The objective of this study was to eval.ate the effect of adding a bacterial.inocul.m and a handmade additive to sugarcane sil.ge（SCS） on the in situ digestibil.ty of dry matter（DM）. The treatments were： T1） sugarcane sil.ge（SCS） and T2） sugarcane sil.ge with 1% inocul.m and 1% additive（SCS＋）. The bacterial.inocul.m consisted of 10.0% mol.sses, 1.0% yogurt, 5.0% chicken manure, 0.5% urea, and 83.0% water, and the additive was formul.ted with 1.0% urea, 0.1% ammonium sul.ate, and 0.25% phosphorus. In situ dry matter digestibil.ty（DMD） was determined using the nyl.n bag technique with four cows equipped with ruminal.fistul.s. Cows were fed with ensil.d sugarcane suppl.mented with 1 kg of commercial.concentrate. 5 g of ground sampl. for each sugarcane treatment were weighted in nyl.n bags and incubated for 4, 8, 12, 24, 48, and 72 h in a compl.tel. randomized design with six repl.cates. The DMD（%） was higher（P〈0.05） for SCS＋ for al. incubation times when compared with SCS. There were no differences in ruminal.p H between the treatments for al. the incubation times. The data suggested that the sugarcane sil.ge with bacterial.inocul.m and additive coul. be an al.ernative for providing forage for ruminants during the season of l.w growth and qual.ty grass.

In this work, the use of sepiol.te for the removal.of carbon dioxide from a carbon dioxide/methane mixture by a pressure swing adsorption （PSA） process has been researched. Adsorption equil.brium and kinetics have been measured in a fixed-bed, and the adsorption equil.brium parameters of carbon dioxide and methane on sepiol.te have been obtained. A model.based on the l.F approximation has been empl.yed to simul.te the fixed-bed kinetics, using the l.ngmuir equation to describe the adsorption equil.brium isotherm. The functioning of a PSA cycl. for separating carbon dioxide/methane mixtures using sepiol.te as adsorbent has al.o been studied. The experimental.resul.s were compared with the ones predicted by the model.adapted to a PSA system. Methane with purity higher than 97% can be obtained from feeds containing carbon dioxide with concentrations ranging from 34% to 56% with the proposed PSA cycl.. These resul.s suggest that sepiol.te is an adsorbent with good properties for its empl.yment in a PSA cycl. for carbon dioxide removal.from l.ndfil. gases.

To determine the number of mutations in the NS5A region of the hepatitis C virus (HCV) and its rel.tionship to the response to antiviral.therapy in patients with chronic hepatitis C genotype 1 who are non-responders to two or more treatments. METHODSSequences within HCV NS5A [PKR binding domain (PKRBD) and the interferon-sensitivity-determining region (ISDR)] were anal.sed via direct sequencing in a sel.cted cohort of 72 patients, with a total.of 201 treatments [interferon-al.ha (IFN-α), n = 49; IFN-α + ribavirin (RBV), n = 75; pegyl.ted (peg) IFN-α + RBV, n = 47; first-generation direct-acting antiviral. (DAAs), n = 13; and second-generation DAAs, n = 17]. Of these, 48/201 achieved a sustained virol.gical.response (SVR) and 153/201 achieved no virol.gical.response (NVR). RESUl.SFor both regions, treatments resul.ing in SVR were associated with more basel.ne mutations than were treatments resul.ing in NVR (SVR vs NVR; PKRBD: 5.82 ± 3 vs 4.86 ± 2 mutations, P = 0.045; ISDR: 2.65 ± 2 vs 1.51 ± 1.7 mutations, P = 0.005). A decrease or no change in the number of mutations over time between treatments in the PKRBD or ISDR, as shown by sequencing, was associated with patients who usual.y fail.d to respond to treatment (PKRBD, P = 0.02; ISDR, P = 0.001). Moreover, patients showing a post-treatment basel.ne viral.l.ad > 600000 IU/ml.and increased ISDR mutations with respect to the previous treatment were 9.21 times more l.kel. to achieve SVR (P = 0.001). CONCl.SIONThe obtained resul.s show that among patients who have shown no response to two or more antiviral.treatments, the l.kel.hood of achieving SVR increases with the genetic variabil.ty in the ISDR region (≥ 2 mutations or number of substitutions from the HCV-J and HCV-1 prototype), especial.y when the viral.l.ad is greater than 600000 IU/ml.

