Bone marrow microRNA-34a is a good indicator for response to treatment in acute myeloid leukemia

Background:microRNA 34a(miR 34a)had been reported to have a diagnostic role in acute myeloid leukemia(AMl).However,its value in the bone marrow(BM)of AMl patients,in addition to its role in response to therapy is still *** current study was designed to assess the diagnostic,prognostic,and predictive significance of miR 34a in the BM of AMl ***:The miR.34a was assed in BM aspirate of 82 AMl patients in relation to 12 normal control subjects using *** data were assessed for correlation with the relevant dinical critenia,response to therapy,disease-free survival(DFS),and overall survival(OS)***:miR.34a was significantly downregulated in AMl patients[0.005(3.3×10^(-6)-1.32)],compared to the control subjects[0.108(3.2× 10^(-4)-1.64),p=0.021].*** relative quantification(RQ)of miR-34a was 0.106(range;0-32.12).The specifaity,sensitivity,and area under the curve(AUC)for the diagnosis of AMl were(58.3%,69.5%,0.707,respectively,p=0.021).patients with upregulated miR-34a showed decreased platelets countl,and achieved early complete remission(CR,p=0.031,p=0.044,respectively).Similarly,patients who were refractory to therapy showed decreased miR 34a levels in comparison to those who achieved CR[0.002(0-0.01)and 0.12(0-32.12),respectively,p=0.002].Therefore,miR 34a could significantly identify patients with CR with a specificity of 75%and sensitivity of 100%at a cut-off of 0.014(AUC=0.927,p=0.005).There was no considerable association between miR-34a expression and survival rates of the induded AMl ***:miR-34a could be a beneficial diagnostic biomarker for AMl *** addition,it serves as a good indicator for response to therapy,which could possibly identify patients who are refractory to treatment with 100%sensitivity and 75%specificity.

In this paper,we extend our previous result from l.vy(2016).We prove that transport equations with rough coefficients do possess a uniqueness property,even in the presence of *** method relies strongly on duality and bears a strong resemblance to the well-known DiPerna-lions theory first developed by DiPerna and lions(1989).This uniqueness result allows us to reprove the celebrated theorem of Serrin(1962)in a novel *** a byproduct of the techniques,we derive an l^(1) bound for the vorticity in terms of a critical lebesgue norm of the velocity *** also show that the zero solution is unique for the 2D Euler equations on the torus under a mild integrability assumption.

Obesity is one of the diseases with severe health consequences and rapidly increasing worldwide *** the complex network of food intake and energy balance regulation is an essential prerequisite for pharmacological intervention with obesity.G protein-coupled receptors(GPCRs)are among the main modulators of metabolism and energy ***,for instance,regulate appetite and satiety in certain hypothalamic neurons,as well as glucose and lipid metabolism and hormone secretion from *** in some GPCRs,such as the melanocortin receptor type 4(MC4R),have been associated with early-onset ***,we identified the adhesion GPCR latrophilin 1(ADGRl1/lPHN1)as a member of the regulating network governing food intake and the maintenance of energy *** of the highly conserved receptor in mice results in increased food consumption and severe obesity,accompanied by dysregulation of glucose ***,we identified a partially inactivating mutation in human ADGRl1/lPHN1 in a patient suffering from ***,we propose that lPHN1 dysfunction is a risk factor for obesity development.

Oxygen(O_(2))-sensing matrices are promising tools for the live monitoring of extracellular O_(2) consumption levels in long-term cell *** this study,ratiometric O_(2)-sensing membranes were prepared by electrospinning,an easy,low-cost,scalable,and robust method for fabricating ***(ε-caprolactone)and poly(dimethyl)siloxane polymers were blended with tris(4,7-diphenyl-1,10-phenanthroline)ruthenium(II)dichloride,which was used as the O_(2)-sensing probe,and rhodamine B isothiocyanate,which was used as the reference *** functionalized scaffolds were morphologically characterized by scanning electron microscopy,and their physicochemical profiles were obtained by Fourier transform infrared spectroscopy,thermogravimetric analysis,and water contact angle *** sensing capabilities were investigated by confocal laser scanning microscopy,performing photobleaching,reversibility,and calibration curve studies toward different dissolved O_(2)(DO)*** sensing nanofibers showed a high response to changes in DO concentrations in the physiological-pathological range from 0.5%to 20%and good stability under ratiometric *** addition,the sensing systems were highly biocompatible for cell growth promoting adhesiveness and growth of three cancer cell lines,namely metastatic melanoma cell line SK-mel2,breast cancer cell line MCF-7,and pancreatic ductal adenocarcinoma cell line Panc-1,thus recreating a suitable biological environment in *** O_(2)-sensing biomaterials can potentially measure alterations in cell metabolism caused by changes in ambient O_(2)content during drug testing/validation and tissue regeneration processes.

