Nano-encapsulation is a platform which offers a promising application for control release and the delivery of drugs in pharmaceuticals and antioxidant/ antimicrobial in food systems. Poly (lactic-co-glycolide acid) (PLGA) is a biodegradable and biocompatible co-polymer of lactic acid and glycolic acid which is used for synthesizing food based polymeric nanoparticles (NP). The aim of this study was to evaluate the morphological and physicochemical properties and the controlled release of bioactive components derived from Aloe vera gel loaded PLGA NP. The results shows the mean hydrodynamic diameter of the unloaded NP is 103 nm which is significantly (p han the loaded freeze dried powered gel (FDG) (147 nm) and liquid gel (LG) (221 nm) and the particle size distribution given by the Poly-dispersity Index were 0.2, 0.2 and 0.3, respectively. The zeta potential for unloaded, FDG and LG NP were ±60, ±28 and ±22 mV, respectively, hence were electrokinetically stable NP. No significant (p > 0.05) inhibition of the antioxidant potential was observed with loaded NP. The entrapment efficiency for the FDG synthesized was 87%, and the burst effect was observed after 4 h as a result of the encapsulation effect. The release kinetics of bioactive is govern by the combination of mass diffusion and capillary action.

Biosynthesis of silver nanoparticles (AgNP) using biomolecular extracts of plant origin is of interest to many researchers because of their potential to produced stable product. It is hypothesized that laser ablation of the AgNP in solution will enhance the biomolecules such as aliphatic amines, alkenes (=C-h), alkanes (C-h), alcohol (O-h) and unsaturatedesters (C-O) in the Nano emulsion. hence, the objective of this study was to evaluate the effect of laser oblation on the physicochemical properties and stability of AgNP reduced with Aloe vera (Aloe barbadensis ). Experiments were conducted with the pre-made AgNP were subjected to the three laser oblation treatments: 1) Control no laser treatment of AgNP, 2) Laser ablation treatment for 5 minutes and 3) Laser ablation treatment for 10 minutes. The results of the analysis show that laser oblation treatment has significant effect (p he concentration of AgNP. The intensity of the absorption peak significantly (p h laser exposure time. While 214 ppm was observed with no laser treatment, 224 and 229 ppm increase of concentration was observed when laser treated for 5 and 10 min. The rates of reaction of restructuring the particles sizes were 0.384, 0.408 and 0.4288 min-1 at different laser exposure treatments times 0, 5 and 10 min, respectively. The FTIR results show significant (p hatic amines, alkenes (=C-h), alkanes (C-h), alcohol (O-h) and unsaturatedesters (C-O).

The Relative Prevalence of Typhoid and Malaria in Febrile Patients in Freetown, Sierra Leone

Malaria and typhoid fever are debilitating diseases responsible for the deaths of thousands of lives annually. Over the last decade both diseases have received increasing attention with particular focus on malaria. however, recent reports indicate an increasing co-infection rate between mala- ria and typhoid fever. Familiar signs and symptoms of both diseases have led to the belief in some communities that it’s a new disease. In this study, we investigated the relationship between mala- ria and typhi O and h antigens among patients reporting with fever. Patients reporting with fever from April 2013-March 2014 were recruited for the study. Our results showed that there was no association between having malaria and typhoid fever infection. however, having fever was asso- ciated with having both diseases. Also, fever among patients was more likely to be caused by Sal- monella typhi O and h antigens than plasmodium parasites. Infections of both diseases were higher in wet season than in dry season. The study recommends that malaria and typhoid fever intervention programs are increased in endemic areas. Also attention should be paid to typhoid fever infection rates in the country.

The present study aims to investigate the equation of state(EOS)parameters of CaMg3 in aReCh(D09),AIFR(DO3),CU3A11(LI2)and CuTi3(L60)structures,using full potential linear muffin-tin orbitals(FP-LMTO)approach based on the density functional theory(DFT).The local density approximation(LDA)and the generalized gradient approximation(GGA)were both applied for the exchange-correlation potential *** calculated equation of slate parameters at equilibrium,in general,agreed well with the available data of the *** calculations showed that under compression CaMg3 transforms from DO3 to DO9 at about 29.96GPa,and 25.1 GPa using LDA and GGA,*** elastic constants C,y,aggregate moduli,Vickers hardness,sound velocity,and Debye temperature of CaMg3 in D03 structure were also reported,discussed and *** LDA(GGA),the calculated values of hv andθD were found at around 5.80GPa(5.93GPa)and 393.44 K(389.91 K),*** band structure,total density of states(TDOS)as well as the partial density of states(PDOS)have been also *** electronic band structure confirms the metallic behavior of CaMg3 in DO3 phase,the valence bands are dominated by the maximum contribution of‘d’like states of Ca in the energy ranging from 2 to 3 eV for GGA,and from 4.5 to 5 eV for LDA,respectively.

