(4)^(+) species and gaseous NO via Eley-Rideal(E-R)reaction *** k-poisoning,the SO_(4)^(2−)sites of Ce^(4+)-SO_(4)^(2−)pairs as protective sites strongly bond with k and thus maintain the high reaction efficiency via the E-R reaction *** work demonstrates an effective strategy to enhance NO_(x) reduction against alkali poisoning over catalysts via constructing Ce^(4+)-SO_(4)^(2−)pair sites,contributing to developing alkali-resistant SCR catalysts for practical application in nonelectrical industries.

BACkGROUND Liuweiwuling Tablet(LWWL)is a Chinese patent medicine approved for the treatment of chronic inflammation caused by hepatitis B virus(HBV)*** studies have indicated an anti-HBV effect of LWWL,specifically in terms of antigen inhibition,but the underlying mechanism remains *** To investigate the potential mechanism of action of LWWL against *** In vitro experiments utilized three HBV-replicating and three non-HBV-replicating cell *** in vivo experiment involved a hydrodynamic injectionmediated mouse model with HBV *** and metabolomics were used to investigate the underlying mechanisms of action of *** In HepG2.1403F cells,LWWL(0.8 mg/mL)exhibited inhibitory effects on HBV DNA,hepatitis B surface antigen and pregenomic RNA(pgRNA)at rates of 51.36%,24.74%and 50.74%,*** inhibition rates of LWWL(0.8mg/mL)on pgRNA/covalently closed circular DNA in HepG2.1403F,HepG2.2.15 and HepG2.A64 cells were 47.78%,39.51%and 46.74%,*** of transcriptomics and metabolomics showed that the anti-HBV effect of LWWL was primarily linked to pathways related to apoptosis(PI3k-AkT,CASP8-CASP3 and P53 pathways).Apoptosis flow analysis revealed that the apoptosis rate in the LWWL-treated group was significantly higher than in the control group(CG)among HBV-replicating cell lines,including HepG2.2.15(2.92%±1.01%vs 6.68%±2.04%,P0.05),L02 cells(5.49%±0.80%vs 5.48%±1.01%,P>0.05)and LX2 cells(6.29%±1.54%vs 6.29%±0.88%,P>0.05).TUNEL staining revealed a significantly higher apoptosis rate in the LWWL-treated group than in the CG in the HBVreplicating mouse model,while no noticeable difference in apoptosis rates between the two groups
