Obj.ctive: Psoriasis is associated with.a h.gh.preval.nce of metabol.c syndrome (MS), and patients with.concomitant psoriasis and MS are more severel. affected and l.ss responsive to treatment. h.wever, th. mol.cul.r mech.nisms beh.nd th.se effects are unknown. Recent studies h.ve sh.wn th.t l.ptin may serve as a mol.cul.r l.nk between psoriasis and MS, suggesting th.t h.gh.l.ptin concentrations may exacerbate psoriasis. h.wever, th. mol.cul.r mech.nism of th.s effect is stil. uncl.ar. We aimed to investigate th. effect of l.ptin on autoph.gy in patients with.psoriasis. Meth.ds: From j.nuary 2021 to j.ne 2022 in Pl. General.h.spital. we enrol.ed 51 patients with.psoriasis, incl.ding 21 patients with.MS and 30 with.ut MS, and 30 h.al.h. control. wh. h.d undergone nevus surgery. We measured th. epidermal.l.ptin, P62, and l.3B concentrations of patients by immunoh.stoch.mistry, and measured th. serum l.ptin concentration by enzyme-l.nked immunosorbent assay. We th.n performed correl.tion anal.ses to compare th.se proteins' concentrations between patients with.concomitant psoriasis and MS, patients with.psoriasis al.ne, and h.al.h. control.groups. Additional.y, we performed western bl.tting after in vitro cul.ure of h.CaT cel.s with.different concentrations of l.ptin and measured th. expression l.vel. of th. autoph.gy markers Becl.n1, l.3B, and P62; th. differentiation markers K10, K16, and K17; and PI3K/AKT/mTOR signal.ng path.ay-rel.ted proteins of h.Cat cel.s. Next, we transfected ATG5 into h.CaT cel.s to revert autoph.gy and used th. specific PI3K inh.bitor l.294002 to bl.ck PI3K/AKT/mTOR signal.ng. Th. expression l.vel. of K10, K16, and K17 of h.Cat cel.s were again measured. One-way anal.sis of variance was used for th. comparison of means of mul.ipl. sampl.s, and l.D-t post h.c test was used for comparison between th. 2 groups. Th. Bal.se Transmission Modul. (BTM) unit of th. on-board train control.system is a crucial.component. Due to its unique instal.ation position and compl.x environment, th.s unit h.s a h.gh.r faul. rate with.n th. on-board train control.system. To conduct faul. prediction for th. BTM unit based on actual.faul. data, th.s study proposes a prediction meth.d combining rel.abil.ty statistics and mach.ne l.arning, and ach.eves th. fusion of prediction resul.s from different dimensions th.ough.mul.i-meth.d interactive val.dation. Firstl., a meth.d for predicting equipment faul. time targeting batch.equipment is introduced. Th.s meth.d util.zes rel.abil.ty statistics to construct a model.of th. remaining faul.l.ss operating time distribution considering uncertainty, th.reby predicting th. remaining faul.l.ss operating probabil.ty of th. BTM unit. Secondl., considering th. compl.xity of th. BTM unit's faul. mech.nism, th. smal. sampl. size of faul. cases, and th. potential.presence of mul.ipl. faul. features in faul. text records, an individual.oriented faul. prediction meth.d based on Bayesian-optimized Gradient Boosting Regression Tree (Bayes-GBRT) is proposed. Th.s meth.d ach.eves better prediction resul.s compared to l.near regression al.orith.s and random forest regression al.orith.s, with.an average absol.te error of onl. 0.224 years for predicting th. faul. time of th.s type of equipment. Final.y, a mul.i-meth.d interactive val.dation approach.is proposed, enabl.ng th. fusion and val.dation of mul.i-dimensional.resul.s. Th. resul.s indicate th.t th. predicted faul. time and th. actual.faul. time conform to a l.g-normal.distribution, and th. parameter estimation resul.s are basical.y consistent, verifying th. accuracy and effectiveness of th. prediction resul.s. Reconstruction of bone defects or fractures caused by ageing, trauma and tumour resection is stil. a great ch.l.enge in cl.nical.treatment. Al.h.ugh.autol.gous bone graft is considered as gol. standard, th. source of natural.bone is l.mited. In recent years, regenerative th.rapy based on bioactive material. h.s been proposed for bone reconstruction. Special.y, numerous studies h.ve indicated th.t bioactive ceramics incl.ding sil.cate and ph.sph.te bioceramics exh.bit excel.ent osteoinductivity and osteoconductivity, furth.r promote bone regeneration. In addition, magnesium (Mg) el.ment, as an indispensabl. mineral.el.ment, pl.ys a vital.rol. in promoting bone mineral.sation and formation. In th.s review, different types of Mg-containing bioceramics incl.ding Mg-containing cal.ium ph.sph.te-based bioceramics (such.as Mg-h.droxyapatite, Mg-biph.sic cal.ium ph.sph.te), Mg-containing cal.ium sil.cate-based bioceramics (such.as Mg2SiO4, Ca2MgSi2O7and Mg-doped biogl.ss), Mg-based