YBa_(2)Cu_(3)O_(y)(YBCO)foam samples show an open,porous foam structure,which may have benefits for many applications of high-Tc *** the basic material of these foams is a pseudo-single crystalline material with the directional growth initiated by a seed crystal similar to standard melt-textured samples,the texture of YBCO is a very important ***,we analysed the local texture and grain orientation of the individual struts forming the foam by means of atomic force microscopy(AFM)and electron backscatter diffraction(EBSD).Due to the processing route starting with Y_(2)BaCuO_(5)(211),a two-phase analysis must be performed,so a high surface quality is necessary to enable an automated EBSD *** quality Kikuchi patterns were obtained from both the YBCO and 211 *** found an inhomogeneous distribution of the residual 211 particles,which are mainly randomly oriented and have sizes ranging between 200 nm and 15μ*** contrast to this,the YBCO matrix shows a dominating orientation with cracks with a typical distance of 1-10μ***,the analysis of strut cross-sections reveals that the entire strut is converted to the YBCO phase.

Background: There is considerable interest in implementing mobile scanning technology for on-farm body composition analysis on live *** experiments evaluated the use of dual energy X-ray absorptiometry(DXA) as an accurate method of total body fat measurement in live ***: In Exp.1,visceral and whole body fat analysis was undertaken in sheep with body condition scores(BCS)in the range 2 to 3.25(scale 1: thin to 5: fat).The relationship of BCS was moderately correlated with visceral fat depot mass(r = 0.59,p p p.2,sheep with BCS in the range 2.25 to 3.75 were blood sampled to analyse circulating leptin concentrations,and were DXA scanned immediately post mortem for total body *** leptin concentrations had low correlations with BCS(r = 0.50,p p p > 0.05,n = 9).There was a moderate correlation between DXA body fat and BCS(r = 0.70,p p p.3,a series of five DXA scans,at 8-week intervals,was performed on growing sheep over a 32-week *** average BCS ranged from 2.39 ± 0.07(S.E.M.) to 3.05 ± 0.11 and the DXA body fat(%) ranged from 16.8 ± 0.8 to 24.2 ± *** was a moderate correlation between DXA body fat and BCS over the 32 weeks(r = 0.61,p periments indicated that there was good agreement between BCS,DXA and chemical analysis for measuring total body fat in sheep,and that DXA scanning is a valid method for longitudinal measurement of total body fat in live sheep.

Insect herbivory can seriously hinder plant performance and reduce crop yield. Thrips are minute cell-content-feeding insects that are important vectors of viral plant pathogens, and are serious crop pests. We investigated the role of a sweet pepper (Capsicum annuum) lipoxygenase gene, CaLOX2, in the defense of pepper plants against Western flower thrips (Frankliniella occidentalis). This was done through a combination of in-silico, transcrip-tional, behavioral *** datashowthat CaLOX2 is involved in jasmonic acid (jA) biosynthesis and mediates plant resistance. Expression of the jA-related marker genes, CaLOX2 and CapIN II, was induced by thrips feeding. Silencing of CaLOX2 in pepper plants through virus-induced gene silencing (VIGS) resulted in low levels of CaLOX2 transcripts, as well as significant reduction in the accumulation of jA, and its derivatives, upon thrips feeding compared to control plants. CaLOX2-silenced pepper plants exhibited enhanced susceptibility to thrips. This indicates that CaLOX2 mediates jA-dependent signaling, resulting in defenseagainstthrips. Furthermore,exogenous application of jA to pepper plants increased plant resistance to thrips, constrained thrips population development and made plants less attractive to thrips. Thus, a multidisciplinary approach shows that an intact lipoxygenase pathway mediates various components of sweet pepper defense against F. occidentalis.

