The effects of anti-vascular endothelial growth factor agents on human retinal pigment epithelial cells under high glucose conditions

AIM： To investigate the effects of high glucose levels and anti-vascular endothelial growth factor（VEGF） agents（bevacizumab,ranibizumab and aflibercept） on retinal pigment epithelium（RPE） ***： ARPE-19 cells were cultured at different glucose levels（5.5 mmol/L,25 mmol/L,and 75 mmol/L）.Cell viability was evaluated by MTT assay at 3d after treatment with *** migration ability was measured by wound healing assay at 3d.A cell death detection kit was used to assess apoptosis at 3 and 14 *** proliferation was assessed by EdU assay at *** culture medium was treated with anti-VEGF agents at clinically relevant *** experiment was then repeated at a different glucose ***： The viability and migration of ARPE-19 cells were significantly decreased in the presence of 75 mmol/L as compared to 5.5 mmol/L *** percentage of TUNEL-positive cells was significantly increased and the proliferative potential was decreased with 75 mmol/L compared to 5.5 mmol/L *** were no significant differences in the results between 25 mmol/L and 5.5 mmol/L *** the presence of 75 mmol/L glucose,the groups treated with anti-VEGF showed decreased cell viability and proliferation and increased ***,there were no significant differences between the anti-VEGF ***： High glucose level decreases the viability,wound healing ability,and proliferation of RPE cells,while increasing ***,anti-VEGF agents interfered with the physiological functions of RPE cells under high-glucose conditions,accompanied by decreases in cell viability and proliferation.

Effects of foundation damage and water-level change on vibration modal parameters of gravity-type caisson structure

In this paper,the effects of foundation damage and water-level change on vibration characteristics of gravity-type caisson structure are examined by analyzing modal parameters extracted from output-only *** achieve the objective,the following approaches are ***,vibration response analysis methods are selected to estimate power spectral density and modal parameters such as natural frequency,damping ratio and mode shape of a lab-scale caisson structural ***,vibration tests on the lab-scale caisson system are performed under a series of test scenarios which include three water-level changes and three damage ***,experimental modal parameters corresponding to the damaging cases as well as the water level cases are extracted by frequency domain decomposition method and stochastic subspace identification ***,the effects of the water-level variation and foundation damage on the extracted modal parameters are examined to assess the feasibility of the vibration-based damage detection in gravity-type caisson structures under water-level uncertainty.

Comparative detection of syndecan-2 methylation in preoperative and postoperative stool DNA in patients with colorectal cancer

BACKGROUND Early detection of colorectal cancer(CRC)is essential to reduce cancer-related morbidity and *** DNA(sDNA)testing is an emerging method for early CRC ***-2(SDC2)methylation is a potential biomarker for the sDNA *** DNA methylation is an early epigenetic event during tumorigenesis and can occur in the normal colonic mucosa during aging,which can compromise the sDNA test *** In this prospective study,we enrolled 151 patients with CRC who underwent curative surgical resection between September 2016 and May *** stool samples were collected from 123 patients and postoperative samples were collected from 122 patients.A total of 104 samples were collected from both preoperative and postoperative *** promoter methylation of SDC2 in sDNA was assessed using linear target enrichment quantitative methylation-specific real-time polymerase chain *** parameters were analyzed using the results of SDC2 *** Detection rates of SDC2 methylation in the preoperative and postoperative stool samples were 88.6%and 19.7%,*** tumor size(3 cm,P=0.019)and advanced T stage(T3–T4,P=0.033)were positively associated with the detection rate of SDC2 methylation before *** sex was associated with false positives after surgery(P=0.030).Cycle threshold(CT)values were significantly decreased postoperatively compared with preoperative values(P<0.001).The postoperative negative conversion rate for preoperatively methylated SDC2 was 79.3%(73/92).CONCLUSION Our results suggested that the SDC2 methylation test for sDNA has acceptable sensitivity and ***,small size and early T stage tumors are associated with a low detection rate of SDC2 *** the cycle threshold values significantly decreased after surgery,SDC2 methylation test for sDNA might have a diagnostic value for CRC.

A simple, flexible and high-throughput cloning system for plant genome editing via CRISPR-Cas system

CRISPR-Cas9 system is now widely used to edit a target genome in animals and plants. Cas9 protein derived from Streptococcus pyogenes（Sp Cas9） cleaves double-stranded DNA targeted by a chimeric single-guide RNA（sg RNA）. For plant genome editing, Agrobacterium-mediated T-DNA transformation has been broadly used to express Cas9 proteins and sg RNAs under the control of Ca MV 35 S and U6/U3 promoter, respectively. We here developed a simple and high-throughput binary vector system to clone a 19 20 bp of sg RNA, which binds to the reverse complement of a target locus, in a large T-DNA binary vector containing an Sp Cas9 expressing cassette. Twostep cloning procedures：（1） annealing two target-specific oligonucleotides with overhangs specific to the Aar I restriction enzyme site of the binary vector; and（2） ligating the annealed oligonucleotides into the two Aar I sites of the vector, facilitate the high-throughput production of the positive clones. In addition, Cas9-coding sequence and U6/U3 promoter can be easily exchanged via the GatewayTMsystem and unique Eco RI/Xho I sites on the vector, respectively. We examined the mutation ratio and patterns when we transformed these constructs into Arabidopsis thaliana and a wild tobacco, Nicotiana attenuata. Our vector system will be useful to generate targeted large-scale knock-out lines of model as well as non-model plant.

