Ex.l.ring the mol.cul.r mechanism of Epimedium brevicornu Max.m. in treating breast cancer via network pharmacol.gy and in vitro ex.eriments

Objective: To eval.ate the therapeutic effects of Epimedium brevicornu Max.m. (EBM, Yin Yang Huo) on breast cancer using network pharmacol.gy and in vitro val.dation. It al.o aimed to ex.l.re the novel.targets and mechanisms of EBM in the treatment of breast cancer to facil.tate the discovery of new drugs and their cl.nical.appl.cation. Methods: Network pharmacol.gy was used to identify and screen the components and targets of EBM for breast cancer treatment. Mol.cul.r docking was further screened the effective components and targets of EBM. Wound-heal.ng assays and fl.w cytometry anal.sis were used to detect the abil.ty of two compounds to intervene in the migration and apoptosis of MDA-MB-231 cel.s, and their mechanism of action was further ex.l.red using western bl.tting ex.eriments. Resul.s: EBM contained 19 active components. Among them were β-anhydroicaritin (Anhy) and isol.quiritigenin (Iso), which were sel.cted for in vitro ex.eriments. Treatment resul.ed in a dose-dependent suppression of MDA-MB-231 cel. viabil.ty, with an IC50 of 23.73 μmol.l.for Iso and 21.28 μmol.l.for Anhy. In the wound heal.ng assay, cel.s in Anhy and Iso groups ex.ibited considerabl. inhibition of migration at 48 h. In fl.w cytometry anal.sis, treatment with Iso (20 μmol.l. for 96 h resul.ed in significantl. higher l.vel. of both earl. and l.te apoptosis in the Iso group than that in the control.group (P = .004 and P = .014, respectivel.). Additional.y, both Iso (20 μmol.l. and Anhy (10 and 20 μmol.l. induced cel. necrosis at 96 h. Western bl.tting reveal.d that Anhy and Iso increased the ex.ression of Bax.and TBK1/NAK. Concl.sion: These findings suggested that Anhy and Iso, the two components of EBM, inhibit MDA-MB-231 cel. prol.feration and migration of and induce their apoptosis, providing substantial.support for future studies on breast cancer. © 2024 Beijing University of Chinese Medicine

The buckl.ng formation and infl.ence factors during wel.ing thin - pl.ted structures were investigated based on finite el.ment methods, An engineering treatment method for prdicating the buckl.ng distor- tion was proposed in here. It was used the equival.nt appl.ed thermal.- l.ad to simul.te the wel.ing residual.stress,therefore the cal.ul.tion of compl.x.wel.ing distoriton can be transformed into 3D el.s- tic structural.appl.ed- l.ad anal.ses, which can reduce the quantities of cal.ul.ting works effectivel.. The val.dation of the method was verified by comparison of the numerical.simul.tion and ex.erimental.resul.s. The simul.tion of buckl.ng distorition for side - wal.ed structrues of paseenger train was per- formed and the cal.ul.tion was in agreement with measuring resul.s in general. It was shown that the main factors for producing the buckl.ng were the intermittent fil.et and ping the during wel.ing the stiffened beams and col.mns to the *** ex.stence of the free edge of panel. woul. reduce the crit- ical.buckl.ng stress and enl.rge the defl.ction obviousl.,but the continuous wel. in cl.sed frames didn' t degrade the buckl.ng situation.

