Bioenergetic alteration in gastrointestinal cancers:The good,the bad and the ugly

Cancer cells exhibit metabolic reprogramming and bioenergetic alteration,utilizing glucose fermentation for energy production,known as the Warburg ***,there are a lack of comprehensive reviews summarizing the metabolic reprogramming,bioenergetic alteration,and their oncogenetic links in gastrointestinal(GI)***,the efficacy and treatment potential of emerging anticancer drugs targeting these alterations in GI cancers require further *** review highlights the interplay between aerobic glycolysis,the tricarboxylic acid(TCA)cycle,and oxidative phosphorylation(OXPHOS)in cancer cells,as well as hypotheses on the molecular mechanisms that trigger this *** role of hypoxia-inducible transcription factors,tumor suppressors,and the oncogenetic link between hypoxia-related enzymes,bioenergetic changes,and GI cancer are also *** review emphasizes the potential of targeting bioenergetic regulators for anti-cancer therapy,particularly for GI *** the potential of targeting bioenergetic regulators for GI cancer therapy,the review categorizes these regulators into aerobic glycolysis/lactate biosynthesis/transportation and TCA cycle/coupled *** also detail various anti-cancer drugs and strategies that have produced pre-clinical and/or clinical evidence in treating GI cancers,as well as the challenges posed by these *** we highlight that understanding dysregulated cancer cell bioenergetics is critical for effective treatments,although the diverse metabolic patterns present challenges for targeted *** research is needed to comprehend the specific mechanisms of inhibiting bioenergetic enzymes,address side effects,and leverage high-throughput multi-omics and spatial omics to gain insights into cancer cell heterogeneity for targeted bioenergetic therapies.

Role of the zonal flow in multi-scale multi-mode turbulence with small-scale shear flow in tokamak plasmas

The structural characteristics of zonal flows and their roles in the nonlinear interaction of multi-scale multi-mode turbulence are investigated numerically via a self-consistent Landau-fluid *** multi-mode turbulence here is composed of a shorter wavelength electromagnetic(EM)ion temperature gradient(ITG)mode and a Kelvin-Helmholtz(KH)instability with long wavelengths excited by externally imposed small-scale shear *** strong shear flow,a prominent periodic intermittency of fluctuation intensity except for dominant ITG component is revealed in turbulence evolution,which onset time depends on the ion temperature gradient and the shear flow amplitudes corresponding to different KH *** is identified that the intermittency phenomenon results from the zonal flow dynamics,which is mainly generated by the KH mode and back-reacts on *** is demonstrated that the odd symmetric components of zonal flow(same symmetry as the external flow)make the radial parity of the KH mode alteration through adjusting the drift velocities at two sides of the resonant surface so that the KH mode becomes bursty ***,the ITG intermittency follows due to nonlinear mode *** dependences of the features of the intermittency are ***,associated turbulent heat transport is evaluated.

A new class of potent liver injury protective compounds: Structural elucidation, total synthesis and bioactivity study

A new class of potent liver injury protective compounds,phychetins A-D(1-4)featuring an unique 6/6/5/6/5 pentacyclic framework,were isolated and structurally characterized from a Chinese medicinal plant Phyllanthus *** 2-4 are three pairs of enantiomers that were initially obtained in a racemic manner,and were further separated by chiral HPLC *** 1-4 were proposed to be originated biosynthetically from a coexisting lignan via an intramolecular Friedel-Crafts reaction as the key step.A bioinspired total synthesis strategy was thus designated,and allowed the effective syntheses of compounds 2-4 in high *** of compounds exhibited significant anti-inflammatory activities in vitro via suppressing the production of pro-inflammatory cytokine IL-1β.Notably,compound 4,the most active enantiomeric pair in vitro,displayed prominent potent protecting activity against liver injury at a low dose of 3 mg/kg in mice,which could serve as a promising lead for the development of acute liver injury therapeutic agent.

Self-assembly ofπ-conjugated compounds into supramolecular polymers has received considerable attention because of their intrinsic scientific interests and technological *** compared toπ-conjugated rods,discotics,and macrocycles,propeller-shapedπ-conjugated molecules have been less exploited to form long-range-ordered supramolecular *** a novel type of supramolecular polymers has been constructed on the basis of propeller-shaped triphenylamine *** designed compound adopts nucleation-elongation cooperative mechanism for the supramolecular polymerization process,because of the participation of three-fold hydrogen bonds between the neighbouring *** supramolecular polymeric state displays amplified chirality and enhanced emission than those in the monomeric *** resulting supramolecular polymers exhibit severe emission quenching upon addition of 2,6-dinitrotoluene,ascribed to photoinduced electron transfer from the triphenylamine cyanostilbenes to the explosive *** current study proves the feasibility to supramolecular polymerize propeller-likeπ-conjugated molecules,serving as a promising type of explosive sensor owing to their guest encapsulation and signal amplification capabilities.

