Phosphorylated p70S6K in urothelial carcinoma of the tumor recurrence noninvasive low-grade bladder： correlation with

We investigated whether inhibiting phosphorylated p70S6K （p-p70S6K） suppresses the proliferation and growth of noninvasive low-grade urothelial carcinoma （LG-URCa） in vitro and whether p-p70S6K can serve as a predictive biomarker for the recurrence of noninvasive LG-URCa of the bladder in patients. We constructed a tissue microarray （TMA） for 95 LG-URCa and 35 benign urothelium samples and performed immunohistochemical staining for p-p70S6K and p-4E-BP1. A Cox regression model was used to investigate the predictive factors for recurrence of LG-URCa. We investigated the dose-dependent antiproliferative effect of rapamycin, its antiproliferative effect and the growth-inhibition effect of p70S6K siRNA transfection in RT4 and 253J cell lines. The pT1 staged group （P 〈 0.05; hazard ratio （HR）, 2.415） and the high p-p70S6K staining group （P 〈 0.05; HR, 2.249） were independent factors for predicting recurrence. Rapamycin inhibited RT4 and 253J cell proliferation in a dose-dependent manner （r = -0.850, P 〈 0.001 in RT4 cells; r = -0.835, P 〈 0.001 in 253J cells）. RT4 and 253J cell proliferation and growth were inhibited by the transfection of p70S6K siRNA and rapamycin, respectively （P 〈 0.05）. Transfection of p70S6K siRNA resulted in inhibitory effects on cell proliferation and growth that were similar to those of rapamycin. Our results suggest that inhibiting p70S6K phosphorylation is important to prevent recurrence and that p70S6K phosphorylation can be used as a molecular biomarker to predict recurrence of certain LG-URCa of the bladder.

Prognosis of hepatocellular carcinoma expressing cytokeratin 19:Comparison with other liver cancers

AIM:To investigate whether expressing biliary phenotype predicted poor outcome after the surgical treatment in primary liver cancers. METHODS:Out of 204 patients that underwent liver resection due to hepatocellular carcinoma (HCC), liver specimens of 70 patients with HCC were evaluated for biliary components by cytokeratin (CK) 19 immunostain (CK19 - HCC and CK19 + HCC). CK19 positivity was defined as membranous and/or cytoplasmic expression in ≥ 5% of tumor cells with moderate or strong intensity. Patients with other primary liver cancers, such as com- bined HCC and cholangiocarcinoma (cHCC-CC), intrahe- patic cholangiocarcinoma (ICC) who received curative liver resection, were also included in the study. Clinical characteristics of CK19-HCC and CK19 + HCC patients, including survival outcome after curative liver resection, were compared with that of cHCC-CC and ICC patients. RESULTS: The overall survival (OS) rate of CK19 - HCC(n = 49) after the curative surgical treatment was 90.7%, and 80.4% at 1 and 5 years after the resection. OS rate of CK19 + HCC (n = 21) was 74.3%, 28.9% and OS rate of cHCC-CC (n = 22) was 66.7%, 32.2% at 1 and 5 years after the surgery. For ICC (n = 19), 1 and 5-year-OS rate was 50.2% and 14.3% after the cura-tive resection. The OS rates of CK19 + HCC and cHCC-CC were significantly lower than that of CK19-HCC, but higher than the OS rate of ICC (P = 0.000). There was no statistically significant difference in OS rate between CK19 + HCC and cHCC-CC. The disease free survival (DFS) rate of CK19-HCC was 72.0% and 54.5% at 1 and 3 years after the surgical treatment. DFS rate of CK19 + HCC was 53.3%, 34.3% and DFS rate of cHCC- CC was 51.5%, 39.2% at 1 and 3 years after the resection. For ICC, 1 and 3-year-DFS rate was 28.0% and 14.0% after the curative resection. DFS rate of CK19-HCC was significantly higher than that of ICC (P = 0.017), but marginally higher than DFS rate of either CK19 + HCC or cHCC-CC (P = 0.097, P = 0.089, respec-tively).

Effect of Hemodilution on Fluctuation of Cerebral Oxygen Saturation during Cardiopulmonary Bypass in Children with Cyanotic Congenital Heart Disease