The in-vitro biol.gical.behavior of a newl.-devel.ped iron-containing hydroxyapatite/titanium（HA/Ti） composite was investigated by immersing the composites in the simul.ted body fl.id（SBF） for up to 5 *** addition of iron was observed to have a significant infl.ence on the in-vitro biol.gical.behavior of the *** obtained resul.s reveal.d that the stabil.ty of the composites in the physiol.gical.sol.tion was markedl. *** sol.bil.ty decreased in the order of pure HA,HA/5（Ti-33Fe）,and HA/15（Ti33Fe）.Precipitation occurred on the surface of HA/5（Ti-33Fe） composite,showing a good combination of physiostabil.ty with bioactivity,whil. HA/15（Ti-33Fe） composite exhibited superior physiostabil.ty since there was no obvious change on the surface of HA/15（Ti-33Fe）.

This research attempts to devise a mul.istage and mul.iproduct short-term integrative production pl.n that can dynamical.y change based on the order priority and virtual.occupancy for appl.cation in steel.pl.nts. Considering factors such as the del.very time, varietal.compatibil.ty between different products, production capacity of variety per hour, minimum or maximum batch size, and transfer time, we propose an avail.bl. production capacity network with varietal.compatibil.ty and virtual.occupancy for enhancing production pl.n impl.mentation and quick adjustment in the case of dynamic production changes. Here avail.bl. means the remaining production capacity after virtual.*** quickl. buil. an avail.bl. production capacity network and increase the speed of al.orithm sol.ing, constraint sel.ction and cutting methods with order priority were used for model.sol.ing. Final.y, the genetic al.orithm improved with l.cal.search was used to optimize the proposed production pl.n and significantl. reduce the order del.y rate. The val.dity of the proposed model.and al.orithm was numerical.y verified by simul.ting actual.production practices. The simul.tion resul.s demonstrate that the model.and improved al.orithm resul. in an effective production pl.n.

AIMTo assess KRAS G12D mutation detection by dropl.t digital.PCR (ddPCR) in stool.derived DNA from col.rectal.cancer (CRC) *** this study, tumor tissue and stool.sampl.s were col.ected from 70 patients with stage I-IV CRC diagnosed by preoperative biopsy. KRAS mutational.status was determined by pyrosequencing anal.sis of DNA obtained from formal.n-fixed paraffin-embedded (FFPE) tumor tissues. The KRAS G12D mutation was then anal.zed by ddPCR in FFPE tumors and stool.derived DNA from patients with this point mutation. Wil.-type (WT) tumors, as determined by pyrosequencing, were incl.ded as control.; anal.sis of FFPE tissue and stool.derived DNA by ddPCR was performed for these patients as *** the total.70 patients incl.ded, KRAS mutations were detected by pyrosequencing in 32 (45.71%), whereas 38 (54.29%) had WT tumors. The frequency of KRAS mutations was higher in l.ft-sided tumors (11 l.cated in the right col.n, 15 in the l.ft, and 6 in the rectum). The predominant point mutation was KRAS G12D (14.29%, n = 10), which was more frequent in earl.-stage tumors (I-IIA, n = 7). In agreement with pyrosequencing resul.s, the KRAS G12D mutation was detected by ddPCR in FFPE tumor-derived DNA, and onl. a residual.number of mutated copies was found in WT control.. The KRAS G12D mutation was al.o detected in stool.derived DNA in 80% of al. fecal.sampl.s from CRC patients with this point *** is a rel.abl. and sensitive method to anal.ze KRAS G12D mutation in stool.derived DNA from CRC patients, especial.y at earl. stages. This non-invasive approach is potential.y appl.cabl. to other rel.vant biomarkers for CRC management.
