Harnessing the potential of gene editing technology using CRISPR in inflammatory bowel disease

The molecular scalpel of clustered regularly interspersed short palindromic repeats/CRISPR associated protein 9(CRISPR/Cas9) technology may be sharp enough to begin cutting the genes implicated in inflammatory bowel disease(IBD) and consequently decrease the 6.3 billion dollar annual financial healthcare burden in the treatment of IBD. For the past few years CRISPR technology has drastically revolutionized DNA engineering and biomedical research field. We are beginning to see its application in gene manipulation of sickle cell disease,human immunodeficiency virus resistant embryologic twin gene modification and IBD genes such as Gatm(Glycine amidinotransferase, mitochondrial),nucleotide-binding oligomerization domain-containing protein 2, KRT12 and other genes implicated in adaptive immune convergence pathways have been subjected to gene editing, however there are very few publications. Furthermore,since Crohn's disease and ulcerative colitis have shared disease susceptibility and share genetic gene profile, it is paramount and is more advantageous to use CRISPR technology to maximize impact. Although, currently CRISPR does have its limitations due to limited number of specific Cas enzymes, off-target activity,protospacer adjacent motifs and crossfire between different target sites. However,these limitations have given researchers further insight on how to augment and manipulate enzymes to enable precise gene excision and limit crossfire between target sites.

Micro RNAs potential utility in colon cancer: Early detection, prognosis, and chemosensitivity

Over the past decade, research has shown that aberrant expression of micro RNA(mi RNA) is involved in colorectal cancer development and progression. Micro RNAs are small sequences of non-coding RNA that regulate expression of genes involved in important cellular functions, such as cell differentiation, multiplication, and apoptosis. A specific mi RNA may display the effects of a tumor suppressor or oncogene. Altered mi RNA expression is found in colorectal cancer(CRC) and patterns of mi RNA expression correlate with CRC detection and outcome. Studies also have examined the use of circulating serum mi RNA and fecal mi RNA expression as non-invasive markers for early detection. Here, we review recent evidence demonstrating the potential role of mi RNA in CRC and the implications of its use in the diagnosis, prognosis, and management of CRC.

Phytosociology of stratification in a lowland tropical rainforest occurring north of the Tropic of Cancer in Meghalaya, India

Stratification in lowland rainforests of Meghalaya,India,which represent the westernmost limit of the rainforests north of the Tropic of Cancer,was studied in horizontal and vertical planes to elucidate the patterns in stacking of species diversity and community attributes,and to draw comparisons with rainforests of‘Indo-Malaya’ecozone(biogeographical realm).All individuals≥10 cm GBH(girth at breast height)were enumerated in six transects of 10 m width and up to 500 m length covering 2.45 ha area.The stratification of whole assemblage of species in vertical plane is referred to as‘storey structure’(=height class distribution),which explains structural complexity.In horizontal plane,it is typically referred to as‘stand structure’(=girth class distribution),which explains structural heterogeneity.The stratification of an individual species in vertical plane is referred to as‘loftiness’and in horizontal plane,it is frequently referred to as‘population structure’.The stand structure was characterized by a negative exponential relationship or a reverse J-shaped curve,which is typical of a well regenerating forest stand.The storey structure was characterized by the low stature of the rainforest(<30 m),subtle layering in the canopy with dearth of discrete multi-stories,narrower widths of the stories and a thick understory.The cluster analysis and‘candlestick charts’showed that the dominant species spatially segregated in canopy height to profile three strata,viz.,an understory of juveniles and shrubs below 5 m,a middle-storey of intermediate trees between 5 and 15 m,and an overstory of large trees between 15 and 25 m.Emergent trees of a few species may reach up to 30 m.The individuals tended to scatter in increasingly wider range of height classes from a lower to the next higher girth class,creating a‘torchlight scatter’pattern,with a tendency to concentrate towards smaller height classes due to stochastic factors,which potentially influence vertical growth proportionate to diametric growth.In comparison to equatorial r