By extending the l.vy wavefunction constrained search to Fock Space,one can define a wavefunction constrained search for electron densities in systems having noninteger number of *** pure-state v-representable densities,the results are equivalent to what one would obtain with the zero-temperature grand canonical *** other cases,the wavefunction constrained search in Fock space presents an upper bound to the grand canonical ensemble *** advantage of the Fock-space wavefunction constrained search functional over the zero-temperature grand-canonical ensemble constrained search functional is that certain specific excited states(i.e.,those that are not ground-statev-representable) are the stationary points of the Fock-space ***,a potential disadvantage of the Fock-space constrained search functional is that it is not convex.

Asymptotic analysis of a mat hematical model of the atherosclerosis development

A mathematical model,which takes into account new experimental results about diverse roles of macrophages in the atherosclerosis development,is *** technic of upper and lower solutions,the existence and uniqueness of its positive solution are *** the nondimensionalization,small parameters are found and the multiscale analysis of the corresponding perturbed problem is performed when those parameters tend to *** particular,the limit two-dimensional problem,which is a coupled system of reaction-diffusion equations and ordinary differential equations,is derived;the asymptotic approximation is constructed;the uniform pointwise estimate for the difference between the solution of the original problem and the solution of the limit problem as well as the respective l^2-estimates for the fluxes are proved.

Gemcitabine in elderly patients with advanced pancreatic cancer

AIM： TO assess feasibility, tolerability and efficacy of gemcitabine-based chemotherapy in patients ≥ 75 years old with advanced pancreatic cancer. METHODS： All consecutive patients ≥ 75 years old with advanced pancreatic adenocarcinoma were included in this retrospective study. Necessary criteria to receive chemotherapy were： performance status 0-2, adequate biological parameters and no serious comorbidities. Other patients received best supportive care （BSC）. RESUlTS： Thirty-eight patients （53% women, median age 78 years, range 75-84） with pancreatic cancer （metastatic： n = 20, locally advanced： n = 18） were studied. Among them, 30 （79%） were able to receive chemotherapy [median number： 9 infusions （1-45）]. Six patients （23%） had at least one episode of grade 3 neutropenia and one patient developed a grade 3 hemolytic-uremic syndrome. No toxic death occurred. Three patients （11%） had a partial tumor response, 13 （46%） had a stable disease and 12 （43%） had a tumor progression. Median survival was 9.1 mo （metastatic： 6.9 too, locally advanced： 11.4 too）. CONClUSION： Tolerance and efficacy of gemcitabinebased chemotherapy is acceptable in elderly patients in good condition, with similar results to younger patients.

Rodents with prevailing subterranean activity usually play an important role in the ecosystems of which they are a part due to the combined effect of herbivory and soil *** is the case for the giant root-rat Tachyoryctes macrocephalus endemic to the Afroalpine ecosystem of the Bale Mountains,*** studied the impact of root-rats on various ecosystem features within a 3.5-ha study locality dominated by Alchemilla pasture,which represents an optimal habitat for this species,in 2 periods of a *** root-rats altered plant species composition,reducing the dominant forb,Alchemilla abyssinica,while enhancing Salvia merjame and a few other species,and reduced vegetation cover,but not the fresh plant *** burrows were abandoned by root-rats,other rodents took them over and *** increased ***-rat burrowing created small-scale heterogeneity in soil compactness due to the backfilling of some unused burrow *** compacted soil tended to be rich in nutrients,including carbon,nitrogen and phosphorus,which likely affected the plant growth on sites where the vegetation has been reduced as a result of root-rat foraging and burrowing.

Testicular sperm extraction(TESE)outcomes in the context of malignant disease:a systematic review

Advances in the oncology field have led to improved survival ***,quality of life after remission is anticipated,which includes the possibility to conceive *** cancer treatments are potentially gonadotoxic,fertility preservation must be *** fertility preservation is mainly based on ejaculated sperm *** this is not possible,testicular sperm extraction(TESE)may be *** identify situations in which TESE has been beneficial,a systematic review was *** search was carried out on the PubMed,Scopus,Google Scholar,and CISMeF databases from 1 January 2000 to 19 March *** Reporting Items for Systematic Reviews and Meta-Analyses(PRISMA)recommendations were followed in selecting items of ***-four articles were included in the systematic review,including 15 articles on oncological testicular sperm extraction(oncoTESE),18 articles on postgonadotoxic treatment TESE and 1 article on both oncoTESE and postgonadotoxic treatment *** sperm freezing was possible for 42.9%to 57.7%of patients before gonadotoxic treatment and for 32.4%to 75.5%of patients after gonadotoxic treatment,depending on the type of malignant *** no formal conclusion could be drawn about the chances to obtain sperm in specific situations,our results suggest that TESE can be proposed before and after gonadotoxic *** treatment,TESE is more often proposed for men with testicular cancer presenting with azoospermia since TESE can be performed simultaneously with tumor removal or *** chemotherapy,TESE may be planned if the patient presents with persistent azoospermia.