Tumor necrosis factor α （TNF-α） has been recognized as an activator of nuclear factor kB （NF-kB）, a factor implicated in the protection of many cell types from apoptosis. We and others have presented evidence to suggest that Fas-induced apoptosis may be an important aspect of the resolution of inflammation, and that delayed resolution of inflammation may be directly associated with NF-kB-dependent resistance to Fas. Because TNF-α activates NF-kB in many cell types including inflammatory cells such as eosinophils, we examined effects of TNF-α signaling on the Fas-mediated killing of an eosinophilic cell line AML14. While agonist anti-Fas （CHII） treatment induced apoptosis in AML14 cells, no significant cell death occurred in response to TNF-α alone. Electrophoretic mobility shift assay （EMSA） revealed that TNF-α induced NF-kB transactivation in AMLI4 cells in a time- and dose-dependent fashion, and subsequent supershift assays indicated that the translocated NF-kB was the heterodimer p65 （RelA）/p50. pre-treatment of cells with TNF-α dramatically decreased the CHll-induced cell death in a transient fashion, accompanied by suppression of activation of caspase-8 and caspase-3 activation. Inhibition of NF-kB transactivation by inhibitors, BAY 11-7085 and parthenolide, reversed the suppression of Fas-mediated apoptosis by TNF-α. Furthermore, TNF-α up-regulated X-linked inhibitor of apoptosis protein （XIAp） transiently and XIAp levels were correlated with the temporal pattern of TNF-α protection against Fas-mediated apoptosis. This finding suggested that TNF-α may contribute to the prolonged survival of inflammatory cells by suppression of Fas-mediated apoptosis, the process involved with NF-kB transactivation, anti-apoptotic XIAp up-regulation and caspase suppression. AIM： To evaluate the protective effect of 2′-p-hydroxy benzoylmussaenosidic acid [negundoside （NG）, against carbon tetrachloride （CCl4）-induced toxicity in HUH-7 cel Is.METHODS： CCI4 is a well characterized hepatotoxin, and inducer of cytochrome p450 2E1 （CYp2E1）-mediated oxidative stress. In addition, lipid peroxidation and accumulation of intracellular calcium are important steps in the pathway involved in CCl4 toxicity. Liver cells （HUH-7） were treated with CCI4, and the mechanism of the cytoprotective effect of NG was assessed. Silymarin, a known hepatoprotective drug, was used as control. RESULTS： NG protected HUH-7 cells against CCl4 toxicity and loss of viability without modulating CYp2E1 activity. prevention of CCl4 toxicity was associated with a reduction in oxidative damage as reflected by decreased generation of reactive oxygen species （ROS）, a decrease in lipid peroxidation and accumulation of intracellular Ca^2＋ levels and maintenance of intracellular glutathione homeostasis. Decreased mitochondrial membrane potential （MMp）, induction of caspases mediated DNA fragmentation and cell cycle arrest, as a result of CCl4 treatment, were also blocked by NG. The protection afforded by NG seemed to be mediated by activation of cyclic adenosine monophosphate （cAMp） synthesis and inhibition of phospholipases （cpLA2）. CONCLUSION： NG exerts a protective effect on CYp2E1-dependent CCl4 toxicity via inhibition of lipid peroxidation, followed by an improved intracellular calcium homeostasis and inhibition of Ca^2＋-dependent proteases.

Background： The objective of this study was to study how changing the ratio of Lys to Thr, Lys to His, and Lys to Val affects the expression of lipogenic genes and microRNA （miRNA） in bovine mammary epithelial cells. Results： Triplicate cultures with the respective ＂optimal＂ amino acid （AA） ratio （OpAA = Lys：Met 2.9：1; Thr：phe 1.05：1; Lys：Thr 1.8：1; Lys：His 2.38：1; Lys：Val 1.23：1） plus rapamycin （OpAARMC; positive control）, OpAA, Lys：Thr 2.1：1 （LT2.1）, Lys：Thr 1.3：1 （LT1.3）, Lys：His 3.05：1 （LH3.0）, or Lys：Val 1.62：1 （LV1.6） were incubated in lactogenic medium for 12 h. The expression of 15 lipogenic genes and 7 miRNA were evaluated. Responses to LT2.1, LT1.3, LH3.0, and LV1.6 relative to the control （OpAARMC） included up-regulated expression ofACSS2, FABp3, ACACA, FASN, SCD, LpIN1, INSIG1, SREBF1, ppARD, and NR1H3 （commonly known as LXR-a）. Furthermore, LV1.6 up-regulated expression of ACSL1, DGAT1, and RXRA and down-regulated ppARG expression. Although no effect of OpAA on expression of ppARG was observed, compared with the control, OpAA up-regulated expression of the ppAR targets ACSS2, FABp3, ACACA, FASN, SCD, LpIN1, INSIG1, and SREBF1. Compared with the control, the expression of the anti-lipogenic MIR27AB was down-regulated by OpAA, LT2.1, LT1.3 and LH3.0. In contrast, compared with the control, the expression of the pro-lipogenic MIR21 was up-regulated by LT2.1, LT1.3, LH3.0, and LV1.6. Conclusions： The observed up-regulation of lipogenic gene networks and the changes in expression of key miRNA involved in the control of lipogenic balance are indicative of a potentially important role of EAA ratios and mTOR signaling in the regulation of milk fat synthesis.

Early Cenozoic Mega Thrusting in the Qiangtang Block of the Northern Tibetan plateau

Recent mapping and seismic survey reveal that intensive compression during the Early Cenozoic in the Qiangtang block of the central Tibetan plateau formed an extensive complex of thrust sheets that moved relatively southward along several generally north-dipping great thrust systems. Those at the borders of the ~450 km wide block show it overrides the Lhasa block to the south and is overridden by the Hohxil-Bayanhar block to the north. The systems are mostly thin-skinned imbricate thrusts with associated folding. The thrust sheets are chiefly floored by jurassic limestone that apparently slid over Triassic sandstone and shale, which is locally included, and ramped upward and over paleocene-Eocene red-beds. Some central thrusts scooped deeper and carried up paleozoic metamorphic rock, permian carbonate and granite to form a central uplift that divides the Qiangtang block into two parts. These systems and their associated structures are unconformably overlain by little deformed Late Eocene-Oligocene volcanic rock or capped by Miocene lake beds. A thrust system in the northern part of the block, as well as one in the northern part of the adjacent Lhasa block, dip to the south and appear to be due to secondary adjustments within the thrust sheets. The relative southward displacement across this Early Cenozoic mega thrust system is in excess of 150 km in the Qiangtang block, and the average southward slip-rate of the southern Qiangtang thrusts ranged from 5.6 mm to 7.4 mm/a during the Late Eocene-Oligocene. This Early Cenozoic thrusting ended before the Early Miocene and was followed by Late Cenozoic crustal extension and strike-slip faulting within the Qiangtang block. The revelation and understanding of these thrust systems are very important for the evaluation of the petroleum resources of the region.