In-plane and out-of-plane quasi-static compression performance enhancement of 3D printed re-entrant diamond auxetic metamaterial with geometrical tuning and fiber reinforcement

Auxetic materials are cellular materials with a unique property of negative Poisson’s ratio.The auxeticity and performance of these metamaterials utterly depend on the geometrical parameters and loading direction.For the first time,the quasi-static uniaxial compression performance of fused filament fabricated re-entrant diamond auxetic metamaterial is evaluated in the x-direction(in-plane)and z-direction(out-of-plane).The most commonly used thermoplastic feedstock,Acrylonitrile butadiene styrene,is considered a material of choice.The effect of influential geometrical parameters of the re-entrant diamond structure and printing parameter is systematically studied using Taguchi’s design of experiments.Grey-based multi-objective optimisation technique has been adopted to arrive at the optimal structure.Efforts are made to improve the stiffness and strength of the structure with fibre reinforcements.Micro glass fibre reinforcements have enhanced specific strength and stiffness in both in-plane and out-ofplane directions.A sevenfold and thirteen times increase in specific strength and energy absorption is evident for glass fibre-reinforced structures in out-of-plane directions compared to in-plane ones.Proper tuning of geometrical parameters of the re-entrant diamond structure can result in a Poisson’s ratio of up to-3.49 when tested in the x-direction.The parametric study has illustrated the tailorability of the structure according to the application requirements.The statistical study has signified each considered parameter’s contribution to the compression performance characteristics of the 3D printed re-entrant diamond auxetic metamaterial.

Alkali activated slag-fly ash concrete incorporating precious slag as fine aggregate for rigid pavements

This study aims to develop the alkali activated slag-fly ash concrete(AASFC) using precious slag(PS) ball as the fine aggregate. The slag-fly ash proportion, sodium silicate/sodium hydroxide(SS/SH) ratio and sodium hydroxide concentration are the prime variables. The fresh concrete properties like workability, setting time and the hardened concrete properties of AASFC developed under ambient curing were analyzed and similar way by replacing with fly ash(FA). The sodium hydroxide concentration of 8 M, 10 M and 12 M, SS/SH ratio of 1.0, 1.5 and 2.0, alkaline liquid to binder ratio of 0.35 and variation in slag-fly ash proportion as major inputs for mix design. The test results showed that, the fresh state of AASFC blend has a shorter setting time of 12-16 min, because of calcium mineral from the FA, the workability and setting time of AASFC mix was enhanced by the addition of the trisodium phosphate as a retarder. At the higher SS/SH extent, the workability and setting time exaggerated, whereas the increased FA content and sodium hydroxide concentration had a greater influence on the fresh state of concrete. Also, the sodium hydroxide concentration, SS/SH ratio and slag-fly ash proportion are the most inducing parameters on the workability, setting time and strength parameters of AASFC.

Dynamics of some new solutions to the coupled DSW equations traveling horizontally on the seabed

The system of(1+1)-coupled Drinfel’d-Sokolov-Wilson equations describes the surface gravity waves travelling horizontally on the seabed.The objective of the present research is to construct a new variety of analytical solutions for the system.The invariants are derived with the aid of Killing form by using the optimal algebra classification via Lie symmetry approach.The invariant solutions involve time,space variables,and arbitrary constants.Imposing adequate constraints on arbitrary constants,solutions are represented graphically to make them more applicable in designing sea models.The behavior of solutions shows asymptotic,bell-shaped,bright and dark soliton,bright soliton,parabolic,bright and kink,kink,and periodic nature.The constructed results are novel as the reported results[26,28,29,30,33,38,42,49]can be deduced from the results derived in this study.The remaining solutions derived in this study,are absolutely different from the earlier findings.In this study,the physical character of analytical solutions of the system could aid coastal engineers in creating models of beaches and ports.

