Asperuloside Promotes Apoptosis of Cervical Cancer Cells through Endoplasmic Reticulum Stress-Mitochondrial Pathway

Objective: To investigate the effects of asperuloside on cervical cancer based on endoplasmic reticulum(ER) stress and mitochondrial pathway. Methods: Different doses(12.5–800 μg/ml) of asperuloside were used to treat cervical cancer cell lines Hela and CaSki to calculate the half maximal inhibitory concentration(IC_(50)) of asperuloside. The cell proliferation was analyzed by clone formation assay. Cell apoptosis, intracellular reactive oxygen species(ROS) and mitochondrial membrane potential were determined by flow cytometry. The protein expressions of cleaved-caspase-3, Bcl-2, Bax, Cyt-c, cleaved-caspase-4 and glucose-regulated protein 78(GRP78) were analyzed by Western blot. And the inhibitor of ER stress, 4-phenyl butyric acid(4-PBA) was used to treat cervical cancer cells to further verify the role of ER stress in the apoptosis of cervical cancer cells induced by asperuloside. Results: Asperuloside of 325, 650, and 1300 μg/ml significantly inhibited the proliferation and promoted apoptosis of Hela and CaSki cells(Pll doses of asperuloside significantly increased intracellular ROS levels, reduced mitochondrial membrane potential, significantly reduced Bcl-2 protein expression level, and increased Bax, Cyt-c, GRP78 and cleaved-caspase-4 expressions(Pl/l 4-PBA treatment significantly promoted cell proliferation and reduced apoptosis(Pl asperuloside could reverse 4-PBA-induced increased cell proliferation, decreased apoptosis and cleaved-caspase-3,-4 and GRP78 protein expressions(Plusion: Our study revealed the role of asperuloside in cervical cancer, suggesting that asperuloside promotes apoptosis of cervical cancer cells through ER stress-mitochondrial pathway.

Noble metal surfaces with intrinsic chirality serve as an ideal candidate for investigating enantioselective chemistry due to their superior chemical durability and high catalytic ***,significant advance has been made in synthesizing metal nanocrystals with intrinsic ***,the majority reports are limited to ***,through a heteroepitaxial growth strategy,the synthesis of metal nanocrystals with intrinsic chirality to palladium was extended for the first time and their application in enantioselective recognition was *** heteroepitaxial growth strategy allows for transferring the chirality of homochiral Au nanocrystals to Au@Pd core–shell *** employing the chiral Au@Pd nanocrystals as enantiomeric recognizing elements,a series of electrochemical sensors for chiral discrimination were *** optimal conditions,the peak potential between D-dihydroxyphenylalanine(D-DOPA)and l-dihydroxyphenylalanine(l-DOPA)is about 80 m V,and the peak current of D-DOPA is 2 times as much as that of l-DOPA,which enables the determination of the enantiomeric excess(EE,%)of ***,this report not only introduces a heteroepitaxial growth strategy to synthesize metal nanocrystals with intrinsic chirality,but also demonstrates the superior capability of integrating intrinsic chirality and catalytic properties into metal nanocrystals for chiral recognition.

Researchers are paying increasing attention to the development of low-cost and microcontroller-based accelerometers,in order to make structural health monitoring feasible for conventional bridges with limited monitoring *** with the low-cost sensor development,the use of the embedded accelerometers of smartphones for eigenfrequency analysis of bridges is becoming popular in the civil engineering *** paper,for the first time in the literature,studies these two promising technologies by comparing the noise density and eigenfrequency analysis of a self-developed,validated and calibrated low-cost Internet of things based accelerometer lARA(low cost adaptable reliable accelerometer)with those of a state of the art smartphone(iPhone XR).The eigenfrequency analysis of a footbridge in San Sebastian,Spain,showed that the embedded accelerometer of the iPhone XR can measure the natural frequencies of the under study bridge.

Following the recent report by Dasenbrock-Gammon et al.[Nature 615,244–250(2023)]of near-ambient superconductivity in nitrogendoped lutetium trihydride(luH_(3-δ)N_(ε)),significant debate has emerged surrounding the composition and interpretation of the observed sharp resistance ***,we meticulously revisit these claims through comprehensive characterization and *** definitively identify the reported material as lutetium dihydride(luH_(2)),resolving the ambiguity surrounding its *** similar conditions(270–295 K and 1–2 GPa),we replicate the reported sharp decrease in electrical resistance with a 30%success rate,aligning with the observations by Dasenbrock-Gammon et ***,our extensive investigations reveal this phenomenon to be a novel pressure-induced metal-to-metal transition intrinsic to luH_(2),distinct from ***,nitrogen doping exerts minimal impact on this *** work not only elucidates the fundamental properties of luH_(2)andluH_(3),but also critically challenges the notion of superconductivity in these lutetium hydride *** findings pave the way for future research on lutetium hydride systems,while emphasizing the crucial importance of rigorous verification in claims of ambient-temperature superconductivity.

