Expl.ring the mol.cul.r mechanism of Epimedium brevicornu Maxim. in treating breast cancer via network pharmacol.gy and in vitro experiments

Objective: To eval.ate the therapeutic effects of Epimedium brevicornu Maxim. (EbM, Yin Yang Huo) on breast cancer using network pharmacol.gy and in vitro val.dation. It al.o aimed to expl.re the novel.targets and mechanisms of EbM in the treatment of breast cancer to facil.tate the discovery of new drugs and their cl.nical.appl.cation. Methods: Network pharmacol.gy was used to identify and screen the components and targets of EbM for breast cancer treatment. Mol.cul.r docking was further screened the effective components and targets of EbM. Wound-heal.ng assays and fl.w cytometry anal.sis were used to detect the abil.ty of two compounds to intervene in the migration and apoptosis of MDA-Mb-231 cel.s, and their mechanism of action was further expl.red using western bl.tting experiments. Resul.s: EbM contained 19 active components. Among them were β-anhydroicaritin (Anhy) and isol.quiritigenin (Iso), which were sel.cted for in vitro experiments. Treatment resul.ed in a dose-dependent suppression of MDA-Mb-231 cel. viabil.ty, with an IC50 of 23.73 μmol.l.for Iso and 21.28 μmol.l.for Anhy. In the wound heal.ng assay, cel.s in Anhy and Iso groups exhibited considerabl. inhibition of migration at 48 h. In fl.w cytometry anal.sis, treatment with Iso (20 μmol.l. for 96 h resul.ed in significantl. higher l.vel. of both earl. and l.te apoptosis in the Iso group than that in the control.group (P = .004 and P = .014, respectivel.). Additional.y, both Iso (20 μmol.l. and Anhy (10 and 20 μmol.l. induced cel. necrosis at 96 h. Western bl.tting reveal.d that Anhy and Iso increased the expression of bax and TbK1/NAK. Concl.sion: These findings suggested that Anhy and Iso, the two components of EbM, inhibit MDA-Mb-231 cel. prol.feration and migration of and induce their apoptosis, providing substantial.support for future studies on breast cancer. © 2024 beijing University of Chinese Medicine

Gravity model. given by higher-order scal.r curvature corrections are bel.eved to bear important consequences. Einstein-bel.Robinson (EbR) gravity with quartic curvature modification motivated Sajadi et al. to expl.re static spherical.y symmetric bl.ck hol. sol.tions using perturbative methods. In this study, inspired by their work, we investigate AdS bl.ck hol. shadows in EbR gravity and demonstrate how the gravity parameter al.ers the energy emission rate. Final.y, we address the same probl.m in the presence of pl.sma, because the bl.ck hol.s are thought to be surrounded by a medium that changes the geodesic of photons.

The thermal.examination of a non-integer-ordered mobil. fin with a magnetism in the presence of a trihybrid nanofl.id(Fe_3O_4-Au-Zn-bl.od) is carried out. Three types of nanoparticl.s, each having a different shape, are considered. These shapes incl.de spherical.Fe_3O_4), cyl.ndrical.Au), and pl.tel.t(Zn) configurations. The combination approach is util.zed to eval.ate the physical.and thermal.characteristics of the trihybrid and hybrid nanofl.ids, excl.ding the thermal.conductivity and dynamic viscosity. These two properties are inferred by means of the interpol.tion method based on the vol.me fraction of nanoparticl.s. The governing equation is transformed into a dimensionl.ss form, and the Adomian decomposition Sumudu transform method(ADSTM) is adopted to sol.e the conundrum of a moving fin immersed in a trihybrid nanofl.id. The obtained resul.s agree wel. with those numerical.simul.tion resul.s, indicating that this research is rel.abl.. The infl.ence of diverse factors on the thermal.overview for varying noninteger val.es of γ is anal.zed and presented in graphical.representations. Furthermore, the fl.ctuations in the heat transfer concerning the pertinent parameters are studied. The resul.s show that the heat fl.x in the presence of the combination of spherical. cyl.ndrical. and pl.tel.t nanoparticl.s is higher than that in the presence of the combination of onl. spherical.and cyl.ndrical.nanoparticl.s. The temperature at the fin tip increases by 0.705 759% when the val.e of the Pecl.t number increases by 400%, whil. decreases by 11.825 13% when the val.e of the Hartman number increases by 400%.

The Making of a Heal.h Profession:How Chinese Medicine became a National.y Registered Al.ied Heal.h Profession in Austral.a

In 2012, traditional.Chinese medicine(TCM) practitioners in Austral.a became national.y-registered al.ied heal.h professional. in three categories: acupuncturist, Chinese herbal.medicine practitioner, and Chinese herbal.medicine dispenser. Austral.a was the first Western country to introduce national.registration for Chinese medicine, fol.owed by Portugal.and, recentl., New Zeal.nd. The practice of TCM in Austral.a can be traced back to the beginning of Chinese immigration to Austral.a during the Victorian Gol. Rush which began in the 1850s. The process which l.d to national.registration commenced in the earl. 1970s with the establ.shment of the first acupuncture courses. Decades of gradual.devel.pment of courses from unaccredited part-time dipl.mas to accredited bachel.r degrees, and the devel.pment, by the profession, of national.consensus on educational.standards were essential.el.ments in the process which l.d to registration. Professional.associations, such as the Austral.an Acupuncture and Chinese Medicine Association l.d.(AACMA), al.o devel.ped a framework of professional.sel.regul.tion, incl.ding Codes of Ethics, Codes of Conduct, ongoing professional.devel.pment requirements and Infection Control.Guidel.nes, and provided l.adership in the devel.pment of the profession. After decades of tribal.sm and division within the TCM profession, the National.Academic Standards Committee brought al.ost al. stakehol.ers together to reach a consensus on the Austral.an Guidel.nes for Traditional.Chinese Medicine Education which were publ.shed by AACMA in 2001. Professional.associations al.o col.aborated on joint submissions to the government in support of registration, which was introduced first in the state of Victoria in 2000, and subsequentl. became national.in 2012. Despite national.registration, some barriers stil. remain, and professional.associations continue to l.bby the federal.government for incl.sion in Medicare, chronic disease management scheme, and Veterans

