Nitrate regulates rhizobial and mycorrhizal symbiosis in common bean(Phaseolus vulgaris L.)

Nitrogen-limited conditions are considered to be a prerequisite for legume-rhizobial symbiosis, but the effects of nitrate-rich conditions on symbiotic status remain poorly understood. We addressed this issue by examining rhizobial （Rhizobim tropici） and arbusclar mycorrhizal （Glomus intraradices） symbiosis in Phaseolus vulgaris L. cv. Negro Jamapa under nitrate pre-incubation and continuous nitrate conditions. Our results indicate that nitrate pre-incubation, independent of the concentration, did not affect nodule development. However, the continuous supply of nitrate at high concentrations impaired nodule maturation and nodule numbers. Low nitrate conditions, in addition to positively regulating nodule number, biomass, and nitrogenase activity, also extended the span of nitrogen-fixing activity. By contrast, for arbuscular mycorrhizae, continuous 1o and 50mmol/L nitrate increased the percent root length colonization, concomitantly reduced arbuscule size, and enhanced ammonia transport without affecting phosphate transport. Therefore, in this manuscript, we have proposed the importance of nitrate as a positive regulator in promoting both rhizobial and mycorrhizal symbiosis in the common bean.

We studied moist deciduous forests of Chhattisgarh, India （1） to assess the effect of four levels of historic wildland fire frequency （high, medium, low, and no-fire） on regeneration of seedlings in fire affected areas during pre and post-fire seasons, （2） to evaluate vegetation struc- ture and diversity by layer in the four fire frequency zones, （3） to evalu- ate the impact of fire frequency on the structure of economically impor- tant tree species of the region, and （4） to quantify fuel loads by fire fre- quency level. We classified fire-affected areas into high, medium, low, and no-fire frequency classes based on government records. Tree species were unevenly distributed across fire frequency categories. Shrub density was maximum in zones of high fire frequency and minimum in low- frequency and no-fire zones. Lower tree density after fires indicated that regeneration of seedlings was reduced by fire. The population structure in the high-frequency zone was comprised of seedlings of size class （A） and saplings of size class （B）, represented by Diospyros melanoxylon, Dalbergia sissoo, Shorea robusta and Tectona grandis. Younger and older trees were more abundant for Tectona grandis and Dalbargia sis- soo after fire, whereas intermediate-aged trees were more abundant pre- fire, indicating that the latter age-class was thinned by the catastrophic effect of fire. The major contributing components of fuel load included duff litter and small woody branches and twigs on the forest floor. Total fuel load on the forest floor ranged from 2.2 to 3.38 Mg/ha. The netchange in fuel load was positive in high- and medium-frequency fire zones and negative under low- and no-fire zones. Repeated fires, how- ever, slowly reduced stand stability. An ecological approach is needed for fire management to restore the no-fire spatial and temporal structure of moist deciduous forests, their species composition and fuel loads. To evaluate the response to lime on cultivars of ricebean(Vigna umbellata), a field experiment was conducted during the two consecutive rabi seasons of 2010–2011 and 2011–2012 in the Nagaland foothills, India. The experiment used a split-plot design with four levels of lime(control, 0.2, 0.4and 0.6 t ha-1) in main plots and four ricebean cultivars(RBS-16, RBS-53, PRR-2, and RCRB-4) in sub-plots with three replicates. The results revealed that increasing levels of lime(in the furrow)from 0 to 0.6 t ha-1significantly increased growth, yield attributes and yield. The quality parameters of ricebean were also influenced significantly by the application of lime. Maximum gross return(INR 39,098 ha-1), net return(INR 27,281 ha-1), benefit:cost(B:C) ratio(2.29),production efficiency, and economic efficiency were also realized with the application of lime at 0.6 t ha-1. Among the ricebean cultivars, RBS-53 produced significantly higher growth, yield attributes, grain yield, straw yield, biological yield, and harvest index. Similarly, yield and protein content were higher in RBS-53. Maximum gross return, net return, B:C ratio, production efficiency, and economic efficiency were observed for RBS-53.

