This study wa. concentra.ed on the biosynthesis of silver na.opa.ticles from Penicillium a.ra.tiogresium (IMI 89372) with a.focus on its cytotoxicity in MCF-7 a.d MCT ca.cer cell lines a. well a. Vero (norma.) cell line tha. wa. a.sessed by crysta. violet a.sa. a.ter trea.ment with va.ious concentra.ions (0.44 – 145 μg/ml) for 24 h. The cell morphology wa. exa.ined by inverted light microscopy. Further, the ra.iosensitizing effect of silver na.opa.ticles (a.NPs) on MCF-7 wa. a.so demonstra.ed by a.sessing cell morphology, cell prolifera.ion of MTT a.sa., LDH a.tivity a.d induction of a.optosis through checking of some a.optotic genes tha. a.tered during ca.cinogenesis, including ca.pa.e-3, Ba. a.d Bcl-2. Ca.pa.e-3 a.tivity wa. a.so estima.ed. Synthesis of a.NPs wa. determined by UV-Visible spectrum a.d it wa. further cha.a.terized by TEM, FT-IR a.d X-Ra. a.a.ysis (EDX, XRD). The biosynthesized a.NPs were spherica. a.d of 12.7 nm in size a. recorded by direct electron microscopy visua.iza.ion. The biosynthesized a.NPs showed va.ia.ion in cytotoxicity a.a.nst MCF-7, MCT a.d Vero cell lines in a.concentra.ion dependa.t response with a.va.ied degree of a.tera.ion in cell morphology. The result showed tha. a.NPs were highly toxic towa.ds MCF-7 with IC50 va.ue of 10.5 μg/ml. Trea.ment of MCF-7 (10.5 μg/ml) prior to irra.ia.ion improved the effect of irra.ia.ion dose (6 Gy) via.increa.ing a.tera.ion of cell morphology, inhibition of cell prolifera.ion, a.tiva.ion of the la.ta.e dehydrogena.e (LDH) a.d ca.pa.e-3 lea.ing to induction of a.optosis which wa. further confirmed through increa.ing nuclea. DNa.da.a.e a.d up regula.ion of ca.pa.e 3 a.d Ba. genes a.d downrgula.ion of Bcl-2 genes. In conclusion, the present findings clea.ly indica.ed tha. a.NPs showed dose dependa.t cytotoxicity a.d verified tha. a.NPs a.ted a. a.potent ra.iosensitizer a.d could enha.ce ga.ma.irra.ia.ion induced killing of MCF-7 brea.t ca.cer cells.

Introduction: There is growing evidence tha. genetic a.d environmenta. fa.tors pla. a. importa.t role in the development a.d progression of non-a.coholic fa.ty liver disea.e (Na.LD). We investiga.ed the a.socia.ion of single nucleotide polymorphism (SNP) rs738409 in PNPLa. gene a.d TNF-α G238a.polymorphism with the development a.d severity of Na.LD in a. overweight a.d obese Egyptia. popula.ion. Ma.eria. a.d Methods: 100 overweight a.d obese pa.ients with Na.LD a.d 30 control subjects were enrolled. a.l Na.LD pa.ients underwent a.confirma.ory biopsy. La.ora.ory investiga.ions included fa.ting pla.ma.glucose, kidney a.d liver function tests, liver enzymes, lipid profile a.d hepa.itis ma.kers. a.domina. ultra.ound wa. performed a.d a.l subjects were genotyped for (rs738409) PNPLa. a.d (rs361525) TNF-α gene polymorphisms using polymera.e cha.n rea.tion a.d restriction fra.ment length polymorphism (PCR-RFLP). Results: The homozygous GG genotype of the PNPLa. wa. most frequent a.ong pa.ients with Na.H (26%) a. compa.ed to borderline Na.H (20.5%) a.d simple stea.osis (20%). Higher serum levels of tra.sa.ina.es were observed in Na.LD pa.ients a.d controls who were ca.riers of the G a.lele of rs738409, but this wa. not sta.istica.ly significa.t. Rega.ding the TNF-α G238a.SNP;the frequency of the a.a.lele wa. significa.tly higher in Na.LD pa.ients (20%) compa.ed to controls (5%) (p va.ue = 0.006). The highest TNF G a.lele frequency wa. observed in the Na.H group (88%) a.d this wa. sta.istica.ly significa.t (p va.ue = 0.009). Conclusion: Our study confirmed the a.socia.ion of the PNPLa. (rs738409) a.d TNF-α promoter region G238a.polymorphisms with susceptibility to Na.LD a.d its progression.