Machine learning and bioinformatics to identify biomarkers in response to Burkholderia pseudomallei infection in mice

Synthesis of a High-Efficiency Demulsifier and Study on Optimal Demulsification Conditions

Oil and water separation has always been a top priority in the oil *** this study,a series of hyperbranched fluorinated polyamine-amine polymers(HFPA1-5)were synthesized directly using an improved“one-pot method.”The highly active fluorinated p-trifluoromethylaniline was used as the core raw material,while diethylenetriamine and methyl acrylate were used as the chain segment.A hyperbranched fluorine-containing polyamine-amine demulsifier(NHFPA6)was obtained through nano-grafting copolymerization of *** enhance the demulsification and dehydration performance,the copolymerized HFPA6 was modified and ***,the effects of the combination ratio,demulsifier concentration,demulsification time,and demulsification temperature on the demulsification effect were *** results revealed that a combination ratio of DE-401:NHFPA6=1:1,a demulsification temperature of 50℃,a demulsification time of 60 min,and a demulsifier concentration of 150 mg/l yielded a dehydration rate as high as 99.80%.A response surface optimization design of demulsification conditions was *** model verified that the optimal demulsification conditions were 50℃,300 mg/l,and 90 ***,considering the economic benefits of factories,it is more favorable to select demulsification conditions with a shorter time and lower concentration when the dehydration standard is ***,the demulsification conditions were selected as 50℃,150 mg/l,and 60 *** to existing demulsifiers,the demulsifier developed in this study exhibits a lower demulsification temperature and higher demulsification efficiency.

Dynamic analysis of a latent HIV infection model with CTl immune and antibody responses

This paper develops a mathematical model to investigate the Human Immunodeficiency Virus(HIV)infection *** model includes two transmission modes(cell-to-cell and cell-free),two adaptive immune responses(cytotoxic T-lymphocyte(CTl)and antibody),a saturated CTl immune response,and latent HIV *** existence and local stability of equilibria are fully characterized by four reproduction *** sensitivity analyses,we assess the partial rank correlation coefficients of these reproduction numbers and identify that the infection rate via cell-to-cell transmission,the number of new viruses produced by each infected cell during its life cycle,the clearance rate of free virions,and immune parameters have the greatest impact on the reproduction ***,we compare the effects of immune stimulation and cell-to-cell spread on the model's *** findings highlight the significance of adaptive immune responses in increasing the population of uninfected cells and reducing the numbers of latent cells,infected cells,and ***,cell-to-cell transmission is identified as a facilitator of HIV *** analytical and numerical results presented in this study contribute to a better understanding of HIV dynamics and can potentially aid in improving HIV management strategies.

Comparative transcriptomic analysis of ovary and testis reveals sex-related genes in roughskin sculpin Trachidermus fasciatus

The mechanisms underlying sex determination and differentiation have long intrigued researchers in the fields of development and evolutionary *** roughskin sculpin(Trachidermus fasciatus Heckel),displaying sexual dimorphism,provides an ideal model for studying the ***,both genetic and genomic information concerning sex determination and differentiation,such as gonadal transcriptome data in roughskin sculpin,are ***,we present the first gonadal transcriptomes of roughskin sculpin and identify sex-related *** identified 8531 differentially expressed genes(DEGs),among them 4065 were upregulated in the ovary and 4466 upregulated in the *** sex-related gene ontology(GO)terms were enriched in ovary-biased genes,including“binding of sperm to zona pellucida”,“egg coat formation”,“positive regulation of acrosome reaction”,“cell division”,and“cell cycle”,while the GO terms such as“spermatogenesis”,“sperm axoneme assembly”,“cilium assembly”,“cilium movement”,and“cilium movement involved in cell motility”were enriched in testis-biased ***,six KEGG pathways were significantly enriched in the ovary,whereas only one was enriched in the *** these DEGs,40 sex-related genes were identified,which including 26 testis-biased genes(such as Dmrtb 1,Gsdf,Sox 9 b,Wnt 4 b,Tcp 11 l 2,and Efhb),and 14 ovary-biased genes(such as Cyp 19 a 1 a,Foxh 1,Foxr 1,Gdf 3,Hsd 17 b 12,and Igf 2 bp 3).This gonadal transcript dataset would broaden our understanding of sex determination and differentiation mechanisms in roughskin sculpin,expand the genomic database,support future studies on sex-related gene functions,and facilitate molecular biology research into roughskin sculpin.

How interfacial electron-donating defects influence the structure and charge of gold nanoparticles on TiO_(2)support

The reduction degree of TiO_(2)support is critical to the performances of metal *** many previous theoretical calculations,only the bridge oxygen vacancy(Ov)was considered as the electron-donating defect on reduced rutile TiO_(2)(r-TiO_(2−x))***,titanium adatoms(Tiad.),oxidized titanium islands(***),and acid hydroxyls(ObrH)also exist at the metal/support *** conducting density functional theory(DFT)calculations and ab initio molecular dynamics(AIMD)simulations,we compared r-TiO_(2−x)surfaces with Ov,Tiad.,***,and ObrH sites loaded with Au nanoparticles(NPs).The results showed the Au NPs were oxygen-phobic but titanium-philic,resulting in wetting of Ov and *** short contact with *** and *** Bader charges of Au NPs(QM)showed a good linear relationship with the ideal number of donating electrons(Ne)from the defective sites(QM=−KeNe+QM,S),demonstrating the intrinsic electron allocation at the *** Ov,Tiad.,and *** exhibited similar slopes(Ke),relatively steeper than that of *** means in the scope of Au NP charge state,the *** *** have a close electron-donating ability with Ov,but the ObrH donates relatively fewer *** linear relationship can be extended approximately to other *** higher the metal work function,the steeper the Ke for easier electron donation from defective *** stronger the metal oxygen affinity,the more positive the intercept(QM,S).That explains the easy generation of metallic or negative Pt and Au NPs on r-TiO_(2−x),but hard for Cu and Zn in *** provides theoretical guidance for regulating the charge of metal NPs over TiO_(2−x)supports.