Survivin expression induced by doxorubicin in cholangiocarcinoma

AIM:To study the role of survivin expression induced by chemotherapy agent (doxorubicin) in the development and anti-chemotherapy of ***:Expression of survivin was detected by SP immunohistochemical technique in 33 cases of cholangiocarcinorna,28 cases of adjacent noncancerous bile duct, and 5 cases of benign bile duct lesions. Low concentration of doxorubicin (0.05mg/l) was added in cultured cholangiocarcinoma cell line (QBC939). The expression of survivin was detected by RT-PCR and Western blot at 24h and 48h after adding ***:Survivin was expressed in 24 of 33 cholangiocarcinoma cases (72.7%).In contrast,no expression of survivin in adjacent noncancerous and benign bile duct lesions was observed (P<0.01). No correlation was found between survivin expression and clinical *** could markedly (P<0.001) up-regulate survivin mRNA and protein expression of QBC939 ***:Overexpression of survivin in cholangiocarcinomas may play an important role in the development of cholangiocarcinoma, its relationship with prognosis of cholangiocarcinoma deserves further investigation. AIM: To investigate the therapeutic effect of somatostatin receptor type 2 (SSTR2) gene transfection on pancreatic carcinoma xenograftsin vivo in experimental ***: Human pancreatic cancer cell line Panc-1 was inoculated subcutaneously into the back of nude mice. When tumor nodules were grown as large as about 5 mm×5 mm days after inoculation, the mice were randomly divided into 3 groups (6 mice in each group). Group Ⅰ served as untreated control group. Group Ⅱ received an intratumoral injection of a combination of human cytomegalovirus promoter-6C (pCMV-6C) and lipofectamine 2000. Group Ⅲ received an intratumoral injection of a combination of pCMV-6C-SSTR2 and lipofectamine 2000. The rate of tumor growth was compared among these three groups. The expression of SSTR2 in these tumors was detected by immunohistochemistry and Western-blot. Apoptosis index (AI) in these tumors was examined by using TUNEL in ***: Intratumoral injection of a combination of pCMV-6C-SSTR2 and lipofectamine 2000 resulted in the expression of SSTR2 protein. The tumor size and weight in group Ⅲ (0.318±0.098 cm3, and 0.523±0.090 g,respectively) were significantly lower than those in group Ⅰ (2.058±0.176 cm3, and 1.412±0.146 g, respectively) and group Ⅱ (2.025±0.163 cm3, and 1.365±0.116 g, respectively)(P＜0.05) The AI in group Ⅲ (1.47±0.13%) was significantly higher than that in group Ⅰ (0.56±0.09%) and group Ⅱ (0.57±0.11%) (P＜0.05). But there were no significant differences between groups Ⅰ and Ⅱ.CONCLUSION: Our data demonstrate that re-expression of SSTR2 gene has antitumor effects on experimental pancreatic cancer. Restoration of SSTR2 gene expression through gene transfer in vivo might be a potential gene therapy strategy for human pancreatic cancer.

Background and Aims: Lifestyle (exercise and dietary) modification is the mainstay of treatment for non-alcoholic fatty liver disease (NAFLD). However, there is paucity of data on effect of intensity of exercise in management of NAFLD, and we aimed to study the effect of variable intensities of exercise on NAFLD. Methods: The study was performed in the Department of Gastroenterology of the SCB Medical Col-lege, Cuttack and the Biju Patnaik State Police Academy, Bhu-baneswar. The subjects were police trainees [18 in a moderate intensity exercise group (MIG) and 19 in a low in-tensity exercise group (LIG)] recruited for a 6-month physical training course (261.8 Kcalorie, 3.6 metabolic equivalent in MIG and 153.6 Kcalorie, 2.1 metabolic equivalent in LIG). NAFLD was diagnosed by ultrasonography, with exclusion of all secondary causes of steatosis. All participants were eval-uated by anthropometry (weight, height, body mass index (BMI), waist circumference), assessed for blood pressure and biochemical parameters (blood glucose, liver function test, lipid profile, serum insulin), and subjected to transabdo-minal ultrasonography before and after 6 months of physical training, and the results were compared. Results: Both the groups had similar BMI, fasting plasma glucose, AST, gamma-glutamyl transpeptidase, insulin, and homeostatic model as-sessment-insulin resistance (known as HOMA-IR) (p>0.05). However, subjects in the LIG were older and had lower alanine transaminase, higher triglycerides and lower high-density lip-oproteins than MIG subjects. There was a significant reduc-tion in BMI (27.0±2.1 to 26.8±2.0;p=0.001), fasting blood glucose (106.7±21.6 to 85.8±19.0;p<0.001), serum trigly-cerides (167.5±56.7 to 124.6±63.5;p=0.017), total choles-terol (216.8±29.2 to 196.7±26.6;p=0.037), low-density lipoprotein cholesterol (134.6±21.4 to 130.5±21.9;p=0.010), serum aspartate transaminase (39.3±32.2 to 30.9±11.4;p<0.001), serum alanine transaminase (56.6 ±28.7 to 33.0±11.3;p<0.

Objective:To investigate two cyanobacteria isolated from different origins *** aestuarii(***)from brackish water and Aphanothece bullosa(***)from fresh water paddy fields for antifungal and antileishmanila activity taking Candida albicans and Leishmania donovain as ***:Biomass of *** and *** were harvested after 40 and60 d respectively and lyophilized twice in methanol(100%)and redissolved in methanol(5%)for *** bioassay was done by agar well diffusion method while antileishmanial,by counting cell numbers and flageller motility observation of promastigotes and amastigotes from *** and 5%methanol were used as ***:Both the cyanobacteria were found to be potent source of antifungal activity keeping fluconazole as positive control,however,methanolic crude extract(15 mg/mL)of *** was found more potent(larger inhibition zone)over that of methanolic crude extract of ***,Similarly antileishmanial activity of crude extract(24.0 mg/mL)of *** was superior over that of methanolic crude extract of ***(25.6 mg/mL).Conclusions:Antifungal and antileishmanial drugs are still limited in the *** of microbes possessing antifungal and antileishmanial activity drug is of prime *** are little explored in this context because most of the drugs in human therapy are derived from microorganisms,mainly bacterial,fungal and *** in the present study two cyanobacterial strains from different origins showed potent source of antifungal and antileishmanial biomolecules.