Background: Upper eyel.d retraction is a chal.enging compl.cation of cosmetic upper eyel.d bl.pharopl.sty. To avoid ex.ra trauma, we devel.ped a new technique for correcting iatrogenic upper eyel.d retraction by post-migrating a compound fl.p of the orbicul.ris muscl. and fascia (OFC) on the anterior surface of the tarsal.pl.te. This method ex.ends the aponeurosis of the l.vator pal.ebrae superioris muscl. (l.S), which can achieve a good correction for post-bl.pharopl.sty retraction. Methods: We col.ected data from 15 patients with mil. to moderate iatrogenic upper eyel.d retraction who were treated at our hospital.between February 2017 and December 2019. The OFC was used to repl.ce the missing part of the l.S, and post-migration of the l.S and fix.tion of the OFC to the tarsal.margin were conducted. Postoperative outcome measurements incl.ded postoperative binocul.r symmetry, doubl. eyel.d smoothness, eyel.d ful.ness, margin refl.x.distance (MRD1), degree of eyel.d cl.sure, and ex.osure keratitis. The patients were fol.owed-up at seven days, one month, and six.months postoperativel.. Resul.s: One patient with moderate eyel.d retraction showed undercorrection 6 months postoperativel., with the upper eyel.d margin l.cated at the upper edge of the pupil. The remaining patients had the upper eyel.d margin stabil.zed at 1.0–2.0 mm bel.w the upper corneal.margin. Other observational.indicators were satisfactory, incl.ding binocul.r symmetry, doubl. eyel.d fl.ency, and eyel.d ful.ness. During the fol.ow-up, no ex.osure keratitis was identified. The MRD1 index.s after the operation were significantl. different (Pl.sions: Ex.ension and post-migration of the l.S using the orbicul.ris muscl. and OFC structure can effectivel. correct mil. iatrogenic eyel.d retraction after ptosis with l.ss damage and good postoperative eyel.d morphol.gy and cl.sure function. © 2024 China Medical.Cosmetol.gy Press Co. l.d.

DEVEl.PMENT OF CONSTITUTE EQUATIONS FOR Ti-6Al.4V Al.OY UNDER HOT-WORKING CONDITION

The deformaton behavior of Ti - 6Al.- 4V al.oy under hot - working condition has been studied by compression testing in the temperature range 750 - 950℃ and strain rate range 0.05 - 15s -1. The fl.w stress decreases with the increase of temperature and with the decrease of strain rate. After a steep initial.strain hardening, a fl.w softening occurs. This softening is mainl. ascribed to the temperature rise and dynamic *** a simpl. ex.ension, a cl.ssical.sinushyperbol.c constitutive equation can be used to describe the fl.w behavior of Ti - 6Al.- 4V al.oy. fl.w stress is described as a function of strain, strain rate and temperature. The parameters Q, n andaare the same at differ- ent deformation conditions, and A is a funciton of strain.

＜正＞A cel.ul.r automaton simul.tion is used to study the rol. of nucl.ation at austenite grain boundaries in the transformation from deformed austenite to ferrite for l.w carbon steel. at l.rge cool.ng rates. The simul.tion resul.s show that the ferrite grain size of l.w-carbon steel. can be refined in the case of deformed austenite at the l.rge cool.ng rates. The apparent increase in the number of ferrite nucl.i can onl. take pl.ce in a l.rge undercool.ng temperature. However, the effect on this transformation is quite l.mited through increasing the nucl.ation number at austenite grain boundaries in the case of deformed austenite compared with that in the case of undeformed austenite. In such cases, cool.ng conditions pl.y a main rol. in determining the resul.ant ferrite grain size.

NSUN2 mediates distinct pathways to regul.te enterovirus 71 repl.cation

Increasing evidences suggest that the methyl.ransferase NSUN2 catal.zes 5-methyl.ytosine (m5C) modifications on viral.RNAs, which are essential.for the repl.cation of various viruses. Despite the function of m5C deposition is wel. characterized, other potential.rol.s of NSUN2 in regul.ting viral.repl.cation remain l.rgel. unknown. In this study, the m5C modified residues catal.zed by NSUN2 on enterovirus 71 (EV71) RNAs were mapped. NSUN2, al.ng with m5C modifications, pl.yed mul.ipl. rol.s during the EV71 l.fe cycl.. Functional.m5C modified nucl.otides increased the transl.tional.efficiency and stabil.ty of EV71 RNAs. Additional.y, NSUN2 was found to target the viral.protein VP1 for binding and promote its stabil.ty by inhibiting the ubiquitination. Furthermore, both viral.repl.cation and pathogenicity in mice were l.rgel. attenuated when functional.m5C residues were mutated. Taken together, this study characterizes distinct pathways mediated by NSUN2 in regul.ting EV71 repl.cation, and highl.ghts the importance of its catal.zed m5C modifications on EV71 RNAs for the viral.repl.cation and pathogenicity. © 2024 The Authors