目的:制备hutchinson-Gilford早老症转基因小鼠模型,观察突变的核纤层蛋白A对小鼠受精卵着床和成活情况的影响。方法:实验于2004-09/2007-04在北京大学医学部医学遗传学系和中国农业大学生物技术国家重点实验室完成。①实验方法:将缺失了第11外显子末端150个碱基的LMNA基因cDNA克隆到pcDNA3.1载体,应用原核显微注射方法将线性化的pcDNA3.1-mutant LMNA注射到小鼠受精卵的雄原核,再将注射后的受精卵移植至假孕鼠的输卵管。②实验评估:观察受精卵的着床和成活情况。通过聚合酶链反应检测着床的胚胎,鉴定有无阳性转基因动物。结果:①178只注射pcDNA3.1-mutant LMNA后的存活受精卵移植至假孕鼠输卵管后,出生13只子代鼠,出生率只有7%;58只注射pcDNA3.1-mutant LMNA后的存活受精卵移植到假孕鼠输卵管后,在移植后12￣15d的受体母鼠子宫内收集到1只活胚和5只死胚。②聚合酶链反应方法检测所收集到的胚胎,没有得到pcDNA3.1-mutant LMNA阳性转基因鼠。在着床的胚胎中也没有检测到pcDNA3.1-mutant LMNA。结论:过量表达的突变核纤层蛋白A对小鼠受精卵有致死性,只有无目的基因整合的受精卵才能存活至出生。

Restrictive dermopathy (RD) is a lethal human genetic disorder characterized by very tight, thin, easily eroded skin, rocker bottom feet, and joint contractures. This disease was recently reported to be associated with a single heterozygous mutation in ZMPSTE24 and hypothesized to be a digenic disorder (Navarro et al, lamin A and ZMPSTE24 (FACE-1) defects cause nuclear disorganization and identify restrictive dermopathy as a lethal neonatal laminopathy. hum Mol Genet 13:2493- 2503, 2004). ZMPSTE24 encodes an enzyme necessary for the correct processing and maturation of lamin A, an intermediate filament component of the nuclear envelope. here we present four unrelated patients with homozygous mutations in ZMPSTE24 and a fifth patient with compound heterozygous mutations in ZMPSTE24. Two of the three different mutations we found are novel, and all are single base insertions that result in messenger RNA frameshifts. As a consequence of the presumed lack of ZMPSTE24 activity, prelamin A, the unprocessed toxic form of lamin A, was detected in the nuclei of both cultured cells and tissue from RD patients, but not in control nuclei. Abnormally aggregated lamin A/C was also observed. These results indicate that RD is an autosomal recessive laminopathy caused by inactivating ZMPSTE24 mutations that result in defective processing and nuclear accumulation of prelamin A.

lamin A and C are components of the nuclear envelope, located at the nucleoplasmatic surface of the inner nuclear membrane within cells. Recently,mutations within LMNA encoding lamin A/C have been associated with various disease entities including cardiomyopathy. We screened heart transplant recipients suffering from dilated cardiomyopathy(DCM) with a positive family history of LMNA mutations. Four index patients and one relative belonging to four unrelated families carrying LMNA mutations were identified. The mutations p.Q355X and p.S22L have not been reported before, whereas p.R190W has already been reported in other studied DCM cohorts. In the patients of the present study, the mean age at manifestation of heart disease was 37.6 years(range 30- 45 years), with progression to end- stage heart failure requiring transplantation at a mean age of 45.8 years(range 35- 54 years). Three patients presented initially with atrial fibrillation. These data confirm the involvement of LMNA mutations in patients with DCM and extend the mutational spectrum of LMNA. The p.R190W mutation has been reported in different populations and may therefore be useful for analyzing the impact of a specific LMNA mutation on the phenotype of muscle disease.