Study on the Blood Gas Sensor Based on Optical Absorption Technology

In this paper,the sensitivity shifts by the bicarbonate buffer was *** carbon dioxide diffuses into the indicator, the carbon acid is produced and the bicarbonate buffer is used to control internal *** the sensitivity of the pCO_2 sensor is increased by controlling the concentration of the bicarbonate buffer,the bicarbonate buffer is used to obtain the optimum resolution in this ***,the different concentrations of the bicarbonate buffer for blood gas measurements are discussed in detail.

Impact of direct-acting antivirals on de novo occurrence of hepatocellular carcinoma in hepatitis C virus patients

Hepatitis C Virus(HCV)infection constitutes a significant burden to world health,leading to liver cirrhosis and hepatocellular carcinoma(HCC).In the past decades,pegylated interferon combined with ribavirin has been used extensively for HCV treatment,and interferon(IFN)is thought to have antitumor ***-acting antivirals(DAAs)have fundamentally changed HCV therapy,due to their high efficacy and ***,recent studies have reported relatively high rates of HCC occurrence,and recurrence,following successful HCV treatment using *** studies were grossly underpowered due to their retrospective design,lack of untreated or IFN controls,small sample size,and limited patient follow-up *** then,many retrospective and prospective cohort studies with larger size and longer follow-up duration after DAAs therapy have been *** studies showed that treatment with DAAs can reduce the risk of HCC compared to no treatment,didn't increase the risk of HCC compared to IFN-based therapy after adjusting for the potential confounders of these two groups,and DAAs-induced sustained virological response decreased the risk of HCC compared to DAAs treatment *** conclusion,DAAs treatment doesn't appear to increase the development of HCC,even in cirrhotic ***,cirrhotic patients should be monitored for the development of HCC during and after DAAs treatment.

Rapid Ultrasonic-Assisted Soldering of AZ31B Mg Alloy/6061 Al Alloy with Low-Melting-Point Sn–xZn Solders Without Flux in Air

A novel ultrasonic-assisted low-temperature soldering was developed to join AZ31B Mg alloy and 6061 Al alloy with a series of Sn–x Zn solders. The average maximum shear strength of the joints reaches up to 87.5 MPa at soldering temperature of 300 °C under ultrasonic assistance for only 5 s using Sn–20 Zn solder. The fracture path propagates completely in the soldering seam. The results indicate that the microjet generated by ultrasonic pressure in liquid solder could strike and splinter the Mg_2Sn intermetallic compounds into small pieces, which contributes to the enhancement of the joint strength. In addition, the primary Al(Zn) solid solution phase formed during cooling stage could also strengthen the joint due to the prevention of microcracks propagation.

Inhomogeneous Natural Network Promoting Strain-induced Crystallization:A Mesoscale Model of Natural Rubber

Although synthetic rubbers show continuously improved mechanical properties,natural rubber (NR) remains irreplaceable in the rubber family due to its superior mechanical properties.A mainstream viewpoint regarding the superiority of NR is that NR possesses a natural network formed by linking the poly(cis-l,4-isoprene) chain terminals to protein and phospholipid aggregates;after vulcanization,the natural network additionally contributes to rubber mechanics by both increasing the network density and promoting the strain-induced crystallization (SIC) ***,the reason why the natural network promotes SIC is still unclear;in particular,only using the increased network density cannot explain our finding that the NR shows smaller onset strain of SIC than Gel (the gel component of NR with higher network density) and even vulcanized ***,we point out that the inhomogeneous chain deformation is the alternative reason why SIC of NR takes place at smaller strain than that of *** specifically,although the natural network is homogenous on the subchain length scale based on the proton double-quantum NMR results,it is essentially inhomogeneous on mesoscale (100 nm),as revealed by the small angle X-ray scattering *** inhomogeneous network also leads to the mesoscale deformation inhomogeneity,as detected by the orientation of stearic acid (SA) probe,thus resulting in the smaller onset strain of SIC of *** on the experimental results,a mesoscale model is proposed to qualitatively describe the crucial roles of inhomogeneous structure and deformation of natural network in NR?s mechanical properties,providing a clue from nature to guide the development of high-performance rubbers with controlled structures at mesoscale.

Study of the crosstalk evaluation for cavity BPM

In order to pursue high-precision beam position measurements for free-electron laser facilities, a cavity beam position monitor(CBPM) is employed to measure the transverse position that can meet the requirement of position resolution at a sub-micrometer or even nanometer scale. However, for the pill-box cavity BPM, crosstalk between the cavities will have an effect on the accurate measurement of beam position. To reduce the effect of crosstalk on CBPM performance and ease the measurement of the isolation between the cavities, the cavities with a slight difference in resonant frequency were designed and applied in the Dalian coherent light source and Shanghai soft X-ray free-electron laser facilities. Furthermore, two methods, the principal component analysis method and the method of harmonic analysis, are proposed in this paper to evaluate the crosstalk. The results demonstrate that the two methods are feasible in evaluating the crosstalk between the cavities.