Objective: To eva.ua.e the a.tioxida.t, immunomodula.ory a.d a.ti-infla.ma.ory a.tivities of pyrrolidine dithioca.ba.a.e a.d sa.a.liptin in ra.s with thioa.eta.ide-induced ulcera.ive colitis. Methods: a.ima.s were ora.ly a.ministered with a.vehicle, sulfa.a.a.ine(500 mg/kg), pyrrolidine dithioca.ba.a.e(100 mg/kg), a.d sa.a.liptin(10 mg/kg) for two weeks. Ulcera.ive colitis wa. induced by a.single intra.ecta. instilla.ion of thioa.eta.ide on da. 8. Colon sa.ples were collected to a.sess mitogen-a.tiva.ed protein kina.e(Ma.K), phosphoryla.ed extra.ellula. signa.-regula.ed kina.e(ERK), c a.P response element-binding protein(CREB), interleukin-12(IL-12), ca.pa.e-3, β-defensin, inducible nitric oxide syntha.e(i NOS) a.d gluca.on like peptide-1(GLP-1). Moreover, histopa.hologica. exa.ina.ion wa. performed. Results: Ra.s trea.ed with thioa.eta.ide ca.sed increa.es in colonic Ma.K, phosphoryla.ed ERK, CREB, ca.pa.e-3, IL-12, β-defensin, i NOS, a. well a. decrea.es in body weight a.d GLP-1. In a.dition, distortion of colonic structure wa. found by histopa.hologica. exa.ina.ion. Pyrrolidine dithioca.ba.a.e a.d sa.a.liptin mitiga.ed colitis severity by improving body weight decrea.e a.d GLP-1, a.d reducing colonic Ma.K, phosphoryla.ed ERK, CREB, ca.pa.e-3, IL-12, β-defensin a.d i NOS. a.M: To correla.e a.genetic polymorphism of the low-density lipoprotein(LDL) receptor with a.tivira. responses in Egyptia. chronic hepa.itis C virus(HCV) ***: Our study included 657 HCV-infected pa.ients with genotype 4 who received interferonba.ed combina.ion thera.y. Pa.ients were divided into two groups ba.ed on their response to thera.y: 356 were responders, a.d 301 were non-responders. Introduction: Insulin resista.ce (IR) is documented in pa.ients with chronic hepa.itis C(CHC), pla.s a. importa.t fa.tor in disea.e progression a.d predicting poor response to trea.ment. In chronic liver disea.e, levels of insulin-like growth fa.tor-1 (IGF-1) were correla.ed with IR. a.m: To eva.ua.e IR a.d serum levels of IGF-1 in Egyptia. pa.ients with CHC a.ter a.ti-vira. thera.y. Pa.ients a.d Methods: Forty biopsy-proven, non-dia.etic, CHC pa.ients, who received combined IFN/riba.irin thera.y, in a.dition to 10 hea.thy controls were studied. Serum levels of IGF-1, growth hormone (GH) were mea.ured a.d HOMa.IR(homeosta.is model a.sessment of IR), body ma.s index (BMI) were ca.cula.ed. Ba.eline da.a. retrieved from pa.ients’ files before initia.ion of thera.y, together with response to a.tivira. thera.y were a.a.yzed with respect to the mea.ured va.ia.les. Results: a.l pa.ients possessed a.significa.t higher HOMa.IR score (p = 0.02), GH levels (p a.d blood glucose levels (p a. controls while significa.tly lower levels of IGF-1 were found in pa.ients (p = 0.008)with no significa.t difference between responders a.d non-responders a. rega.ds the previously mentioned va.ia.les. HOMa.IR score wa. significa.tly correla.ed with GH levels, p = 0.003 a.d with IGF-1 levels in both responders a.d non responders. Low IGF-1 levels were a.socia.ed with increa.e in fibrosis sta.es a.d were significa.tly correla.ed with insulin(r = –0.499, p = 0.001). Conclusion: CHC pa.ients exhibit a. increa.ed HOMa.IR score reflecting the existence of IR irrespective of trea.ment response. Low IGF-1 levels were a.socia.ed with a.va.ced sta.es of fibrosis a.d thus could contribute to the progression of hepa.ic fibrosis in CHC.
