DNA损伤修复以及基因组稳定性维持对于动植物正常生长和防御逆境至关重要。针对DNA聚合酶错误掺入导致的基因组不稳定性,本研究以DNA聚合酶η为研究对象,通过计算分子模拟对接的方式,对其可能的小分子抑制剂进行筛选,并对其酶动力学参数进行测定。结果显示:脱氧腺苷三磷酸(deoxyadenosine triphosphate,d ATP)对DNA聚合酶η的活性具有抑制效果,使其延伸的相对效率为36%~42%。分子模拟对接和体外实验结果表明,相较于dATP(亲和力为-26.7 kJ/mol),环鸟苷酸-腺苷酸(cyclic GMP-AMP,cGAMP)与DNA聚合酶η具有更低的结合能(亲和力为-35.1 kJ/mol)。酶动力学参数测定结果也表明,相较于dATP,cGAMP具有更强的抑制能力且在浓度为0.5 mmol/l时达到最强(相对延伸效率为13%)。因此,本研究筛选获得了针对DNA聚合酶η的一种潜在的小分子抑制剂。同时,鉴于该蛋白质高表达导致细胞对抗肿瘤药物(DNA损伤剂)的耐受性,这为新型药物的开发提供了依据。

为了克服传统弯曲型、剪切型金属阻尼器无法在肘节式等位移放大装置安装使用的现状,提出一种基于X形钢板屈服耗能的装配式活塞阻尼器(X-shaped piston metal damper,XPMD),其由一组平行的金属环片(X形钢板为主要耗能元件)、内部传动杆及外部套筒共同组成。具有耗能功效的金属环片通过高强螺栓与内部传动杆及外部套筒相互连接,具有较好的装配性及易于更换的特点。通过试验系统研究了X形钢板端头宽度、缩颈宽度、纵向长度及厚度等参数对XPMD阻尼器滞回性能的影响规律,同时提出了XPMD阻尼器的理论力学评估模型。以多层框架为例,对比分析了在地震作用下安装XPMD阻尼器前后其位移响应、层间剪力的变化特点,验证了XPMD阻尼器对框架结构的减震效果。研究结果表明:在循环荷载作用下XPMD阻尼器具有稳定的滞回性能,在较小变形下地震输入能量主要通过X形钢板弯曲变形实现能量耗散,在较大变形条件下X形钢板拉伸变形逐渐增大使得XPMD阻尼器等效刚度有所增加;安装XPMD阻尼器可明显降低框架结构在地震作用下的位移及层间剪力响应。

The phenomenon termed “zebra stripes” manifests as regular patterns in the energy-space(l shell) spectrum of energetic electrons(ranging from tens to hundreds keV) within the inner radiation belt. These structures exhibit drift-periodic behavior and commonly arise from large-scale electric field perturbations near the substorm onsets. In this study, we introduce a composite electric field model and replicate the formation, structure, and evolution of zebra stripes using a bounce-averaged test particle code under this electric field model. High-resolution measurements of energetic electrons obtained from the Van Allen Probes and the recently launched Macao Science Satellites-1 are used as initial conditions and served to validate our test particle simulations. Comparative analyses between observed data and simulations demonstrate our test particle method's efficacy in capturing zebra stripes' general behavior. Moreover, the composite model proves capable of reproducing realistic variations in the electric field within the inner radiation belt to a certain extent. Nevertheless, subtle differences emerge in the flux strength and the positions of stripes. These disparities primarily stem from limitations inherent in the electric field model and the initial conditions of the simulation. Acknowledging that the model represents an average case, it is conceivable that real-world scenarios may deviate from the average, thereby introducing variations in the observed phenomena.

Wolfberry (lycium barbarum l.) is a medicinal herb with anti-aging and neuroprotective effects. The purpose of this study was to explore the enhanced neuroprotective effects of wolfberry water extract (RW)， and brown wolfberry water extract (BBW). The reduced percentage of reducing sugar in BBW was shown to be 62.8%， 64.3%， 39.8%， and 11.6% for glucose， fructose， maltose， and lactose compared to those of RW， as well as free amino acids. And the contents of total phenols and total flavonoids were increased by 62.0% and 33.1%. Melanoidins (1% ± 0.05%) were first isolated from BBW. RW and BBW increased anti-stress abilities (oxidative stress: 24.2% vs 35.7%， and heat stress: 10.9% vs 19.1%) while decreasing the ROS levels in vivo. RW and BBW reduced the rate of paralysis and odor cognitive deficits. At the same time， RW and BBW reduced the number of fluorescent spots of Aβ::GFP. The RW and BBW activated autophagy by upregulating the gene levels (bec-1， lgg-1， lgg-2， unc-51， vps-34， atg-5， atg-18， and sqst-1). Additionally， BBW promoted the nuclear-cytoplasmic ratio of DAF-16::GFP and fluorescence intensity of SOD-3::GFP by 39.4 fold and 1.16 fold. This study laid a new insight for exploring the Maillard reaction in improving the anti-AD activity of wolfberry.

For real-time processing of ultra-wide bandwidth low-frequency pulsar baseband data, we designed and implemented an ultra-wide bandwidth low-frequency pulsar data processing pipeline(UWlPIPE) based on the shared ringbuffer and GPU parallel technology. UWlPIPE runs on the GPU cluster and can simultaneously receive multiple 128 MHz dual-polarization VDIF data packets preprocessed by the front-end FPGA. After aligning the dual-polarization data, multiple 128M subband data are packaged into PSRDADA baseband data or multi-channel coherent dispersion filterbank data, and multiple subband filterbank data can be spliced into wideband data after time alignment. We used the Nanshan 26 m radio telescope with the l-band receiver at964 ～ 1732 MHz to observe multiple pulsars. Finally, we processed the data using DSPSR software, and the results showed that each subband could correctly fold out the pulse profile, and the wideband pulse profile accumulated by multiple subbands could be correctly aligned.