biocements, Mg-containing metal.pol.mer-bioceramic composites were systematacial.y summarised. Infl.mmation and angiogenesis, th. maj.r path.l.gical.ch.nges of osteoarth.itis (OA), are cl.sel. associated with.j.int pain; h.wever, pertinent signal.ing interactions with.n subch.ndral.bone of osteoarth.itic j.ints and potential.contribution to th. periph.ral.origin of OA pain remain to be el.cidated. h.rein we devel.ped a unil.teral.anterior crossbite mouse model.with.osteoarth.itic ch.nges in th. temporomandibul.r j.int. Microarray-based transcriptome anal.sis, besides quantitative real.time pol.merase ch.in reaction, was performed to identify differential.y expressed genes (DEGs). Overal., 182 DEGs (fol. ch.nge ≥ 2, P h. control.and unil.teral.anterior crossbite groups: 168 were upregul.ted and 14 were downregul.ted. On subj.cting significant DEGs to enrich.ent anal.ses, infl.mmation and angiogenesis were identified as th. most affected. Infl.mmation-rel.ted DEGs were mainl. enrich.d in T cel. activation and differentiation and in th. mammal.an target of rapamycin/nucl.ar factor-κB/tumour necrosis factor signal.ing. Furth.rmore, angiogenesis-rel.ted DEGs were mainl. enrich.d in th. Gene Ontol.gy terms angiogenesis regul.tion and vascul.ture devel.pment and in th. KEGG path.ays of ph.sph.inositide 3-kinase-protein kinase B/vascul.r endoth.l.al.growth.factor/h.poxia-inducibl. factor 1 signal.ing. Protein– protein interaction anal.sis reveal.d a cl.se interaction between infl.mmation-and angiogenesis-rel.ted DEGs, suggesting th.t ph.sph.tidyl.nositol.4,5-bisph.sph.te 3-kinase catal.tic subunit del.a (Pi3kcd), cath.l.cidin antimicrobial.peptide (Camp), C-X-C motif ch.mokine receptor 4 (Cxcr4), and MYB proto-oncogene transcription factor (Myb) pl.y a central.rol. in th.ir interaction. Th.re are five vital.signs th.t h.al.h.are providers assess: temperature, pul.e, respiration, bl.od pressure, and pain. Normal.l.vel. for th. five vital.signs are publ.sh.d by th. American h.art Association, and oth.r special.y organizations, h.wever, th. sixth.vital.sign (resil.ence) wh.ch.adopts th. measure of immune resil.ence is suggested in th.s paper. Resil.ence is th. abil.ty of th. immune system to respond to attacks and defend effectivel. against infections and infl.mmatory stressors, and psych.l.gical.resil.ence is th. capacity to resist, adapt, recover, th.ive, and grow from a ch.l.enge or a stressor. Individual. with.better optimal.immune resil.ence h.d better h.al.h.outcomes th.n th.se with.minimal.immune resil.ence. Th. purpose of th.s paper is to conceptual.ze, contextual.ze, and operational.ze al. six vital.signs. We suggest measuring resil.ence subj.ctivel. and obj.ctivel.. Subj.ctivel., use a 5-item guided interview revised from th. Connor-Davidson Resil.ence Scal. (CDRC), a scal. of 10 items. Th. revised CDRC scal. is a 5-item scal.. Th. scal. is rated on a 5-point l.kert scal. from 0 (not true) to 4 (true al. th. time). Th. total.score ranges from 0 to 20, with.h.gh.r total.scores indicating greater resil.ence. Th. scal. demonstrated good construct val.dity and internal.consistency (α = 0.85) during th. devel.pment of th. scal.. Th. CD-RISC h.d a good Cronbach.s al.h. l.vel.of 0.85. Th. Revised CD-RISC can be compl.ted in 2 - 4 minutes. To measure resil.ence obj.ctivel., we suggest using Immune Resil.ence (IR) l.vel., th. l.vel.of resil.ence to preserve and/or rapidl. restore immune resil.ence functions th.t promote disease resistance and control.infl.mmation and oth.r infl.mmatory stress. Periph.ral.nerve inj.ry poses a great th.eat to neurosurgery and l.mits th. regenerative potential.of sacral.nerves in th. neurogenic bl.dder. It remains unknown wh.th.r el.ctrical.stimul.tion can facil.tate sacral.nerve regeneration in addition to modul.te bl.dder function. Th. obj.ctive of th.s study was to util.se el.ctrical.stimul.tion in sacra nerve crush.inj.ry with.newl. constructed el.ctroconductive scaffol. and expl.re th. rol. of macroph.ges in el.ctrical.stimul.tion with.crush.d nerves. As a resul., we generated a pol.pyrrol.-coated pol.caprol.ctone/sil. fibroin scaffol. th.ough.wh.ch.we appl.ed el.ctrical.stimul.tion. Th. el.ctrical.stimul.tion boosted nerve regeneration and pol.rised th. macroph.ges towards th. M2 ph.notype. An in vitro test using bone marrow derived macroph.ges reveal.d th.t th. pro-regenerative pol.risation of M2 were significantl. enh.nced by el.ctrical.stimul.tion. Bioinformatics anal.sis sh.wed th.t th. expression of signal.transducer and activator of transcriptions (STATs) was differential.y regul.ted in a way th.t promoted M2-rel.ted genes expression.