Background:In patients with cyanotic congenital heart disease(CHD),cerebral oxygenation may be maintained by elevations in hematocrit(Hct).Hemodilution accompanying cardiopulmonary bypass(CPB),however,can disrupt cerebral oxygen balance,leading to fluctuations in cerebral oxygen saturation(ScO_(2)).The present study investigated the effects of Hct changes on the fluctuation of ScO_(2)during CPB in cyanotic CHD using performance measurement(PM).Methods:Children with CHD(51 acyanotic and 46 cyanotic)who had undergone cardiac surgery using CPB were *** performance error(MDPE),median absolute performance error(MDAPE),and wobble parameters of ScO_(2)were calculated before(reference value),during,and after *** of PM parameters with Hct and reductions in Hct(ΔHct)were also ***:Before CPB,patients with cyanotic CHD had lower MDPE and larger wobble than those with acyanotic CHD,although mean ScO_(2)did not differ significantly between the two *** CPB,ScO_(2)of acyanotic CHD increased asΔHct increased,but PM variables were not associated withΔ*** cyanotic CHD,MDPE(r=−0.324,p=0.032)and MDAPE(r=0.339,p=0.024)correlated significantly withΔHct during *** CPB,MDPE(r=0.574,p=0.025)and MDAPE(r=−0.543,p=0.036)were significantly correlated with Hct in children with cyanotic CHD who underwent palliative ***:Therefore,ScO_(2)fluctuation during CPB in children with cyanotic CHD may be affected by the decrease in Hct,suggesting that excessive hemodilution can negatively influence the maintenance of cerebral oxygenation in these patients.

p73 G4C14 to A4T14 polymorphism is associated with colorectal cancer risk and survival

AIM:To analyze the association between the p73 G4C14-to-A4T14 polymorphism(a.k.a.,the GC/AT variation) and colorectal cancer risk and survival in the Korean population,and to evaluate the relationships between p73 polymorphism and the p73 protein expression or clinicopathological characteristics of colorectal ***:Three hundred and eighty-three histologically confirmed cases and 469 healthy controls,recruited at one teaching hospital in Pusan,Korea from 2001 and 2007,were genotyped for p73 G4C14-to-A4T14 by PCR with confronting two-pair primers(PCR-CTPP) and the expression profile of p73 in cancer tissues(n=383) was analyzed by ***:Odds ratios(ORs) and 95% confidence intervals(CIs) were calculated by unconditional logistic regression model adjusted for age and *** with the GC/GC genotypes,the GC/AT and AT/AT genotypes were significantly associated with colorectal cancer risk(GC/AT vs GC/GC:OR = 1.46,95% CI:1.10-1.94;AT/AT vs GC/GC:1.72,0.98-3.03;Ptrend=0.01).When stratified by age and gender,the association was restricted to those less than 60 years of age(GC/AT or AT/AT vs GC/GC:2.22,1.39-3.55) and male(GC/AT or AT/AT vs GC/GC:1.91,1.31-2.77).The expression of p73 was associated with invasion depth(P = 0.003) and advanced Duke's stage(P = 0.06) of colorectal *** patients with the GC/GC genotype were associated with worse survival compared with those with the other genotypes(P = 0.02).However,no signif icant relationship was observed between the p73 G4C14-to-A4T14 polymorphism and p73 protein expression in cancer ***:Our results suggest that the p73 GC/AT polymorphism is associated with an increased colorectal cancer risk and survival in the Korean population.

Effects of Melandrium firmum methanolic extract on testosterone-induced benign prostatic hyperplasia in Wistar rats

Benign prostatic hyperplasia （BPH） is an age-related disease of unknown aetiology characterized by prostatic enlargement coincident with distinct alterations in tissue histomorphology. Instead of therapeutic agents that can cause severe side effects, plant extracts are frequently used to treat BPH. In this study, we investigated whether the Melandrium firmum methanolic extract （MFME） improves BPH, using the testosterone propionate （TP）-induced BPH rat model. Castration was performed via the scrotal route under sodium pentobarbital anaesthesia. BPH in castrated rats was generated via daily subcutaneous injections of TP （3 mg kg-1） dissolved in corn oil, for 4 weeks. MFME was administered daily by oral garage at a dose of 200 mg kg-1 for 4 weeks, along with the TP injections. The control group received injections of corn oil subcutaneously. At the scheduled termination of the experiment, all rats were killed and their prostates weighed; the relative prostate weight （prostate/body weight ratio） was calculated, and histomorphological changes in the prostate were examined. Additionally, we measured the levels of testosterone and dihydrotestosterone （DHT） in the serum and the prostate. Experimentally induced BPH led to marked decreases in the relative prostate weight and the DHT levels in the serum and the prostate. Histologically, BPH was evident in the ventral lobe of the prostate, and MFME treatment suppressed the severity of the lesions. These results indicate that MFME effectively inhibits the development of BPH induced by testosterone in a rat model. Further studies will be needed to identify the compound（s） responsibility for inducing the protective effect against BPH and determine its mechanism of action,

Effect of Fe_2P in LiFePO_4/Fe_2P composite on electrochemical properties synthesized by MA and control of heat condition

In order to control the size and distribution of the high conductive Fe2P in LiFePO4/Fe2P composite, two different cooling rates (Fast: 15 ℃·min-1, Slow: 2 ℃·min-1) were employed after mechanical alloying. The discharge capacity of the fast cooled was 83 mAh·g-1 and the slow cooled 121 mAh·g-1. The particle size of the synthesized powder was examined by transmission electron microscopy and distribution of Fe2P was characterized using scanning electron microscopy (SEM). In addition, two-step heat treatment was carried out for better distribution of Fe2P. X-ray diffraction (XRD) and Rietveld refinement reveal that LiFePO4/Fe2P composite consists of 95.77% LiFePO4 and 4.33% of Fe2P.