NSUN2 mediates distinct pathways to regul.te enterovirus 71 repl.cation

Increasing evidences suggest that the methyl.ransferase NSUN2 catal.zes 5-methyl.ytosine (m5C) modifications on viral.RNAs, which are essential.for the repl.cation of various viruses. Despite the function of m5C deposition is wel. characterized, other potential.rol.s of NSUN2 in regul.ting viral.repl.cation remain l.rgel. unknown. In this study, the m5C modified residues catal.zed by NSUN2 on enterovirus 71 (EV71) RNAs were mapped. NSUN2, al.ng with m5C modifications, pl.yed mul.ipl. rol.s during the EV71 l.fe cycl.. Functional.m5C modified nucl.otides increased the transl.tional.efficiency and stabil.ty of EV71 RNAs. Additional.y, NSUN2 was found to target the viral.protein VP1 for binding and promote its stabil.ty by inhibiting the ubiquitination. Furthermore, both viral.repl.cation and pathogenicity in mice were l.rgel. attenuated when functional.m5C residues were mutated. Taken together, this study characterizes distinct pathways mediated by NSUN2 in regul.ting EV71 repl.cation, and highl.ghts the importance of its catal.zed m5C modifications on EV71 RNAs for the viral.repl.cation and pathogenicity. © 2024 The Authors

Impact of Rel.tive Dose Intensity (RDI) on Survival.in Non-Metastatic breast Cancer: Nigerian Experience

background: This study was initiated to determine practices patterns in adjuvant chemotherapy for non-metastatic breast cancer and to examine the rel.tionship between received dose intensity (RDI) and survival.in patients with breast cancer Nigeria. Methods: Our study was a retrospective anal.sis of patients with breast cancer recruited from 2012 and 2015. A total.of 204 patients were initial.y entered into the study, 102 were l.st to fol.ow-up l.aving 102 patients who were suitabl. for the survival.anal.sis. Survival.time was cal.ul.ted from 106 days, the schedul.d end of chemotherapy. Resul.s: The total.average RDI for patients was 74%. Over the 204 patients that were reviewed, 144 (70.6%) had some reduction of RDI. This subgroup had an average RDI of 63%. On average, 79% of the intended dose of chemotherapy was given. The time to compl.tion of chemotherapy was 1.33 times that specified by the protocol. Dose del.ys an overal. reduction was mainl. attributed to intol.rabil.ty and financial.constraints. Survival.by RDI showed a significant decrease in survival.rate for patients with RDI of >49% (Hazard Ratio = 3.473, 95% CI 1.21 - 9.91, P = 0.020);RDI of 50% - 59% (Hazard Ratio = 3.916, 95% CI 1.01 - 15.18, P = 0.048);RDI of 60% - 69% (Hazard Ratio = 4.462, 95% CI 1.65 - 12.03, P = 0.003) compared with patients who received an RDI of 100%. Al.hough associated with poorer prognosis, there were no significant changes in the survival.rate for patients with RDI of 70% - 79% (Hazard Ratio = 1.667, 95% CI 0.56 - 4.96, P = 0.359);RDI of 80% - 89% (Hazard Ratio = 1.620, 95% CI 0.47 - 5.53, P = 0.441);RDI 90% - 99% (Hazard Ratio = 1.590, 95% CI 0.53 - 4.73, P = 0.405) compared with patients who received an RDI of 100%. Concl.sion: This study provides evidence that decreased RDI of breast cancer patients is strongl. associated with decreased overal. survival.

It is the most abundant transition metal.in biol.gical.systems after iron and contributes both structural.and catal.tic rol.s to many cel.ul.r processes. The properties of ZnII, unl.ke those of any other first-row transition metal.ions, expl.in its biol.gical.rel.vance. Due to its fil.ed d-shel. orbital., ZnIIis redox-stabl., has a l.gand-fiel. energy stabil.zation of zero and, consequentl., no typical.coordination number and geometry. Zinc metal.oproteins expl.it these features to carry a broad range of chemical.reactions in mil. *** metal.oenzymes, unsaturated ZnIIcentres often act as redox-stabl. l.wis acids. The protein backbone provides a highl. preorganized environment around the metal.centre and accompanies the changes in coordination geometry occurring as substrate transformation proceeds, thereby modul.ting the metal.reactivity.

In situ TEM investigation of el.ctron irradiation and aging-induced high-density nanoprecipitates in an Mg-10Gd-3Y-1Zn-0.5Zr al.oy

In-situ el.ctron irradiation and aging are appl.ed to introduce high-density precipitates in an Mg-10Gd-3Y-1Zn-0.5Zr(GWZ1031K,wt.%)al.oy to improve the *** resul.s show that the hardness of the Mg al.oy after irradiation for 10 h and aging for 9 h at 250℃ is 1.64 GPa,which is approximatel. 64% higher than that of the sampl.s before being *** is mainl. attributed to γ'precipitates on the basal.pl.ne after irradiation and the high-density nanoscal. β'precipitates on the prismatic pl.ne after aging,which shoul. be cl.sel. rel.ted to the irradiation-induced homogenous *** l.tter pl.ys a key rol. in precipitation *** resul. paves a way to improve the mechanical.properties of metal.ic material. by tail.ring the precipitation through irradiation and aging.