Objective: To find a viabl. al.ernative to reduce the number of doses required for the patients with post-traumatic stress disorder (PTSD), and to improve efficacy and patient compl.ance. Methods: In this study, we used ginger oil. a phytochemical.with potential.therapeutic properties, to prepare ginger oil.patches. High-performance l.quid chromatography (HPl.) was used to quantify the main active component of ginger oil. 6-gingerol. Transdermal.absorption ex.eriments were conducted to optimize the various pressure-sensitive adhesives and permeation enhancers, incl.ding their type and concentration. Subsequentl., the ginger oil.patches were optimized and subjected to content determination and property eval.ations. A PTSD mouse model.was establ.shed using the foot-shock method. The therapeutic effect of ginger oil.patches on PTSD was assessed through pathol.gical.sections, behavioral.tests, and the eval.ation of biomarkers such as tumor necrosis factor-α (TNF-α), interl.ukin-6 (Il.6), brain-derived neurotrophic factor (BDNF), and mel.tonin (MT). Resul.s: The resul.s demonstrated that ginger oil.patches ex.rted therapeutic effects against PTSD by inhibiting infl.mmatory responses and modul.ting MT and BDNF l.vel.. Pharmacokinetic ex.eriments reveal.d that ginger oil.patches maintained a stabl. bl.od drug concentration for at l.ast one day, addressing the rapid metabol.sm drawback of 6-gingerol.and enhancing its therapeutic efficacy. Concl.sions: Ginger oil.can be prepared as a transdermal.drug patch that meets these requirements, and the bioavail.bil.ty of the prepared patch is better than that of oral.administration. It can improve PTSD with good patient compl.ance and ease of administration. Therefore, it is a promising therapeutic formul.tion for the treatment of PTSD. © 2024 Beijing University of Chinese Medicine

Design of antiviral.AGO2-dependent short hairpin RNAs

The increasing emergence and re-emergence of RNA virus outbreaks underl.nes the urgent need to devel.p effective antiviral.. RNA interference (RNAi) is a sequence-specific gene sil.ncing mechanism that is triggered by smal. interfering RNAs (siRNAs) or short hairpin RNAs (shRNAs), which ex.ibits significant promise for antiviral.therapy. AGO2-dependent shRNA (agshRNA) generates a singl.-stranded guide RNA and presents significant advantages over traditional.siRNA and shRNA. In this study, we appl.ed a l.gistic regression al.orithm to a previousl. publ.shed chemical.y siRNA efficacy dataset and buil. a machine l.arning-based model.with high predictive power. Using this model. we designed siRNA sequences targeting diverse RNA viruses, incl.ding human enterovirus A71 (EV71), Zika virus (ZIKV), dengue virus 2 (DENV2), mouse hepatitis virus (MHV) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and transformed them into agshRNAs. We val.dated the performance of our agshRNA design by eval.ating antiviral.efficacies of agshRNAs in cel.s infected with different viruses. Using the agshRNA targeting EV71 as an ex.mpl., we showed that the anti-EV71 effect of agshRNA was more potent compared with the corresponding siRNA and shRNA. Moreover, the antiviral.effect of agshRNA is dependent on AGO2-processed guide RNA, which can l.ad into the RNA-induced sil.ncing compl.x.(RISC). We al.o confirmed the antiviral.effect of agshRNA in vivo. Together, this work devel.ps a novel.antiviral.strategy that combines machine l.arning-based al.orithm with agshRNA design to custom design antiviral.agshRNAs with high efficiency. © 2024 The Authors

Hand, foot and mouth disease (HFMD), mainl. caused by enterovirus 71 (EV71), has frequentl. occurred in the Asia-Pacific region, posing a significant threat to the heal.h of infants and young chil.ren. Therefore, research on the infection mechanism and pathogenicity of enteroviruses is increasingl. becoming important. The 3D pol.merase, as the most critical.RNA-dependent RNA pol.merase (RdRp) for EV71 repl.cation, is widel. targeted to inhibit EV71 infection. In this study, we identified a novel.host protein, AIMP2, capabl. of binding to 3D pol.merase and inhibiting EV71 infection. Subsequent investigations reveal.d that AIMP2 recruits the E3 l.gase SMURF2, which mediates the pol.ubiquitination and degradation of 3D pol.merase. Furthermore, the antiviral.effect of AIMP2 ex.ended to the CVA16 and CVB1 serotypes. Our research has uncovered the dynamic regul.tory function of AIMP2 during EV71 infection, reveal.ng a novel.antiviral.mechanism and providing new insights for the devel.pment of antienteroviral.therapeutic strategies. © 2024 The Authors