Codimension-2 bifurcation in a discrete predator-prey system with constant yield predator harvesting

This work investigates the bifurcation analysis in a discrete-time Leslie-Gower predatorprey model with constant yield predator harvesting.The stability analysis for the fixed points of the discretized model is shown briefy.In this study,the model undergoes codimension-1 bifurcation such as fold bifurcation(limit point),flip bifurcation(perioddoubling)and Neimark-Sacker bifurcation at a positive fixed point.Further,the model exhibits codimension-2 bifurcations,including Bogdanov-Takens bifurcation and generalized fip bifurcation at the fixed point.For each bifurcation,by using the critical normal form coefficient method,various critical states are calculated.To validate our analytical findings,the bifurcation curves of fixed points are drawn by using MATCONTM.The system exhibits interesting rich dynamics including limit cycles and chaos.Moreover,it has been shown that the predator harvesting may control the chaos in the system.

Decision Tree Based Key Management for Secure Group Communication

Group communication is widely used by most of the emerging network applications like telecommunication,video conferencing,simulation applications,distributed and other interactive systems.Secured group communication plays a vital role in case of providing the integrity,authenticity,confidentiality,and availability of the message delivered among the group members with respect to communicate securely between the inter group or else within the group.In secure group communications,the time cost associated with the key updating in the proceedings of the member join and departure is an important aspect of the quality of service,particularly in the large groups with highly active membership.Hence,the paper is aimed to achieve better cost and time efficiency through an improved DC multicast routing protocol which is used to expose the path between the nodes participating in the group communication.During this process,each node constructs an adaptive Ptolemy decision tree for the purpose of generating the contributory key.Each of the node is comprised of three keys which will be exchanged between the nodes for considering the group key for the purpose of secure and cost-efficient group communication.The rekeying process is performed when a member leaves or adds into the group.The performance metrics of novel approach is measured depending on the important factors such as computational and communicational cost,rekeying process and formation of the group.It is concluded from the study that the technique has reduced the computational and communicational cost of the secure group communication when compared to the other existing methods.

How do project-oriented organizations enhance innovation?An institutional theory perspective

A project-oriented organization is a hybrid form of organization where a functional hierarchy is augmented with structures to manage projects strategically across the organization.Six project-oriented organizations from diverse industries that emphasize innovation in their strategies were selected for this study.We use the three pillars of institutional theory—regulative,normative and cultural-cognitive—and institutional entrepreneurship to analyze interview data from executive and project,program and portfolio management personnel in the project-oriented organizations to investigate how innovation is facilitated through external influences and internal responses.Our findings indicate that processes and new structures provide effective ways for innovation and,while individuals are important,processes are more effective than individuals at enabling innovation.We put forward some lessons for practice that emerge directly from the findings,including suggestions on improving allocation of resources,a need to focus on processes,reconceptualization of“failure”,and dedicated investment in market knowledge,customer knowledge,performance metrics and flexible governance structures.

A hierarchical analytical model for performance management of integrated logistics

In this paper,a holistic hierarchical analytical model is proposed to assess the performance of enablers in an integrated logistics system.Due to the ambiguous and complex environment,various refinements are needed to assess enablers and prioritize for the criteria such as economic,operational,and environment.The proposed hierarchical model is developed by a systematic approach that includes fuzzy analytical hierarchy process(FAHP),triangular fuzzy numbers(TFN),an evidential reasoning algorithm(ERA),and expected utility theory(EUT).The FAHP is used to analyze and obtain the weights of the criteria by considering the expert’s opinions.ERA is used to synthesize the enablers based on the selected criteria.These enablers are represented using subjective assessment along with a set of evaluation grades for a qualitative attribute.EUT helps in obtaining crisp values of enablers for their performance estimation.With these set of methodologies,a hierarchical model is proposed that prevent low flexibility and inadequate appropriateness of the proposed model.Further,the model helps in scenario generation for the logistics professionals who are facing various problems in integrating logistics and incorporating sustainability due to lack of appropriate methodologies and evaluation techniques.Finally,sensitivity analysis is used for overall model validation.