Programmed death ligand-1 expression and its prognostic role in esophageal squamous cell carcinoma

AIM To investigate the expression and prognostic role of programmed death ligand-1(PD-L1) in locally advanced esophageal squamous cell carcinoma(ESCC).METHODS A total of 200 patients with ESCC who underwent radical esophagectomy with standard lymphadenectomy as the initial definitive treatment in Seoul National University Hospital from December 2000 to April 2013 were eligible for this analysis. Tissue microarrays were constructed by collecting tissue cores from surgical specimens, and immunostained with antibodies directed against PD-L1, p16, and c-Met. Medical records were reviewed retrospectively to assess clinical outcomes. Patients were divided into two groups by PD-L1 status, and significant differences in clinicopathologic characteristics between the two groups were assessed. RESULTS Tumor tissues from 67 ESCC patients(33.5%) were PDL1-positive. Positive p16 expression was observed in 21 specimens(10.5%). The H-score for c-Met expression was ≥ 50 in 42 specimens(21.0%). Although PDL1-positivity was not significantly correlated with any clinical characteristics including age, sex, smoking/alcoholic history, stage, or differentiation, H-scores for c-Met expression were significantly associated with PDL1-positivity(OR = 2.34, 95%CI: 1.16-4.72, P = 0.017). PD-L1 expression was not significantly associated with a change in overall survival(P = 0.656). In contrast, the locoregional relapse rate tended to increase(P = 0.134), and the distant metastasis rate was significantly increased(HR = 1.72, 95%CI: 1.01-2.79, P = 0.028) in patients with PD-L1-positive ESCC compared to those with PD-L1-negative *** PD-L1 expression is positively correlated with c-Met expression in ESCC. PD-L1 may play a critical role in distant failure and progression of ESCC.

Ultimate strength of single shear bolted connections with cold-formed ferritic stainless steel

This paper is focused on the structural behavior of the single shear bolted connections with thin-walled ferritic stainless *** purpose of this study is to investigate the ultimate behaviors,such as ultimate strength and fracture mode of the single shear bolted connections of thin-walled ferritic stainless steel(low cost steel) rather than austenitic stainless steel(high cost steel).Bolt arrangement and end distance parallel to the direction of applied load are considered as main variables of the test specimens for bolted *** have a constant dimension of edge distance perpendicular to the loading direction,bolt diameter,pitch,and gauge.A monotonic tensile test for specimens has been carried out and some bolted connections with long end distance showed curling(out of plane deformation) occurrence which led to strength *** ultimate behaviors such as fracture mode,ultimate strength are compared with those predicted by current design ***,conditions of curling occurrence and the strength reduction due to curling are investigated and modified strength equations are suggested considering the curling effect.

Surface treatment to improve corrosion resistance of Al plate heat exchangers

The correlations between thermal and physical properties were studied through thermal conductivity measurements, hardness tests, salt spray tests (AASS) among the surface treatment samples named K20, K40 with thickness of 20, 40 μm respectively and raw sample named K00. In thermal conductivity measurements, there are little differences among the samples as K00, K20 and K40, they exhibit 153.39, 150.69 and 149.76 W/(m·K), respectively. According to hardness tests, K00, K20 and K40 exhibit 87.9, 259.7 and 344.8 in Vickers values. In the result of salt spray tests to examine the effects on corrosion resistance, K00, K20 and K40 exhibit the grade of 3?5, 2.0?9.8 and 10, respectively. The mutual relation of the above results was analyzed. It is found that the surface treatments do not affect the thermal conductivity of aluminum and result in the improvement of physical properties. As a result of the technology, the surface improvement of aluminum alloy specimen is achieved without thermal degradation. It validates the ability of the aluminum plate heat exchangers with surface treatment to enhance the corrosion resistance. Present work is performed as the first fundamental threshold in the process of aluminum plate heat exchangers development to check out its possibility, therefore the next step-experimental and numerical study of practical aluminum plate heat exchangers will be made.

Mechanical properties of high-strength concrete subjected to high temperature by stressed test

Recently, the effects of high temperature on compressive strength and elastic modulus of high strength concrete were experimentally investigated. The present study is aimed to study the effect of elevated temperatures ranging from 20 ℃ to 700 ℃ on the material mechanical properties of high-strength concrete of 40, 60 and 80 MPa grade. During the strength test, the specimens are subjected to a 25% of ultimate compressive strength at room temperature and sustained during heating, and when the target temperature is reached, the specimens are loaded to failure. The tests were conducted at various temperatures (20-700 ℃) for concretes made with W/B ratios of 46%, 32% and 25%, respectively. The results show that the relative values of compressive strength and elastic modulus decrease with increasing compressive strength grade of specimen.
