＜正＞ The novel.mechanism of functional.sel.ctivity （differential.activation of pathways l.nked to a singl. receptor） has been shown for dopamine hD2l.receptor regul.ted endpoints such as GIRK. MAPK. ACase. etc. The current work expl.red the hypothesis that functional.y sel.ctive l.gands induce unusual.receptor conforma-tional.states that l.ad to differential.activation. We docked the propyl.orapo-morphine enantiomers [ RNPA ＆ SNPA] to the D2R receptor to identify potential.residues of importance, and then made and expressed sel.cted mutant receptors. WT D2R binding confirmed that RNPA forms p - OH H bonds with both S5.46 and T3.37 whereas the m OH interacts with S5.42, whil. SNPA H- bonds to S5.42 （p ＆ m OH）. When mutated, differential.effects were seen on functional.endpoints （e.g.. S5.42A caused l.ss of D2R mediated effects on ACase. but not MAPK or AA rel.ase）. These data demonstrate that singl. point receptor mutations can make a "normal. l.gand become functional.y sel.ctive, or change the character of a functional.y sel.ctive compound. We hypothesize that l.gandspecific residue interactions contribute to the conformational.changes needed to resul. in activation of specific heterotrimeric compl.xes.

Antoine de Ginestiere, Nice, Cé dex 03, France]Background: Al.facept, a ful.y human l.A-3/IgG1 fusion protein, is a sel.ctive biol.gical.agent approved in theUnited States for the treatment of chronic pl.que psoriasis. In phase 3 trial., cl.nical.improvement and prol.nged off-treatment remission of psoriasis correl.ted with reductions in circul.ting memory T cel.s. Reductions in pathogenic epidermal.T cel.s in psoriatic l.sions al.o have been noted fol.owing phototherapy with ul.raviol.t B (UVB) l.ght. Because al.facept and UVB target T cel.s in different ways, combination therapy with these two agents may l.ad to greater efficacy. Obj.ctives: To determine the safety, tol.rabil.ty, and efficacy trends of combination therapy with al.facept pl.s UVB l.ght in patients with chronic pl.que psoriasis. Methods: In an open-l.bel. paral.el.group study conducted at two sites, one in France and one in the United States, patients with chronic pl.que psoriasis who were candidates for phototherapy received 12-weekl. intramuscul.r inj.ctions of al.facept, 15 mg. In addition, patients were randomized to one of three treatment arms: no UVB treatment, 6-week UVB treatment, and 12-week UVB treatment. UVB treatment consisted of narrowband (NB) UVB at the site in France and broadband (BB) UVB at the site in the United States. The 12-week treatment period was fol.owed by a 12-week fol.ow-up period. Cl.nic visits occurred weekl. during treatment and every 2-4 weeks during fol.ow-up. Resul.s: A total.of 60 patients (n =30/site)were enrol.ed in the study. Al.facept was wel. tol.rated when administered in combination with UVB treatment and as monotherapy. There was no evidence of increased phototoxicity or photosensitivity with the combination. The concept of“robust dynamics”describes the incorporation of mechanical.y interl.cked mol.cul.s(MIMs)into metal.organic framework(MOF)material. such that l.rge ampl.tude motions(e.g.,rotation or transl.tion of a macrocycl.)can occur inside the free vol.me pore of the *** aid in the preparation of such material.,reticul.r synthesis was used herein to design rigid mol.cul.r buil.ing bl.cks with predetermined ordered structures starting from the wel.-known MOF *** l.nkers were synthesized that have a T-shape,based on a triphenyl.ne tetra-carboxyl.te strut,and their incorporation into Cu(II)-based MOFs was *** singl.-crystal.structures of three new MOFs,UWCM-12(fof),β-UWCM-13(l.z),UWCM-14(l.l.,with naked T-shaped l.nkers were determined;β-UWCM-13 is the first reported exampl. of the l.z topol.gy.A fourth MOF,UWDM-14(l.l.is anal.gous to UWCM-14(l.l.but contains a[2]rotaxane ***-temperature,^(2)H sol.d-state NMR was used to probe the dynamics of a 24-membered macrocycl. threaded onto the MOF skel.ton.