Effect of Doubling the Ensemble Size on the Performance of Ensemble Prediction in the Warm Season Using MOGREPS Implemented at the KMA

Using the Met Office Global and Regional Ensemble Prediction System （MOGREPS） implemented at the Korea Meteorological Administration （KMA）, the effect of doubling the ensemble size on the performance of ensemble prediction in the warm season was evaluated. Because a finite ensemble size causes sampling error in the full forecast probability distribution function （PDF）, ensemble size is closely related to the efficiency of the ensemble prediction system. Prediction capability according to doubling the ensemble size was evaluated by increasing the number of ensembles from 24 to 48 in MOGREPS implemented at the KMA. The initial analysis perturbations generated by the Ensemble Transform Kalman Filter （ETKF） were integrated for 10 days from 22 May to 23 June 2009. Several statistical verification scores were used to measure the accuracy, reliability, and resolution of ensemble probabilistic forecasts for 24 and 48 ensemble member forecasts. Even though the results were not significant, the accuracy of ensemble prediction improved slightly as ensemble size increased, especially for longer forecast times in the Northern Hemisphere. While increasing the number of ensemble members resulted in a slight improvement in resolution as forecast time increased, inconsistent results were obtained for the scores assessing the reliability of ensemble prediction. The overall performance of ensemble prediction in terms of accuracy, resolution, and reliability increased slightly with ensemble size, especially for longer forecast times.

Fluorescence recovery after photobleaching(FRAP)and single particle tracking(SPT)techni-ques determine the diffusion coefficient from average diffusive motion of high-concentration molecules and from trajectories of low-concentration single molecules,*** dif-fusion coefficients measured by FRAP and SPT techniques for the same biomolecule on cell membrane have exhibited inconsistent values across laboratories and platforms with larger dif-fusion coefficient determined by FRAP,but the sources of the inconsistency have not been investigated ***,we designed an image-based FRAP-SPT system and made a direct comparison between FRAP and SPT for diffusion coefficient of submicron particles with known theoretical values derived from Stokes-Einstein equation in aqueous *** combined iFRAP-SPT technique allowed us to measure the diffusion coefficient of the same fluorescent particle by utilizing both techniques in a single platform and to scrutinize inherent errors and artifacts of *** results reveal that diffusion coefficient overestimated by FRAP is caused by inaccurate estimation of the bleaching spot size and can be corrected by simple image *** iFRAP-SPT technique can be potentially used for not only cellular membrane dynamics but also for quantitative analysis of the spatiotemporal distribution of the solutes in small scale analytical devices.

Intermediate-scale Analysis of Landscape Characteristics Affecting Edge Formation in Burned Forests in Samcheok,Korea

Compared to burn interiors, edges exhibit distinct biotic and abiotic conditions that include microclimate, wind speed, sunlight levels, soil composition, moisture content, nutrient availability, population density, and species diversity. This study characterized the landscapes in which burned forest edges formed in Samcheok, Korea. Over the study area, 500-m2 grid cells were generated to capture landscape characteristics. Grid cells intersecting burn boundary lines were designated as edge, while cells without these lines were classified as the interior of burned areas. Topographic variables including slope, elevation, topographic wetness index, solar radiation index, and proportions of fuel and land use types within grid cells were computed in a geographical information system(GIS). Correlation analysis with modified t-test and regression tree analysis were performed to explore the influences of landscape variables on edge formation with avoiding spatial autocorrelation problems. The results indicated that edges formed at low elevations with mild slopes, high topographic wetness, and low solar radiation. Edges were unlikely to form in areas dominated by Japanese red pines at low elevations. Moreover, heterogeneous land use/cover types contributed significantly to edge formation. Different forest management strategies for different landscape conditions can be more effective for enhancing resilience of forests to fire. Reducing susceptible fuel types might be effective at low elevations, while enhancing forest heterogeneity might be more effective at high elevations.

Densification simulation of compacted Al powders using multi-particle finite element method

The powder compaction simulations were performed to demonstrate deformation behavior of particles and estimate the effect of different punch speeds and particle diameters on the relative density of powder by a multi-particle finite element model(MPFEM). Individual particle discretized with a finite element mesh allows for a full description of the contact mechanics. In order to verify the reliability of compaction simulation by MPFEM, the compaction tests of porous aluminum with average particle size of 20 μm and 3 μm were performed at different ram speeds of 5, 15, 30 and 60 mm/min by MTS servo-hydraulic tester. The results show that the slow ram speed is of great advantage for powder densification in low compaction force due to sufficient particle rearrangement and compaction force increases with decrease in average particle size of aluminum.
