Hypoxic and temporal variation in the endocrine disrupting toxicity of perfluorobutanesulfonate in marine medaka(Oryzias melastigma)

Perfluorobutanesulfonate(PFBS)is an emerging pollutant capable of potently disrupting the sex and thyroid endocrine systems of ***,the hypoxic and temporal variation in PFBS endocrine disrupting toxicity remain largely *** the present study,adult marine medaka were exposed to environmentally realistic concentrations of PFBS(0 and 10μg/L)under normoxia or hypoxia conditions for 7 days,aiming to explore the interactive behavior between PFBS and *** addition,PFBS singular exposure was extended till 21days under normoxia to elucidate the time-course progression in PFBS *** results showed that hypoxia inhibited the growth and caused the suspension of egg spawn regardless of PFBS *** regard to the sex endocrine system,7-day PFBS exposure led to an acute stimulation of transcriptional profiles in females,which,subsequently,recovered after the 21-day *** potency of hypoxia to disturb the sex hormones was much stronger than PFBS.A remarkable increase in estradiol concentration was noted in medaka blood after hypoxia *** in sex endocrinology of coexposed fish were largely determined by hypoxia,which drove the formation of an estrogenic *** further enhanced the endocrine disrupting effects of ***,the hepatic synthesis of vitellogenin and choriogenin,two commonly used sensitive biomarkers of estrogenic activity,failed to initiate in response to the estrogen *** to sex endocrine system,disturbances in thyroidal axis by PFBS or hypoxia were relatively ***,the present findings will advance our toxicological understanding about PFBS pollutant under the interference of hypoxia.

Traditional Chinese medicine （TCM） relies on the combined effects of herbs within prescribed formulae. However, given the combinatorial explosion due to the vast number of herbs available for treatment, the study of these combined effects can become computationally intractable. Thus feature selection has become increasingly crucial as a pre-processing step prior to the study of combined effects in TCM informatics. In accord with this goal, a new feature se- lection algorithm known as a co-evolving memetic wrapper （COW） is proposed in this paper. COW takes advantage of recent research in genetic algorithms （GAs） and memetic al- gorithms （MAs） by evolving appropriate feature subsets for a given domain. Our empirical experiments have demonstrated that COW is capable of selecting subsets of herbs from a TCM insomnia dataset that shows signs of combined effects on the prediction of patient outcomes measured in terms of classification accuracy. We compare the proposed algorithm with results from statistical analysis including main effects and up to three way interaction terms and show that COW is capable of correctly identifying the herbs and herb by herb effects that are significantly associated to patient outcome prediction.

Leptin, a protein hormone produced by the adipocytes, has long been recognized to regulate metabolism, neuroendorine and other physiological functions. Early findings of increased leptin production during infection and inflammation and dysregulated immune response in leptin signaling-deficient mice provide strong evidence for the involvement of leptin in the immune responses. Recent data have established the regulatory function for leptin in immunity similar to the function of a pro-inflammatory cytokine, while gene-targeting studies also demonstrated an essential role of leptin in regulating hematopoiesis and lymphopoiesis. Moreover, there has been increasing evidence that leptin is involved in the pathogenesis of various autoimmune diseases. This review discusses recent advances in understanding the role of leptin in immunity and leptin-signaling pathways involved in modulating immune homeostasis and autoimmune pathogenesis. S5-1 Effects of persistent organic pollutants(POPs) in water on Chongqing population health CAO Jia, TIAN Huan-Jun, SHU Wei-Qun(Preventive Medicine College, the Third Military Medical University, Chongqing 400038, China)Abstract:Many organic pollutants occur in drinking water in considerable amounts, and mimic or interfere with reproductive development and function. Furthermore, it is not known whether multiple organic pollutants (MOP) from drinking water induce uterotrophic effects. In this study, we compared the effects of MOP on uterine wet weight by different exposure approaches (ip, sc, po) on prepubertal 21-d-old Wistar rats and Kunming mice. The multiple organic pollutants in drinking water were concentrated by solid-phase extraction (SPE), and dissolved in DMSO.

Steroid hormones, alkylphenol ethoxylates（APEOs） and phthalic acid esters（PAEs） are emerging endocrine disrupting chemicals（EDCs） that can interfere with the endocrine function in organisms at low concentrations. The occurrence, distribution behavior, removal rate and the fate of 31 target EDCs in sewage treatment plants, which consist of various treatment facilities and receiving water in Hong Kong, were investigated. Estrone,nonylphenol-di-ethoxylate and diethyl phthalate were found to be dominant in each group of influent samples with concentrations ranging from 11–33, 747–3945 and 445–4635 ng/L,respectively. Conversely, progesterone, nonylphenol-mono-ethoxylate and bis（2-ethylhexyl）phthalate were the most abundant in dewatered sludge, with concentrations ranging from 0.9–237, 75–19,743 and 4310–37,016 ng/g（dry weight）. The removal rates of primary sedimentation and disinfection approaches were lower than 30% for most of the chemicals,while those of activated sludge and reverse osmosis were greater than 80% for more than two-thirds of the compounds, noticeably decreasing the estrogenic risk of sewage discharged into the environment. Steroid hormones were removed via biological degradation, while some APEOs and PAEs adsorbed to the sludge. Victoria Harbor poses a low to medium estrogenic risk mainly contributed by estrone and estradiol and deserves attention.

Prevacuolar compartments （PVCs） and endosomal compartments are membrane-bound organelles mediating protein traffic to vacuoles in the secretory and endocytic pathways of plant cells. Over the years, great progress has been made towards our understanding in these two compartments in plant cells. In this review, we will summarize our contributions toward the identification and characterization of plant prevacuolar and endosomal compartments. Our studies will serve as important steps in future molecular characterization of PVC biogenesis and PVC-mediated protein traffickinq in plant cells.

BACKGROUND: Chemoembolization with doxorubucin-eluting beads (DEB) has been used to treat hepatocellular carcinoma (HCC) since 2007. This study compared the efifcacy and sur-vival between transarterial chemoembolization (TACE) with DEB and conventional approach (cTACE) in HCC treatment. METHODS: This retrospective case-control study compared the overall survival and tumor response of HCC patients to cTACE (n=190) and DEB (n=143) by the reassessment of com-puted tomography and serum alpha-fetoprotein (AFP). Mul-tivariate analysis was used to determine the factors affecting tumor response. RESULTS: The median post-treatment to pre-treatment AFP level was 0.8 for a DEB session (n=258) and 1.0 for a cTACE session (n=452), showing a signiifcantly greater decrease in AFP after DEB (P80 months after the ifrst TACE session in the DEB group. CONCLUSION: DEB is a safe alternative to cTACE in HCC pa-tients with better therapeutic efifcacy.
